| 1 | Prognostic factors of refractory NSCLC patients receiving anlotinib hydrochloride as the third-or further-line treatment显示文摘Objective:Anlotinib hydrochloride is a multitarget tyrosine kinase inhibitor that targets vascular endothelial growth factor receptor,fibroblast growth factor receptor,platelet-derived growth factor receptor,c-Kit,and c-MET;therefore,it exhibits both antitumor and anti-angiogenetic activities.A phase III trial has shown that anlotinib improved progression-free survival(PFS)and overall survival(OS)in patients with advanced non-small cell lung cancer(NSCLC),who presented with progressive disease or intolerance after standard chemotherapy.This study aimed to analyze the characteristics of patients receiving anlotinib treatment to determine the dominant populations who are fit for the treatment.Methods:Data were collected from March 2015 to January 2017 from a randomized,double-blind,placebo-controlled,multicenter,phase III trial of anlotinib(ALTER0303).A total of 437 patients were enrolled and randomly allocated(2:1)to the anlotinib and placebo groups.Kaplan–Meier analysis and log-rank test were performed to compare PFS and OS.Cox proportional hazards model was adopted for multivariate prognostic analysis.Results:Multivariate analysis indicated that high post-therapeutic peripheral blood granulocyte/lymphocyte ratio and elevated alkaline phosphatase levels were independent risk factors for PFS.Meanwhile,elevated thyroid-stimulating hormone,blood glucose,and triglyceride levels;hypertension;and hand–foot syndrome were independent protective factors of PFS.High posttherapeutic peripheral blood granulocyte/lymphocyte ratio,an Eastern Cooperative Oncology Group(ECOG)score≥2,and the sum of the maximal target lesion length at baseline were independent risk factors of OS,and hypertriglyceridemia was an independent protective factor of OS.Conclusions:This study preliminarily explored the possible factors that affected PFS and OS after anlotinib treatment in patients with advanced refractory NSCLC,and the baseline characteristics of the therapeutically dominant populations were then identified. | Jing Wang Yizhuo Zhao Qiming Wang Li Zhang Jianhua Shi Zhehai Wang Ying Cheng Jianxing He Yuankai Shi Hao Yu Yang Zhao Weiqiang Chen Yi Luo Xiuwen Wang Kejun Nan Faguang Jin Jian Dong Baolan Li Zhujun Liu Baohui Han Kai Li | 2018 | Cancer Biology & Medicine2018,15,4: | 46 |
| 2 | Dental stem cell and dental tissue regeneration显示文摘The teeth are highly differentiated chewing organs formed by the development of tooth germ tissue located in the jaw and consist of the enamel, dentin, cementum, pulp, and periodontal tissue. Moreover, the teeth have a complicated regulatory mechanism, special histologic origin, diverse structure, and important function in mastication, articulation, and aesthetics. These characteristics, to a certain extent, greatly complicate the research in tooth regeneration. Recently, new ideas for tooth and tissue regeneration have begun to appear with rapid developments in the theories and technologies in tissue engineering. Numerous types of stem cells have been isolated from dental tissue, such as dental pulp stem cells (DPSCs), stem cells isolated from human pulp of exfoliated deciduous teeth (SHED), periodontal ligament stem cells (PDLSCs), stem cells from apical papilla (SCAPs), and dental follicle cells (DFCs). All these cells can regenerate the tissue of tooth. | 3 | BMAL1 knockout macaque monkeys display reduced sleep and psychiatric disorders显示文摘Circadian disruption is a risk factor for metabolic, psychiatric and age-related disorders, and non-human primate models could help to develop therapeutic treatments. Here, we report the generation of BMAL1 knockout cynomolgus monkeys for circadian-related disorders by CRISPR/Cas9 editing of monkey embryos. These monkeys showed higher nocturnal locomotion and reduced sleep, which was further exacerbated by a constant light regimen. Physiological circadian disruption was reflected by the markedly dampened and arrhythmic blood hormonal levels. Furthermore, BMAL1-deficient monkeys exhibited anxiety and depression, consistent with their stably elevated blood cortisol, and defective sensory processing in auditory oddball tests found in schizophrenia patients. Ablation of BMAL1 up-regulated transcriptional programs toward inflammatory and stress responses, with transcription networks associated with human sleep deprivation, major depressive disorders, and aging. | 4 | Effects of dietary arginine levels on growth performance,body composition, serum biochemical indices and resistance ability against ammonia-nitrogen stress in juvenile yellow catfish(Pelteobagrus fulvidraco)显示文摘This experiment was conducted to investigate the effects of dietary arginine levels on growth performance, body composition, serum biochemical indices and resistance ability against ammonia-nitrogen stress in juvenile yellow catfish(Pelteobagrus fulvidraco). Five isonitrogenous and isolipidic diets(42%protein and 9% lipid) were formulated to contain graded levels of arginine(2.44%, 2.64%, 2.81%, 3.01% and3.23% of diet), by supplementing L-Arginine HCI. Seven hundred juvenile yellow catfish with an initial average body weight of 1.13 ± 0.02 g were randomly divided into 5 groups with 4 replicates of 35 fish each and each group was fed one of the diets. After 56 d feeding, fish were exposed to 100 mg/L of ammonia-nitrogen for 72 h. The results showed that weight gain(WG) and specific growth rate(SGR) in2.64% and 2.81% groups were significantly higher than those in 3.23% group(P < 0.05). The feed conversation ratio(FCR) in 2,64%, 2,81% and 3.01% groups was significantly decreased when compared with3,23% group. The protein efficiency ratio(PER) in 2.64% group was significantly higher than that in 2.44%and 3.23% groups(P < 0.05). The condition factor(CF) of fish was significantly higher in 2.81% group than that in 2,44% group(P < 0.05). Dietary arginine levels had no significant effect on hepatosomatic index(HSI), viscerosomatic index(VSI), and whole-body dry matter, crude protein, crude lipid, ash contents, as well as serum total protein(TP), triglyceride(TG), glucose(GLU), urea nitrogen(UN) contents and aspartate aminotransferase(AST), alanine aminotransferase(ALT) activities(P> 0.05). After the fish were challenged to ammonia-nitrogen for 72 h, their cumulative mortality rate in 2.81% group was significantly lower than that in 2.44% group(P < 0.05). The results suggested that dietary arginine level at 2.81%could optimize anti-ammonia-nitrogen stress ability of juvenile yellow catfish and a level of 3.23%arginine seemed to depress the growth performance of fish and decreased their tolerance to the ammonia-nitrogen stress under current study. | 5 | Fluid Inclusion and H-O Isotope Geochemistry of the Phapon Gold Deposit, NW Laos: Implications for Fluid Source and Ore Genesis显示文摘The Phapon gold deposit, located in northern Laos, is a unique large-scale gold deposit in Luang Prabang-Loei metallogenic belt. It is hosted in the Lower Permian limestone and controlled by a NE-trending ductile-brittle fault system. There are three types of primary ore including auriferous calcite vein type, disseminated type, and breccia type, and the first two are important in the Phapon gold deposit. Based on fluid inclusion petrography and microthermometry, three types of primary fluid inclusions including type 1 liquid-rich aqueous, type 2 vapor-rich aqueous and type 3 daughter mineralbearing aqueous were identified in hydrothermal calcite grains. The ore-forming fluids are normally homogeneous, as indicated by the widespread type 1 inclusions with identical composition. The coexistence of type 1 and type 2 inclusions, showing similar final homogenization temperature but different compositions, indicate that fluid immiscibility did locally take place in both two types of ores. The results of microthermometry and H-O isotopes geochemistry indicate that there are little differences on ore-fluid geochemistry between the auriferous calcite vein-type and disseminated type ores. The oreforming fluids are characterized by medium-low temperatures(157–268 oC) and low salinity(1.6 wt.%–9.9 wt.% NaCl eq.). It is likely to have a metamorphic-dominant mixed source, which could be associated with dehydration and decarbonisation of Lower Permian limestone and Middle–Upper Triassic sandstones during the dynamic metamorphism. The fluid-wallrock interaction played a major role, and the locally occurred fluid-immiscible processes played a subordinate role in gold precipitation. | 6 | Commissioning progress of the FAST显示文摘The Five-hundred-meter Aperture Spherical radio Telescope(FAST) was completed with its main structure installed on September 25, 2016, after which it entered the commissioning phase. This paper aims to introduce the commissioning progress of the FAST over the past two years. To improve its operational reliability and ensure effective observation time, FAST has been equipped with a real-time information system for the active reflector system and hierarchical commissioning scheme for the feed support system, which ultimately achieves safe operation of the two systems. For meeting the high-performance indices, a highprecision measurement system was set up based on the effective control methods that were implemented for the active reflector system and feed support system. Since the commissioning of the FAST, a low-frequency ultra-wideband receiver and 19-beam1.05-1.45 GHz receiver have been mainly used. Telescope efficiency, pointing accuracy, and system noise temperature were completely tested and ultimately achieved the acceptance indices of the telescope. The FAST has been in the process of national acceptance preparations and has begun to search for pulsars. | 9 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. Here, we identified a recessive mutant gene, grain length and awn 1 (gla1), which caused a significant increase in grain length and weight, and was associated with long awns. The gla1 mutation was mapped to a single-nucleotide polymorphism in a gene encoding a cytoplasmically-localized mitogen-activated protein kinase (MAPK) phosphatase. Overexpression of GLA1 caused a decrease in grain length, and the GLA1 protein interacted with OsMAPK6. | 11 | Transformative hyaluronic acid-based active targeting supramolecular nanoplatform improves long circulation and enhances cellular uptake in cancer therapy显示文摘Hyaluronic acid(HA) is a natural ligand of tumor-targeted drug delivery systems(DDS) due to the relevant CD44 receptor overexpressed on tumor cell membranes. However, other HA receptors(HARE and LYVE-1) are also overexpressing in the reticuloendothelial system(RES). Therefore,polyethylene glycol(PEG) modification of HA-based DDS is necessary to reduce RES capture.Unfortunately, pegylation remarkably inhibits tumor cellular uptake and endosomal escapement,significantly compromising the in vivo antitumor efficacy. Herein, we developed a Dox-loaded HA-based transformable supramolecular nanoplatform(Dox/HCVBP) to overcome this dilemma. Dox/HCVBP contains a tumor extracellular acidity-sensitive detachable PEG shell achieved by a benzoic imine linkage.The in vitro and in vivo investigations further demonstrated that Dox/HCVBP could be in a 'stealth' state at blood stream for a long circulation time due to the buried HA ligands and the minimized nonspecific interaction by PEG shell. However, it could transform into a 'recognition' state under the tumor acidic microenvironment for efficient tumor cellular uptake due to the direct exposure of active targeting ligand HA following PEG shell detachment. | 15 | Anlotinib as third- or further-line therapy for short-term relapsed small-cell lung cancer: subgroup analysis of a randomized phase 2 study (ALTER1202)显示文摘Patients with small-cell lung cancer (SCLC) relapse within months after completing previous therapies. This study aimed to investigate the efficacy and safety of anlotinib as third- or further-line therapy in patients with short-term relapsed SCLC from ALTER1202. Patients with short-term relapsed SCLC (disease progression within 3 months after completing ≥ two lines of chemotherapy) in the anlotinib (n = 67) and placebo (n = 34) groups were analyzed. The primary endpoint was progression-free survival (PFS). The secondary endpoints included overall survival, objective response rate (ORR), disease control rate, and safety. Anlotinib significantly improved median PFS/OS (4.0 vs. 0.7 months, P < 0.0001)/(7.3 vs. 4.4 months, P = 0.006) compared with placebo. The ORR was 4.5%/2.9% in the anlotinib/placebo group (P = 1.000). The DCR in the anlotinib group was higher than that in the placebo group (73.1% vs. 11.8%, P < 0.001). The most common adverse events (AEs) were hypertension (38.8%), loss of appetite (28.4%), and fatigue (22.4%) in the anlotinib group and gamma-glutamyl transpeptidase elevation (20.6%) in the placebo group. No grade 5 AEs occurred. For patients with short-term relapsed SCLC, third- or further-line anlotinib treatment was associated with improved survival benefit. | 17 | Direct Comparative Analyses of 10X Genomics Chromium and Smart-seq2显示文摘Single-cell RNA sequencing(scRNA-seq)is generally used for profiling transcriptome of individual cells.The droplet-based 10X Genomics Chromium(10X)approach and the plate-based Smart-seq2 full-length method are two frequently used scRNA-seq platforms,yet there are only a few thorough and systematic comparisons of their advantages and limitations.Here,by directly comparing the scRNA-seq data generated by these two platforms from the same samples of CD45cells,we systematically evaluated their features using a wide spectrum of analyses.Smart-seq2 detected more genes in a cell,especially low abundance transcripts as well as alternatively spliced transcripts,but captured higher proportion of mitochondrial genes.The composite of Smart-seq2 data also resembled bulk RNA-seq data more.For 10X-based data,we observed higher noise for mRNAs with low expression levels.Approximately 10%30%of all detected transcripts by both platforms were from non-coding genes,with long non-coding RNAs(lncRNAs)accounting for a higher proportion in 10X.10X-based data displayed more severe dropout problem,especially for genes with lower expression levels.However,10X-data can detect rare cell types given its ability to cover a large number of cells.In addition,each platform detected distinct groups of differentially expressed genes between cell clusters,indicating the different characteristics of these technologies.Our study promotes better understanding of these two platforms and offers the basis for an informed choice of these widely used technologies. | Xiliang Wang Yao He Qiming Zhang Xianwen Ren Zemin Zhang | 2021 | Genomics, Proteomics & Bioinformatics2021,19,2: | 4 |
| 18 | The diversity of denitrifying bacteria in the alpine meadow soil of Sanjiangyuan natural reserve in Tibet Plateau显示文摘This is the first time to describe the diversity of denitrifying bacteria in Sanjiangyuan natural reserve in Tibet Plateau by investigating the molecular diversity and phylogenetic analysis of nirK and nosZ genes using PCR-RFLP and sequencing. Four soil samples were collected from alpine meadow communities from over an altitude of 4600 m which had different physicochemical properties by principal component analysis (PCA). For the genes fragment of nirK and nosZ, the diverse PCR products were characterized by cloning, restriction fragment length polymorphism (RFLP) analysis and sequenced. A total of 253 nirK clones and 283 nosZ clones were received in four samples, and 78 operational taxonomic units (OTUs) of nirK and 120 OTUs of nosZ by the restriction enzymes MspI and RsaI digested. The analysis of environmental factors showed that altitude and C/N ratio in soil may be the key factors to the denitrifying bacteria community. 36 nirK clones and 17 nosZ clones were sequenced, and their levels of nucleotide identity were from 69% to 98% and 57% to 97%, respectively. The phylogenetic tree was constructed using the Clustal W and Mega softwares, and all the sequenced clones could be subdivided into 4 groups. Some of clone sequences were related to the nirK and nosZ genes belonging to three phylogenetic subdivisions (α-, β- and γ subclasses of the Proteobacteria), while most of the clones were closely related to the genes of the uncultured bacteria. | 19 | Co-implantation of magnesium and zinc ions into titanium regulates the behaviors of human gingival fibroblasts显示文摘Soft tissue sealing around implants acts as a barrier between the alveolar bone and oral environment,protecting implants from the invasion of bacteria or external stimuli.In this work,magnesium(Mg)and zinc(Zn)are introduced into titanium by plasma immersed ion implantation technology,and their effects on the behaviors of human gingival fibroblasts(HGFs)as well as the underlying mechanisms are investigated.Surface characterization confirms Mg and Zn exist on the surface in metallic and oxidized states.Contact angle test suggests that surface wettability of titanium changes after ion implantation and thus influences protein adsorption of surfaces.In vitro studies disclose that HGFs on Mg ion-implanted samples exhibit better adhesion and migration while cells on Zn ion-implanted samples have higher proliferation rate and amounts.The results of immunofluorescence staining and real-time reverse-transcriptase polymerase chain reaction(RT-PCR)suggest that Mg mainly regulates the motility and adhesion of HGFs through activating the MAPK signal pathway whereas Zn influences HGFs proliferation by triggering the TGF-βsignal pathway.The synergistic effect of Mg and Zn ions ensure that HGFs cultured on co-implanted samples possessed both high proliferation rate and motility,which are critical to soft tissue sealing of implants. | Lanyu Wang Qiming Luo Xianming Zhang Jiajun Qiu Shi Qian Xuanyong Liu | 2021 | Bioactive Materials2021,6,1: | 4 |
| 20 | Underwater Sonar Image Detection:A Novel Quantum-Inspired Shuffled Frog Leaping Algorithm显示文摘Object detection plays an important role in the underwater object recognition technology of sonar equipment. We propose a Novel quantum-inspired shuffled frog leaping algorithm(NQSFLA) to obtain more accurate detection results in this paper. The proposed NQSFLA adopts a fitness function combining intra-class difference with inter-class difference to evaluate the frog position more accurately and a new quantum evolution update strategy to improve the searching ability in the searching process. In order to avoid the disadvantages of Quantuminspired shuffled frog leaping algorithm(QSFLA), a fuzzy membership matrix with spatial information model is developed, which can remove isolated regions and further improve the detection accuracy. Segmentation, distribution and noise entropy(SDNE) model is also proposed to quantitatively evaluate the detection results.
