| 1 | Network Pharmacology-based Approaches Capture Essence of Chinese Herbal Medicines显示文摘Traditional Chinese Medicine(TCM), a crucial component of the current medical system, has been extensively used in clinical practice due to its valuable therapeutic efficacy, and its potentials as an important source of new pharmacophores. TCM is characterized by holistic theory, which emphasizes maintaining the balance of the patient's whole body using herbal formulae(fangji in Chinese) composed of mixtures of herbs with multiple bioactive ingredients. Because of the complex nature of these formulae, it is necessary to develop systematic methods to identify their bioactive ingredients and to clarify their mechanisms of action. With the rapid progress in bioinformatics, systems biology, and polypharmacology, 'network pharmacology', which shifts the 'one target, one drug' paradigm to the 'network target, multi-component' strategy, has attracted the attention because it can not only reveal the underlying complex interactions between a herbal formula and cellular proteins but detect the influence of their interactions on the function and behavior of the system. Growing evidence shows that the network pharmacology strategy can be a powerful approach to modern research on TCM. | 3 | A comprehensive classification method for VOC emission sources to tackle air pollution based on VOC species reactivity and emission amounts显示文摘In China, volatile organic compound(VOC) control directives have been continuously released and implemented for important sources and regions to tackle air pollution. The corresponding control requirements were based on VOC emission amounts(EA), but never considered the significant differentiation of VOC species in terms of atmospheric chemical reactivity. This will adversely influence the effect of VOC reduction on air quality improvement. Therefore,this study attempted to develop a comprehensive classification method for typical VOC sources in the Beijing–Tianjin–Hebei region(BTH), by combining the VOC emission amounts with the chemical reactivities of VOC species. Firstly, we obtained the VOC chemical profiles by measuring 5 key sources in the BTH region and referencing another 10 key sources, and estimated the ozone formation potential(OFP) per ton VOC emission for these sources by using the maximum incremental reactivity(MIR) index as the characteristic of source reactivity(SR). Then, we applied the data normalization method to respectively convert EA and SR to normalized EA(NEA) and normalized SR(NSR) for various sources in the BTH region.Finally, the control index(CI) was calculated, and these sources were further classified into four grades based on the normalized CI(NCI). | 4 | Oridonin induces apoptosis in gastric cancer through Apaf-1,cytochrome c and caspase-3 signaling pathway显示文摘AIM:To investigate the effect and mechanism of oridonin on the gastric cancer cell line HGC-27 in vitro.METHODS:The inhibitory effect of oridonin on HGC-27 cells was detected using the 3-(4,5-dimethylthiazol2-yl)-2,5-diphenyl tetrazolium bromide assay.After treatment with 10 μg/mL oridonin for 24 h and 48 h,the cells were stained with acridine orange/ethidium bromide.The morphologic changes were observed under an inverted fluorescence microscope.DNA fragmen-tation(a hallmark of apoptosis) and lactate dehydrogenase activity were examined using DNA ladder assay and lactate dehydrogenase-release assay.After treated with oridonin(0,1.25,2.5,5 and 10 μg/mL),HGC-27 cells were collected for anexin V-phycoerythrin and 7-amino-actinomycin D double staining and tested by flow cytometric analysis,and oridonin-induced apoptosis in HGC-27 cells was detected.After treatment with oridonin for 24 h,the effects of oridonin on expression of Apaf-1,Bcl-2,Bax,caspase-3 and cytochrome c were also analyzed using reverse-transcript polymerase chain reaction(RT-PCR) and Western blotting.RESULTS:Oridonin significantly inhibited the proliferation of HGC-27 cells in a dose-and time-dependent manner.The inhibition rates of HGC-27 treated with four different concentrations of oridonin for 24 h(1.25,2.5,5 and 10 μg/mL) were 1.78% ± 0.36%,4.96% ± 1.59%,10.35% ± 2.76% and 41.6% ± 4.29%,respectively,which showed a significant difference(P < 0.05).The inhibition rates of HGC-27 treated with oridonin at the four concentrations for 48 h were 14.77% ± 4.21%,21.57% ± 3.75%,30.31% ± 4.91% and 61.19% ± 5.81%,with a significant difference(P < 0.05).The inhibition rates of HGC-27 treated with oridonin for 72 h at the four concentrations were 25.77% ± 4.85%,31.86% ± 3.86%,48.30% ± 4.16% and 81.80% ± 6.72%,with a significant difference(P < 0.05).Cells treated with oridonin showed typical apoptotic features with acridine orange/ethidium bromide staining.After treatment with oridonin,the cells became round,shrank,and developed small buds around the nuclear membrane while forming apoptotic bodies.Lactate dehydrogenase(LDH) release assay showed that after treated with 1.25 μg/mL and 20 μg/mL oridonin for 24 h,LDH release of HGC-27 caused by apoptosis increased from 22.94% ± 3.8% to 52.68% ± 2.4%(P < 0.001).However,the change in the release of LDH caused by necrosis was insignificant,suggesting thatthe major cause of oridonin-induced HGC-27 cell death was apoptosis.Flow cytometric analysis also revealed that oridonin induced significant apoptosis compared with the controls(P < 0.05).And the apoptosis rates of HGC-27 induced by the four different concentrations of oridonin were 5.3% ± 1.02%,12.8% ± 2.53%,28.5% ± 4.23% and 49.6% ± 3.76%,which were in a dose-dependent manner(P < 0.05).After treatment for 24 h,DNA ladder showed that oridonin induced a significant increase in DNA fragmentation in a dosedependent manner.RT-PCR revealed that mRNA expression levels were up-regulated compared with the controls in caspase-3(0.917 ± 0.103 vs 0.357 ± 0.019,P < 0.05),cytochrome c(1.429 ± 0.111 vs 1.002 ± 0.014,P < 0.05),Apaf-1(0.688 ± 0.101 vs 0.242 ± 0.037,P < 0.05) and Bax(0.856 ± 0.101 vs 0.278 ± 0.027,P < 0.05)(P < 0.05),whereas down-regulated in Bcl-2(0.085 ± 0.012 vs 0.175 ± 0.030,P < 0.05).Western blotting analysis also confirmed this result.CONCLUSION:Apoptosis of HGC-27 induced by oridonin may be associated with differential expression of Apaf-1,caspase-3 and cytochrome c,which are highly dependent upon the mitochondrial pathway. | Ke-Wang Sun Ying-Yu Ma Tian-Pei Guan Ying-Jie Xia Chang-Ming Shao Le-Gao Chen Ya-Jun Ren Hai-Bo Yao Qiong Yang Xu-Jun He | 2012 | World Journal of Gastroenterology2012,18,48: | 25 |

| 6 | Genome analysis of Taraxacum kok-saghyz Rodin provides new insights into rubber biosynthesis显示文摘The Russian dandelion Taraxacum kok-saghyz Rodin(TKS), a member of the Composite family and a potential alternative source of natural rubber(NR) and inulin, is an ideal model system for studying rubber biosynthesis. Here we present the draft genome of TKS, the first assembled NR-producing weed plant. The draft TKS genome assembly has a length of 1.29 Gb, containing 46 731 predicted protein-coding genes and68.56% repeats, in which the LTR-RT elements predominantly contribute to the genome enlargement. We analyzed the heterozygous regions/genes, suggesting its possible involvement in inbreeding depression.Through comparative studies between rubber-producing and non-rubber-producing plants, we found that enzymes of the mevalonate(MVA) pathway and rubber elongation might be critical for rubber biosynthesis, and several key isoforms have been isolated and shown to be predominantly expressed in the latex, indicating their crucial functions in rubber biosynthesis. Moreover, for two important families in rubber elongation, the CPT/CPTL and REF/SRPP families, diverse evolutionary tracks have been revealed. | 10 | Progress in miRNA target prediction and identification显示文摘Recently, the identification of miRNA targets has received much attention. The strategies to determine miRNA targets include bioinformatic prediction and experimental assays. The bioinformatic prediction methods are mainly based on the confirmed rules of interaction between miRNAs and their targets, and are carried out by programs, such as miRanda, TargetScan, TargetScanS, RNAhybrid, DIANA-microT, PicTar, RNA22 and FindTar, which follow well-known principles. The experimental assays to find miRNA targets employ immunoprecipitation of AGO proteins to identify interacting mRNAs, or the analysis of mRNA or protein levels to identify genes which can be regulated by miRNAs. The improvement of current bioinformatic and experimental assays and the development of novel assays will enable greater efficiency in the identification of miRNA targets and thus facilitate miRNA research. | 12 | Metal distribution characteristic of MSWI bottom ash in view of metal recovery显示文摘Bottom ash is the major by-product of municipal solid waste incineration(MSWI), and is often reused as an engineering material, such as road-base aggregate. However, some metals(especially aluminum) in bottom ash can react with water and generate gas that could cause expansion and failure of products containing the ash; these metals must be removed before the ash is utilized. The size distribution and the chemical speciation of metals in the bottom ash from two Chinese MSWI plants were examined in this study, and the recovery potential of metals from the ash was evaluated. The metal concentrations in these bottom ashes were lower than that generated in other developed countries. Specifically, the contents of Al,Fe, Cu and Zn were 18.9–29.2, 25.5–32.3, 0.7–1.0 and 1.6–2.5 g/kg, respectively. Moreover,44.9–57.0 wt.% of Al and 55.6–75.4 wt.% of Fe were distributed in bottom ash particles smaller than 5 mm. Similarly, 46.6–79.7 wt.% of Cu and 42.9–74.2 wt.% of Zn were concentrated in particles smaller than 3 mm. | 13 | Electroacupuncture regulates glucose-inhibited neurons in treatment of simple obesity显示文摘The glucose-inhibited neurons present in the lateral hypothalamic area are regarded as glucose detectors. This structure is involved in the regulation of food intake through extracellular blood glucose concentrations, and plays a crucial role in obesity onset. In the present study, obesity models established with high fat feeding were treated with electroacupuncture at Zusanli (ST36)/ Inner Court (ST44) on the left side and Tianshu (ST25) bilaterally. We found that electroacupuncture could effectively reduce body weight and the fat-weight ratio, and decrease serum leptin, resistin, tumor necrosis factor alpha, and neuropeptide Y levels, while increase serum adiponectin and cholecystokinin-8 levels. This treatment altered the electrical activity of glucose-inhibited neurons in the lateral hypothalamic area, with electroacupuncture at Zusanli/ Inner Court exerting an inhibitory effect, while electroacupuncture at bilateral Tianshu exerting an excitatory effect. | 16 | Multi-scale traffic vehicle detection based on faster ReCNN with NAS optimization and feature enrichment显示文摘It well known that vehicle detection is an important component of the field of object detection.However,the environment of vehicle detection is particularly sophisticated in practical processes.It is comparatively difficult to detect vehicles of various scales in traffic scene images,because the vehicles partially obscured by green belts,roadblocks or other vehicles,as well as influence of some low illumination weather.In this paper,we present a model based on Faster ReCNN with NAS optimization and feature enrichment to realize the effective detection of multi-scale vehicle targets in traffic scenes.First,we proposed a Retinex-based image adaptive correction algorithm(RIAC)to enhance the traffic images in the dataset to reduce the influence of shadow and illumination,and improve the image quality.Second,in order to improve the feature expression of the backbone network,we conducted Neural Architecture Search(NAS)on the backbone network used for feature extraction of Faster ReCNN to generate the optimal cross-layer connection to extract multi-layer features more effectively.Third,we used the object Feature Enrichment that combines the multi-layer feature information and the context information of the last layer after cross-layer connection to enrich the information of vehicle targets,and improve the robustness of the model for challenging targets such as small scale and severe occlusion.In the implementation of the model,K-means clustering algorithm was used to select the suitable anchor size for our dataset to improve the convergence speed of the model.Our model has been trained and tested on the UN-DETRAC dataset,and the obtained results indicate that our method has art-of-state detection performance. | Ji-qing Luo Hu-sheng Fang Fa-ming Shao Yue Zhong Xia Hua | 2021 | Defence Technology(防务技术)2021,17,4: | 10 |
| 17 | Mycoplasma pneumoniae Macrolide Resistance and MLVA Typing in Children in Beijing,China,in 2016:Is It Relevant?显示文摘Objective The aim of this study is to investigate the macrolide resistance rate and molecular type with multiple-locus variable-number tandem-repeat analysis(MLVA)of Mycoplasma pneumoniae of Beijing in 2016 in pediatric patients.Methods Real-time quantitative polymerase chain reaction(PCR)was used to identify M.pneumoniae,and MLVA was performed.The domain V of the 23 S rRNA was sequenced to detect macrolide-resistant point mutations.We also investigated the activities of antibiotics against M.pneumoniae isolates in vitro.Results The PCR detection rate of M.pneumoniae in children in Beijing was 40%,and the macrolide resistance rate was 66%.The A2063 G mutation in the 23 S rRNA V region is the dominant mutation(137/146,93.84%),whereas the A2064 G mutation is rare(9/146,6.16%).Seventy-three samples were typed successfully by MLVA typing,including 86.3%(63/73)were MLVA type 4-5-7-2,and 13.7%(10/73)were MLVA type 3-5-6-2.No other types were found.No strains were resistant to levofloxacin or tetracycline.Conclusion In 2016,a specific decrease in the macrolide resistance rate occurred in Beijing.The detection rate and macrolide resistance rate of outpatients are lower than those of inpatients.The A2063 G mutants M.pneumoniae have high levels of resistance to erythromycin and azithromycin.The primary MLVA type is 4-5-7-2,followed by 3-5-6-2.No other MLVA types were detected.No strains resistant to tetracycline or levofloxacin were found in vitro. | DOU Hai Wei TIAN Xiu Jun XIN De Li WEI Ran ZHOU Wei WANG Hong QIN Xuan Guang SHAO Jun Yan XU Bao Ping GE Li Xia SHI Da Wei | 2020 | Biomedical and Environmental Sciences2020,33,12: | 9 |

| 19 | Effect of Cyclocarya paliurus polysaccharides on streptozotocin-induced diabetic nephropathy in rats显示文摘OBJEVTIVE:To investigate the efficacy of Cyclocarya paliurus(C.paliurus)polysaccharides on streptozotocin-induced diabetic nephropathy in rats.METHODS:Rats were divided into 6 groups,including group of normal control,group of diabetic control,group of metformin treatment,low-dose group of C.paliurus polysaccharides treatment,middle-dose group of C.paliurus polysaccharides treatment and high-dose group of C.paliurus polysaccharides treatment.Histological analysis of kidney was analyzed using hematoxilin and eosin.Levels of blood glucose,creatinine,urea,uric acid were determined by spectrophotometry.Anti-oxidative enzymes were measured by real-time polymerase chain reaction(PCR)and enzyme-linked immunosorbent assay(ELISA).Advanced glycation end products(AGEs)and transforming growth factor-β1(TGF-β1)level was measured by ELISA.RESULTS:Abnormal changes were observed in the group of diabetic control characterized by atrophy of the renal glomeruli with hypercellularity,congestion of glomerular tufts,dilation of the renal spaces,and degeneration of renal tubule.Compared with that of normal group,blood glucose,creatinine,urea,uric acid level was significantly increased in the group of diabetic control.Superoxide dismutase,catalase,glutathione peroxidase,glutathione reductase level was significantly decreased,but AGEs and TGF-β1 level was significantly increased.By contrast,administration of C.paliurus polysaccharides and metformin could reverse the above-mentioned results of the group of diabetic control,especially in the high-dose group of C.paliurus polysaccharides.CONCLUSION:Our findings suggest that C.paliurus polysaccharides may play a protecting role for nephropathy of diabetic rats by lowering glucose,creatinine,urea,uric acid level,enhancing the antioxidative ability,and reducing AGEs and TGF-β1 expression. | Xia Xichao Mao Dongxue Dai Hongmei Wu Xi Zhang Zuyuan Wang Huaying Zhou Wenwen Dong Yanmei Wang Mengqi Li Yuan Shao Xiangyang Ouyang Jingfeng | 2020 | Journal of Traditional Chinese Medicine2020,40,6: | 9 |
