1The estimate of sea ice resources quantity in the Bohai Sea based on NOAA/AVHRR data显示文摘The research on sea ice resources is the academic base of sea ice exploitation in the Bohai Sea. According to the ice-water spectrum differences and the correlation between ice thickness and albedo, this paper comes up with a sea ice thickness inversion model based on the NOAA/AVHRR data. And then a sea ice resources quantity (SIQ) time series of Bohai Sea is established from 1987 to 2009. The results indicate that the average error of inversion sea ice thickness is below 30%. The maximum sea ice resources quantity is about 6 × 10 9 m 3 and the minimum is 1.3 × 10 9 m 3 . 3PARylation regulates stress granule dynamics, phase separation, and neurotoxicity of disease-related RNA-binding proteins显示文摘Mutations in RNA-binding proteins (RBPs) localized in ribonucleoprotein (RNP) granules, such as hnRNP A1 and TDP-43, promote aberrant protein aggregation, which is a pathological hallmark of various neurodegenerative diseases, such as amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Protein posttranslational modifications (PTMs) are known to 佗gulate RNP granules. In this study, we investigate the function of poly(ADP-ribosyl)ation (PARylation), an important PTM involved in DNA damage repair and cell death, in RNP granule-related neurodegeneration. We reveal that PARylation levels are a major regulator of the assembly-disassembly dynamics of RNP granules containing disease-related RBPs, hnRNP A1 and TDP-43. We find that hnRNP A1 can both be PARylated and bind to PARylated proteins or poly(ADP-ribose)(PAR). We further uncover that PARylation of hnRNP A1 at K298 controls its nucleocytoplasmic transport, whereas PAR-binding via the PAR-binding motif (PBM) of hnRNP Al regulates its association with stress granules. Moreover, we reveal that PAR not only dramatically enhances the liquid-liquid phase separation of hnRNP A1, but also promotes the co-phase separation of hnRNP A1 and TDP-43 in vitro and their interaction in vivo. Finally, both genetic and pharmacological inhibition of PARP mitigates hnRNP Al- and TDP-43-mediated neurotoxicity in cell and Drosophila models of ALS. 5Laser Alloyed Coatings of TiB_2/Graphite on 9Cr18 Stainless Steel Surface显示文摘Modified coatings including carbide of iron,nickel,chromium,silicon,and titanium are obtained on 9Cr18 stainless steel surface by laser alloying.The processing method,the microstructure,the interface,the tribological properties,and the forming mechanisms of the coatings are analyzed.The results show that the microstructure of the alloyed coatings is mainly irregular FeC crystals.Carbides of chromium and iron are around the FeC crystals.Small granular TiC disperses in the alloyed coatings.The microhardness of the alloyed coatings is greatly improved because of the occurrence of carbide with high hardness.At the same time,the wear resistance of the alloyed coatings are higher than that of 9Cr18 stainless steel.YING Li-xia WANG Li-qin JIA Xiao-mei GU Le 2007Journal of Iron and Steel Research(International)2007,14,3:5
6Score model for predicting acute-on-chronic liver failure risk in chronic hepatitis B显示文摘AIM: To establish a clinical scoring model to predict risk of acute-on-chronic liver failure(ACLF) in chronic hepatitis B(CHB) patients.METHODS: This was a retrospective study of 1457 patients hospitalized for CHB between October 2008 and October 2013 at the Beijing Ditan Hospital, Capital Medical University, China. The patients were divided into two groups: severe acute exacerbation(SAE) group(n = 382) and non-SAE group(n = 1075). The SAE group was classified as the high-risk group based on the higher incidence of ACLF in this group than in the non-SAE group(13.6% vs 0.4%). Two-thirds of SAE patients were randomly assigned to risk-model derivation and the other one-third to model validation. Univariate risk factors associated with the outcome were entered into a multivariate logistic regression model for screening independent risk factors. Each variable was assigned an integer value based on the regression coefficients, and the final score was the sum of these values in the derivation set. Model discrimination and calibration were assessed using area under the receiver operating characteristic curve and the Hosmer-Lemeshow test. RESULTS: The risk prediction scoring model includedthe following four factors: age ≥ 40 years, total bilirubin ≥ 171 μmol/L, prothrombin activity 40%-60%, and hepatitis B virus DNA > 107 copies/m L. The sum risk score ranged from 0 to 7; 0-3 identified patients with lower risk of ACLF, whereas 4-7 identified patients with higher risk. The Kaplan-Meier analysis showed the cumulative risk for ACLF and ACLF-related death in the two risk groups(0-3 and 4-7 scores) of the primary cohort over 56 d, and log-rank test revealed a significant difference(2.0% vs 33.8% and 0.8% vs 9.4%, respectively; both P < 0.0001). 7Fluorescent Organohydrogel with Thermal-Induced Color Change for Anti-counterfeiting显示文摘Smart fluorescent patterns enable dynamic color variation under external stimuli,showing a much higher security level in the field of anti-counterfeiting.However,there is still lacking of a simple and convenient way to achieve dynamic fluorescence changes.Herein,a fluorescent organohydrogel made up of a poly(N,/N-dimethylacrylamide-co-isopropylacrylamide)(p(DMA-NIPAM))hydrogel network and a polyflauryl methacrylate)(PLMA)organogel network was fabricated via a two-step interpenetrating technique.The former network bears naphthalimide moieties(DEAN,green fluorescent monomer)and the later contains 6-acrylamidopicolinic acid(6APA,fluorescent ligand),leading to emitting green fluorescence.When Eu^(3+) was introduced and coordinated with 6APA,the organohydrogel displays red fluorescence,which can further emit yellow after applying thermal stimulus.Furthermore,by adjusting the proportion of comonomers,various organohydrogels can be obtained,which can be programmed and act as an effective platform for the encryption and decryption of secret information.Xiaoxia Le Hui Shang Shirong Gu Guangqiang Yin Fuqing Shan Danyang Li Tao Chen 2022Chinese Journal of Chemistry2022,40,3:3
8Bispecific antibody simultaneously targeting PD1 and HER2 inhibits tumor growth via direct tumor cell killing in combination with PD1/PDL1 blockade and HER2 inhibition显示文摘Immune checkpoint blockade has shown significant clinical benefit in multiple cancer indications,but many patients are either refractory or become resistant to the treatment over time.HER2/neu oncogene overexpressed in invasive breast cancer patients associates with more aggressive diseases and poor prognosis.Anti-HER2 mAbs,such as trastuzumab,are currently the standard of care for HER2-overexpressing cancers,but the response rates are below 30%and patients generally suffer relapse within a year.In this study we developed a bispecific antibody(BsAb)simultaneously targeting both PD1 and HER2 in an attempt to combine HER2-targeted therapy with immune checkpoint blockade for treating HER2-positive solid tumors.The BsAb was constructed by fusing scFvs(anti-PD1)with the effector-functional Fc of an IgG(trastuzumab)via a flexible peptide linker.We showed that the BsAb bound to human HER2 and PD1 with high affinities(EC50 values were 0.2 and 0.14 nM,respectively),and exhibited potent antitumor activities in vitro and in vivo.Furthermore,we demonstrated that the BsAb exhibited both HER2 and PD1 blockade activities and was effective in killing HER2-positive tumor cells via antibody-dependent cellular cytotoxicity.In addition,the BsAb could crosslink HER2-positive tumor cells with T cells to form PD1 immunological synapses that directed tumor cell killing without the need of antigen presentation.Thus,the BsAb is a new promising approach for treating late-stage metastatic HER2-positive cancers.Chang-ling Gu Hai-xia Zhu Lan Deng Xiao-qing Meng Kai Li Wei Xu Le Zhao Yue-qin Liu Zhen-ping Zhu Hao-min Huang 2022Acta Pharmacologica Sinica2022,43,3:3
9Activation of Rac1-PI3K/Akt is required for epidermal growth factorinduced PAK1 activation and cell migration in MDA-MB-231 breast cancer cells显示文摘Epidermal growth factor (EGF) may increase cell motility,an event implicated in cancer cell invasion and metastasis.However,the underlying mechanisms for EGF-induced cell motility remain elusive.In this study,we found that EGF treatment could activate Ras-related C3 botulinum toxin substrate 1 (Rac1),PI3K/Akt and p21actived kinase (PAK1) along with cell migration.Ectopic expression of PAK1 K299R,a dominant negative PAK1 mutant,could largely abolish EGF-induced cell migration.Blocking PI3K/Akt signalling with LY294002 or Akt siRNA remarkably inhibited both EGF-induced PAK1 activation and cell migration.Furthermore,expression of dominant-negative Rac1 (T17N) could largely block EGF-induced PI3K/Akt-PAK1 activation and cell migration.Interestingly,EGF could induce a significant production of ROS,and N-acetyl-L-cysteine,a scavenger of ROS which abolished the EGF-induced ROS generation,cell migration,as well as activation of PI3K/Akt and PAK,but not Rac1.Our study demonstrated that EGF-induced cell migration involves a cascade of signalling events,including activation of Rac1,generation of ROS and subsequent activation of PI3K/Akt and PAK1.Yu Yang Jun Du Zhenzhen Hu Jiaojing Liu Yinhui Tian Yichao Zhu Le Wang Luo Gu 2011The Journal of Biomedical Research2011,25,4:3

11A possible family of Ni-based high temperature superconductors显示文摘We suggest that a family of Ni-based compounds, which contain [Ni_2M_2O]~2à(M = chalcogen) layers with an antiperovskite structure constructed by mixed-anion Ni complexes, Ni M_4O_2, can be potential high temperature superconductors(high-Tc) upon doping or applying pressure. The layer structures have been formed in many other transitional metal compounds such as La_2B_2Se_2O_3(B = Mn, Fe, Co). For the Ni-based compounds, we predict that the parental compounds host collinear antiferromagnetic states similar to those in iron-based high temperature superconductors. The electronic physics near Fermi energy is controlled by two egd-orbitals with completely independent in-plane kinematics. 12Quasi Dynamic Calculation of Local Stiffness of Angular Contact Ball Bearings显示文摘In order to describe the performance of thin wall bearing on rotor system more accurate,the simplified model of bearing local stiffness was proposed. The load distribution and local contact deformation in angular contact ball bearings were calculated using quasi dynamic calculation method. Based on the relationship of local load to contact deformation,the calculation model of local bearing stiffness was subsequently built to get radial and axial components of local stiffness. Effects of external loads on the local bearing stiffness were analyzed. The results showed that local stiffness in bearings is symmetric to the axis of radial load,and its value has a maximum on the symmetry axis along the radial load direction. 14The establishment of polypeptide PSMA-targeted chimericantigen receptor-engineered natural killer cells forcastration-resistant prostate cancer and the induction of ferroptosis-related cell death显示文摘Background:The mortality of castration-resistant prostate cancer(CRPC)is high due to lack of an effective treatment.Chimeric antigen receptor(CAR)-based therapy is a promising immunotherapeutic strategy.Here,we aimed to design a novel CAR-natural killer(NK)cells with a clinically significant tumoricidal effect on CRPC.Methods:We constructed novel CAR-NK92MI cells with a CD244-based recombinant lentiviral vector.Different intracellular segments(CD244,NKG2D,or CD3ζ)were screened to identify the best candidate according to cell lysis assay and CD107a expression levels.To enhance the affinity of the CAR to the tumor antigen,we compared an antibody specific for prostate-specific membrane antigen(anti-PSMA)with PSMA-targeted polypeptide(p-PSMA),which was screened by phage display combinatorial library.Then,CAR-NK92MI cells with both a high affinity for PSMA and a strong tumoricidal capacity were generated.In addition,we verified their tumor-killing effect in vitro and in vivo.The release of cytokine by NK92MI cells was compared with that by CAR-NK92MI cells through flow cytometry and enzyme-linked immunosorbent assay.Moreover,ferroptosis-related cell death was explored as a possible underlying mechanism.Results:Three different CAR intracellular regions CAR1(CD244),CAR2(CD244,NKG2D)and CAR3(CD244,NKG2D,and CD3ζ)were constructed.CAR2 was chosen to confer a stronger tumoricidal ability on CAR-NK92MI cells.Compared with anti-PSMA,p-PSMA exhibited enhanced affinity for the tumor antigen.Thus,p-PSMA-CAR-NK92MI cells,which expressed CAR with a polypeptide-based antigen-binding region,an intracellular CD244 and a NKG2D costimulatory domain,were generated.They could selectively and successfully kill PSMA+target cells and exhibited specific lysis rate of 73.19%for PSMA-positive C4-2 cells and 33.04%for PSMA-negative PC3 cells.Additionally,p-PSMA-CAR-NK92MI cells had significantly higher concentrations of IFN-γ,TNF-αand granzyme B than NK92MI cells.In a CRPC cancer xenograft model,p-PSMA-CAR-NK92MI cells significantly inhibited tumor growth and exerted a more consistent killing effect than NK92MI cells.Moreover,ferroptosis is a potential mechanism through which CAR-NK92MI cells attack cancer cells,and is triggered by IFN-γ.Conclusions:p-PSMA-CAR-NK92MI cells can effectively kill CRPCPSMA+cells in vitro and in vivo.This strategy may provide additional treatment options for patients with CRPC.Liyuan Wu Fei Liu Le Yin Fangming Wang Hui Shi Qinxin Zhao Feiya Yang Dong Chen Xiying Dong Yuchun Gu Nianzeng Xing 2022Cancer Communications2022,42,8:2
