1Simultaneous virus identification and characterization of severe unexplained pneumonia cases using a metagenomics sequencing technique显示文摘Many viruses can cause respiratory diseases in humans.Although great advances have been achieved in methods of diagnosis,it remains challenging to identify pathogens in unexplained pneumonia(UP) cases.In this study,we applied next-generation sequencing(NGS) technology and a metagenomic approach to detect and characterize respiratory viruses in UP cases from Guizhou Province,China.A total of 33 oropharyngeal swabs were obtained from hospitalized UP patients and subjected to NGS.An unbiased metagenomic analysis pipeline identified 13 virus species in 16 samples.Human rhinovirus C was the virus most frequently detected and was identified in seven samples.Human measles virus,adenovirus B 55 and coxsackievirus A10 were also identified.Metagenomic sequencing also provided virus genomic sequences,which enabled genotype characterization and phylogenetic analysis.For cases of multiple infection,metagenomic sequencing afforded information regarding the quantity of each virus in the sample,which could be used to evaluate each viruses' role in the disease.Our study highlights the potential of metagenomic sequencing for pathogen identification in UP cases.Xiaohui Zou Guangpeng Tang Xiang Zhao Yan Huang Tao Chen Mingyu Lei Wenbing Chen Lei Yang Wenfei Zhu Li Zhuang Jing Yang Zhaomin Feng Dayan Wang Dingming Wang Yuelong Shu 2017Science China(Life Sciences)2017,60,3:14

3Berberine mediates root remodeling in an immature tooth with apical periodontitis by regulating stem cells from apical papilla differentiation显示文摘Once pulp necrosis or apical periodontitis occurs on immature teeth,the weak root and open root apex are challenging to clinicians.Berberine(BBR)is a potential medicine for bone disorders,therefore,we proposed to apply BBR in root canals to enhance root repair in immature teeth.An in vivo model of immature teeth with apical periodontitis was established in rats,and root canals were filled with BBR,calcium hydroxide or sterilized saline for 3 weeks.The shape of the roots was analyzed by micro-computed tomography and histological staining.In vitro,BBR was introduced into stem cells from apical papilla(SCAPs).Osteogenic differentiation of stem cells from apical papilla was investigated by alkaline phosphatase activity,mineralization ability,and gene expression of osteogenic makers.The signaling pathway,which regulated the osteogenesis of SCAPs was evaluated by quantitative real time PCR,Western blot analysis,and immunofluorescence.In rats treated with BBR,more tissue was formed,with longer roots,thicker root walls,and smaller apex diameters.In addition,we found that BBR promoted SCAPs osteogenesis in a time-dependent and concentration-dependent manner.BBR induced the expression ofβ-catenin and enhancedβ-catenin entering into the nucleus,to up-regulate more runt-related nuclear factor 2 downstream.BBR enhanced root repair in immature teeth with apical periodontitis by activating the canonical Wnt/β-catenin pathway in SCAPs.Yujia Cui Jing Xie Yujie Fu Chuwen Li Liwei Zheng Dingming Huang Changchun Zhou Jianxun Sun Xuedong Zhou 2020International Journal of Oral Science2020,12,2:5
4NUMB maintains bone mass by promoting degradation of PTEN and GLI1 via ubiquitination in osteoblasts显示文摘The adaptor protein NUMB is involved in asymmetric division and cell fate determination and recognized as an antagonist of Notch.Previous studies have proved that Notch activation in osteoblasts contributes to a high bone mass. In this study, however, an osteopenic phenotype was found in 9-week-old mice using osteoblastic specific Col1a1–2.3-Cre to ablate both Numb and its homologue Numbl. The trabecular bone mass decreased dramatically while the cortical bone mass was unaffected. Here, the Notch signal was not activated,while the tensin homologue deleted on human chromosome 10(PTEN), which dephosphorylates phosphatidylinositide 3-kinases, was elevated, attenuating protein kinase B(Akt). The ubiquitination assay revealed that NUMB may physiologically promote PTEN ubiquitination in the presence of neural precursor cell-expressed developmentally downregulated protein 4–1. In addition, the deficiency of Numb/Numbl also activated the Hedgehog pathway through GLI1. This process was found to improve the ratio of the receptor activator of nuclear factor-k B ligand to osteoprotegerin, which enhanced the differentiation of osteoclasts and bone resorption. 5Expert consensus on digital guided therapy for endodontic diseases显示文摘Digital guided therapy(DGT)has been advocated as a contemporary computer-aided technique for treating endodontic diseases in recent decades.The concept of DGT for endodontic diseases is categorized into static guided endodontics(SGE),necessitating a meticulously designed template,and dynamic guided endodontics(DGE),which utilizes an optical triangulation tracking system.Based on cone-beam computed tomography(CBCT)images superimposed with or without oral scan(OS)data,a virtual template is crafted through software and subsequently translated into a 3-dimensional(3D)printing for SGE,while the system guides the drilling path with a real-time navigation in DGE.DGT was reported to resolve a series of challenging endodontic cases,including teeth with pulp obliteration,teeth with anatomical abnormalities,teeth requiring retreatment,posterior teeth needing endodontic microsurgery,and tooth autotransplantation.Case reports and basic researches all demonstrate that DGT stand as a precise,time-saving,and minimally invasive approach in contrast to conventional freehand method.This expert consensus mainly introduces the case selection,general workflow,evaluation,and impact factor of DGT,which could provide an alternative working strategy in endodontic treatment.Xi Wei Yu Du Xuedong Zhou Lin Yue Qing Yu Benxiang Hou Zhi Chen Jingping Liang Wenxia Chen Lihong Qiu Xiangya Huang Liuyan Meng Dingming Huang Xiaoyan Wang Yu Tian Zisheng Tang Qi Zhang Leiying Miao Jin Zhao Deqin Yang Jian Yang Junqi Ling 2023International Journal of Oral Science2023,15,4:0
6Survey of Distributed Computing Frameworks for Supporting Big Data Analysis显示文摘Distributed computing frameworks are the fundamental component of distributed computing systems.They provide an essential way to support the efficient processing of big data on clusters or cloud.The size of big data increases at a pace that is faster than the increase in the big data processing capacity of clusters.Thus,distributed computing frameworks based on the MapReduce computing model are not adequate to support big data analysis tasks which often require running complex analytical algorithms on extremely big data sets in terabytes.In performing such tasks,these frameworks face three challenges:computational inefficiency due to high I/O and communication costs,non-scalability to big data due to memory limit,and limited analytical algorithms because many serial algorithms cannot be implemented in the MapReduce programming model.New distributed computing frameworks need to be developed to conquer these challenges.In this paper,we review MapReduce-type distributed computing frameworks that are currently used in handling big data and discuss their problems when conducting big data analysis.In addition,we present a non-MapReduce distributed computing framework that has the potential to overcome big data analysis challenges.Xudong Sun Yulin He Dingming Wu Joshua Zhexue Huang 2023Big Data Mining and Analytics2023,6,2:0
7Expert consensus on difficulty assessment of endodontic therapy显示文摘Endodontic diseases are a kind of chronic infectious oral disease. Common endodontic treatment concepts are based on the removal of inflamed or necrotic pulp tissue and the replacement by gutta-percha. However, it is very essential for endodontic treatment to debride the root canal system and prevent the root canal system from bacterial reinfection after root canal therapy(RCT). Recent research, encompassing bacterial etiology and advanced imaging techniques, contributes to our understanding of the root canal system’s anatomy intricacies and the technique sensitivity of RCT. Success in RCT hinges on factors like patients, infection severity, root canal anatomy, and treatment techniques. Therefore, improving disease management is a key issue to combat endodontic diseases and cure periapical lesions. The clinical difficulty assessment system of RCT is established based on patient conditions, tooth conditions, root canal configuration, and root canal needing retreatment, and emphasizes pre-treatment risk assessment for optimal outcomes. The findings suggest that the presence of risk factors may correlate with the challenge of achieving the high standard required for RCT. 8The potential regulatory role of BMP9 in inflammatory responses显示文摘Inflammation is a protective response of the body to pathogens and injury.Hence,it is particularly important to explore the pathogenesis and key regulatory factors of inflammation.BMP9 is a unique member of the BMP family,which is widely known for its strong osteogenic potential and insensitivity to the inhibition of BMP3.Recently,several studies have reported an underlying pivotal link between BMP9 and inflammation.What is clear,though not well understood,is that BMP9 plays a role in inflammation in a carefully choreographed manner in different contexts.In this review,we have summarized current studies focusing on BMP9 and inflammation in various tissues and the latest advances in BMP9 expression,signal transduction,and crystal structure to better understand the relationship between BMP9 and inflammation.In addition,we also briefly summarized the inflammatory characteristics of some TGF-b superfamily members to provide better insights and ideas for the study of BMP9 and inflammation.Tianzhu Song Dingming Huang Dongzhe Song 2022Genes & Diseases2022,9,6:0
