| 2 | Onset of Coronary Heart Disease is Associated with HCMV Infection and Increased CD14^+CD16^+Monocytes in a Population of Weifang,China显示文摘Objective To investigate the relationship between human cytomegalovirus(HCMV)infection and peripheral blood CD14+CD16+monocytes in the pathogenesis of coronary heart disease(CHD),and to elucidate the mechanism of pathogenesis in CHD by analyzing the correlation between infection,inflammation,and CHD,to provide a basis for the prevention,evaluation,and treatment of the disease.Methods In total,192 patients with CHD were divided into three groups:latent CHD,angina pectoris,and myocardial infarction.HCMV-IgM and-IgG antibodies were assessed using ELISA;CD14+CD16+monocytes were counted using a five-type automated hematology analyzer;mononuclear cells were assessed using fluorescence-activated cell sorting;and an automatic biochemical analyzer was used to measure the levels of triglyceride,cholesterol,high-and low-density lipoprotein cholesterols,lipoprotein,hs-CRp and Hcy.Results The positive rates of HCMV-IgM and-IgG were significantly higher in the CHD groups than in the control group.HCMV infection affects lipid metabolism to promote immune and inflammatory responses.Conclusion HCMV infection has a specific correlation with the occurrence and development of CHD.The expression of CD14+CD16+mononuclear cells in the CHD group was increased accordingly and correlated with acute HCMV infection.Thus,HCMV antibody as well as peripheral blood CD14+CD16+mononuclear cells can be used to monitor the occurrence and development of CHD. | LI Hong Zheng WANG Qin ZHANG Yi Yuan WANG Jin Dong WU Hong Juan ZHANG Mo Gen LI Ji Chen LIU Zhi Jun | 2020 | Biomedical and Environmental Sciences2020,33,8: | 8 |

| 5 | Qualitative analysis for a Wolbachia infection model with diffusion显示文摘We consider a reaction-diffusion model which describes the spatial Wolbachia spread dynamics for a mixed population of infected and uninfected mosquitoes. By using linearization method, comparison principle and Leray-Schauder degree theory, we investigate the influence of diffusion on the Wolbachia infection dynamics.After identifying the system parameter regions in which diffusion alters the local stability of constant steadystates, we find sufficient conditions under which the system possesses inhomogeneous steady-states. | 7 | MAP4K4 induces early blood-brain barrier damage in a murine subarachnoid hemorrhage model显示文摘Sterile-20-like mitogen-activated protein kinase kinase kinase kinase 4(MAP4 K4)is expressed in endothelial cells and activates inflammatory vascular damage.Endothelial cells are important components of the blood-brain barrier.To investigate whether MAP4 K4 plays a role in the pathophysiology of subarachnoid hemorrhage,we evaluated the time-course expression of MAP4 K4 after subarachnoid hemorrhage.A subarachnoid hemorrhage model was established using the intravascular perforation method.The model mice were assigned to four groups:MAP4 K4 recombinant protein,scramble small interfering RNA,and MAP4 K4 small interfering RNA were delivered by intracerebroventricular injection,while PF-06260933,a small-molecule inhibitor of MAP4 K4,was administrated orally.Neurological score assessments,brain water assessments,Evans blue extravasation,immunofluorescence,western blot assay,and gelatin zymography were performed to analyze neurological outcomes and mechanisms of vascular damage.MAP4 K4 expression was elevated in the cortex at 24 hours after subarachnoid hemorrhage,and colocalized with endothelial markers.MAP4 K4 recombinant protein aggravated neurological impairment,brain edema,and blood-brain barrier damage;upregulated the expression of phosphorylated nuclear factor kappa B(p-p65)and matrix metalloproteinase 9(MMP9);and degraded tight junction proteins(ZO-1 and claudin 5).Injection with MAP4 K4 small interfering RNA reversed these effects.Furthermore,administration of the MAP4 K4 inhibitor PF-06260933 reduced blood-brain barrier damage in mice,promoted the recovery of neurological function,and reduced p-p65 and MMP9 protein expression.Taken together,the results further illustrate that MAP4 K4 causes early blood-brain barrier damage after subarachnoid hemorrhage.The mechanism can be confirmed by inhibiting the MAP4 K4/NF-κB/MMP9 pathway.All experimental procedures and protocols were approved by the Experimental Animal Ethics Committee of General Hospital of Northern Theater Command(No.2018002)on January 15,2018. | Zheng Zou Yu-Shu Dong Dong-Dong Liu Gen Li Guang-Zhi Hao Xu Gao Peng-Yu Pan Guo-Biao Liang | 2021 | Neural Regeneration Research2021,16,2: | 5 |
