| 1 | Transgenic expression of MYB15 confers enhanced sensitivity to abscisic acid and improved drought tolerance in Arabidopsis thaliana显示文摘Abiotic stresses cause serious crop losses. Knowledge on genes functioning in plant responses to adverse growth conditions is essential for developing stress tolerant crops. Here we report that transgenic expression of MYB15, encoding a R2R3 MYB transcription factor in Arabidopsis thaliana, conferred hypersensitivity to exogenous abscisic acid (ABA) and improved tolerance to drought and salt stresses. The promoter of MYB15 was active in not only vegetative and reproductive organs but also the guard cells of stomata. Its transcript level was substantially upregulated by ABA, drought or salt treatments. Compared with wild type (WT) control, MYB15 overexpression lines were hypersensitive to ABA in germination assays, more susceptible to ABA-elicited inhibition of root elongation, and more sensitive to ABA-induced stomatal closure. In line with the above findings, the transcript levels of ABA biosynthesis (ABA1, ABA2), signaling (ABI3), and responsive genes (AtADH1, RD22, RD29B, AtEM6) were generally higher in MYB15 overexpression seedlings than in WT controls after treatment with ABA. MYB15 overexpression lines displayed improved survival and reduced water loss rates than WT control under water deficiency conditions. These overexpression lines also displayed higher tolerance to NaCl stress. | 2 | A Review on Lithium-ion Power Battery Thermal Management Technologies and Thermal Safety显示文摘Lithium-ion power battery has become one of the main power sources for electric vehicles and hybrid electric vehicles because of superior performance compared with other power sources. In order to ensure the safety and improve the performance, the maximum operating temperature and local temperature difference of batteries must be maintained in an appropriate range. The effect of temperature on the capacity fade and aging are simply investigated. The electrode structure, including electrode thickness, particle size and porosity, are analyzed. It is found that all of them have significant influences on the heat generation of battery. Details of various thermal management technologies, namely air based, phase change material based, heat pipe based and liquid based, are discussed and compared from the perspective of improving the external heat dissipation. The selection of different battery thermal management(BTM) technologies should be based on the cooling demand and applications, and liquid cooling is suggested being the most suitable method for large-scale battery pack charged/discharged at higher C-rate and in high-temperature environment. | 3 | Contemporary crustal movement of continental China obtained by global positioning system(GPS) measurements显示文摘We obtain a unified horizontal velocity field of continental China and vicinity through measurements at 81 GPS stations provided by the National Key Infrastructure Project, Crustal Movement Observation Network of China. The velocity field delineates patterns of movements and deformation of active crustal blocks in continental China under the Eurasia-fixed reference frame. The space-based geodesy also clearly shows for the first time the horizontal movementand deformation of continental China induced by indentation of the Indian plate. | 5 | Precise nanomedicine for intelligent therapy of cancer显示文摘Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. | 6 | Effect of L-NAME on nitric oxide and gastrointestinal motility alterations in cirrhotic rats显示文摘AIM: To invsstigare the effect of L-NAME on nitric oxide andgastriubtestubal motility alterations in cirrhotic ratsMETHODS: Rats with cirrhosis induced by carbontetrachloride were randomly divided into two groups, one( n= 13) receiving 0. 5 mg@ kg-1 per clay of NG-nitro-L-argininemethyl ester (L-NAME), a nitric oxide synthase inhibitor,for 10 days, whereas the other group ( n = 13) and control( n = 10) rats were administrated the same volume of 9 g@ L-1saline.Half gastric emptying time and 2 h residual rate weremeasured by SPECT, using 99m Tc-DTPA-labeled bariumsuifate as test meal. Gastrointestinal transition time wasrecorded simultaneously. Serum concentration of nitrcoxide (NO) was determined by the kinetic cadmiunreduction and colorimetric methods. ImmunohistochemicalSABC method was used to observe the expression anddistribution of three types of nitric oxide synthase (NOS)isoforms in the mt gastrointestinal tract. Western blot wasused to detect expression of gastrointestinal NOS isoforms.RESULTS: Half gastric emptying time and trans-gastrointestinal time were significantly prolonged( 124.0 ± 26.4min; 33.7± 8.9min;72.1 ± 15.3 min; P<0.01), (12.4±0.5h; 9.5±0.3 h; 8.2±0.8 h; P<0.01), 2h residual rate wasraised in cirrhotic rots than in controls and cirrhotic ratstreated with L-NAME(54.9± 7.6 % ,13.7 ± 3.2 %, 34.9± 10.3%, P< 0.01). Serum concentration of NO was significantlyincreased in cirrhotic rots than in the other groups (8.20 ± 2.48)μmol@L-1, (5.94± 1.07) μmol@L-1 ,and control (5.66± 1.60) tμmol@L-1, P< 0.01. NOS staining intensities which weremainly located in the gastrointestinal tissues were markedlylower in cirrhotic rats than in the controls and cirrhotic ratsafter treated with L- NAME.CONCLUSION: Gastrointestinal motility was remarkablyinhibited in cirrhotic rats, which could he alleviated by L-NAME. | 7 | Identification of Hercynian shoshonitic intrusive rocks in central Hainan Island and its geotectonic implications显示文摘An identification has been made of some shoshonitic intrusive rocks in central Hainan Island recently. These rocks are K-rich (K2O=2.9%―5.1%, K2O/Na2O=0.95―2.12), distinctly enriched in LILE and LREE, strongly depleted in Nb, Ta, and moder- ately depleted in Sr and Ti, with (87Sr/86Sr)i = 0.70859―0.71425 and ε Nd(t) = (?2.77―?7.49). They were derived from an EMⅡ-type mantle source. The enrichment process is related to metasomatism of depleted mantle caused by a great amount of fluid-melt released from oceanic crust and terri- genous sediments at great depth (eclogite facies) during the subduction of the South China plate under the Indochina-South China Sea plate in the Carbon- iferous–Early Permian. A SHRIMP U-Pb zircon dating yields a crystallization age of 272±7 Ma for the sho- shonitic intrusions, which is coeval with the strongly peraluminous granites found in central Hainan Island. These two kinds of rocks generally possess syn-intrusion ductile deformation structures. Thus they are considered to have been generated during the early stage (syn-thrust phase) of a post-collisional event. The primary magma of shoshonitic rocks was produced at a depth > 80 km by decompression- dehydration melting of previously enriched litho- spheric mantle wedge, phlogopite-bearing garnet peridotite, which was in turn caused by the break-off of a descendent slab and upwelling of a hot as- thenosphere. The rising of melts was accompaniedby crustal contamination and crystallization fractiona- tion (AFC). Combining with other related data, it is proposed that the southwards subduction and amal- gamation of the South China plate with the Indo- China-South China Sea plate took place at ca. 287- 278 Ma, which was a part of the convergence proc- ess of the Pangea supercontinent. | 9 | Effect of dilution holes on the performance of a triple swirler combustor显示文摘A triple swirler combustor is considered to be a promising solution for future high temperature rise combustors. The present paper aims to study dilution holes including primary dilution holes and secondary dilution holes on the performance of a triple swirler combustor. Experimental investigations are conducted at different inlet airflow velocities(40–70 m/s) and combustor overall fuel–air ratio with fixed inlet airflow temperature(473 K) and atmospheric pressure. The experimental results show that the ignition is very difficult with specific performance of high ignition fuel–air ratio when the primary dilution holes are located 0.6H(where H is the liner dome height)downstream the dome, while the other four cases have almost the same ignition performance. The position of primary dilution holes has an effect on lean blowout stability and has a large influence on combustion efficiency. | 11 | Anti-HBV effect of TAT-HBV targeted ribonuclease显示文摘AIM: To prepare and purify TAT-HBV targeted ribonuclease fusion protein, evaluate its transduction activity and investigate its effect on HBV replication in 2.2.15 cells.METHODS: The prokaryotic expression vector pTAT containing TR gene was used in transforming E. coli BL21(DE3) LysS and TR was expressed with the induction of IPTG. The TAT-TR fusion protein was purified using Ni-NTA-agrose and PD-10 desalting columns, and analyzed by SDSPAGE. Transduction efficiency of TAT-TR was detected with immunofluorescence assay and the concentration of HBeAg in the supernatant of the 2.2.15 cells was determined via solid-phase radioimmunoassay (spRIA). MTT assay was used to detect the cytotoxicity of TAT-TR.RESULTS: The SDS-PAGE showed that the TAT-TR fusion protein was purified successfully, and the purity of TAT-TR was 90 %. | 12 | Aberrant expression of ether à go-go potassium channel in colorectal cancer patients and cell lines显示文摘AIM: To study the expression of ether à go-go (Eag1) potassium channel in colorectal cancer and the relation- ship between their expression and clinico-pathological features. METHODS: The expression levels of Eag1 protein were determined in 76 cancer tissues with paired non- cancerous matched tissues as well as 9 colorectal adenoma tissues by immunohistochemistry. Eag1 mRNA expression was detected in 13 colorectal cancer tissues with paired non-cancerous matched tissues and 4 colorectal adenoma tissues as well as two colorectal cancer cell lines (LoVo and HT-29) by reverse transcription PCR. RESULTS: The frequency of positive expression of Eag1 protein was 76.3% (58/76) and Eag1 mRNA was 76.9% (10/13) in colorectal cancer tissue. Expression level of Eag1 protein was dependent on the tumor size, lymphatic node metastasis, other organ metastases and Dukes’ stage (P < 0.05), while not dependent on age, sex, site and degree of differentiation. Eag1 protein and mRNA were negative in normal colorectal tissue, and absolutely negative in colorectal adenomas except that one case was positively stained for Eag1 protein. CONCLUSION: Eag1 protein and mRNA are aberrantly expressed in colorectal cancer and occasionally expressed in colorectal adenoma. | 13 | Evidences for vagus nerve in-maintenance of immune balance and transmission of immune information from gut to brain in STM-infected rats显示文摘AIM:To determine whether salmonella Typhimurium(STM) in gastrointestinal tract can induce the functional activation of brain,whether the vagus nerve involves in signaling immune infomation from gastrointestinal tract to brain and how it influences the immune function under natural infection condition.METHODS:Animal model of gastrointestinal tract infection in the rat was established by an intuation of Salmonella Typhimurium(STM) into stomach to minic the condition of natural bacteria infection,Subdiagphragmatic vagotomy was performed in some of the animals 28 days before infection The changes of Fos expression visualized with immunohistochemistry technique in hypothalamic paraventricular nucleus(pVN) and superaoptic nuclesu (SON)were conted.Meanwhilie,the percentage and the Mean Intensities of Flurescent (MIFs)of CD4+ and CD8+ T cells in peripheral blood were measured by using flow cytometry(FCM),and the pathological changes in ileum and mesenteric lymph node were observed in HE stained sections.RESULTS:In bacteria-stimulated groups,inflammatory pathological changes were seen in ileum and mesenteric lymph node,The percentages of CD4+ T cells in peripheral blood were decreased from 43%±4.5%to 34%±4.9%(P<0.5)and MIFs of CD8+ T cells were also decreased from 2.9±0.39to 2.1±0.36(P<0.05)with STM stimulation.All of them proved that our STM-infection model was reliable Fos immunoreactive(Fos-ir) cells in PVN and SON incerased significantly with STM stimulation,from 189±41 to467±62(P<0.05)and from 64±21 to 282±47(P<0.05)individually,which suggested that STM in gastointestinal tract induced the functional activation of brain Subdiagphragmatic vagotomy attenuated FOs expression in PVN and SON induced by STM,from 467±62to 226±45(P<0.05)and from 282±47to 71±19(P<0.05)individually,and restored the decreased percentages of CD4+ T cells induced by STM from 34%±4.9% to original level 44%±6.0%(P<0.05).In addition,subdiagphragmatic vagotomy itself also decreased the percentages of CD8+ T cells (from 28%±3.0%to 21%±5.9%),P<0.05 and MIFs of CD4+(from 6.6±0.6to4.9±1.0P<0.05)and CD8+ T cells (from 2.9±0.39to 1.4±0.34,P<0.05).Both of them manifested the important role of vagus nerve in transmitting immune information from gut to brain and maintaining the immune balance of the organism.CONCLUSION:Vagus nerve dose involve in transmitting adbominal immune informaton into the brain in STM infection condition and play an important role in maintenance of the immune balance of the oranism. | Xi Wang Bai-Ren Wang Zhen Xu Yu-Qiang Ding Gong Ju,Institute of Neuroscience,Fourth Military Medical University,Xi’an 710032,Shaanxi Province,China Xi-Jing Zhang,Department of Anesthesiology,Xi Jing Hospital,Fourth Military Medical University,Xi’an 710032,Shaanxi Province,China | 2002 | World Journal of Gastroenterology2002,8,3: | 10 |

| 15 | Stress evolution and support mechanism of a bolt anchored in a rock mass with a weak interlayer显示文摘By applying experimental method, the bolt stress and supporting mechanism is studied during the deformation process of a rock mass containing a weak interlayer. The force measuring bolt is installed manually and instrumented five pairs of symmetrical strain gauges. The experimental results show that the fully grouted bolt suffers tensile, compressive, bending and shear stress at the same time. The bolt stress evolution is closely related to the deformation stages of the rock mass which are very gradually varying stage, gradually varying stage at the pre-peak and suddenly varying stage at the post peak stage.The axial compressive stress in the bolt is mainly induced by the moment. Thus, in most cases the axial compressive stress is distributed on one side of the bolt. For axial stresses, induced by the axial force and the bending moment at the post-peak stage, three types of changing are observed, viz. increasingincreasing type, decreasing-increasing type and increasing-decreasing type. The stress characteristics of the bolt section in the weak interlayer are significantly different from those in the hard rock. The failure models of the anchored bolt are tensile failure and shear failure, respectively. The bolt not only provides constraints on the free surface of the rock mass, but also resists the axial and lateral loading by the bending moment. | 16 | Preparation of single chain variable fragment of MG_7 mAb by phage display technology显示文摘AIM To develop the single chain variable fragment of MG7 murine anti-human gastric cancer monoclonal antibody using the phage display technology for obtaining a tumor-targeting mediator.METHODS mRNA was isolated from MG7-producing murine hybridoma cell line and converted into cDNA. The variable fragments of heavy and light chain were amplified separately and assembled into ScFv with a specially constructed DNA linker by PCR. The ScFvs DNA was ligated into the phagmid vector pCANTAB5E and the ligated sample was transformed into competent E. Coli TG1. The transformed cells were infected with M13K07 helper phage to form MG7 recombinant phage antibody library. The volume and recombinant rate of the library were evaluated by means of bacterial colony count and restriction analysis. After two rounds of panning with gastric cancer cell line KATOⅢ of highly expressing MG7binding antigen, the phage clones displaying ScFv of the antibody were selected by ELISA from the enriched phage clones. The antigen-binding affinity of the positive clone was detected by competition ELISA. HB2151 E. Coli was transfected with the positive phage clone demonstrated by competition ELISA for production of a soluble form of the MG7 ScFv. ELISA assay was used to detect the antigenbinding affinity of the soluble MG7 ScFv. Finally, the relative molecular mass of soluble MG7 ScFv was measured by SDS-PAGE.RESULTS The VH, VL and ScFv DNAs were about 340bp,320bp and 750bp, respectively. The volume of the library was up to 2 × 106 and 8 of 11 random clones were recombinants. Two phage clones could strongly compete with the original MG7 antibody for binding to the antigen expressed on KATO Ⅲ cells. | 19 | Impact of CYP3A4*1G Polymorphism on Fentanyl Analgesia Assessed by Analgesia Nociception Index in Chinese Patients Undergoing Hysteroscopy显示文摘Background:The clinical efficacy of fentanyl for pain control differs greatly across individuals.The purpose of this study was to investigate the impact of CYP3A4*1G polymorphism including wild-type homozygote (CYP3A4*1/*1,GG),mutant heterozygote (CYP3A4*1/*1G,GA),and mutant homozygote (CYP3A4*1G/*1G,AA)on fentanyl analgesia in Chinese patients undergoing hysteroscopy by the assessment of analgesia nociception index (ANI). Methods:A total of 200 gynecologic patients scheduled for elective hysteroscopy under general anesthesia at Peking University People's Hospital from May to December in 2017 were enrolled in this study.Venous blood was withdrawn for genotyping of CYP3A4*1G before operation.Fentanyl 1μg/kg was administered preoperatively followed by target-controlled infusion of propofol for induction and maintenance.Intraoperative analgesic efficacy of fentanyl was assessed by ANI monitoring at T0(entering room),T1(cervical dilation), T2(start of cervical aspiration),and T3(end of cervical aspiration)time points.The duration of propofol infusion and total dosage of propofol were recorded as well. Results:The patients were divided into three groups according to CYP3A4*1G polymorphism,including 143 in GG group,47in GA group, and 10 in AA group.There was no significant difference in clinical demographics among three groups.The frequency of CYP3A4*1G variant alleles accounted for 16.8%and the distribution of variant alleles was consistent with Hardy-Weinberg equilibrium.Using a multilevel model,ANI values at T1(63.81±19.61),T2(63.63±17.82),and T3(65.68±17.79)were significantly lower than that at T0 (77.16±12.93)in the study population (F =23.50,P <0.001),suggesting that higher levels of pain at T 1,T2,and T3 than T0.Patients with GG genotype showed significantly lower ANI than those with GA or AA genotypes during hysteroscopy under the same dose of fentanyl.
