1Anti-tumor effect of the extract from Qingyihuaji formula on pancreatic cancer by down-regulating Notch-4 and Jagged-1显示文摘OBJECTIVE: To investigate, in terms of Notch signaling pathway, the effect on pancreatic cancer of the extract of an anti-tumor prescription — Qingyihuaji formula(QYHJ) — from Traditional Chinese Medicine(TCM).METHODS: Nude mice were implanted subcutaneously with human pancreatic cancer cell line SW1990 and then randomly divided into four groups: Control, QYHJ extract, Gemcitabine, and Combination of QYHJ extract and gemcitabine.Treatments were given for 21 days and tumor growth was evaluated simultaneously. Then, expression of Notch receptors(Notch-1, Notch-2,Notch-3, and Notch-4) and their Jagged ligands(Jagged-1 and Jagged-2) in dissected tumor tissuewere detected by real-time quantitative reverse transcription-polymerase chain reaction and Western blot. Finally, immunohistochemistry was performed to detect CD133, a marker of pancreatic cancer stem cells(CSCs), to evaluate the impact of QYHJ extract on pancreatic CSCs.RESULTS: QYHJ extract treatment effectively inhibited the tumor growth in nude mice. The expression of both Notch-4 and Jagged-1 were decreased significantly in QYHJ treatment groups(P < 0.05), while gemcitabine alone had no significant effect in down-regulating Jagged-1(P > 0.05).No significant difference was observed in the expression of Notch-1, Notch-2, Notch-3, and Jagged-2 between three treatment groups and control group(P > 0.05). Moreover, immunohistochemical analysis showed that the number of CD133 positive cells was significantly reduced by QYHJ treatment(P < 0.05), and the combined treatment was more effective than gemcitabine alone(P < 0.05).CONCLUSION: The role of the extract in pancreatic cancer treatment was associated with down-regulation of Notch-4 and Jagged-1 in Notch signaling pathway. 5Asymmetric ferromagnetic criticality in pyrochlore ferromagnet Lu2V2O7显示文摘We study the ferromagnetic criticality of the pyrochlore magnet Lu2 V2 O7 at the ferromagnetic transition TC% 70 K from the isotherms of magnetization MeHT via an iteration process and the Kouvel-Fisher method. The critical exponents associated with the transition are determined: b = 0.32(1), c = 1.41(1),and d ? 5:38. The validity of these critical exponents is further verified by scaling all the MeHT data in the vicinity of TConto two universal curves in the plot of M=jejbversus H=jejbtc, where e ? T=TCà 1.The obtained b and c values show asymmetric behaviors on the T < TCand the T > TCsides, and are consistent with the predicted values of 3 D Ising and cubic universality classes, respectively. This makes Lu2 V2 O7 a rare example in which the critical behaviors associated with a ferromagnetic transition belong to different universality classes. 10Investigating the prescription compatibility of Rhodiolarosea,Cordycepsmilitaris,and Rhubarbroot for the treatment of metabolic syndrome显示文摘OBJECTIVE To investigate the compatibility of the prescription Rhodiolarosea,Cordycepsmilitaris,and Rhubarb root.METHODS The DIO mice accompanying with insulin resistance,hypercholesterolemia and obesity were induced by high fat diet(HFD)in C57BL/6mice.For evaluating the insulin sensitivity in vivo,the values of serum insulin,HOMA-IR,and the glucose infuse rate(GIR)in hyperinsulinemiceuglycemic clamp test were determined;the time-dependent blood glucose changes were measured after insulin loading in insulin tolerance test(ITT),or after glucose loading in glucose tolerance test(GTT),respectively.Recombinant human protein tyrosine phosphatase 1B(PTP1B)was overexpressed by hGST-PTP1BBL21 E.coli and purified by GST affinity chromatography.The activities of PTP1 Band glutamine:fructose 6-phosphate amidotransferase(GFAT)were measured by the pNPP and GDH method,respectively.The activity ofα-glucosidase was determined with the substrates of p-nitrophenyl-alpha-D-glucopyranoside(pNPG).The IC50 value was calculated using the software GraphPad Prism 5.RESULTS 1st,the standard for the raw materials and for the extraction process of Rhodiolarosea,Cordycepsmilitaris,and Rhubarbroot are formulated according to the inhibition on PTP1 B,α-glucosidase,and GFAT in vitro,and the improvement on impaired glucose tolerance(IGT),hypercholesterolemia,and obesity in vivo,respectively.2nd,the composition was determined by the synergistic effects of Rhodiolarosea,Cordycepsmilitaris,and Rhubarb root on insulin resistance in DIO mice.3rd,in accordance with the IC50 values from the L9(34)and L8(27)orthogonal experiments targeting on PTP1 B and α-glucosidase,respectively,the best rate of Rhodiolarosea∶Cordycepsmilitaris∶Rhubarbroot was selected.4th,the effective dose for the treatment of metabolic syndrome was assessed based on the effects on IGT,hypercholesterolemia,obesity,insulin resistance,respectively,in DIO mice.CONCLUSION The prescription compatibility of Rhodiolarosea,Cordycepsmilitaris,and Rhubarbroot with the rate of 20∶1∶1was formed to treat the metabolic syndrome,such as IGT,hypercholesterolemia,and obesity,with the effective dose of 200 mg·kg-1 by the major mechanism of the improvement on insulin sensitivity.Monarch drug,Rhodiola,can clear away the lung-heat,tonify Qi,resolve stasis and nourish the heart.Adjuvant drug,Cordycepsmilitaris,can tonify the lung Qi and the kidney essence,strong waist and knee,compatibility with Rhodiola to enhance the function of invigorating lung and kidney.Assistant drug,Rhubarb,can clear heat,detoxify,remove blood stasis.These three herbal are compatible to show the effects of tonifying Qi,nourishing essence,cleaning heat,reducing phlegm and resolving masses for the treatment of metabolic syndrome.Jin-yingTIAN LingCHEN Xiao-linZHANG JingHAN Pei-chengZHANG FeiYE 2015中国药理学与毒理学杂志2015,29,S1:0

12Privacy Preserving Demand Side Management Method via Multi-Agent Reinforcement Learning显示文摘The smart grid utilizes the demand side management technology to motivate energy users towards cutting demand during peak power consumption periods, which greatly improves the operation efficiency of the power grid. However, as the number of energy users participating in the smart grid continues to increase, the demand side management strategy of individual agent is greatly affected by the dynamic strategies of other agents. In addition, the existing demand side management methods, which need to obtain users’ power consumption information,seriously threaten the users’ privacy. To address the dynamic issue in the multi-microgrid demand side management model, a novel multi-agent reinforcement learning method based on centralized training and decentralized execution paradigm is presented to mitigate the damage of training performance caused by the instability of training experience. In order to protect users’ privacy, we design a neural network with fixed parameters as the encryptor to transform the users’ energy consumption information from low-dimensional to high-dimensional and theoretically prove that the proposed encryptor-based privacy preserving method will not affect the convergence property of the reinforcement learning algorithm. We verify the effectiveness of the proposed demand side management scheme with the real-world energy consumption data of Xi’an, Shaanxi, China. Simulation results show that the proposed method can effectively improve users’ satisfaction while reducing the bill payment compared with traditional reinforcement learning(RL) methods(i.e., deep Q learning(DQN), deep deterministic policy gradient(DDPG),QMIX and multi-agent deep deterministic policy gradient(MADDPG)).
