1TALEN or Cas9-Rapid,Efficient and Specific Choices for Genome Modifications显示文摘Precise modifications of complex genomes at the single nucleotide level have been one of the big goals for scientists working in basic and applied genetics,including biotechnology,drug development,gene therapy and synthetic biology.However,the relevant techniques for making these manipulations in model organisms and human cells have been lagging behind the rapid high throughput studies in the post-genomic era with a bottleneck of low efficiency,time consuming and laborious manipulation,and off-targeting problems.Recent discoveries of TALEs(transcription activator-like effectors) coding system and CRISPR(clusters of regularly interspaced short palindromic repeats) immune system in bacteria have enabled the development of customized TALENs(transcription activator-like effector nucleases) and CRISPR/Cas9 to rapidly edit genomic DNA in a variety of cell types,including human cells,and different model organisms at a very high efficiency and specificity.In this review,we first briefly summarize the development and applications of TALENs and CRISPR/Cas9-mediated genome editing technologies;compare the advantages and constraints of each method;particularly,discuss the expected applications of both techniques in the field of site-specific genome modification and stem cell based gene therapy;finally, propose the future directions and perspectives for readers to make the choices.Chuanxian Wei Jiyong Liu Zhongsheng Yu Bo Zhang Guanjun Gao Renjie Jiao 2013Journal of Genetics and Genomics2013,40,6:52

4The origin of Wx^(la) provides new insights into the improvement of grain quality in rice显示文摘Appearance and taste are important factors in rice(Oryza sativa) grain quality. Here, we investigated the taste scores and related eating-quality traits of533 diverse cultivars to assess the relationships between—and genetic basis of—rice taste and eating-quality. A genome-wide association study highlighted the Wx gene as the major factor underlying variation in taste and eating quality. Notably, a novel waxy(Wx) allele, Wx^(la), which combined two mutations from Wx^(b) and Wx^(in), exhibited a unique phenotype. Reduced GBSSI activity conferred Wx^(la) rice with both a transparent appearance and good eating quality. Haplotype analysis revealed that Wx^(la) was derived from intragenic recombination. In fact,the recombination rate at the Wx locus was estimated to be 3.34 kb/c M, which was about 75-fold higher than the genome-wide mean, indicating that intragenic recombination is a major force driving diversity at the Wx locus. Based on our results, we propose a new network for Wx evolution, noting that new Wx alleles could easily be generated by crossing genotypes with different Wx alleles. 5Kinematics and trajectory planning of a cucumber harvesting robot manipulator显示文摘In order to reduce cucumber harvesting cost and improve economic benefits,a cucumber harvesting robot was developed.The cucumber harvesting robot consists of a vehicle,a 4-DOF articulated manipulator,an end-effector,an upper monitor,a vision system and four DC servo drive systems.The Kinematics of the cucumber harvesting robot manipulator was constructed using D-H coordinate frame model.And the inverse kinematics which provides a foundation for trajectory planning has been solved with inverse transform technique.The cycloidal motion,which has properties of continuity and zero velocity and acceleration at the ports of the bounded interval,was adopted as a feasible approach to plan trajectory in joint space of the cucumber harvesting robot manipulator.Moreover,hardware and software based on CAN-bus communication between the upper monitor and the joint controllers have been designed.Experimental results show that the upper monitor communicates with the four joint controllers efficiently by CAN-bus,and the integrated errors of four joint angles do not exceed four degrees.Probable factors resulting in the errors were analyzed and the corresponding solutions for improving precision are proposed.Zhang Libin Wang Yan Yang Qinghua Bao Guanjun Gao Feng Xun Yi 2009International Journal of Agricultural and Biological Engineering2009,2,1:7
6Overview of research on agricultural robots in China显示文摘As the representation of new concept agricultural machinery,agricultural robots possess great advantages of improving agricultural productivity,enhancing production environment and solving the problem of labor shortage.Therefore,the strategy for application of agricultural robots and precision agriculture to improve the intelligence and information level of agriculture is the inevitable trend for China’s agriculture in the twentieth century.Based on the developmental status of agricultural robots in China,the agricultural robots are categorized and the performances,structures and characteristics of various agricultural robots such as vegetable grafting robots,transplanting robots,spraying robots,mowing robots,harvesting robots,grading and detecting robots are amply introduced.It can be seen that vegetable grafting robots,spraying robots,harvesting robots,grading and detecting robots have already been put into production while others are still at experimental stage.At present,there are several problems such as low popularization,great limitations,high cost and low intelligence,which greatly restrict the development of agricultural robots in China.Thus,open agricultural robot system with good expansibility,generality and flexibility should be developed and adopted to decrease its cost and shorten developing cycle.The mechanical structure of robots should also be designed as simply as possible.Finally,multi-robot system would become another important development direction of agricultural robots in the future.Zhang Libin Yang Qinghua Bao Guanjun Wang Yan Qi Liyong Gao Feng Xu Fang 2008International Journal of Agricultural and Biological Engineering2008,1,1:4
7Genetic architecture and key genes controlling the diversity of oil composition in rice grains显示文摘Rice grain oil is a valuable nutrient source.However,the genetic basis of oil biosynthesis in rice grains remains unclear.In this study,we performed a genome-wide association study on oil composition and oil concentration in a diverse panel of 533 cultivated rice accessions.High variation for 11 oil-related traits was observed,and the oil composition of rice grains showed differentiation among the subpopulations.We identified 46 loci that are significantly associated with grain oil concentration or composition,16 of which were detected in three recombinant inbred line populations.Twenty-six candidate genes encoding enzymes involved in oil metabolism were identified from these 46 loci,four of which(PAL6,LIN6,MYR2,and ARA6)were found to contribute to natural variation in oil composition and to show differentiation among the subpopulations.Interestingly,population genetic analyses revealed that specific haplotypes of PAL6 and LIN6 have been selected in japonica rice.Based on these results,we propose a possible oil biosynthetic pathway in rice grains.Collectively,our results provide new insights into the genetic basis of oil biosynthesis in rice grains and can facilitate marker-based breeding of rice varieties with enhanced oil and grain quality.Hao Zhou Duo Xia Pingbo Li Yiting Ao Xiaodong Xu Shanshan Wan Yanhua Li Bian Wu Huan Shi Kaiyue Wang Guanjun Gao Qinglu Zhang Gongwei Wang Jinghua Xiao Xianghua Li Sibin Yu Xingming Lian Yuqing He 2021Molecular Plant2021,14,3:4
8Effect and mechanism of YB-1 knockdown on glioma cell growth,migration,and apoptosis显示文摘Ybox binding protein 1(YB-1)is manifested as its involvement in cell proliferation and differ-entiation and malignant cell transformation.Overexpression of YB-1 is associated with glioma progression and patient survival.The aim of this study is to investigate the influence of YB-1 knockdown on glioma cell progression and reveal the mechanisms of YB-1 knockdown on glioma cell growth,migration,and apoptosis.It was found that the knockdown of YB-1 decreased the mRNA and protein levels of YB-1 in U251 glioma cells.The knockdown of YB-1 significantly inhibited cell proliferation,colony formation,and migration in vitro and tumor growth in vivo.Proteome and phosphoproteome data revealed that YB-1 is involved in glioma progression through regulating the expression and phosphorylation of major proteins involved in cell cycle,adhesion,and apoptosis.The main regulated proteins included CCNB1,CCNDBP1,CDK2,CDK3,ADGRG1,CDH-2,MMP14,AIFM1,HO-1,and BAX.Furthermore,it was also found that YB-1 knockdown is associated with the hypo-phosphorylation of ErbB,mTOR,HIF-1,cGMP-PKG,and insulin signaling pathways,and proteoglycans in cancer.Our findings indicated that YB-1 plays a key role in glioma progression in multiple ways.including regulating the expression and phosphorylation of major proteins associated with cell cycle,adhesion,and apoptosis.Huilin Gong Shan Gao Chenghuan Yu Meihe Li Ping Liu Guanjun Zhang Jinning Song Jin Zheng 2020Acta Biochimica et Biophysica Sinica2020,52,2:2
9Engineering Deinococcus radiodurans into biosensor to monitor radioactivity and genotoxicity in environment显示文摘Based on a genetically modified radioresistant bacteria Deinococcus radiodurans, we constructed a real time whole cell biosensor to monitor radioactivity and genotoxicity in highly radioactive environ-ment. The enhanced green fluorescence protein (eGFP) was fused to the promoter of the crucial DNA damage-inducible recA gene from D. radiodurans, and the consequent DNA fragment (PrecA-egfp) car-ried by plasmid was introduced into D. radiodurans R1 strain to obtain the biosensor strain DRG300. This engineered strain can express eGFP protein and generate fluorescence in induction of the recA gene promoter. Based on the correlation between fluorescence intensity and protein expression level in live D. radiodurans cells, we discovered that the fluorescence induction of strain DRG300 responds in a remarkable dose-dependent manner when treated with DNA damage sources such as gamma radiation and mitomycin C. It is encouraging to find the widely detective range and high sensitivity of this re-constructed strain comparing with other whole cell biosensors in former reports. 13Variation of airborne quartz in air of Beijing during the Asia-Pacific Economic Cooperation Economic Leaders' Meeting显示文摘Quartz particles are a toxic component of airborne paniculate matter(PM).Quartz concentrations were analyzed by X-ray diffraction in eighty-seven airborne PM samples collected from three locations in Beijing before,during,and after the Asia-Pacific Economic Cooperation(APEC) Leaders' Meeting in 2014.The results showed that the mean concentrations of quartz in PM samples from the two urban sites were considerably higher than those from the rural site.The quartz concentrations in samples collected after the APEC meeting,when the pollution restriction lever was lifted,were higher than those in the samples collected before or during the APEC meeting.The quartz concentrations ranged from 0.97 to 13.2 μg/m^3,which were among the highest values amid those reported from other countries.The highest quartz concentration exceeded the Californian Office of Environmental Health Hazard Assessment reference exposure level and was close to the occupational threshold limit values for occupational settings.Moreover,a correlation analysis showed that quartz concentrations were positively correlated with concentrations of pollution parameters PM_(10),PM_(2.5),SO_2 and NO_x,but were negatively correlated with O_3 concentration.The results suggest that the airborne quartz particles may potentially pose health risks to the general population of Beijing.Gang Li Yingming Li Hongxing Zhang Honghua Li Guanjun Gao Qian Zhou Yuan Gao Wenjuan Li Huizhong Sun Xiaoke Wang Qinghua Zhang 2016Journal of Environmental Sciences2016,28,1:1

19Thermofluidic investigations of oil natural transformer:Closed-loop modelling and experimental validation显示文摘Excessively high temperatures inside a power transformer can accelerate the ageing rate and even cause unplanned outages.Accurately estimating the hot-spot temperature(HST)of a power transformer is important for regulating the load and ensuring long-term operation.In this study,a closed-loop model of a 110-kV product transformer with an oil natural/air forced(ONAF)cooling mode is built to predict HST,in which the compression ratio of windings,equivalent heat source and radiator geometry are considered.According to the flow field of the winding,the‘dead oil zone’and reverse oil flow are observed in the horizontal oil channels.The accuracy of the proposed model is experimentally validated by temperature-rise tests.The results show that the absolute errors between the measured results and simulation are below 5°C,and the average error is~2°C.Furthermore,the arrangements of windings and radiators are studied to enhance the oil circulation.By adding 5 washers to the high-voltage winding,the average temperature can be decreased by 6°C compared with the case without a washer.Elevating the radiator can also mitigate the characteristic temperatures,and the optimal height difference between the radiator and winding centre is suggested to be 0.66 m.Yuan Li Yiran Gao Chen Wang Chao Zhu Ying Li Yuankai Wang Guanjun Zhang 2024High Voltage2024,9,1:0
20Microstructure and properties of Si_3N_4 ceramics obtained from addition of β-phase seeds and celsian显示文摘The silicon nitride based ceramics obtained by the addition of β-Si3N4 seeds was fabricated using celsian as the additive and by pressureless sintering. The microstructure of the material was observed and analyzed using XRD,SEM,and TEM and the mechanical response of this array microstructure was characterized for flexural strength and fracture behavior. The result shows that the growth of the larger aspect ratio grains initiate mainly from the seeds through the heterogeneous nucleation mechanism during the process of phase transformation,and the intergranular phase BaAl2Si2O8 services as good refractory materials.
