| 2 | BLT1 in dendritic cells promotes Th1/Th17 differentiation and its deficiency ameliorates TNBS-induced colitis显示文摘Leukotriene B4(LTB4)synthesis is enhanced in the colonic mucosa in patients with inflammatory bowel disease(IBD).BLT1,a highaffinity receptor for LTB4,exhibits no effect on the progression of dextran sodium sulfate(DSS)-induced colitis,which mostly relies on innate immunity.Here,we reported that BLT1 regulates trinitrobenzene sulfonic acid(TNBS)-induced colitis,which reflects CD4+T-cell-dependent adaptive immune mechanisms of IBD.We found that BLT1 signaling enhanced the progression of colitis through controlling the production of proinflammatory cytokines by dendritic cells(DCs)and modulating the differentiation of Th1 and Th17.BLT1−/−mice displayed an alleviated severity of TNBS-induced colitis with reduced body weight loss and infiltrating cells in the lamina propria.BLT1 deficiency in DCs led to reduced production of proinflammatory cytokines,including IL-6,TNF-α,and IL-12,and these results were further confirmed via treatment with a BLT1 antagonist.The impaired cytokine production by BLT1−/−DCs subsequently led to reduced Th1 and Th17 differentiation both in vitro and in vivo.We further performed a conditional DC reconstitution experiment to assess whether BLT1 in DCs plays a major role in regulating the pathogenesis of TNBS-induced colitis,and the results indicate that BLT1 deficiency in DCs also significantly reduces disease severity.The mechanistic study demonstrated that BLT1-regulated proinflammatory cytokine production through the Gαiβγsubunit-phospholipase Cβ(PLCβ)-PKC pathway.Notably,we found that treatment with the BLT1 antagonist also reduced the production of proinflammatory cytokines by human peripheral blood DCs.Our findings reveal the critical role of BLT1 in regulating adaptive immunity and TNBS-induced colitis,which further supports BLT1 as a potential drug target for adaptive immunity-mediated IBD. | Jinfeng Zhou Weiming Lai Wanjie Yang Juping Pan Hu Shen Yingying Cai Cuixia Yang Ningjia Ma Yue Zhang Ru Zhang Xin Xie Zhongjun Dong Yuan Gao Changsheng Du | 2018 | Cellular & Molecular Immunology2018,15,12: | 9 |
| 3 | Growth of the Sayram Lake and retreat of its water-supplying glaciers in the Tianshan Mountains from 1972 to 2011显示文摘Inland lakes and alpine glaciers are important constituents of water resources in arid and semiarid regions. Understanding their variations is critical for both an accurate evaluation of the dynamic changes of water resources and the retrieval of climatic information. On the basis of earlier researches, this study investigated the growth of the Sayram Lake and the retreat of its water-supplying glaciers in the Tianshan Mountains using long-term sequenced remote sensing images. Our results show that over the past 40 years, the surface area and the water level of the lake has increased by 12.0±0.3 km2 and 2.8 m, respectively, and the area of its water-supplying glaciers has decreased continuously since the early 1970 s with a total reduction of about –2.13±0.03 km2. | 9 | Dynamic interaction of neutrophils and RFP-labelled Vibrio parahaemolyticus in zebrafish(Danio rerio)显示文摘Vibrio parahaemolyticus(Vp)is an aquatic zoonotic pathogen that causes vibriosis in marine animals as well as sepsis,gastroenteritis and wound infection in human.In vertebrates,the innate immune system plays a critical defense mechanism against Vp infection including transmigration of neutrophils.In this study,we have examined the genetic recombination and infectious process of Vp in the zebrafish(Danio rerio),a vertebrate model system extensively used for studying host-pathogen interactions.A pathogenic Vp strain,Vp57,tagged with red fluorescent protein(RFP)(Vp57^(RFP))was introduced into larval zebrafish at 3 days post fertilization(dpf),in which the innate immunity is present whereas the adaptive immunity has not yet developed.Vp57 and Vp57^(RFP)showed similar LD50 and induced similar symptoms and pathological changes in the hosts.We microinjected 579 colony-forming units(CFU)Vp57^(RFP)into the zebrafish caudal vein,and observed that neutrophils were recruited to the injection site and within 3 h post infection(hpi)Vp57^(RFP)were mainly distributed in the tail,eyes,heart and optic vesicle.After 3 hpi,the fish died with slight spine bending.Several Vp57^(RFP)were also detected in somites,the phagocytosis of neutrophils was activated through the progress of bacterial infections.We found that chemotaxis and phagocytosis of neutrophils occurs when zebrafish is infected by Vp57^(RFP),whereas Vp57^(RFP)can escape from neutrophils and colonize other remote regions using the blood circulation system. | Qiuyue Zhang An Ding Qianwen Yue Weiming Li Yao Zu Qinghua Zhang | 2017 | Aquaculture and Fisheries2017,2,6: | 1 |
