3Volatile composition and classification of Lilium flower aroma types and identification,polymorphisms,and alternative splicing of their monoterpene synthase genes显示文摘Lily is a well-known ornamental plant with a diversity of fragrant types.Basic information on lily floral scent compounds has been obtained for only a few accessions,and little is known about Lilium aroma types,the terpene synthase genes that may play roles in the production of key volatiles,or the range of monoterpenes that these genes produce.In this study,41 cultivars were analyzed for volatile emissions,and a total of 46 individual volatile compounds were identified,16 for the first time in lilies.Lily accessions were classified into six groups according to the composition of major scent components:faint-scented,cool,fruity,musky,fruity-honey,and lily.Monoterpenes were one of the main groups of volatiles identified,and attention was focused on terpene synthase(TPS)genes,which encode enzymes that catalyze the last steps in monoterpene synthesis.Thirty-two candidate monoterpene synthase cDNAs were obtained from 66 lily cultivars,and 64 SNPs were identified.Two InDels were also shown to result from variable splicing,and sequence analysis suggested that different transcripts arose from the same gene.All identified nucleotide substitution sites were highly correlated with the amounts of myrcene emitted,and InDel site 230 was highly correlated with the emission of all major monoterpenoid components,especially(E)-β-ocimene.Heterologous expression of five cDNAs cloned from faint-scented and strong-scented lilies showed that their corresponding enzymes could convert geranyl diphosphate to(E)-β-ocimene,α-pinene,and limonene.The findings from this study provide a major resource for the assessment of lily scent volatiles and will be helpful in breeding of improved volatile components.Fang Du Ting Wang Jun-miao Fan Zhi-zhi Liu Jia-xin Zong Wei-xin Fan Yuan-huai Han Donald Grierson 2019Horticulture Research2019,6,1:12
4应用免缝SILO袋分期治疗新生儿腹裂临床研究显示文摘目的 介绍应用免缝SILO袋分期治疗新生儿腹裂。方法 收集芝加哥大学儿童医院 1998年 1月至2 0 0 2年 7月应用免缝SILO袋分期治疗 15例腹裂患儿。患儿体重 2 1~ 3 5kg ,根据腹壁缺损大小选择环口直径 3~ 5cm的SILO袋。脏器回纳腹腔后择期关闭腹壁。结果 15例患儿全部顺利分期关闭腹壁缺损。 2例 (13 3% )SILO袋在使用中滑出腹壁 ,立即于床边重新放置SILO袋。SILO袋平均使用天数为 3 7d ,完全经口进食平均为2 2d。 1例患儿伴有小肠闭锁做肠吻合后再安置SILO袋。随访 3个月至 4年 ,15例全部存活。结论 应用免缝SILO袋分期关闭腹壁缺损是一种安全、可靠、有效的方法。吴晔明 Donald C Liu 2004中国实用儿科杂志2004,19,1:8

6Estimating the prevalence of Echinococcus in domestic dogs in highly endemic for echinococcosis显示文摘Background:Cystic echinococcosis(CE)and alveolar echinococcosis(AE)are highly endemic in Xiji County of Ningxia Hui Autonomous Region(NHAR)in China where the control campaign based on dog de-worming with praziquantel has been undertaken over preceding decades.This study is to determine the current prevalence of Echinococcus granulosus and E.multilocularis in domestic dogs and monitor the echinococcosis transmission dynamics.Methods:Study villages were selected using landscape patterns(Geographic Information System,GIS)for Echinococcus transmission“hot spots”,combined with hospital records identifying risk areas for AE and CE.A survey of 750 domestic dogs,including copro-sampling and owner questionnaires,from 25 selected villages,was undertaken in 2012.A copro-multiplex PCR assay was used for the specific diagnosis of E.granulosus and E.multilocularis in the dogs.Data analysis,using IBM SPSS Statistics,was undertaken,to compare the prevalence of the two Echinococcus spp.in dogs between four geographical areas of Xiji by theχ^(2)test.Univariate analysis of the combinations of outcomes from the questionnaire and copro-PCR assay data was carried out to determine the significant risk factors for dog infection.Results:The highest de-worming rate of 84.0%was found in the northwest area of Xiji County,and significant differences(P<0.05)in the de-worming rates among dogs from the four geographical areas of Xiji were detected.The highest prevalence(19.7%,59/300)of E.multilocularis occurred in northwest Xiji,though the highest prevalence(18.1%,38/210)of E.granulosus occurred in southwest Xiji.There was no significant difference(P>0.05)in the prevalence of E.granulosus in dogs from the northwest,southwest,northeast,and southeast of Xiji,but there were significant differences(P<0.05)between dogs infected with E.multilocularis from the four areas.None of the other independent variables was statistically significant.Conclusions:The results from this study indicate a high prevalence of both E.granulosus and E.muiltilocularis in dogs in Xiji County,NHAR.Transmission of E.multilocularis was more impacted by geographical risk-factors in Xiji County than that of E.granulosus.Dogs have the potential to maintain the transmission of both species of Echinococcus within local Xiji communities,and the current praziquantel dosing of dogs appears to be ineffective or poorly implemented in this area.Cong-Nuan Liu Yang-Yang Xu Angela M.Cadavid-Restrepo Zhong-Zi Lou Hong-Bin Yan Li Li Bao-Quan Fu Darren J.Gray Archie A.Clements Tamsin S.Barnes Gail M.Williams Wan-Zhong Jia Donald P.McManus Yu-Rong Yang 2018Infectious Diseases of Poverty2018,7,1:3
7Chimeric protein probes for C5a receptors through fusion of the anaphylatoxin C5a core region with a small-molecule antagonist显示文摘Blockade of the interaction of anaphylatoxin C5a with its receptor C5aR1 has been actively studied as a potential treatment for many inflammatory diseases;but current C5a antagonists exhibit inadequate potency and poor species cross-reactivity, and novel biochemical tools are needed to investigate whether the core region of C5a contains important interaction epitopes that can explain these limitations. Herein, we report the development of chimeric protein C5a probes containing both the complete core region of rat or human C5a, and the small-molecule antagonist PMX53-1. These probes were chemically synthesized through hydrazide-based native chemical ligation of a linear peptide hydrazide with the requisite cyclopeptidic antagonist, both of which were made by solid-phase synthesis. Quasi-racemic X-ray crystallography established that attachment of PMX53-1 did not affect the structure of the core region of C5a. Subsequent C5aR1 activity assays demonstrated the probes can provide valuable insights into the development of C5a antagonists;for example, they exhibited significantly better binding affinity and much improved species cross-reactivity than PMX53-1, supporting the notion that the effect of some epitopes outside the C-terminus of C5a should be taken into consideration when designing better C5a antagonists. Surprisingly, the core region of C5a was found to partially agonize C5aR1, suggesting the presence of more than one agonistic interaction in the binding of C5a to C5aR1. 8SIMYB1 and SIMYB2, two new MYB genes from tomato, transcriptionally regulate cellulose biosynthesis in tobacco显示文摘Cellulose,a major constituent of plant biomass,is synthesized by a cellulose synthase complex.It has been demonstrated that MYB genes transcriptionally regulate cellulose synthase in Arabidopsis.However,little is known about this process in tomato.Here,two MYB(SIMYB1/2) and three cellulose synthase(CESA)(SICESA4/5/6) genes were isolated.SIMYB1/2and SICESA4I5I6 accumulation was found to correspond to cellulose accumulation in different tissues of tomato.Dual luciferase assays indicated that these two MYSs were transcriptional activators that interact with promoters of SICESA4/5/6.Moreover,SIMYB2 could also activate promoters of SIMYB1/2,suggesting the possible underlying auto-activation mechanisms for MYB transcription factors.Transient over-expression of SIMYB1/2 in Nicotiana tabacum up-regulated tobacco endogenous NtCESA genes and increased cellulose accumulation.The function of SIMYB1/2 was further investigated using stable transformation and the results indicated that N.tabacum lines heterologous expressing SIMYB1/2 displayed a pleiotropic phenotype,long and narrow leaves,with NtCESA induced and significant increase of cellulose.In conclusion,our data suggest that tomato SIMYB1I2 have transcriptional regulatory roles in cellulose biosynthesis and SIMYB2 was more effective than SIMYB1,which may due to the transcriptional activation by SIMYB2 on SIMYB1 and itself.SHI Yan-na LIU Xiao-fen LI Xue DONG Wen-cheng Donald Grierson YIN Xue-ren CHEN Kun-song 2017Journal of Integrative Agriculture2017,16,1:2
