1Numerical simulation of overburden and surface movements for Wongawilli strip pillar mining显示文摘The Wongawilli strip pillar mining technique,which combines the strip pillar mining layout and Wongawilli mining technology,is a new high efficient mining technology for mining under surface structures. The Wongawilli strip pillar mining technique was studied in this paper using theoretical analysis and numerical simulation. As an example,the geological and mining conditions of a coal mine were used to design the Wongawilli strip pillar plans,including the support parameters of the entries and the mining technology. In order to control the surrounding rocks and manage the roof effectively during coal mining,the stress fields,displacement fields and plastic zones were studied by numerical simulation. The stress fields,displacement fields,and plastic zones generated by Wongawilli strip pillar mining were obtained. And the surface movement and deformation were also determined after mining was completed and its effects on surface structures were analyzed and evaluated. The results demonstrate that it is feasible to mine under surface structures with the Wongawilli strip pillar mining technique. 2A novel procedure for pollen-based quantitative paleoclimate reconstructions and its application in China显示文摘Traditionally, the evaluation of pollen-based quantitative paleoclimate reconstructions focuses on the ability of calibration sets to infer present climatic conditions and/or the similarity between fossil and modern assemblages. Objective criteria for choosing the most appropriate climate parameter(s) to be reconstructed at a specific site are thus lacking. Using a novel approach for testing the statistical significance of a quantitative reconstruction using random environmental data, in combination with the advantageous large environmental gradients, abundant vegetation types and comprehensive modern pollen databases in China, we describe a new procedure for pollen-based quantitative paleoclimatic reconstructions. First, the most significant environmental variable controlling the fossil pollen assemblage changes is identified. Second, a calibration set to infer changes in this targeted variable is built up, by limiting the modern ranges of other environmental variables. Finally, the pollen-based quantitative reconstruction is obtained and its statistical significance assessed. This novel procedure was used to reconstruct the mean annual precipitation(Pann) from Gonghai Lake in the Lvliang Mountains, and Tianchi Lake in the Liupan Mountains, on the eastern and western fringe of the Chinese Loess Plateau, respectively. Both Pann reconstructions are statistically significant(p<0.001), and a sound and stable correlation relationship exists in their common period, showing a rapid precipitation decrease since 3300 cal yr BP. 3Targeting circular RNAs as a therapeutic approach: current strategies and challenges显示文摘Significant progress has been made in circular RNA(circRNA)research in recent years.Increasing evidence suggests that circRNAs play important roles in many cellular processes,and their dysregulation is implicated in the pathogenesis of various diseases.CircRNAs are highly stable and usually expressed in a tissue-or cell type-specific manner.Therefore,they are currently being explored as potential therapeutic targets.Gain-of-function and loss-of-function approaches are typically performed using circRNA expression plasmids and RNA interference-based strategies,respectively.These strategies have limitations that can be mitigated using nanoparticle and exosome delivery systems.Furthermore,recent developments show that the cre-lox system can be used to knockdown circRNAs in a cell-specific manner.While still in the early stages of development,the CRISPR/Cas13 system has shown promise in knocking down circRNAs with high specificity and efficiency.In this review,we describe circRNA properties and functions and highlight their significance in disease.We summarize strategies that can be used to overexpress or knockdown circRNAs as a therapeutic approach.Lastly,we discuss major challenges and propose future directions for the development of circRNA-based therapeutics.Alina T.He Jinglei Liu Feiya Li Burton B.Yang 2021Signal Transduction and Targeted Therapy2021,6,6:5
4Evaluation of the reliability of twin variant analysis in Mg alloys by in situ EBSD technique显示文摘Electron backscatter diffraction(EBSD)is an orientation mapping technique,which has been widely used to study texture evolution and twinning behavior.This work aims to study the variant of{10¯12}twins during tensile deformation of AZ31 Mg alloys.An in-situ tensile stage was used,and coupled with EBSD technique,the variants of twins can be analyzed without releasing the tensile stress.Based on the EBSD maps,the first and second smallest misorientation angles(M_(1)and M 2)between each active twin and respective potential variants were calculated,and the distribution of M_(1)values with tensile strain was obtained.The results indicated that{10¯12}twins were deviated from their ideal orientations with strain increased.The criteria of M_(1)<4°and M 2−M_(1)>2°were suggested to be applied to select the data points for reliable variant identification.Moreover,it confirmed that the deviation of twin relationship was ascribed to the profuse basal glide in matrices and/or twins.Feiya Liu Changfa Guo Renlong Xin Guilin Wu Qing Liu 2019Journal of Magnesium and Alloys2019,7,2:2
5Influence of traffic on the temperature field of tunnel in cold region:A case study on the world’s longest highway spiral tunnel显示文摘The impact of the piston wind caused by vehicle traffic on the temperature field distribution of the tunnel cannot be ignored,especially in spiral tunnels,where the temperature field distribution is more complicated than that in straight tunnels.Based on the world’s longest highway spiral tunnel,this study explores the influence of traffic on the temperature fields of spiral and straight tunnels,and proposes a new design method for laying length of insulation layer.The results show that traffic has a significant effect on the temperature field dis-tribution in the tunnel.The influence degree of different traffic elements decreases in the following order:average vehicle speed>daily traffic>morning rush hour>evening rush hour.The temperature field distribution of spiral tunnels is asymmetry on the left and right compared with straight tunnels,while the influence degree of traffic on the outside of spiral tunnels is similar to that of straight tunnels.Due to the difference of wind direction and traffic direction at different tunnel openings,an asymmetric insulation layer laying method is proposed.It is suggested that the lengths of the insulation layers of the Jinjiazhuang Tunnel should be 1200 m for the right tunnel inlet,700 m for the right tunnel outlet,700 m for the left tunnel inlet,and 1400 m for the left tunnel outlet.Yifan Jiang Jianyou Yu Ping Zhou Feicong Zhou Jiayong Lin Jinyi Li Ming Lin Feiya Lei Zhijie Wang 2023Underground Space2023,,1:2
6The establishment of polypeptide PSMA-targeted chimericantigen receptor-engineered natural killer cells forcastration-resistant prostate cancer and the induction of ferroptosis-related cell death显示文摘Background:The mortality of castration-resistant prostate cancer(CRPC)is high due to lack of an effective treatment.Chimeric antigen receptor(CAR)-based therapy is a promising immunotherapeutic strategy.Here,we aimed to design a novel CAR-natural killer(NK)cells with a clinically significant tumoricidal effect on CRPC.Methods:We constructed novel CAR-NK92MI cells with a CD244-based recombinant lentiviral vector.Different intracellular segments(CD244,NKG2D,or CD3ζ)were screened to identify the best candidate according to cell lysis assay and CD107a expression levels.To enhance the affinity of the CAR to the tumor antigen,we compared an antibody specific for prostate-specific membrane antigen(anti-PSMA)with PSMA-targeted polypeptide(p-PSMA),which was screened by phage display combinatorial library.Then,CAR-NK92MI cells with both a high affinity for PSMA and a strong tumoricidal capacity were generated.In addition,we verified their tumor-killing effect in vitro and in vivo.The release of cytokine by NK92MI cells was compared with that by CAR-NK92MI cells through flow cytometry and enzyme-linked immunosorbent assay.Moreover,ferroptosis-related cell death was explored as a possible underlying mechanism.Results:Three different CAR intracellular regions CAR1(CD244),CAR2(CD244,NKG2D)and CAR3(CD244,NKG2D,and CD3ζ)were constructed.CAR2 was chosen to confer a stronger tumoricidal ability on CAR-NK92MI cells.Compared with anti-PSMA,p-PSMA exhibited enhanced affinity for the tumor antigen.Thus,p-PSMA-CAR-NK92MI cells,which expressed CAR with a polypeptide-based antigen-binding region,an intracellular CD244 and a NKG2D costimulatory domain,were generated.They could selectively and successfully kill PSMA+target cells and exhibited specific lysis rate of 73.19%for PSMA-positive C4-2 cells and 33.04%for PSMA-negative PC3 cells.Additionally,p-PSMA-CAR-NK92MI cells had significantly higher concentrations of IFN-γ,TNF-αand granzyme B than NK92MI cells.In a CRPC cancer xenograft model,p-PSMA-CAR-NK92MI cells significantly inhibited tumor growth and exerted a more consistent killing effect than NK92MI cells.Moreover,ferroptosis is a potential mechanism through which CAR-NK92MI cells attack cancer cells,and is triggered by IFN-γ.Conclusions:p-PSMA-CAR-NK92MI cells can effectively kill CRPCPSMA+cells in vitro and in vivo.This strategy may provide additional treatment options for patients with CRPC.Liyuan Wu Fei Liu Le Yin Fangming Wang Hui Shi Qinxin Zhao Feiya Yang Dong Chen Xiying Dong Yuchun Gu Nianzeng Xing 2022Cancer Communications2022,42,8:2
7Proteomic Analysis of High Temperature Stress-Responsive Proteins in Chrysanthemum Leaves显示文摘Chrysanthemum is one of the most important ornamental flowers in the world,and temperature has a significant influence on its field production.In the present study,differentially expressed proteins were investigated in the leaves of Dendranthema grandiflorum‘Jinba’under high temperature stress using label-free quantitative proteomics techniques.The expressed proteins were comparatively identified and analyzed.A total of 1,463 heat-related,differentially expressed proteins were successfully identified by Liquid Chromatography-tandem Mass Spectrometry(LC-MS/MS),and 1,463 heat-related,differentially expressed proteins were successfully identified by mass spectrometry after a high temperature treatment.Among these,701 proteins were upregulated and 762 proteins were downregulated.The in-depth bioinformatics analysis of these differentially expressed proteins revealed that these were involved in energy metabolism pathways,protein metabolism,and heat shock.In the present study,the investigators determined the changes in the levels of some proteins,and their expression at the protein and molecular levels in chrysanthemum to help reveal the mechanism of heat resistance in chrysanthemum.Furthermore,the present study elucidated some of the proteins correlated to heat resistance in chrysanthemum,and their expression changes at the protein and molecular levels to help reveal the mechanism of heat resistance in this flower species.These results provide a theoretical basis for the selection of new heat resistant varieties of chrysanthemum in the field.Xin Li Feiya Liao Qiqi Ma Beibei Jiang Yuanzhi Pan Cheng Luo Xinjie Wang Aining Ran 2021Phyton-International Journal of Experimental Botany2021,90,5:1

14A hierarchical vascularized engineered bone inspired by intramembranous ossification for mandibular regeneration显示文摘Mandibular defects caused by injuries,tumors,and infections are common and can severely affect mandibular function and the patient's appearance.However,mandible reconstruction with a mandibular bionic structure remains challenging.Inspired by the process of intramembranous ossification in mandibular development,a hierarchical vascularized engineered bone consisting of angiogenesis and osteogenesis modules has been produced.Moreover,the hierarchical vascular network and bone structure generated by these hierarchical vascularized engineered bone modules match the particular anatomical structure of the mandible.The ultra-tough polyion complex has been used as the basic scaffold for hierarchical vascularized engineered bone for ensuring better reconstruction of mandible function.According to the results of in vivo experiments,the bone regenerated using hierarchical vascularized engineered bone is similar to the natural mandibular bone in terms of morphology and genomics.The sonic hedgehog signaling pathway is specifically activated in hierarchical vascularized engineered bone,indicating that the new bone in hierarchical vascularized engineered bone underwent a process of intramembranous ossification identical to that of mandible development.Thus,hierarchical vascularized engineered bone has a high potential for clinical application in mandibular defect reconstruction.Moreover,the concept based on developmental processes and bionic structures provides an effective strategy for tissue regeneration.Xin Ye Jianxiang He Shaolong Wang Qianglong Han Dongqi You Bin Feng Feiya Zhao Jun Yin Mengfei Yu Huiming Wang Huayong Yang 2022International Journal of Oral Science2022,14,3:1

16Simultaneous determination of ten nucleosides and bases in Ganoderma by micellar electrokinetic chromatography显示文摘Ganoderma(lingzhi)is a famous herbal medicine and edible supplement in oriental countries for a long history.In this study,a simple micellar electrokinetic chromatography(MEKC)method was established for the analysis of nucleosides and bases,the major bioactive components in Ganoderma for the first time.By optimizing the borate concentration,the sodium dodecyl sulfate(SDS)concentration and the pH value of running buffer,10 nucleosides and bases achieved an ideal separation.In real sample analysis,the developed method was successfully used to determine the 10 target analytes in 23 batches of Ganoderma samples from different regions.Results indicated that contents of 10 investigated analytes in each sample showed obvious variation.The principal components analysis(PCA)and hierarchical cluster analysis(HCA)analysis classified the samples into three groups,and the HCA tree visualized the relationships which was mainly contributed by geographical partition.The results indicated geographical origin to be an important factor that affect the accumulation of nucleosides and bases in Ganoderma.In summary,this study provides a simple and practical strategy for quality assessment and cultivation reference of Ganoderma.Feiya Sheng Songsong Wang Xiao Luo Jianbo Xiao Linfeng Hu Peng Li 2022Food Science and Human Wellness2022,11,2:1

18Effects of packaging materials and types on postharvest nutritional quality of mini Pakchoi Brassica chinensis显示文摘The baby leaves of mini Pakchoi Brassica chinensis are gaining popularity due to its pleasant appearance,flavour,tender texture and nutrition.Effects of retail packaging on the nutritional quality of Brassica chinensis at 4℃were studied.Samples were packaged using polyethylene bags(PE-B),polystyrene foam tray covered with polyethylene film(PE-C/PS-T),perforated oriented polypropylene bags(POPP-B)and biaxially oriented polystyrene box(BOPS-B).Weight losses,respiration rate,chlorophyll and carotenoid,vitamin C and nitrate were determined during storage period.After 10 d storage,the weight loss for samples packaged in PE-B was 0.89%,followed by PE-C/PS-T with weight losses of 6%.A significant reduction(>10%)in moisture was observed for the samples packaged in POPP-B and BOPS-B.The variation in respiration rate during storage seemed to form a trend with a sharp decline up to the fourth day.The difference in the reduction of chlorophyll and carotenoid was not significant among the four packages.BOPS-B had a better protective effect on vitamin C loss.The nitrate concentration during storage was within the official limit.Wu Shimin Shu Feiya Huang Danfeng 2016International Journal of Agricultural and Biological Engineering2016,9,6:0
