| 1 | Serum levels of microRNAs can specifically predict liver injury of chronic hepatitis B显示文摘AIM:To investigate whether circulating microRNAs (miRNAs) can serve as molecular markers to predict liver injury resulted from chronic hepatitis B (CHB). METHODS:The profiles of serum miRNA expression were first generated with serum samples collected from 10 patients with CHB and 10 healthy donors (Ctrls) by microarray analysis. The levels of several miRNAs were further quantitated by real-time reverse transcriptionpolymerase chain reaction with serum samples from another 24 CHB patients and 24 Ctrls. Serum samples of 20 patients with nonalcohlic steatohepatitis (NASH) were also included for comparison. The comparison in the levels of miRNAs between groups (CHB, NASH and Ctrl) was analyzed with Mann-Whitney Utest. The correlation between miRNAs and clinical pathoparameters was analyzed using Spearman correlation analysis or canonical correlation analysis. The receiver-operator characteristic (ROC) curves were also generated to determine the specificity and sensitivity of each individual miRNA in distinguishing patients with CHB from Ctrls. RESULTS:miRNA profile analysis showed that 34 miRNAs were differentially expressed between CHB and Ctrl subjects, in which 12 were up-regulated and 22 down-regulated in CHB subject (fold change > 2.0 and P < 0.01). The median levels of miR-122, -572, -575 and -638 were significantly higher (P < 1.00 × 10 -5 ) while miR-744 significantly lower (P < 1.00 × 10 -6 ) in CHB compared with the Ctrl. The levels of miR-122, -572 and -638 were also higher (P < 1.00 × 10 -3 ) while the level of miR-744 lower in CHB (P < 0.05) than in NASH, although the difference between them was not as significant as that between CHB and Ctrl. ROC curve analysis revealed that the levels of miR-122, -572, -575, -638 and -744 in serum were sensitive and specific enough to distinguish CHB, NASH and Ctrl. Multivariate analysis further showed that the levels of these miRNAs were correlated with the liver function parameters. Most significantly, it was the scatter plot of principal component with the levels of these miRNAs, but not the parameters of liver function, which clearly distinguished CHB, NASH and Ctrl subjects. CONCLUSION:Serum levels of miR-122, -572, -575, -638 and -744 are deregulated in patients with CHB or NASH. | 4 | Development and utilization of a new chemically-induced soybean library with a high mutation density显示文摘Mutagenized populations have provided important materials for introducing variation and identifying gene function in plants. In this study, an ethyl methanesulfonate(EMS)-induced soybean(Glycine max) population,consisting of 21,600 independent M_2 lines, was developed.Over 1,000 M_(4(5))families, with diverse abnormal phenotypes for seed composition, seed shape, plant morphology and maturity that are stably expressed across different environments and generations were identified. Phenotypic analysis of the population led to the identification of a yellow pigmentation mutant, gyl, that displayed significantly decreased chlorophyll(Chl) content and abnormal chloroplast development. Sequence analysis showed that gyl is allelic to Minn Gold, where a different single nucleotide polymorphism variation in the Mg-chelatase subunit gene(ChlI1a) results in golden yellow leaves. A cleaved amplified polymorphic sequence marker was developed and may be applied to marker-assisted selection for the golden yellow phenotype in soybean breeding. | 13 | Rational application of gefitinib in NSCLC patients with sensitive EGFR mutations based on pharmacokinetics and metabolomics显示文摘Gefitinib has been available in the market for 20 years,but its pharmacokinetic mechanism of response is little known.In this study,we examined the pharmacokinetic and metabolomic profiles in non-small cell lung cancer(NSCLC)patients with sensitive EGFR mutations.A total of 216 advanced NSCLC patients were enrolled,and administered gefitinib at the standard dosage of 250 mg/day,which was established in heterogeneous subjects with non-sensitive mutations.We identified and quantified three main metabolites(named as M1,M2 and M3)in the plasma of patients,the correlations between the concentration of gefitinib/metabolites and efficacy were analyzed.In exploratory and validation set,gefitinib concentration was not correlated with clinical effects.Considering the result that the therapeutic effects of 250 mg/2-day was better than that of 250 mg/day in a multiple center clinical trial,the standard dose might be higher than that for maximal efficacy according to the hypothetical dose-response curve.Among the three metabolites,the IC50 of M2 in HCC827 and PC9 cell lines was significantly lower,and Conc.brain/Conc.plasma of M2 in mice was significantly higher than those of gefitinib,suggesting its higher potential to penetrate blood–brain barrier and might be more effective in the treatment of brain metastatic tumor than gefitinib.Consistently and attractively,higher M2 plasma concentration was found to be correlated with better clinical outcome in patients with brain metastases(the median PFS of CM2<12 ng/mL and CM2≥12 ng/mL were 17.0 and 27.1 months,respectively,P=0.038).The plasma concentration of M2≥12 ng/mL was a strong predictor of the PFS of NSCLC patients.In conclusion,for NSCLC patients with EGFR sensitive mutations,the standard dose is suspectable and could be decreased reasonably.M2 plays an important role in efficacy and may be more effective in the treatment of metastatic tumor than gefitinib. | Wei Feng Xi Chen Shao-xing Guan Hong-lian Ruan Yan Huang Hui-zhen Zhang Yun-peng Yang Wen-feng Fang Hong-yun Zhao Wei Zhuang Shuang Xin You-hao Chen Fei Wang Yue Gao Min Huang Xue-ding Wang Li Zhang | 2022 | Acta Pharmacologica Sinica2022,43,7: | 1 |

| 18 | Differentially expressed long noncoding RNAs and regulatory mechanism of LINC02407 in human gastric adenocarcinoma显示文摘BACKGROUND Long noncoding RNAs(lncRNAs)have been identified to play important roles in the development and progression of various tumors,including gastric cancer(GC).However,the molecular role of lncRNAs in GC progression remains unclear.AIM To investigate the differential expression of lncRNAs in human GC and elucidate the function and regulatory mechanism of LINC02407.METHODS The Cancer Genome Atlas database was used to investigate the involvement of lncRNAs in GC.Quantitative real-time polymerase chain reaction was used to estimate the relative expression level of LINC02407 in GC tissues and cells.Functional experiments including CCK8 assay,apoptosis assay,wound healing assay,and transwell assay were used to investigate the effect of LINC02407 on GC cells.Some microRNAs were predicted and verified via bioinformatics analysis and the luciferase reporter system.Predictive analysis and Western blot assay were used to analyze the expression of related proteins.RESULTS Many differentially expressed lncRNAs were identified in GC,and some of them including LINC02407 can affect the survival.LINC02407 was upregulated in tumor tissues compared with adjacent tissues.HGC-27 cells showed the highest LINC02407 expression and HaCaT cells exhibited the lowest expression.Different experiment groups were constructed using LINC02407 overexpressing plasmids and related siRNAs.The results of functional experiments showed that LINC02407 can promote the proliferation,migration,and invasion of GC cells but inhibit apoptosis.Luciferase reporter assay showed that hsa-miR-6845-5p and hsa-miR-4455 was downstream regulated by LINC02407.Western blot analysis showed that adhesion G protein-coupled receptor D1(ADGRD1)was regulated by the LINC02407-miR-6845-5p/miR-4455-ADGRD1 pathways.CONCLUSION LINC02407 plays a role in GC through the LINC02407-miR-6845-5p/miR-4455-ADGRD1 pathways,and thus,it may be an important oncogene and has potential value in GC diagnosis and treatment. | Li-Li Zhou Yan Jiao Hong-Mei Chen Li-Hua Kang Qi Yang Jing Li Meng Guan Ge Zhu Fei-Qi Liu Shuang Wang Xue Bai Yan-Qiu Song | 2019 | World Journal of Gastroenterology2019,25,39: | 1 |

| 20 | Development of a New Cold-Tolerant Maize(Zea mays L.)Germplasm Using the ICE1 Gene from Arabidopsis thaliana显示文摘To develop cold-tolerant maize germplasms and identify the activation of INDUCER OF CRT/DRE-BINDING FACTOR EXPRESSION(ICE1)expression in response to cold stress,RT-PCR was used to amplify the complete open reading frame sequence of the ICE1 gene and construct the plant expression vector pCAMBIA3301-ICE1-Bar.Immature maize embryos and calli were transformed with the recombinant vector using Agrobacterium tumefaciens-mediated transformations.From the regenerated plantlets,three T1 lines were screened and identi-fied by PCR.A Southern blot analysis showed that a single copy of the ICE1 gene was integrated into the maize(Zea mays L.)genomes of the three T1 generations.Under low temperature-stress conditions(4°C),the relative conductivity levels decreased by 27.51%–31.44%,the proline concentrations increased by 12.50%–17.50%,the malondialdehyde concentrations decreased by 16.78%–18.37%,and the peroxidase activities increased by 19.60%–22.89%in the T1 lines compared with those of the control.A real-time quantitative PCR analysis showed that the ICE1 gene was ectopically expressed in the roots,stems,and leaves of the T1 lines.ICE1 positively regulates the expression of the CBF genes in response to cold stress.Thus,this study showed the successful transformation of maize with the ICE1 gene,resulting in the generation of a new maize germplasm that had increased tolerance to cold stress. | Jing Qu Shuang Liu Peng Jiao Zhenzhong Jiang Jianbo Fei Shuyan Guan Yiyong Ma | 2022 | Phyton-International Journal of Experimental Botany2022,91,8: | 0 |
