1A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. 2Long-term storage of lipid-like nanoparticles for mRNA delivery显示文摘Lipid-like nanoparticles(LLNs)have been extensively explored for messenger RNA(mRNA)delivery in various biomedical applications.However,the long-term storage of these nanoparticles is still a challenge for their clinical translation.In this study,we investigated a series of conditions for the long-term storage of LLNs with encapsulation of mRNA.We evaluated the stability of LLNs with different concentrations of cryoprotectants(sucrose,trehalose or mannitol)under the conditions of freezing or lyophilization processes.Through in vitro and in vivo mRNA delivery studies,we identified the optimal storage condition,and found that the addition with 5%(w/v)sucrose or trehalose to LLNs could remain their mRNA delivery efficiency for at least three months in the liquid nitrogen storage condition.Pengxuan Zhao Xucheng Hou Jingyue Yan Shi Du Yonger Xue Wenqing Li Guangya Xiang Yizhou Dong 2020Bioactive Materials2020,5,2:3
3A smart O_(2)-generating nanocarrier optimizes drug transportation comprehensively for chemotherapy improving显示文摘Drug transportation is impeded by various barriers in the hypoxic solid tumor,resulting in compromised anticancer efficacy.Herein,a solid lipid monostearin(MS)-coated CaO_(2)/MnO_(2) nanocarrier was designed to optimize doxorubicin(DOX)transportation comprehensively for chemotherapy enhancement.The MS shell of nanoparticles could be destroyed selectively by highly-expressed lipase within cancer cells,exposing water-sensitive cores to release DOX and produce O_(2).After the cancer cell death,the core-exposed nanoparticles could be further liberated and continue to react with water in the tumor extracellular matrix(ECM)and thoroughly release O_(2) and DOX,which exhibited cytotoxicity to neighboring cells.Small DOX molecules could readily diffuse through ECM,in which the collagen deposition was decreased by O_(2)-mediated hypoxia-inducible factor-1 inhibition,leading to synergistically improved drug penetration.Concurrently,DOX-efflux-associated P-glycoprotein was also inhibited by O_(2),prolonging drug retention in cancer cells.Overall,the DOX transporting processes from nanoparticles to deep tumor cells including drug release,penetration,and retention were optimized comprehensively,which significantly boosted antitumor benefits.Xiaojuan Zhang Chuanchuan He Yun Sun Xiaoguang Liu Yan Chen Chen Chen Ruicong Yan Ting Fan Tan Yang Yao Lu Jun Luo Xiang Ma Guangya Xiang 2021Acta Pharmaceutica Sinica B2021,11,11:1
