| 5 | Expression of a New hemA Gene from Agrobacterium radiobacter in Escherichia coli for 5-Aminolevulinate Production显示文摘A new hemA gene encoding 5-aminolevulinate (ALA) synthase was cloned from Agrobacterium ra- diobacter zju-0121. The ALA synthase catalyzes the pyridoxal phosphate-dependent condensation of succinyl coen- zyme A (succinyl-CoA) and glycine to produce ALA. Four plasmids carrying the A, radiobacter hemA gene were transformed into different E. coli strains. The effects of both genetic and physiological factors on the expression of ALA synthase and ALA production were studied. The results indicated that the final intracellular activity of ALA synthase and the production of ALA in different expression systems varied largely. Among them, the recombinant E. coli BL21 (DE3) harboring the expression plasmid pET28-A. R-hemA was the most suitable one. The effects of isopropyl-β-D-thiogalactopyranoside (IPTG) addition time, IPTG concentration, culture temperature and the initial concentration of precursors and glucose on the ALA production were also evaluated. The expressed ALA synthase accounted for about 23.7% of the intracellular soluble protein. The highest specific activity of ALA syn- thase was 13.8nmol·min-1·mg-1 of intracellular soluble protein. | 6 | MiR-301a transcriptionally activated by HIF-2αpromotes hypoxiainduced epithelial-mesenchymal transition by targeting TP63 in pancreatic cancer显示文摘BACKGROUND Pancreatic cancer(PC)is one of the deadliest cancers worldwide.PC metastasis involves a complex set of events,including epithelial-mesenchymal transition(EMT),that increase tumor cell invasiveness.Recent evidence has shown that hypoxia is a major EMT regulator in pancreatic cancer cells and facilitates metastasis;however,the mechanisms remain elusive.AIM To investigate the role of miR-301a in hypoxia-induced EMT in PC cells.METHODS Real-time PCR and Western blot analysis were used to detect the expression of miR-301a and EMT markers in PDAC cells cultured in hypoxic and normoxic conditions.Western blot analysis was used to detect the expression of EMT markers in PDAC cells with miR-301a overexpression.Wound healing assay and Transwell assay were used to detect the migration capabilities of PDAC cells with miR-301a overexpression and knockout.Luciferase assay was used to detect the miR-301a promoter and the 3’untranslated region activity of TP63.Orthotopic PC mouse models were used to study the role of miR-301a in metastasis of PDAC cells in vivo.In situ hybridization assay was used to detect the expression of miR-301a in PDAC patient samples(adjacent paratumor and paired tumor tissues).RESULTS Hypoxic environment could directly promote the EMT of PC cells.The expression level of miR-301a was increased in a HIF2αdependent manner in hypoxia-cultured CFPAC-1 and BxPC-3 cells.Overexpression of miR-301a enhanced the hypoxia-induced EMT of PC cells,while knocking out miR-301a result in the suppression of hypoxia-induced EMT.TP63 was a direct target of miR-301a and involved in the metastatic process of PC cells.Furthermore,miR-301a upregulation facilitated PDAC distant metastasis and lymph node metastasis in vivo.Additionally,miR-301a overexpression was indicative of aggressive clinicopathological behaviors and poor prognosis.CONCLUSION The newly identified HIF-2α-miR301a-TP63 signaling pathway may play a crucial role in hypoxia-induced EMT in PDAC cells. | Kun-Dong Zhang Bin Hu Gang Cen Yu-Han Yang Wei-Wei Chen Zeng-Ya Guo Xiao-Feng Wang Qian Zhao Zheng-Jun Qiu | 2020 | World Journal of Gastroenterology2020,26,19: | 4 |
| 7 | Fluidized-bed pyrolysis of waste bamboo显示文摘Bamboo was a popular material substituting for wood,especially for one-off commodity in China. In order to recover energy and materials from waste bamboo,the basic characteristics of bamboo pyrolysis were studied by a thermogravimetric analyzer. It implied that the reaction began at 190~210 °C,and the percentage of solid product deceased from about 25% to 17% when temperature ranged from 400 °C to 700 °C. A lab-scale fluidized-bed furnace was setup to research the detailed properties of gaseous,liquid and solid products respectively. When temperature increased from 400 °C to 700 °C,the mass percent of solid product decreased from 27% to 17% approximately,while that of syngas rose up from 19% to 35%. When temperature was about 500°C,the percentage of tar reached the top,about 31%. The mass balance of these experiments was about 93%~95%. | 18 | Morphine Re-arranges Chromatin Spatial Architecture of Primate Cortical Neurons显示文摘The expression of linear DNA sequence is precisely regulated by the three-dimensional(3D)architecture of chromatin.Morphine-induced aberrant gene networks of neurons have been extensively investigated;however,how morphine impacts the 3D genomic architecture of neurons is still unknown.Here,we applied digestion-ligation-only high-throughput chromosome conformation capture(DLO Hi-C)technology to investigate the effects of morphine on the 3D chromatin architecture of primate cortical neurons.After receiving continuous morphine administration for 90 days on rhesus monkeys,we discovered that morphine re-arranged chromosome territories,with a total of 391 segmented compartments being switched.Morphine altered over half of the detected topologically associated domains(TADs),most of which exhibited a variety of shifts,followed by separating and fusing types.Analysis of the looping events at kilobase-scale resolution revealed that morphine increased not only the number but also the length of differential loops.Moreover,all identified differentially expressed genes from the RNA sequencing data were mapped to the specific TAD boundaries or differential loops,and were further validated for changed expression.Collectively,an altered 3D genomic architecture of cortical neurons may regulate the gene networks associated with morphine effects.Our finding provides critical hubs connecting chromosome spatial organization and gene networks associated with the morphine effects in humans. | Liang Wang Xiaojie Wang Chunqi Liu Wei Xu Weihong Kuang Qian Bu Hongchun Li Ying Zhao Linhong Jiang Yaxing Chen Feng Qin Shu Li Qinfan Wei Xiaocong Liu Bin Liu Yuanyuan Chen Yanping Dai Hongbo Wang Jingwei Tian Gang Cao Yinglan Zhao Xiaobo Cen | 2023 | Genomics, Proteomics & Bioinformatics2023,21,3: | 0 |
| 19 | Research on low emission MSW gasification and melting system显示文摘In order to eliminate secondary pollution caused by municipal solid waste(MSW)incineration,a MSW gasification and melting process is proposed.The process is expected to reduce the emission of pollutants,especially heavy-metals and dioxins.In this paper,the combustible components of MSW and simulated MSW were gasified in a lab-scale fluidized bed at 400°C-700°C when the excess air ratio(ER)was between 0.2 and 0.8.The experimental results indicated that the MSW could be gasified effectively in a fluidized bed at approximately 600°C-700°C when excess air ratio was 0.2-0.4.The melting characteristics of two typical fly ash samples from MSW incinerators were investigated.The results indicated that fly ash of pure MSW incineration could be melted at approximately 1,300°C and that of MSW and coal co-combustion could be melted at approximately 1,400°C.When temperature was over 1,100°C,more than 99.9%of the dioxins could be decomposed and most of the heavy-metals could be solidified in the slag.Based on the above experiments,two feasible MSW gasification and mel-ting processes were proposed for low calorific value MSW:(1)sieved MSW gasification and melting system,which was based on an idea of multi-recycle;(2)gasification and melting scheme of MSW adding coal as assistant fuel. | XIAO Gang NI Mingjiang CHI Yong JIN Yuqi ZHANG Jiaquan MIAO Qi CEN Kefa | 2007 | Frontiers of Environmental Science & Engineering2007,1,4: | 0 |
| 20 | HIV-1 Transmission among Injecting Drug Users is Principally Derived from Local Circulating Strains in Guangxi, China显示文摘Objective The mode of human immunodeficiency virus(HIV) transmission via injection drug use(IDU)still exists, and the recent shift in IDU-related transmission of HIV infection is largely unknown. The purpose of this study was to analyze the spatiotemporal sources and dynamics of HIV-1 transmission through IDU in Guangxi.Methods We performed a molecular epidemiological investigation of infections across Guangxi from2009 to 2019. Phylogenetic and Bayesian time-geographic analyses of HIV-1 sequences were performed to confirm the characteristics of transmission between IDUs in combination with epidemiological data.Results Among the 535 subjects, CRF08_BC(57.4%), CRF01_AE(28.4%), and CRF07_BC(10.7%) were the top 3 HIV strains;72.6% of infections were linked to other provinces in the transmission network;93.6% of sequence-transmitted strains were locally endemic, with the rest coming from other provinces,predominantly Guangdong and Yunnan;92.1% of the HIV transmission among people who inject drugs tended to be transmitted between HIV-positive IDUs.Conclusion HIV recombinants were high diversity, and circulating local strains were the transmission sources among IDUs in Guangxi.
