1Genome-wide Analysis of Plant-specific Dof Transcription Factor Family in Tomato显示文摘The Dof (DNA binding with One Finger) family encoding single zinc finger proteins has been known as a family of plant-specific transcription factors. These transcription factors are involved in a variety of functions of importance for different biological processes in plants. In the current study, we identified 34 Dof family genes in tomato (Solanum lycopersicum L.), distributed on 11 chromosomes. A complete overview of SlDof genes in tomato is presented, including the gene structures, chromosome locations, phylogeny, protein motifs and evolution pattern. Phylogenetic analysis of 34 SlDof proteins resulted in four classes constituting six clusters. In addition, a comparative analysis between these genes in tomato, Arabidopsis (Arabidopsis thaliana L.) and rice (Oryza sativa L.) was also performed. The tomato Dof family expansion has been dated to recent duplication events, and segmental duplication is predominant for the SlDof genes. Furthermore, the SlDof genes displayed differential expression either in their transcript abundance or in their expression patterns under normal growth conditions. This is the first step towards genome-wide analyses of the Dof genes in tomato. 2Spatial distribution and focal mechanism solutions of the Wenchuan earthquake series:Results and implications显示文摘We relocate the spatial distribution of the devastating 12 May 2008 Wenchuan earthquake and its aftershocks.The relocation database is obtained from 89 stations deployed by the China Earthquake Administration,including 54 525 seismograms from 1 376 local earthquakes over MS3.5 between 12 May 2008 and 3 August 2008.The cross-correlation technique used in this paper has greatly improved the relocation precision by giving much more accurate P-wave differential travel-time measurements than those obtained from routinely picked phase onsets.At the same time,we pick P-wave polarity observations of the Wenchuan earthquake series (hereafter referred to as WES) from 1 023 stations in China and 59 IRIS (Incorporated Research Institutions of Seismology) stations.Then,employing a newly developed program CHNYTX,we obtain 83 well-determined focal mechanism solutions (hereafter referred to as FMSs).Based on spatial distribution and FMSs of the WES,we draw following conclusions:(1) The region near the main shock exhibits a buried low-angle northwest-dipping seismic zone with the main shock at its upper end and two conjugated seismic zones dipping southeast with roughly equal dip-angle;(2) The compressional directions of all kinds of FMSs of the WES are subhorizontal,which reflects the dominant stress in this area is compressional;(3) The principal compressional direction of the regional stress around Wenchuan is roughly perpendicular to the strike of Beichuan-Yingxiu fault,while around Qingchuan it is roughly parallel to the strike of Qingchuan fault.In intermediate part of the Longmenshan area,the principal compressional direction of the stress should be in-between;(4) The possibly existed molten materials in the lower crust of Songpan-Garze terrain have small contribution to the local stress state in Longmenshan area.The listric geometries of the Longmenshan faults most probably resulted from subhorizontal compression along NW-SE direction in history.Chen Cai Chunquan Yu Kai Tao Xingping Hu Yuan Tian Hao Zhang Xiaofeng Cui Jieyuan Ning 2011Earthquake Science2011,24,1:13

4Mechanisms underlying attenuation of apoptosis of cortical neurons in the hypoxic brain by flavonoids from the stems and leaves of Scutellaria baicalensis Georgi显示文摘Flavonoids from the stems and leaves of Scutellaria baicalensis Georgi, an antioxidant, markedly improve memory impairments and neuronal injuries. In the present study, primary cortical neurons of rats were exposed to potassium cyanide to establish a model of in vitro neural cell apoptosis. Inhibition of apoptosis by flavonoids from the stems and leaves of Scutellaria baicalensis Georgi at concentrations of 18.98, 37.36, and 75.92 μg/mL was detected using this model. These flavonoids dramatically increased cell survival, inhibited cell apoptosis and excessive production of malondialdehyde, and increased the activities of superoxide dismutase, glutathione peroxidase, and Na+-K+-ATPase in primary cortical neurons exposed to potassium cyanide. The flavonoids from the stems and leaves of Scutellaria baicalensis Georgi were originally found to have a polyhydric structure and to protect against cerebral hypoxia in in vitro and in vivo models, including hypoxia induced by potassium cyanide or cerebral ischemia. 5Development and validation of an ocean wave retrieval algorithm for VV-polarization Sentinel-1 SAR data显示文摘The purpose is to study the accuracy of ocean wave parameters retrieved from C-band VV-polarization Sentinel-1Synthetic Aperture Radar(SAR) images, including both significant wave height(SWH) and mean wave period(MWP), which are both calculated from a SAR-derived wave spectrum. The wind direction from in situ buoys is used and then the wind speed is retrieved by using a new C-band geophysical model function(GMF) model,denoted as C-SARMOD. Continuously, an algorithm parameterized first-guess spectra method(PFSM) is employed to retrieve the SWH and the MWP by using the SAR-derived wind speed. Forty–five VV-polarization Sentinel-1 SAR images are collected, which cover the in situ buoys around US coastal waters. A total of 52 subscenes are selected from those images. The retrieval results are compared with the measurements from in situ buoys. The comparison performs good for a wind retrieval, showing a 1.6 m/s standard deviation(STD) of the wind speed, while a 0.54 m STD of the SWH and a 2.14 s STD of the MWP are exhibited with an acceptable error.Additional 50 images taken in China's seas were also implemented by using the algorithm PFSM, showing a 0.67 m STD of the SWH and a 2.21 s STD of the MWP compared with European Centre for Medium-range Weather Forecasts(ECMWF) reanalysis grids wave data. 6Self-Q-switched and wavelength-tunable tungsten disulfide-based passively Q-switched Er:Y2O3 ceramic lasers显示文摘We report on diode-pumped Er:Y_2O_3 ceramic lasers at about 2.7 μm in the tunable continuous-wave, selfQ-switching and tungsten disulfide(WS2)-based passively Q-switching regimes. For stable self-Q-switched operation, the maximum output power reaches 106.6 m W under an absorbed power of 2.71 W. The shortest pulse width is measured to be about 1.39 μs at a repetition rate of 26.7 kHz at maximum output. Using a spin-coated WS2 as a saturable absorber, a passively Q-switched Er:Y_2O_3 ceramic laser is also realized with a maximum average output power of 233.5 m W(for the first time, to the best of our knowledge). The shortest pulse width decreases to 0.72 μs at a corresponding repetition rate of 29.4 kHz, which leads to a pulse energy of 7.92 μJ and a peak power of 11.0 W. 8Qiguiyishen decoction reduced the accumulation of extracellular matrix in the kidneys of rats with adriamycin-induced nephropathy显示文摘OBJECTIVE: To investigate the effect of Qiguiyishen decoction(QGYS) on the severity of nephropathy.METHODS: Twenty-four rats were randomly divided into four groups(Ⅰ, Ⅱ, Ⅲ, Ⅳ) according to the random number table. Group Ⅰ as control group did not establish nephropathy model. Groups Ⅱ,Ⅲ, and Ⅳ were intravenously administered Adriamycin(7.5 mg/kg) through the tail vein to establish nephropathy model. QGYS was prepared with the extracts of Huangqi(Radix Astragali Mongolici),Danggui(Radix Angelicae Sinensis),Niuxi(RadixAchyran-this Bidentatae), and Chuanxiong(Rhizoma Chuanxiong).Group Ⅳ was administered QGYS (2mL·kg-1·d-1), group Ⅲ was administered benazepril(10 mL·kg-1·d-1), and group Ⅰ, Ⅱ was administered water(2mL·kg-1·d-1)once daily for eight weeks.RESULTS: QGYS reduced the excretion of urinary protein and N-acetyl-β-D-glucosaminidase and alleviated the accumulation of extracellular matrix(ECM) in renal tissue. 9Conversion of hepatoma cells to hepatocyte-like cells by defined hepatocyte nuclear factors显示文摘Normal cells become cancer cells after a malignant transformation,but whether cancer cells can be reversed to normal status remains elusive.Here,we report that the combination of hepatocyte nuclear factor IA (HNFIA),HNF4A and forkhead box protein A3 (FOXA3)synergistically reprograms hepatocellular carcinoma (HCC)cells to hepatocyte-like cells (reprogrammed hepatocytes, rHeps).Our results show that rHeps lose the malignant phenotypes of cancer cells and retrieve hepatocyte-specific characteristics including hepatocyte-like morphology;global expression pattern of genes and specific biomarkers of hepatocytes;and the unique hepatic functions of albumin (ALB)secretion,glycogen synthesis,low-density lipoprotein (LDL)uptake,urea production, cytochrome P450 enzymes induction and drug metabolism.Intratumoral injection of these three factors efficiently shrank patientderived tumor xenografts and reprogrammed HCC cells in vivo.Most importantly,transplantation of rHeps in the liver of fumarylacetoacetate hydrolase-deficient (Fah^-/-)mice led to the reconstruction of hepatic lobules and the restoration of hepatic function.Mechanistically,exogenous expression of HNF1A,HNF4A and FOXA3 in HCC cells initiated the endogenous expression of numerous hepatocyte nuclear factors,which promoted the conversion of HCC cells to hepatocyte-like cells.Collectively,our results indicate the successful conversion of hepatoma cells to hepatocyte-like cells,not only extending our current knowledge of cell reprogramming but also providing a route towards a novel therapeutic strategy for cancer.Zhuo Cheng Zhiying He Yongchao Cai Cheng Zhang Gongbo Fu Hengyu Li Wen Sun Changcheng Liu Xiuliang Cui Beifang Ning Daimin Xiang Tengfei Zhou Xiaofeng Li Weifen Xie Hongyang Wang Jin Ding 2019Cell Research2019,29,2:3
10NLRP3 Deficiency Attenuates Secondary Degeneration of Visual Cortical Neurons Following Optic Nerve Injury显示文摘In the visual pathway, optic nerve(ON) injury may cause secondary degeneration of neurons in distal regions, such as the visual cortex. However, the role of the neuroinflammatory response in regulating secondary impairment in the visual cortex after ON injury remains unclear. The NOD-like receptor family pyrin domain containing 3(NLRP3) is an important regulator of neuroinflammation. In this study, we established a mouse model of unilateral ON crush(ONC) and showed that the expression of NLRP3 was significantly increased in the primary visual cortex(V1) as a response to ONC and that the NLRP3 inflammasome was activated in the contralateral V1 1 days–14 days after ONC. Ablation of the NLRP3 gene significantly decreased the trans-neuronal degeneration within 14 days. Visual electrophysiological function was improved in NLRP3-/- mice. 13Comparison of diagnostic accuracy between ^(18)F-FDG PET and PET/CT for pulmonary neoplasm显示文摘Aimed at comparing diagnostic accuracy of 18F-FDG PET with PET/CT for pulmonary neoplasm, a study based on multi-center clinical trial of the diagnoses, in randomized and semi-blind ways, was executed from January 2006 to June 2007. It included 55 patients, i.e. 16 with histopathologically proved lung tumors, 16 with tuberculosis and 23 with benign lesions (inflammation, pseudotumor, granuloma, fibrosis and others). The histopathologic and clinic results were served as reference standard. Statistical significances in pulmonary nodule diagnosis between 18F-FDG PET and PET/CT were determined with 95% confidence interval obtained by ROC analysis. The 18F-FDG PET detected lung neoplasm with a sensitivity of 87.5% (14/16), a specificity of 59.0% (23/39), an accuracy of 67.3% (37/55) and a positive-likelihood ratio of 2.13. The 18F-FDG PET/CT detected lung neoplasm with a sensitivity of 93.8% (15/16), a specificity of 61.5% (24/39), an accuracy of 70.9% (39/55) and a positive-likelihood ratio of 2.43. The area under curves (AUC) of 18F-FDG PET and PET/CT were 0.803±0.068 and 0.799±0.063, respectively. 14Bacteroides ovatus-mediated CD27^(−)MAIT cell activation is associated with obesity-related T2D progression显示文摘Type 2 diabetes(T2D)is highly associated with obesity.However,the factors that drive the transition from excessive weight gain to glucose metabolism disruption are still uncertain and seem to revolve around systemic immune disorder.Mucosal-associated invariant T(MAIT)cells,which are innate-like T cells that recognize bacterial metabolites,have been reported to be altered in obese people and to lead to metabolic dysfunction during obesity.By studying the immunophenotypes of blood MAIT cells from a cross-sectional cohort of obese participants with/without T2D,we found an elevation in CD27^(-)negative(CD27−)MAIT cells producing a high level of IL-17 under T2D obese conditions,which could be positively correlated with impaired glucose metabolism in obese people.We further explored microbial translocation caused by gut barrier dysfunction in obese people as a triggering factor of MAIT cell abnormalities.Specifically,accumulation of the bacterial strain Bacteroides ovatus in the peripheral blood drove IL-17^(-)producing CD27−MAIT cell expansion and could be associated with T2D risk in obese individuals.Overall,these results suggest that an aberrant gut microbiota–immune axis in obese people may drive or exacerbate T2D.Importantly,CD27−MAIT cell subsets and Bacteroides ovatus could represent targets for novel interventional strategies.Our findings extend current knowledge regarding the clinical relevance of body mass index(BMI)-associated variation in circulating MAIT cells to reveal the role of these cells in obesity-related T2D progression and the underlying cellular mechanisms.Yue Li Yi Yang Jin Wang Peihong Cai Mei Li Xixiang Tang Ying Tan Yuchan Wang Fan Zhang Xiaofeng Wen Qiaoxing Liang Yuanpeng Nie Tufeng Chen Xiang Peng Xuemin He Yanhua Zhu Guojun Shi Wai WCheung Lai Wei Yanming Chen Yan Lu 2022Cellular & Molecular Immunology2022,19,7:2

16Microplastic pollution and enrichment of distinct microbiota in sediment of mangrove in Zhujiang River estuary,China显示文摘The microbial communities colonized on microplastics(MPs)have attracted widespread attention.However,few studies focused on the MPs impacts on mangrove ecosystems,particularly on bacterial communities.We investigated the MPs pollution in mangrove of Zhujiang(Pearl)River estuary(ZRE).To study the potential risk posed by MPs to the mangrove ecosystems,the differences in bacterial communities,functions,and complexity between MPs and sediment samples were reported for the first time.Microplastics(2991±1586 items/kg dry weight(dw))in sediment were mainly fibers and polyethylene,mostly transparent,and in size less than 0.5 mm.Bacterial communities and functions significantly differed from MPs in mangrove sediment.Compared with sediment,MPs significantly enriched members of Proteobacteria,Bacteroidetes,and Actinobacteria,as well as the bacteria associated with plastic-degrading and human diseases on their surface,suggesting that microbial communities on MPs may promote MPs degradation and the spread of diseases,posing potential risk to mangrove ecosystems and human health.Although bacteria on MPs exhibited a lower diversity,the co-occurrence network analysis indicated that network of bacteria colonized on MPs was bigger and more complex than those of mangrove sediment,illustrating that MPs can act as a distinct habitat in this special ecosystem.This study provides a new perspective for increasing our understanding of microplastic pollution in mangrove ecosystems.Muting YAN Xiaofeng CHEN Wei CHU Weixin LI Minqian LI Zeming CAI Han GONG 2023Journal of Oceanology and Limnology2023,41,1:1

19Examination of Historical Data of the 1125 Lanzhou M 7.0 Earthquake and Analysis of Seismogenic Structure显示文摘Detailed examination of historical data of earthquakes and field investigations of loess landslide caused by the earthquake and tracing of active faults in Lanzhou area indicate that the Yijitanpu town, one of six towns of Jincheng city, was devastated by the 1125 Lanzhou earthquake. The citly is now located in the Vinylon Factory south of Hekou (River Mouth) in the Xigu district of Lanzhou city. We delermined that the six old towns mentioned in historical records lie in an area stretching from the south of Xigu district to Hekou in Lanzhou. This is consistent with the distribution of loess landslides caused by the earthquake, the extension of Holocene active faults, and the distribution of traces of the seismic rupture zone.
