1Multigene editing via CRISPR/Cas9 guided by a single-sgRNA seed in Arabidopsis显示文摘We report that a solo single-guide RNA(sg RNA) seed is capable of guiding Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)/CRISPRàassociated 9(CRISRP/Cas_9) to simultaneously edit multiple genes At RPL_(10)A, At RPL_(10)B and At RPL_(10)C in Arabidopsis. Our results also demonstrate that it is possible to use CRISPR/Cas_9 technology to create At RPL_(10) triple mutants which otherwise cannot be generated by conventional genetic crossing. Compared to other conventional multiplex CRISPR/Cas systems, a single sg RNA seed has the advantage of reducing off-target gene-editing. 2Perilla oil and exercise decrease expressions of tumor necrosis factor-α,plasminogen activator inhibitor-1 and highly sensitive C-reactive protein in patients with hyperlipidemia显示文摘OBJECTIVE:To verify the effects of perilla oil on the regulation of blood lipid levels in patients with hyperlipidemia.METHODS:Blood was taken from patients prior to and 8 weeks following treatment with perilla oil.Different ways to test for indexes which correlate to hyperlipidemia were performed.Some indexes,which correlate with inflammation and injury to endothelial cells,were tested using enzyme linked immunosorbent assays.RESULTS:Serum lipid levels [triglyceride(TG),total cholesterol(TC),and low-density lipoprotein-cholesterol(LDL-C)] changed significantly after 56 days of treatment.Differences were noted as early as 28 days after treatment began(P<0.05).Treatment with perilla oil showed statistically significant recovery levels of high-density lipoprotein-cholesterol(HDL-C) after 28 and 56 days of treatment.Plasma lipids levels were significantly lower after 56 days of treatment(P<0.05).Perilla oil reduced blood lipid levels in patients,and the regulation of cell signaling factor levels had no adverse effects on patients' liver or kidney function,or blood routine examinations.CONCLUSION:Perilla oil treatment is safe in clinical use,can regulate blood lipid levels and protects the function of endothelial cells.Minggang Wei Peihua Xiong Ling Zhang Mei Fei Aiping Chen Fengling Li 2013Journal of Traditional Chinese Medicine2013,33,2:10
3Carbon Stock of Larch Plantations and Its Comparison with an Old-growth Forest in Northeast China显示文摘The overall goal of this study was to understand carbon(C) stock dynamics in four different-aged Japanese larch(Larix kaempferi) plantations in Northeast China that were established after clear-cutting old-growth Korean pine deciduous forests. Four Japanese larch plantations which were at 10, 15, 21, and 35 years old and an old-growth Korean pine deciduous forest which was 300 years old in Northeast China were selected and sampled. We compared the C pools of biomass(tree, shrub and herb), litterfall(LF), and soil organic carbon(SOC) among them. The biomass C stock of larch plantation at 10, 15, 21, and 35 years old was 26.8, 37.9, 63.6, and 83.2 Mg/ha, respectively, while the biomass C stock of the old-growth Korean pine deciduous forest was 175.1 Mg/ha. The SOC stock of these larch plantations was 172.1, 169.7, 140.3, and 136.2 Mg/ha respectively, and SOC stock of 170.4 Mg/ha in the control of old-growth forest. The biomass C stock increased with stand age of larch plantations, whereas SOC stock decreased with age, and C stock of LF did not change significantly(P > 0.05). The increase of biomass C offset the decline of SOC stock with age, making total carbon stock(TCS) of larch plantations stable from stand ages of 10–35 years. 5Anti-tumor Activity of Biodegradable Polymer-paclitaxel Conjugated Micelle Against Mice U14 Cervical Cancers显示文摘Two kinds of paclitaxel(PTX) conjugate micelles,of which one contained 25%(mass fraction) PTX [M(PTX)] and the other contained 22.5%(mass fraction) of PTX and 1.4%(mass fraction) of folate(FA)[FA-M(PTX)],were prepared for cell apoptosis and anti-tumor activity evaluation on U14 cervical cancer mouse models in comparison with 0.9%(mass fraction) saline(control) and equivalent Taxol.Seven days after tail intravenous injection of the drugs,the mice were sacrificed to measure the tumor masses.The average tumor masses were 4.26,2.89,2.63,and 2.17 g for the control,Taxol,M(PTX) and FA-M(PTX) groups,respectively.The inhibition rates of tumor growth calculated for the three drug groups were 32%,38% and 49%,respectively.Flow cytometry(FC) analysis and terminal deoxynucleotidyl transferase(TdT)-mediated deoxyuridine triphosphate(dUTP) nick end labeling(TUNEL) assay were conducted on the cancer tissues.The cell apoptosis rates based on the FC data and the TUNEL data were 20%,31%,37%,42%,and 10%,22%,26%,34%,respectively,both showing statistically significant differences(P<0.05) between three drug groups and the control group,and between the FA-M(PTX) group and the other two drug groups.In conclusion,the composite FA-M(PTX) micelles can be used for U14 cervical cancer treatment.CHEN Fengl DONG Danl FU Yan ZHENG Yong-hui LIU Shi CHANG Ming-xin JING Xia-bin 2012Chemical Research in Chinese Universities2012,28,4:4
6Qiguiyishen decoction reduced the accumulation of extracellular matrix in the kidneys of rats with adriamycin-induced nephropathy显示文摘OBJECTIVE: To investigate the effect of Qiguiyishen decoction(QGYS) on the severity of nephropathy.METHODS: Twenty-four rats were randomly divided into four groups(Ⅰ, Ⅱ, Ⅲ, Ⅳ) according to the random number table. Group Ⅰ as control group did not establish nephropathy model. Groups Ⅱ,Ⅲ, and Ⅳ were intravenously administered Adriamycin(7.5 mg/kg) through the tail vein to establish nephropathy model. QGYS was prepared with the extracts of Huangqi(Radix Astragali Mongolici),Danggui(Radix Angelicae Sinensis),Niuxi(RadixAchyran-this Bidentatae), and Chuanxiong(Rhizoma Chuanxiong).Group Ⅳ was administered QGYS (2mL·kg-1·d-1), group Ⅲ was administered benazepril(10 mL·kg-1·d-1), and group Ⅰ, Ⅱ was administered water(2mL·kg-1·d-1)once daily for eight weeks.RESULTS: QGYS reduced the excretion of urinary protein and N-acetyl-β-D-glucosaminidase and alleviated the accumulation of extracellular matrix(ECM) in renal tissue. 7Genome-wide identification of agronomically important genes in outcrossing crops using OutcrossSeq显示文摘Many important crops(e.g.,tuber,root,and tree crops)are cross-pollinating.For these crops,no inbred lines are available for genetic study and breeding because they are self-incompatible,clonally propagated,or have a long generation time,making the identification of agronomically important genes difficult,particularly in crops with a complex autopolyploid genome.In this study,we developed a method,OutcrossSeq,for mapping agronomically important loci in outcrossing crops based on whole-genome low-coverage resequencing of a large genetic population,and designed three computation algorithms in OutcrossSeq for different types of outcrossing populations.We applied OutcrossSeq to a tuberous root crop(sweet potato,autopolyploid),a tree crop(walnut tree,highly heterozygous diploid),and hybrid crops(double-cross populations)to generate high-density genotype maps for the outcrossing populations,which enable precise identification of genomic loci underlying important agronomic traits.Candidate causative genes at these loci were detected based on functional clues.Taken together,our results indicate that OutcrossSeq is a robust and powerful method for identifying agronomically important genes in heterozygous species,including polyploids,in a cost-efficient way.The OutcrossSeq software and its instruction manual are available for downloading at www.xhhuanglab.cn/tool/OutcrossSeq.html.Mengjiao Chen Weijuan Fan Feiyang Ji Hua Hua Jie Liu Mengxiao Yan Qingguo Ma Jiongjiong Fan Qin Wang Shufeng Zhang Guiling Liu Zhe Sun Changgeng Tian Fengling Zhao Jianli Zheng Qi Zhang Jiaxin Chen Jie Qiu Xin Wei Ziru Chen Peng Zhang Dong Pei Jun Yang Xuehui Huang 2021Molecular Plant2021,14,4:4
8Serum ficolin-2 concentrations are significantly changed in patients with hepatitis B virus infection and liver diseases显示文摘Human ficolin-2 is an important lectin complement pathway activator that is secreted from liver cells and has been implicated as an anti-infection innate immune molecule. However, the role of ficolin-2 protein and its dynamic changes over the course of and in the prognosis of chronic hepatitis B(CHB) and hepatocellular carcinoma(HCC) remain unclear. In this study, we analyzed ficolin-2 protein expression in a cohort of individuals with CHB infection, HCC and cirrhosis. A sandwich enzyme-linked immunosorbent assay(ELISA) method was used to measure serum ficolin-2 concentrations. Ficolin-2 expression in liver tissues was detected by immunohistochemical staining. Serum ficolin-2 concentrations in CHB patients were significantly higher than in healthy controls and HBV carriers. After 48 weeks of routine amelioration liver function treatment, serum ficolin-2 concentrations decreased and were positively correlated with favorable alanine aminotransferase(ALT), HBV DNA and HBe Ag-seroconversion outcomes. Interestingly, we observed much lower expression of serum and intrahepatic ficolin-2 in HCC and cirrhosis compared with healthy controls. 9Safety of SARS-CoV-2 vaccines:a systematic review and meta-analysis of randomized controlled trials显示文摘Background Various modalities of vaccines against coronavirus disease 2019(COVID-19),based on different platforms and immunization procedures,have been successively approved for marketing worldwide.A comprehensive review for clinical trials assessing the safety of COVID-19 vaccines is urgently needed to make an accurate judgment for mass vaccination.Main text A systematic review and meta-analysis was conducted to determine the safety of COVID-19 vaccine candidates in randomized controlled trials(RCTs).Data search was performed in PubMed,Embase,Cochrane library,Scopus,Web of Science,and MedRxiv.Included articles were limited to RCTs on COVID-19 vaccines.A total of 73,633 subjects from 14 articles were included to compare the risks of adverse events following immunization(AEFI)after vaccinating different COVID-19 vaccines.Pooled risk ratios(RR)of total AEFI for inactivated vaccine,viral-vectored vaccine,and mRNA vaccine were 1.34[95%confidence interval(CI)1.11–1.61,P<0.001],1.65(95%CI 1.31–2.07,P<0.001),and 2.01(95%CI 1.78–2.26,P<0.001),respectively.No significant differences on local and systemic AEFI were found between the first dose and second dose.In addition,people aged≤55 years were at significantly higher risk of AEFI than people aged≥56 years,with a pooled RR of 1.25(95%CI 1.15–1.35,P<0.001).Conclusions The safety and tolerance of current COVID-19 vaccine candidates are acceptable for mass vaccination,with inactivated COVID-19 vaccines candidates having the lowest reported AEFI.Long-term surveillance of vaccine safety is required,especially among elderly people with underlying medical conditions.Musha Chen Yue Yuan Yiguo Zhou Zhaomin Deng Jin Zhao Fengling Feng Huachun Zou Caijun Sun 2021Infectious Diseases of Poverty2021,10,4:2
10Thermally activated delayed fluorescence molecules and their new applications aside from OLEDs显示文摘Thermally activated delayed fluorescence(TADF) organic molecules feature with long-lived delayed fluorescence, because they can undergo not only efficient intersystem crossing(ISC), but also efficient reverse intersystem crossing(RISC) at room temperature. As a new type of luminescent molecules, they have exhibited successful applications in organic light emitting diodes(OLEDs). Aside from OLEDs, they are also found to have potential applications in time-resolved luminescence imaging based on long-lived fluorescence property. Meanwhile, due to their excited triplet characteristic originated from efficient ISC,they were found to be applied in triplet-triplet annihilation upconversion(TTA-UC), photodynamic therapy(PDT) and organic photocatalytic synthesis. 18Bioactive glass promotes the barrier functional behaviors of keratinocytes and improves the Re-epithelialization in wound healing in diabetic rats显示文摘Upon skin injury,re-epithelialization must be triggered promptly to restore the integrity and barrier function of the epidermis.However,this process is often delayed or interrupted in chronic wounds like diabetic foot ulcers.Considering that BG particles can activate multiple genes in various cells,herein,we hypothesized that bioactive glass(BG)might be able to modulate the barrier functional behaviors of keratinocytes.By measuring the transepithelial electrical resistance(TEER)and the paracellular tracer flux,we found the 58S-BG extracts substantially enhanced the barrier function of keratinocyte monolayers.The BG extracts might exert such effects by promoting the keratinocyte differentiation and the formation of tight junctions,as evidenced by the increased expression of critical differentiation markers(K10 and involucrin)and TJ protein claudin-1,as well as the altered subcellular location of four major TJ proteins(claudin-1,occludin,JAM-A,and ZO-1).Besides,the cell scratch assay showed that BG extracts induced the collective migration of keratinocytes,though they did not accelerate the migration rate compared to the control.The in vivo study using a diabetic rat wound model demonstrated that the BG extracts accelerated the process of re-epithelialization,stimulated keratinocyte differentiation,and promoted the formation of tight junctions in the newly regenerated epidermis.Our findings revealed the crucial effects of BGs on keratinocytes and highlighted its potential application for chronic wound healing by restoring the barrier function of the wounded skin effectively.Fengling Tang Junliang Li Weihan Xie Yunfei Mo Lu Ouyang Fujian Zhao Xiaoling Fu Xiaofeng Chen 2021Bioactive Materials2021,6,10:1
19Learning efficient text-to-image synthesis via interstage cross-sample similarity distillation显示文摘For a given text,previous text-to-image synthesis methods commonly utilize a multistage generation model to produce images with high resolution in a coarse-to-fine manner.However,these methods ignore the interaction among stages,and they do not constrain the consistent cross-sample relations of images generated in different stages.These deficiencies result in inefficient generation and discrimination.In this study,we propose an interstage cross-sample similarity distillation model based on a generative adversarial network(GAN)for learning efficient text-to-image synthesis.To strengthen the interaction among stages,we achieve interstage knowledge distillation from the refined stage to the coarse stages with novel interstage cross-sample similarity distillation blocks.To enhance the constraint on the cross-sample relations of the images generated at different stages,we conduct cross-sample similarity distillation among the stages.Extensive experiments on the Oxford-102 and Caltech-UCSD Birds-200-2011(CUB)datasets show that our model generates visually pleasing images and achieves quantitatively comparable performance with state-of-the-art methods.Fengling MAO Bingpeng MA Hong CHANG Shiguang SHAN Xilin CHEN 2021Science China(Information Sciences)2021,64,2:1
20The genome-wide landscape of small insertion and deletion mutations in Monopterus albus显示文摘Insertion and deletion(indel) mutations, which can trigger single nucleotide substitutions on the flanking regions of genes, may generate abundant materials for disease defense, reproduction, species survival and evolution. However, genetic and evolutionary mechanisms of indels remain elusive. We establish a comparative genome-transcriptome-alignment approach for a large-scale identification of indels in Monopterus population. Over 2000 indels in 1738 indel genes, including 1-21 bp deletions and 1-15 bp insertions, were detected. Each indel gene had ~1.1 deletions/insertions, and 2-4 alleles in population. Frequencies of deletions were prominently higher than those of insertions on both genome and population levels. Most of the indels led to in frame mutations with multiples of three and majorly occurred in non-domain regions, indicating functional constraint or tolerance of the indels. All indel genes showed higher expression levels than non-indel genes during sex reversal. Slide window analysis of global expression levels in gonads showed a significant positive correlation with indel density in the genome. Moreover, indel genes were evolutionarily conserved and evolved slowly compared to nonindel genes.
