| 7 | Timing of formation of the Hongdonggou Pb-Zn polymetallic ore deposit,Henan Province,China:Evidence from Rb-Sr isotopic dating of sphalerites显示文摘The Hongdonggou Pb-Zn polymetallic ore deposit,located in the southwestern part of the Luanchuan Mo-W-Pb-Zn-Ag polymetallic ore mineralization in Henan Province,China,is an important part of the East Qinling metallogenic belt.The orebodies in the deposit,which are vein,bedded and lenticular,are mainly hosted in the syenite porphyry,and formed within the carbonate and clastic rocks of the Yuku and Qiumugou formations partially.The genesis of the deposit has previously been argued to be of hydrothermal-vein type or of skarn-hydrothermal type.In this study,we report the results of Rb-Sr isotopic dating based on sphalerites from the main orebody of the Hongdonggou Pb-Zn polymetallic ore deposit,which yield an isochron age of 135.7 ± 3.2 Ma,constraining the timing of mineralization as early Cretaceous.The age is close to those reported for the Pb-Zn deposits in the Luanchuan ore belt.The(^(87)Sr/^(86)Sr),values of the sphalerites(0.71127 + 0.00010) are lower than that of terrigenous silicates(0.720) and higher than the mantle(0.707),suggesting that the metallogenic components were mainly derived through crust-mantle mixing.Combining the results from this study with those from previous work,we propose that the Hongdonggou Pb-Zn polymetallic ore deposit is a hydrothermal-vein deposit associated with the early Cretaceous tectonothermal event,and the mineralization is controlled by NWand near EW-trending faults in the Luanchuan Mo-W-Pb-Zn-Ag polymetallic ore concentration belt. | Fan Yang Gongwen Wang Huawen Cao Ruixi Li Li Tang Yufeng Huang Hao Zhang Fei Xue Wenjuan Jia Nana Guo | 2017 | Geoscience Frontiers2017,8,3: | 2 |
| 8 | Production of transgenic pigs over-expressing theantiviral gene Mx1显示文摘The myxovirus resistance gene (Mx1) has a broad spectrum of antiviral activities. It is therefore an interestingcandidate gene to improve disease resistance in farm animals. In this study, we report the use of somatic cellnuclear transfer (SCNT) to produce transgenic pigs over-expressing the Mx1 gene. These transgenic pigs expressapproximately 15–25 times more Mx1 mRNA than non-transgenic pigs, and the protein level of Mx1 was alsomarkedly enhanced. We challenged fibroblast cells isolated from the ear skin of transgenic and control pigs withinfluenza A virus and classical swine fever virus (CFSV). Indirect immunofluorescence assay (IFA) revealed a profounddecrease of influenza A proliferation in Mx1 transgenic cells. Growth kinetics showed an approximately 10-foldreduction of viral copies in the transgenic cells compared to non-transgenic controls. Additionally, we found thatthe Mx1 transgenic cells were more resistant to CSFV infection in comparison to non-transgenic cells. | 9 | Effects of protein oxidation, cathepsins, and various freezing temperatures on the quality of superchilled sturgeon fillets显示文摘Many aquatic products have been stored using superchilling technology, but rarely used for the storage of sturgeon fillets. In this study, we investigated the effects of protein oxidation, cathepsin, and various freezing temperatures on the quality of superchilled sturgeon fillets. Sensory evaluation results showed that the sensory attributes of superchilled (−3 °C) sturgeon fillets were acceptable three times longer (18 days) than samples stored at refrigeration temperatures (4 °C). The sarcoplasmic protein, carbonyl, myofibrillar protein, total sulfhydryl content and the surface hydrophobicity were determined using fluorescence spectrophotometry and SDS-PAGE. Results showed that superchilling might protect myofibrillar proteins from oxidation compared to refrigeration temperatures. The activity of the three cathepsins (B, L, and H) in terms of myofibrillar, mitochondria, lysosomes, and sarcoplasm demonstrated that superchilling can inhibit cathepsins activity in sturgeon and protect its muscle structure. Microscopic observations showed that as the temperature decreased, the gap area of the muscle fibers decreased, and the deformation of cross-sectional slices was gradually reduced. In addition, the freezing rate of ice crystals produced during the freezing process influenced the muscle structure, texture, and sensory attributes. Superchilled sturgeon fillets showed good hardness, chewiness, and water retention. | 19 | Scutellarin prevents acute alcohol-induced liver injury via inhibiting oxidative stress by regulating the Nrf2/HO-1 pathway and inhibiting inflammation by regulating the AKT,p38 MAPK/NF-κB pathways显示文摘Alcoholic liver disease(ALD)is the most frequent liver disease worldwide,resulting in severe harm to personal health and posing a serious burden to public health.Based on the reported antioxidant and anti-inflammatory capacities of scutellarin(SCU),this study investigated its protective role in male BALB/c mice with acute alcoholic liver injury after oral administration(10,25,and 50 mg/kg).The results indicated that SCU could lessen serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)levels and improve the histopathological changes in acute alcoholic liver;it reduced alcohol-induced malondialdehyde(MDA)content and increased glutathione peroxidase(GSH-Px),catalase(CAT),and superoxide dismutase(SOD)activity.Furthermore,SCU decreased tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),and IL-1βmessenger RNA(mRNA)expression levels,weakened inducible nitric oxide synthase(iNOS)activity,and inhibited nucleotide-binding oligomerization domain(NOD)-like receptor protein 3(NLRP3)inflammasome activation.Mechanistically,SCU suppressed cytochrome P450 family 2 subfamily E member 1(CYP2E1)upregulation triggered by alcohol,increased the expression of oxidative stress-related nuclear factor erythroid 2-related factor 2(Nrf2)and heme oxygenase-1(HO-1)pathways,and suppressed the inflammation-related degradation of inhibitor of nuclear factor-κB(NF-κB)-α(IκBα)as well as activation of NF-κB by mediating the protein kinase B(AKT)and p38 mitogen-activated protein kinase(MAPK)pathways.These findings demonstrate that SCU protects against acute alcoholic liver injury via inhibiting oxidative stress by regulating the Nrf2/HO-1 pathway and suppressing inflammation by regulating the AKT,p38 MAPK/NF-κB pathways. | Xiao ZHANG Zhicheng DONG Hui FAN Qiankun YANG Guili YU Enzhuang PAN Nana HE Xueqing LI Panpan ZHAO Mian FU Jingquan DONG | 2023 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,7: | 0 |
| 20 | Progranulin regulation of autophagy contributes to its chondroprotective effect in osteoarthritis显示文摘Progranulin(PGRN)is a multifunctional growth factor involved in many physiolog-ical processes and disease states.The apparent protective role of PGRN and the importance of chondrocyte autophagic function in the progression of osteoarthritis(OA)led us to investi-gate the role of PGRN in the regulation of chondrocyte autophagy.PGRN knockout chondro-cytes exhibited a deficient autophagic response with limited induction following rapamycin,serum starvation,and IL-1b-induced autophagy.PGRN-mediated anabolism and suppression of IL-1b-induced catabolism were largely abrogated in the presence of the BafA1 autophagy inhibitor.Mechanistically,during the process of OA,PGRN and the ATG5eATG12 conjugate form a protein complex;PGRN regulates autophagy in chondrocytes and OA through,at least partially,the interactions between PGRN and the ATG5eATG12 conjugate.Furthermore,the ATG5eATG12 conjugate is critical for cell proliferation and apoptosis.Knockdown or knockout of ATG5 reduces the expression of ATG5eATG12 conjugate and inhibits the chondroprotective effect of PGRN on anabolism and catabolism.Overexpression of PGRN partially reversed this effect.In brief,the PGRN-mediated regulation of chondrocyte autophagy plays a key role in the chondroprotective role of PGRN in OA.Such studies provide new insights into the pathogen-esis of OA and PGRN-associated autophagy in chondrocyte homeostasis. | Yiming Pan Yuyou Yang Mengtian Fan Cheng Chen Rong Jiang Li Liang Menglin Xian Biao Kuang Nana Geng Naibo Feng Lin Deng Wei Zheng Fengmei Zhang Xiaoli Li Fengjin Guo | 2023 | Genes & Diseases2023,10,4: | 0 |
