| 5 | NUMB maintains bone mass by promoting degradation of PTEN and GLI1 via ubiquitination in osteoblasts显示文摘The adaptor protein NUMB is involved in asymmetric division and cell fate determination and recognized as an antagonist of Notch.Previous studies have proved that Notch activation in osteoblasts contributes to a high bone mass. In this study, however, an osteopenic phenotype was found in 9-week-old mice using osteoblastic specific Col1a1–2.3-Cre to ablate both Numb and its homologue Numbl. The trabecular bone mass decreased dramatically while the cortical bone mass was unaffected. Here, the Notch signal was not activated,while the tensin homologue deleted on human chromosome 10(PTEN), which dephosphorylates phosphatidylinositide 3-kinases, was elevated, attenuating protein kinase B(Akt). The ubiquitination assay revealed that NUMB may physiologically promote PTEN ubiquitination in the presence of neural precursor cell-expressed developmentally downregulated protein 4–1. In addition, the deficiency of Numb/Numbl also activated the Hedgehog pathway through GLI1. This process was found to improve the ratio of the receptor activator of nuclear factor-k B ligand to osteoprotegerin, which enhanced the differentiation of osteoclasts and bone resorption. | 6 | Effect of water on extractive desulfurization of fuel oils using ionic liquids: A COSMO-RS and experimental study显示文摘When evaluating ionic liquids(ILs) for extractive desulfurization(EDS) of fuel oils, the inevitable presence of water in the system may have a significant and in many cases strongly negative effect. However, few studies have considered this particular issue and a promoted water effect on EDS is scarcely reported. In this work,COSMO-RS was firstly employed to calculate the capacity and selectivity for EDS of various IL/H_2 O mixtures,which cover different IL characters and a wide water concentration range. Experiments were then conducted with a representative IL [C_4MIM][H_2PO_4], whose stable and even promoted extraction performance with a small amount of water was suggested by COSMO-RS. Through analyses of the desulfurization ratio, the crosssolubility and the water content in the desulfurized fuel, the promoted effect of water within a certain range(b 10 wt%) was experimentally demonstrated. | 7 | Sn_(4)P_(3)-induced crystalline/amorphous composite structures for enhanced sodium-ion battery anodes显示文摘The optimization of anode materials such as Sn,P and Sn4P3 in terms of capacity and cyclability is crucial to improve the overall performance of sodium-ion batteries.However,the delicate fabrication of these materials,including the balanced crystalline/amorphous domains,reasonable particle size and distribution,complementary components exhibiting synergetic reactions,among others,still greatly retards the realization of maximum performance.Herein,a series of Sn/P-based composite materials with a plum pudding configuration were fabricated to achieve controlled crystalline/amorphous structures as well as optimized size and distribution in a carbon framework.By using a facile and low-cost ball milling method,the structural transformation of Sn4P3 into phase-separated crystalline Sn and amorphous P in a carbonaceous framework can be finely controlled,producing a series of binary(Sn4 P3/C),quaternary(Sn4P3/Sn/P/C) and ternary(Sn/P/C) composites.Due to the complementary components,crystalline/amorphous adjustment,crystallite sizes and well-integrated interfaces,the quaternary Sn4P3/Sn/P/C composite showed the best electrochemical performance,with a noticeable long-cycle performance of 382 mA hg-1 and 86% capacity retention for nearly 300 cycles.Different from binary and ternary composites,the discharge of quaternary composite generates no noticeable signals of Na15Sn4 and Na3 P in the ex-situ X-ray diffraction patterns,suggesting the crystallite growth of sodiation products can be depressed.Moreover,Sn4 P3 in the quaternary composite can be partially regenerated in the desodiation reaction,implying the significant short-range interaction and thus better synergetic reactions between Sn and P components.The results demonstrate that the design and organization of crystalline/amorphous structures can serve as an efficient strategy to develop novel electrode materials for sodium ion batteries. | Jaffer Saddique Xu Zhang Tianhao Wu Heng Su Shiqi Liu Dian Zhang Yuefei Zhang Haijun Yu | 2020 | Journal of Materials Science & Technology2020,52,20: | 3 |
| 8 | Metabolic risk factors associated with sudden cardiac death(SCD)during acute myocardial ischemia显示文摘Sudden cardiac death(SCD)is the leading cause of death worldwide.Myocardial ischemia(MI)is the most common underlying causal disorder for SCD.Metabolic risks leading to SCD during acute MI are still not fully understood.Here,using tissue metabolomics,we aimed to investigate myocardial metabolic alterations relevant to SCD events in an acute MI rat model induced by coronary artery ligation(CAL).Thirty-four rats were successfully performed CAL,of which 13 developed lethal ventricular tachyarrhythmia(LVTA)-SCD and 7 developed severe atrioventricular block(AB)-SCD.Fourteen rats that survived within 70 min after the ligation were served as peer controls.The partial least squares-discriminant analysis plots demonstrated clear separations between the SCD rats and controls,indicating obvious differences in myocardial metabolome between these rats.The levels of isoleucine,lactate,glutamate choline,phosphorylcholine,taurine and asparagine in ischemic myocardia were positively associated with LVTA-SCD events;in contrast,the levels of alanine,urea,phenylalanine,linoleic acid,elaidic acid and stearic acid were inversely correlated with LVTA-SCD events.The levels of glutamate and urea were positively and negatively relevant to AB-SCD events,respectively.The dangerous metabolites indicated that lower levels of energy substrates,severe hypoxia,the inhibition of transamination and hyper sympathetic excitement and reactive oxygen species in myocardia were vulnerable to SCD during acute MI.The results suggest fatal metabolic alterations correlated with SCD events during acute MI,which could offer novel clues for the prevention or treatment of acute MI-related SCD. | Dian Wang Xingxing Wang Jiayan Wu Ruibing Su Jing Kong Xiaojun Yu | 2017 | Forensic Sciences Research2017,2,3: | 2 |

| 10 | Preparation and efficacy evaluation of Paenibacillus polymyxa KM2501-1 microbial organic fertilizer against root-knot nematodes显示文摘Root-knot nematodes(RKNs) cause huge yield losses to agricultural crops worldwide. Meanwhile, livestock manure is often improperly managed by farmers, which leads to serious environmental pollution. To resolve these two problems, this study developed a procedure for the conversion of chicken manure to organic fertilizer by larvae of Hermetia illucens L. and Bacillus subtilis BSF-CL. Chicken manure organic fertilizer was then mixed thoroughly with Paenibacillus polymyxa KM2501-1 to a final concentration of 1.5×10^(8)CFU g^(-1). The efficacy of KM2501-1 microbial organic fertilizer in controlling root-knot nematodes was evaluated in pot and field experiments. In pot experiments, applying KM2501-1 microbial organic fertilizer either as a base fertilizer or as a fumigant at the dose of 40 g/pot suppressed root-knot disease by 61.76 and 69.05% compared to the corresponding control treatments, respectively. When applied as a fumigant at the dose of 1 kg m;in field experiments, KM2501-1 microbial organic fertilizer enhanced the growth of tomato plants, suppressed root-knot disease by 49.97%, and reduced second stage juveniles of RKN in soil by 88.68%. KM2501-1 microbial organic fertilizer controlled RKNs better than commercial bio-organic fertilizer in both pot and field experiments. | 11 | Can gene editing reduce postharvest waste and loss of fruit, vegetables, and ornamentals?显示文摘Postharvest waste and loss of horticultural crops exacerbates the agricultural problems facing humankind and will continue to do so in the next decade.Fruits and vegetables provide us with a vast spectrum of healthful nutrients,and along with ornamentals,enrich our lives with a wide array of pleasant sensory experiences.These commodities are,however,highly perishable.Approximately 33%of the produce that is harvested is never consumed since these products naturally have a short shelf-life,which leads to postharvest loss and waste.This loss,however,could be reduced by breeding new crops that retain desirable traits and accrue less damage over the course of long supply chains.New gene-editing tools promise the rapid and inexpensive production of new varieties of crops with enhanced traits more easily than was previously possible.Our aim in this review is to critically evaluate gene editing as a tool to modify the biological pathways that determine fruit,vegetable,and ornamental quality,especially after storage.We provide brief and accessible overviews of both the CRISPR-Cas9 method and the produce supply chain.Next,we survey the literature of the last 30 years,to catalog genes that control or regulate quality or senescence traits that are'ripe'for gene editing.Finally,we discuss barriers to implementing gene editing for postharvest,from the limitations of experimental methods to international policy.We conclude that in spite of the hurdles that remain,gene editing of produce and ornamentals will likely have a measurable impact on reducing postharvest loss and waste in the next 5-10 years. | Emma N.Shipman Jingwei Yu Jiaqi Zhou Karin Albornoz Diane M.Beckles | 2021 | Horticulture Research2021,8,1: | 2 |

| 16 | Comparative analysis of constitutes and metabolites for traditional Chinese medicine using IDA and SWATH data acquisition modes on LC-Q-TOF MS显示文摘Identification of components and metabolites of traditional Chinese medicines(TCMs)employing liquid chromatography-quadrupole time-of-flight mass spectrometry(LC-Q-TOF MS)techniques with information-dependent acquisition(IDA)approaches is increasingly frequent.A current drawback of IDA-MS is that the complexity of a sample might prevent important compounds from being triggered in IDA settings.Sequential window acquisition of all theoretical fragment-ion spectra(SWATH)is a dataindependent acquisition(DIA)method where the instrument deterministically fragments all precursor ions within the predefined m/z range in a systematic and unbiased fashion.Herein,the superiority of SWATH on the detection of TCMs’components was firstly investigated by comparing the detection efficiency of SWATH-MS and IDA-MS data acquisition modes,and sanguisorbin extract was used as a mode TCM.After optimizing the setting parameters of SWATH,rolling collision energy(CE)and variable Q1 isolation windows were found to be more efficient for sanguisorbin identification than the fixed CE and fixed Q1 isolation window.More importantly,the qualitative efficiency of SWATH-MS on sanguisorbins was found significantly higher than that of IDA-MS data acquisition.In IDA mode,18 kinds of sanguisorbins were detected in sanguisorbin extract.A total of 47 sanguisorbins were detected when SWATH-MS was used under rolling CE and flexible Q1 isolation window modes.Besides,26 metabolites of sanguisorbins were identified in rat plasma,and their metabolic pathways could be deduced as decarbonylation,oxidization,reduction,methylation,and glucuronidation according to their fragmental ions acquired in SWATH-MS mode.Thus,SWATH-MS data acquisition could provide more comprehensive information for the component and metabolite identification for TCMs than IDA-MS. | Dian Kang Qingqing Ding Yangfan Xu Xiaoxi Yin Huimin Guo Tengjie Yu He Wang Wenshuo Xu Guangji Wang Yan Liang | 2020 | Journal of Pharmaceutical Analysis2020,10,6: | 1 |
