2Induction of apoptosis by arsenic trioxide and hydroxycamptothecin in gastric cancer cells in vitro显示文摘AIM To study the effects of arsenic trioxide andHCPT on different degrees of differentiated gastriccancer cells(SGC-7901,MKN-45,MKN-28)withrespect to both cytotoxicity and induction ofapoptosis in vitro.METHODS The cytotoxicity of As2O3 and HCPTon gastric cancer cells was determined by MTTassay.Morphologic changes of apoptosis ofgastric cancer cells were observed by lightmicroscopy and transmission electron microscopy.Apoptosis and cell cycle changes of gastric cancercells induced by HCPT and As2O3 were investigatedby TUNEL method and flow cytometry.RESULTS As2O3 and HCPT had remarkablecytotoxic effects on different degrees ofdifferentiated gastric cancer cells.The IC50ofAs2O3 on well differentiated gastric cancer cellMKN-28,moderately differentiated gastric cancercell SGC-7901,and poorly differentiated gastriccancer cell MKN-28 were 8.91 μmol/L,10.57μmol/L,and 11.65 μmol/L,respectively.The IC50of HCPT on MKN-28,SGC-7901,and MKN-45 were9.35 mg/L,10.21 mg/L,and 12.63 mg/Lrespectively after 48 h treatment.After 12 h ofexposure to both drugs,gastric cancer cellsexhibited morphologic features of apoptosis,including cell shrinkage,nuclear condensation, and formation of apoptotic bodies.A typicalsubdiploid peak before G0/G1 phase was observedby flow cytometry.The apoptotic rates of SGC-7901,MKN-45,and MKN-28 were 13.84%,22.52%,and 9.68%,respectively after 48 hexposure to 10 μmol/L As2O3.The apoptotic ratesof SGC-7901,MKN-45,and MKN-28 were 21.88%,12.35%,and 30.26%,respectively after 48 hexposure to 10 mg/L HCPT.The apoptotic indicewere 7%-15% as assessed by TUNEL method.The effect of As2O3 on SGC-7901 showedremarkable cell cycle specificity,which inducedcell death in G1 phase,and blocked G2/M phase.HCPT also showed a remarkable cell cyclespecificity,by inducing cell death and apoptosis inG1 phase and arrest of proliferation at S phase.CONCLUSION As2O3 and HCPT exhibitsignificant cytotoxicity on gastric cancer cells byinduction of apoptosis.As2O3 and HCPT mighthave a promising prospect in the treatment ofgastric cancer,which needs to be further studied.Tu SP Zhong J Tan JH Jiang XH Qiao MM Wu YX Jiang SH 2000World Journal of Gastroenterology2000,6,4:43

6Precise nanomedicine for intelligent therapy of cancer显示文摘Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. 7Lung transplantation as therapeutic option in acute respiratory distress syndrome for coronavirus disease 2019-related pulmonary fibrosis显示文摘Background:Critical patients with the coronavirus disease 2019(COVID-19),even those whose nucleic acid test results had turned negative and those receiving maximal medical support,have been noted to progress to irreversible fatal respiratory failure.Lung transplantation(LT)as the sole therapy for end-stage pulmonary fibrosis related to acute respiratory distress syndrome has been considered as the ultimate rescue therapy for these patients.Methods:From February 10 to March 10,2020,three male patients were urgently assessed and listed for transplantation.After conducting a full ethical review and after obtaining assent from the family of the patients,we performed three LT procedures for COVID-19 patients with illness durations of more than one month and extremely high sequential organ failure assessment scores.Results:Two of the three recipients survived post-LT and started participating in a rehabilitation program.Pearls of the LT team collaboration and perioperative logistics were summarized and continually improved.The pathological results of the explanted lungs were concordant with the critical clinical manifestation,and provided insight towards better understanding of the disease.Government health affair systems,virology detection tools,and modern communication technology all play key roles towards the survival of the patients and their rehabilitation.Conclusions:LT can be performed in end-stage patients with respiratory failure due to COVID-19-related pulmonary fibrosis.If confirmed positive-turned-negative virology status without organ dysfunction that could contraindicate LT,LT provided the final option for these patients to avoid certain death,with proper protection of transplant surgeons and medical staffs.By ensuring instant seamless care for both patients and medical teams,the goal of reducing the mortality rate and salvaging the lives of patients with COVID-19 can be attained.Jing-Yu Chen Kun Qiao Feng Liu Bo Wu Xin Xu Guo-Qing Jiao Rong-Guo Lu Hui-Xing Li Jin Zhao Jian Huang Yi Yang Xiao-Jie Lu Jia-Shu Li Shu-Yun Jiang Da-Peng Wang Chun-Xiao Hu Gui-Long Wang Dong-Xiao Huang Guo-Hui Jiao Dong Wei Shu-Gao Ye Jian-An Huang Li Zhou Xiao-Qin Zhang Jian-Xing He 2020Chinese Medical Journal2020,,12:19

9Disentangling the effects of topography and space on the distributions of dominant species in a subtropical forest显示文摘Topography and space are two important factors determining plant species assemblages in forest communities.Quantification of the contribution of these two factors in determining species distribution helps us to evaluate their relative importance in determining species assemblages.This study aims to disentangle the effect of topography and space on the distributions of 14 dominant species in a subtropical mixed forest.Spearman correlation analysis and the torustranslation test were used to test the species–habitat associations.Variation partitioning was used to quantify the relative contributions of topography and space at three sampling scales and three life stages.Correlation analyses and torus-translation tests showed species abundance was mostly correlated with topographic wetness index,vertical distance from the channel network and convexity.Variation partitioning showed that pure topography,pure space and spatially structured topography explained about 2.1 %,41.2 % and 13.8 %of the variation in species distributions,respectively.For nine species,total topography fractions peaked in 20 m quadrats.For ten species,the pure space fractions peaked in 50 m quadrats.For many species,the total topography fraction andthe pure space fraction were larger for the most abundant life stages,which reflected the importance of sampling effect.However,some cases did not follow this trend suggesting that the effects of ecological processes such as habitat filtering,density dependence or dispersal limitation may exceed the sampling effects.In conclusion,we found that spatially structured topography and pure space primarily shaped the distribution of dominant tree species.Furthermore,their effects were both scale- and life stage-dependent.Qinggang Wang Yaozhan Xu Zhijun Lu Dachuan Bao Yili Guo Junmeng Lu Kuihan Zhang Haibo Liu Hongjie Meng Xiujuan Qiao Handong Huang Mingxi Jiang 2014Chinese Science Bulletin2014,59,35:16
10Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. 11Effect of Niobium on Microstructure and Properties of the CoCrFeNb_xNi High Entropy Alloys显示文摘A series of CoCrFeNb_xNi(x values in molar ratio, x = 0, 0.25, 0.45, 0.5, 0.75, 1.0 and 1.2) high entropy alloys(HEAs) was prepared to investigate the alloying effect of Nb on the microstructures and mechanical properties. The results indicate that the prepared CoCrFeNb_xNi(x > 0) HEAs consist of a simple FCC solid solution phase and a Laves phase. The microstructures of the alloys change from an initial single-phase FCC solid solution structure(x = 0) to a hypoeutectic microstructure(x = 0.25), then to a full eutectic microstructure(x = 0.45) and finally to a hypereutectic microstructure(0.5 < x < 1.2). The compressive test results show that the Nb0.45(x = 0.45) alloy with a full eutectic microstructure possesses the highest compressive fracture strength of 2558 MPa and a fracture strain of 27.9%. The CoCrFeNi alloy exhibits an excellent compressive ductility, which can reach 50% height reduction without fracture. The Nb0.25 alloy with a hypoeutectic structure exhibits a larger plastic strain of 34.8%. With the increase of Nb content, increased hard/brittle Laves phase leads to a decrease of the plasticity and increases of the Vickers hardness and the wear resistance. 13Geologic characteristics of the Chang'E-3 exploration region显示文摘We present topographic,geomorphologic and compositional characteristics of a 1°×1°(~660 km2)region centered near the landing site of Chang’E-3 using the highest spatial resolution data available.We analyze the topography and slope using Digital Terrain Model(DTM)generated from Terrain Camera(TC)images.The exploration region is overall relatively flat and the elevation difference is less than 300 m,and the slopes of 80%area are less than 5°.Impact craters in the exploration region are classified into four types based on their degradation states.We investigate the wrinkle ridges visible in the exploration region in detail using TC and Lunar Reconnaissance Orbiter(LRO)Narrow Angle Camera(NAC)images.We calculate FeO and TiO2 abundances using Multispectral Imager(MI)data,and confirm two basaltic units:the northern part belongs to Imbrian low-Ti/very-low-Ti mare basalts,and the southern part is Eratosthenian low-Ti/high-Ti mare basalts.Finally,we produce a geological map and propose the geologic evolution of the exploration region.We also suggest several rover traverses to explore interesting targets and maximize the potential scientific output.ZHAO JianNan HUANG Jun QIAO Le XIAO ZhiYong HUANG Qian WANG Jiang HE Qi XIAO Long 2014Science China(Physics,Mechanics & Astronomy)2014,57,3:14
14Variations of geochemical compositions and the paleoclimatic significance of a loess-soil sequence from Garzê County of western Sichuan Province,China显示文摘The West Sichuan Plateau is located in the southeast margin of the Tibetan Plateau, where the climate is mainly influenced by the Indian southwest summer monsoon and the Tibetan Plateau monsoon. In this study, detailed geochemical analysis has been carried out on Ganzisi loess-paleosol sequence in Ganzê County of western Sichuan Province. The results indicate that Ganzê loess and paleosol have experienced the incipient stage of chemical weathering in dust source regions, characterized by the decomposition of plagioclase which caused the depletion of mobile elements Na and Ca. The post-depositional chemical weathering is characterized by carbonate dissolution and oxidation of Fe2+. The variations of some geochemical indexes (such as CIA values, Na/K and Fe2+/ Fe3+ ratios) in Ganzisi loess-paleosol sequence indicate a gradually decreased chemical weathering intensity in the dust source regions and deposition areas since 1.15 Ma BP consistent with the general increase of global ice volume, reflecting that the arid trend since 1.15 Ma BP in the southeast Tibetan Plateau is a regional response to the global climate change. The geochemical indexes in this section also reveal an obvious drying step occurred at about 250 ka BP in this region. We interpret this drying step as a result of decreased influence of the Indian southwest summer monsoon. 16Density dependence and habitat preference shape seedling survival in a subtropical forest in central China显示文摘Aims seedlings are vulnerable to many kinds of fatal abiotic and biotic agents,and examining the causes of seedling dynamics can help understand mechanisms of species coexistence.To disentangle the relative importance of neighborhood densities,habitat factors and phylogenetic relatedness on focal seedling survival,we monitored the survival of 5306 seedlings of 104 species>15 months.We address the following questions:(i)How do neighborhood densities,habitat variables and phylogenetic relatedness affect seedling survival?What is the relative importance of conspecific densities,habitat variables and phylogenetic relatedness to seedling survival?(ii)Does the importance of the neighborhood densities,habitat variables and phylogenetic relatedness vary among growth forms,leaf habits or dispersal modes?specially,does the conspecific negative density dependence inhibit tree and decidu-ous seedlings more compared with shrub and evergreen species?Does density dependence affect the wind and animal-dispersed species equally?Methods We established 135 census stations to monitor seedling dynamics in a 25-ha subtropical forest plot in central China.Conspecific and heterospecific seedling density in the 1-m2 seedling plot and adult basal area within a 20-m radius provided neighborhood density var-iables.mean elevation,convexity and aspect of every 5-×5-m grid with seedling plots were used to quantify habitat characteristics.We calculated the relative average phylodiversity between focal seed-ling and heterospecific neighbors to quantify the species related-ness in the neighborhood.Eight candidate generalized linear mixed models with binominal error distribution were used to compare the relative importance of these variables to seedling survival.akaike’s information criteria were used to identify the most parsimonious models.Important Findingsat the community level,both the neighborhood densities and phylogenetic relatedness were important to seedling survival.We found negative effects of increasing conspecific seedlings,which suggested the existence of species-specific density-dependent mortality.Phylodiversity of heterospecific neighbors was negatively related to survival of focal seedlings,indicating similar habitat preference shared among phylogenetically closely related species may drive seedling survival.The relative importance of neighborhood densities,habitat variables and phylogenetic relatedness varied among ecological guilds.Conspecific densities had significant negative effect for deciduous and wind-dispersed species,and marginally significant for tree seedlings>10 cm tall and animal-dispersed species.Habitat variables had limited effects on seedling survival,and only elevation was related to the sur-vival of evergreen species in the best-fit model.We conclude that both negative density-dependent mortality and habitat preference reflected by the phylogenetic relatedness shape the species coex-istence at seedling stage in this forest.Junmeng Lu Daniel J.Johnson Xiujuan Qiao Zhijun Lu Qinggang Wang Mingxi Jiang 2015Journal of Plant Ecology2015,8,6:12

19Expression of VEGFR2 and NRP-1 in non-small cell lung cancer and their clinical significance显示文摘Objective: Vascular-targeted therapy is gradually becoming more appealing for patients with lung cancer. It is unclear whether vascular endothelial growth factor receptor 2(VEGFR2) and neuropilin-1(NRP-1) can be biomarkers for clinical treatment. We aimed to investigate the expression levels of VEGFR2 and NRP-1 in human non-small cell lung cancer(NSCLC) and their clinical significance by observing patient prognosis. Methods: VEGFR2 and NRP-1 were assessed by immunohistochemistry(IHC) in 40 patients with NSCLC and in 10 patients with benign lesions of lung; kinase insert domain receptor(KDR) and NRP-1 copy number gain(CNG) was assessed by fluorescence in situ hybridization(FISH). The distributions of overall survival(OS) and progression-free survival(PFS) were estimated using the Kaplan-Meier method and compared between groups by log-rank test.Results: Rates of positive immunostaining for VEGFR2 and NRP-1 were 58% and 55%, respectively. KDR and NRP-1 CNG(+) were detected in 32.5% and 30% of tumors, respectively. Levels of both VEGFR2 and NRP-1 in lung tumors were significantly different than in the control tissue(χ2=11.22, P=0.001; χ2=9.82, P=0.001, respectively); similar results were obtained using CNGs(χ2=4.39, P=0.036; χ2=3.95, P=0.046, respectively). Statistically significant correlations were observed with histological grade, clinical TNM stage and the lymph node status(P<0.05), but not age, gender or pathology type(P>0.05). VEGFR2 showed a strong correlation with NRP-1(Rs=0.68, P=0.00); similar results were observed with KDR and NRP-1 CNG(Rs=0.32, P=0.04). Significant differences in OS and PFS were observed between the groups with higher VEGFR2 and NRP-1 and those with lower expression(P<0.05). Conclusions: According to these data, VEGFR2 and NRP-1 are highly expressed in NSCLC. We can conclude that they play a key role in NSCLC occurrence, development and metastasis and are associated with patient prognosis(P<0.05 for OS and PFS). 20Low-Dimensional Halide Perovskites and Their Advanced Optoelectronic Applications显示文摘Metal halide perovskites are crystalline materials originally developed out of scientific curiosity. They have shown great potential as active materials in optoelectronic applications. In the last 6 years, their certified photovoltaic efficiencies have reached 22.1%. Compared to bulk halide perovskites, low-dimensional ones exhibited novel physical properties. The photoluminescence quantum yields of perovskite quantum dots are close to 100%. The external quantum efficiencies and current efficiencies of perovskite quantum dot light-emitting diodes have reached 8% and 43 cd A^(-1),respectively, and their nanowire lasers show ultralow-threshold room-temperature lasing with emission tunability and ease of synthesis. Perovskite nanowire photodetectors reached a responsivity of 10 A W^(-1)and a specific normalized detectivity of the order of 10^(12 )Jones. Different from most reported reviews focusing on photovoltaic applications, we summarize the rapid progress in the study of low-dimensional perovskite materials, as well as their promising applications in optoelectronic devices. In particular, we review the wide tunability of fabrication methods and the state-of-the-art research outputs of low-dimensional perovskite optoelectronic devices.
