2Ameliorating effects of traditional Chinese medicine preparation, Chinese materia medica and active compounds on ischemia/reperfusion-induced cerebral microcirculatory disturbances and neuron damage显示文摘Ischemic stroke and ischemia/reperfusion(I/R) injury induced by thrombolytic therapy are conditions with high mortality and serious long-term physical and cognitive disabilities. They have a major impact on global public health. These disorders are associated with multiple insults to the cerebral microcirculation, including reactive oxygen species(ROS) overproduction, leukocyte adhesion and infiltration, brain blood barrier(BBB) disruption, and capillary hypoperfusion, ultimately resulting in tissue edema, hemorrhage, brain injury and delayed neuron damage. Traditional Chinese medicine(TCM)has been used in China, Korea, Japan and other Asian countries for treatment of a wide range of diseases.In China, the usage of compound TCM preparation to treat cerebrovascular diseases dates back to the Han Dynasty. Even thousands of years earlier, the medical formulary recorded many classical prescriptions for treating cerebral I/R-related diseases. 4Compound Danshen injection improves endotoxin-induced microcirculatory disturbance in rat mesentery显示文摘AIM: To investigate the effect of compound Danshen injection on lipopolysaccharide (LPS)-induced rat mesenteric microcirculatory dysfunctions and the underlying possible mechanism by an inverted intravital microscope and high-speed video camera system. METHODS: LPS was continuously infused through the jugular artery of male Wistar rats at the dose of 2 mg/kg per hour. Changes in mesenteric microcirculation,such as diameters of arterioles and venules,velocity of RBCs in venules,leukocyte rolling,adhesion and emigration,free radicals released from post-capillary venules,FITC- albumin leakage and mast cell degranulation,were observed through an inverted intravital microscope assisted with CCD camera and SIT camera. Meanwhile,the expression of adhesion molecules CD11b/CD18 and the production of free radical in neutrophils,and the expression of intercellular adhesion molecule 1 (ICAM-1) in human umbilical vein endothelial cells (HUVECs) were quantified by flow cytometry (FACS) in vitro. RESULTS: The continuous infusion with LPS resulted in a number of responses in microcirculation,including a significant increase in the positive region of venulestained with Monastral blue B,rolling and adhesion of leukocytes,production of oxygen radical in venular wall,albumin efflux and enhanced mast cell degranulation in vivo,all of which,except for the leukocyte rolling,were attenuated by the treatment with compound Danshen injection. Experiments performed in vitro further revealed that the expression of CD11b/CD18 and the production of oxygen free radical in neutrophils,and the expression of ICAM-1 in HUVECs were increased by exposure to LPS,and they were attenuated by compound Danshen injection. 5Coesite inclusions in zircon from gneisses identified by laser Raman microspectrometer in ultra-high pressure zone of Dabie Mountains,China显示文摘The mineral inclusions in zircon from gneisses in ultra-high pressure (UHP) zone of the Dabie Mountains were identified by using a laser Raman microspectrometer. Coesite occurs as inclusions in zircons from all types of gneiss. Other important minerals, such as jadeite, omphacite, aragonite, barite, and anhydrite were also found as inclusion minerals. 7Identification and differentiation of Panax ginseng,Panax quinquefolium, and Panax notoginseng by monitoring multiple diagnostic chemical markers显示文摘To differentiate traditional Chinese medicines(TCM) derived from congeneric species in TCM compound preparations is usually challenging. The roots of Panax ginseng(PG), Panax quinquefolium(PQ) and Panax notoginseng(PN) are used as popular TCM. They contain similar triterpenoid saponins(ginsenosides) as the major bioactive constituents. Thus far, only a few chemical markers have been discovered to differentiate these three species. Herein we present a multiple marker detection approach to effectively differentiate the three Panax species, and to identify them in compound preparations. Firstly, 85 batches of crude drug samples(including 32 PG, 30 PQ, and 23 PN) were analyzed by monitoring 40 major ginsenosides in the extracted ion chromatograms(EICs) using a validated LC–MS fingerprinting method. Secondly, the samples were clustered into different groups by pattern recognition chemometric approaches using PLS-DA and OPLS-DA models, and17 diagnostic chemical markers were discovered. Aside from the previously known Rf and p-F11, ginsenoside Rs1 could be a new marker to differentiate PG from PQ. 9Effect of notoginsenoside R1 on hepatic microcirculation disturbance induced by gut ischemia and reperfusion显示文摘AIM: To assess the effect of notoginsenoside R1 on hepatic microcirculatory disturbance induced by gut ischemia/reperfusion (I/R) in mice.METHODS: The superior mesenteric artery (SMA) of C57/BL mice was ligated for 15 min to induce gut ischemia followed by 30-min reperfusion. In another set of experiments, R1 was continuously infused (10 mg/kg per hour) from 10 min before I/R until the end of the investigation to study the influence of R1 on hepatic microcirculatory disturbance induced by gut I/R. Hepatic microcirculation was observed by inverted microscopy, and the vascular diameter, red blood cell (RBC) velocity and sinusoid perfusion were estimated. Leukocyte rolling and adhesion were observed under a laser confocal microscope. Thirty and 60 min after reperfusion, lactate dehydrogenase (LDH), alanine aminotransferase (ALT) and aspartate transaminase (AST) in peripheral blood were determined. The expression of adhesion molecules CD11b/CD18 in neutrophils and tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6) and monocyte chemotactic protein-1 (MCP-1) in plasma were evaluated by flow cytometry. E-selectin and intercellular adhesion molecule-1 (ICAM-1) in hepatic tissue were examined by immunofluorescence. RESULTS: After gut I/R, the diameters of terminal portal venules and central veins, RBC velocity and the number of perfused sinusoids were decreased, while the leukocyte rolling and adhesion, the expression of E-selectin in hepatic vessels and CD18 in neutrophils, IL-6, MCP-1, LDH, ALT and AST were increased. R1 treatment attenuated these alterations except for IL-6 and MCP-1.CONCLUSION: R1 prevents I/R-induced hepatic microcirculation disturbance and hepatocyte injury. 10Total salvianolic acid improves ischemia-reperfusion-induced microcirculatory disturbance in rat mesentery显示文摘AIM:To investigate the effect of total salvianolic acid(TSA) on ischemia-reperfusion(I/R)-induced rat mesenteric microcirculatory dysfunctions.METHODS:Male Wistar rats were randomly distributed into 5 groups(n = 6 each):Sham group and I/R group(infused with saline),TSA group,TSA + I/R group and I/R + TSA group(infused with TSA,5 mg/kg per hour).Mesenteric I/R were conducted by a ligation of the mesenteric artery and vein(10 min) and subsequent release of the occlusion.TSA was continuously infused either starting from 10 min before the ischemia or 10 min after reperfusion.Changes in mesenteric microcirculatory variables,including diameter of venule,velocity of red blood cells in venule,leukocyte adhesion,free radicals released from venule,albumin leakage and mast cell degranulation,were observed through an inverted intravital microscope.Meanwhile,the expression of adhesion molecules CD11b/CD18 on neutrophils was evaluated by flow cytometry.Ultrastructural evidence of mesenteric venules damage was assessed after microcirculation observation.RESULTS:I/R led to multiple responses in mesenteric post-capillary venules,including a significant increase in the adhesion of leukocytes,production of oxygen radicals in the venular wall,albumin efflux and enhanced mast cell degranulation in vivo.All the I/R-induced manifestations were significantly reduced by pre-or post-treatment with TSA,with the exception that the I/R-induced increase in mast cell degranulation was inhibited only by pre-treatment with TSA.Moreover,preor post-treatment with TSA significantly attenuated the expression of CD11b/CD18 on neutrophils,reducing the increase in the number of caveolae in the endothelial cells of mesentery post-capillary venules induced by I/R.CONCLUSION:The results demonstrated that TSA protects from and ameliorates the microcirculation disturbance induced by I/R,which was associated with TSA inhibiting the production of oxygen-free radicals in the venular wall and the expression of CD11b/CD18 on neutrophils.Ming-Xia Wang Yu-Ying Liu Bai-He Hu Xiao-Hong Wei Xin Chang Kai Sun Jing-Yu Fan Fu-Long Liao ChuanShe Wang Jun Zheng Jing-Yan Han Ming-Xia Wang 2010World Journal of Gastroenterology2010,16,42:15

12Xiba Granitic Pluton in the Qinling Orogenic Belt, Central China: Its Petrogenesis and Tectonic Implications显示文摘Xiba granitic pluton is located in South Qinling tectonic domain of the Qinling orogenic belt and consists mainly of granodiorite and monzogranite with significant number of microgranular quartz dioritic enclaves. SHRIMP zircon U-Pb isotopic dating reveals that the quartz dioritic enclaves formed at 214±3 Ma, which is similar to the age of their host monzogranite (218±1 Ma). The granitoids belong to high-K calc-alkaline series, and are characterized by enriched LILEs relative to HFSEs with negative Nb, Ta and Ti anomalies, and right-declined REE patterns with (La/Yb) N ratios ranging from 15.83 to 26.47 and δEu values from 0.78 to 1.22 (mean= 0.97). Most of these samples from Xiba granitic pluton exhibit εNd(t) values of 8.79 to 5.38, depleted mantle Nd model ages (T DM ) between 1.1 Ga and 1.7 Ga, and initial Sr isotopic ratios ( 87 Sr/ 86 Sr) i from 0.7061 to 0.7082, indicating a possible Meso-to Paleoproterozoic lower crust source region, with exception of samples XB01-2-1 and XB10-1 displaying higher ( 87 Sr/ 86 Sr) i values of 0.779 and 0.735, respectively, which suggests a contamination of the upper crustal materials. Quartz dioritic enclaves are interpreted as the result of rapid crystallization fractionation during the parent magmatic emplacement, as evidenced by similar age, texture, geochemical, and Sr-Nd isotopic features with their host rocks. Characteristics of the petrological and geochemical data reveal that the parent magma of Xiba granitoids was produced by a magma mingling process. The upwelling asthenosphere caused a high heat flow and the mafic magma was underplated into the bottom of the lower continent crust, which caused the partial melting of the lower continent crustal materials. This geodynamic process generated the mixing parent magma between mafic magma from depleted mantle and felsic magma derived from the lower continent crust. 13How we can improve patients’comfort after Milligan- Morgan open haemorrhoidectomy显示文摘AIM:To demonstrate the value of Diosmin(flavonidic fraction)in the management of post-haemorhoidectomic symptoms.METHODS:Eighty-six consecutive patients with grades ⅢandⅣacute mixed hemorrhoids admitted to the Anorectal Surgical Department of First Affiliated Hospital,Xinjiang Medical University from April 2009 to April 2010,were enrolled in this study.An observerblinded,randomized trial was conducted to compare post-haemorhoidectomic symptoms with use of Diosmin flavonidic fraction vs placebo.Eighty-six patients were randomly allocated to receive Diosmin flavonidic fraction 500 mg for 1 wk(n=43)or placebo(n=43).The Milligan-Morgan open haemorrhoidectomy was performed by a standardized diathermy excision method.Pain,bleeding,heaviness,pruritus,wound edema and mucosal discharge were observed after surgery.The postoperative symptoms and hospitalization time were recorded.RESULTS:The mean age of the Diosmin group and controls was 53.2 and 51.3 years,respectively.In Diosmin group,haemorrhoid piles were of the third degree in 33 patients and the fourth degree in 10;and in the control group,29 were of the third degree and 14 were of the fourth degree.There was no statistically significance in age,gender distribution,degree and number of excised haemorrhoid piles,and the mean duration of haemorrhoidal disease between the two groups.There was a statistically significant improvement in pain,heaviness,bleeding,pruritus from baseline to the 8th week after operation(P<0.05).Patients taking Diosmin had a shorter hospitalization stay after surgery(P< 0.05).There was also a significant improvement on the proctoscopic appearance(P<0.001).However,there was no statistical difference between the two groups in terms of wound mucosal discharge.Two patients experienced minor bleeding at the 8th week in Diosmin group,and underwent surgery.CONCLUSION:Diosmin is effective in alleviating postoperational symptoms of haemorrhoids.Therefore,it should be considered for the initial treatment after haemorrhoid surgery.However,further prospective randomized trials are needed to confirm the findings of this study.Ma-Mu-Ti-Jiang A ba-bai-ke-re Hong-Guo Huang Wen-Ni Re Kai Fan Hui Chu Er-Ha-Ti Ai Mai-Mai-Ti-Tu-Er-Xun KE Li-Mu Yi-Rui Wang Hao Wen 2011World Journal of Gastroenterology2011,17,11:13

17Design and test of tray-seedling sorting transplanter显示文摘In order to improve the automatic sorting and transplanting of tray-seedling and reduce labors strength,a new sorting transplanter was designed.According to the feature of normal seedling tray and manual grading method,the mechanical sorting procedure was defined.The key parts of sorting transplanter are as follows:an efficient transfer mechanism with aerodynamic buffer,a seedling identifying and measuring unit based on linear-structured vision,the extendible multi-grasper with wireless technology and the system controller.The experiments were conducted and showed that the transplanting success rate was 90.0% on the tray of 6×12 holes under the speed of 700 cycles per hour.The vision unit’s failed identifying rate for the superior pepper seedlings on five different trays was less than 10%,and the measurement error of seedling height was about 5 mm.Feng Qingchun Zhao Chunjiang Jiang Kai Fan Pengfei Wang Xiu 2015International Journal of Agricultural and Biological Engineering2015,8,2:12
18TAOK1 negatively regulates IL-17-mediated signaling and inflammation显示文摘Interleukin 17(IL-17)is an important cytokine that can induce tissue inflammation and is involved in the pathogenesis of numerous autoimmune diseases.However,the regulation of its signaling transduction has not been well described.In this study,we report that thousand and one kinase 1(TAOK1)functions as a negative regulator of IL-17-mediated signal transduction and inflammation.TAOK1 knockdown promotes IL-17-induced cytokine and chemokine expression and the activation of mitogen-activated protein kinases and nuclear factor-κB.We further demonstrate that TAOK1 interacts with IL-17 receptor A(IL-17RA)independent of its kinase activity,and TAOK1 dose-dependently prevents the formation of the IL-17R-Act1(nuclear factor activator 1,also known as tumor necrosis factor receptor-associated factor 3 interacting protein 2)complex.Consistent with this,TAOK1 deficiency exacerbates colitis in the 2,4,6-trinitrobenzenesulfonic acid-induced experimental model of inflammatory bowel disease,likely by its promotion of the IL-17-mediated signaling pathway.TAOK1 expression is decreased in the colons of ulcerative colitis patients.In conclusion,these findings suggest that TAOK1 is involved in the development of IL-17-related autoimmune disorders.Zhaoru Zhang Zhen Tang Xianwei Ma Kai Sun Liping Fan Jie Fang Jianping Pan Xiaojian Wang Huazhang An Jun Zhou 2018Cellular & Molecular Immunology2018,15,8:10
19TaqMan Real-time RT-PCR Assay for Detecting and Differentiating Japanese Encephalitis Virus显示文摘Objective To detect Japanese encephalitis virus(JEV) rapidly and distinguish its genotypes, a TaqMan-based reverse transcriptase quantitative polymerase chain reaction(RT-PCR) detection system was developed.Methods By aligning the full-length sequences of JEV(G1-G5), six sets of highly specific TaqMan real-time RT-PCR primers and probes were designed based on the highly conserved NS1, NS2, and M genes of JEV, which included one set for non-specific JEV detection and five sets for the detection of specific JEV genotypes. Twenty batches of mosquito samples were used to evaluate our quantitative PCR assay.Results With the specific assay, no other flavivirus were detected. The lower limits of detection of the system were 1 pfu/mL for JEV titers and 100 RNA copies/μL. The coefficients of variation of this real-time RT-PCR were all < 2.8%. 20Microbial aerosol characteristics in highly polluted and nearpristine environments featuring different climatic conditions显示文摘There is an increasing interest in understanding ambient bioaerosols due to their roles both in health and in climate. Here, we deployed an Ultraviolet Aerodynamic Particle Sizer to monitor viable(fluorescent) bioaerosol concentration levels at city centers(highly polluted) and their corresponding suburbs(near pristine)(total 40 locations) in 11 provinces featuring different climate zones in China between July 16 and 28, 2013. The concentration levels of viable bioaerosol particles(Bio PM) of [0.5 lm were measured, and corresponding percentages of Bio PM%(biological fraction of total PM) and Bio PM2.5%(biological fraction of PM2.5) in particulate matter(PM)and Bio PM, respectively, were determined. For some key cities, indoor viable bioaerosol levels were also obtained.In addition, bacterial structures of the air samples collected across these monitoring locations were studied using pyrosequencing. Bio PM concentration levels ranged from2.1 9 104 to 2.4 9 105/m3 for city centers [Bio PM% =6.4 %(±6.3 %)] and 0.5 9 104 to 4.7 9 105/m3 for suburbs [Bio PM% = 10 %(±8.7 %)]. Distinctive bioaerosol size distribution patterns were observed for different climate zones, e.g., some had fluorescence peaks at 3 lm, while the majority had peaks at 1 lm. Ambient bacterial aerosol community structures were also found different for different geophysical locations. Results suggest that there was a poor overall relationship between PM and Bio PM across 40 monitoring locations(R2= 0.081, two-tailed P value =0.07435). Generally, city centers had higher PM concentrations than suburbs, but not Bio PM and Bio PM%. Indoor bioaerosol levels were found at least tenfold higher than those corresponding outdoors.
