1Report on Childhood Obesity in China (9): Sugar-sweetened Beverages Consumption and Obesity显示文摘Objective To explore the associations between sugar-sweetened beverage (SSB) consumption and obesity as well as obesity-related cardiometabolic disorders among children in China. Methods A total of 6974 (boys 3558, girls 3412) children aged 6-13 years participated in the study. Each participant's height, weight, waist circumference, fasting glucose, triglycerides, total cholesterol, high- density lipoprotein cholesterol, and low-density lipoprotein cholesterol were measured. The type of beverage consumption was determined using a self-administered questionnaire. Results SSBs were consumed regularly by 46.1% of the children. The prevalence [adjusted odds ratio (95% confidence internal (CI)] of obesity was 7.6% [as the reference group (ref.)], 10.1% [1.36(1.07, 1.74)], and 11.6% [1.46(1.21, 1.75)], among children who regularly drank milk, other beverages and SSBs, respectively. Regularly drinking SSBs elevated the likelihood of abdominal obesity [adjusted odds ratio (95% CI): 1.36 (1.17, 1.59)]. The prevalence [adjusted odds ratio (95% CI)] of obesity among children who regularly drank sports/caloric beverages, carbonated beverages, sweet tea, and plant protein beverages was 16.8% [2.00(1.31, 3.07)], 12.7% [1.52(1.23, 1.88)], 11.5% [1.52(1.18, 1.95)], and 10.4% [1.41(1.03, 1.94)], respectively, which was higher than that of regular milk drinkers [7.6 % (ref.)]. The prevalence [adjusted odds ratio (95% CI)] of abdominal obesity among children who regularly drank sweet tea, fruit/vegetable juices, and carbonated beverages was 17.7% [1.55(1.26, 1.90)], 16.2% [1.36(1.09, 1.70)], and 15.3% [1.24(1.03, 1.50)], respectively, which was much higher than that of regular milk drinkers [12.8% (ref.)]. Conclusions Regular SSB consumption was positively related to obesity and abdominal obesity. 2Report on Childhood Obesity in China (10): Association of Sleep Duration with Obesity显示文摘Objectives To explore the association of sleep duration with obesity among children in urban areas of China.Methods A total of 6 576 children(3 293 boys and 3 283 girls) aged 7-11 years were randomly selected from 36 primary schools in 6 metropolitan cities in China.A 7-day Physical Activity Recall was used to assess the sleep duration and physical activity level.The height,weight,waist circumference(WC) and percentage of body fat(%BF,as determined by bioelectrical impedance analysis technique) were measured by following the standardized operation procedures.The information on demography,lifestyle and eating habits was collected with a self-administered questionnaire from participants and their parents.Results The average sleep duration per night in the children was 9.7 h with the decreasing trends along with the increase of age(P<0.05).The sleep duration was negatively associated with body mass index(BMI) and WC in both boys and girls after adjustment for confounders(β value-0.23 and-0.82 for boys,-0.24 and-0.91 for girls,respectively,P<0.01).However,no significant association of sleep duration with %BF was found.Children who slept less than 9.0 h per night had a higher risk for overweight and obesity(OR=1.29,95% CI:1.01,1.64) and abdominal obesity(OR=1.38,95% CI:1.04,1.83) as compared with those who slept for 10.0-10.9 h.Conclusions Short sleep duration is associated with obesity.It is important to ensure adequate sleep duration of children and foster their healthy lifestyle at an early stage of life.MENG Li Ping LIUAi Ling HU Xiao Qi ZHANG Qian DU Song Ming FANG Hong Yun MA Jun XU Gui Fa LI Ying GUO Hong Wei DU Lin MA Guan Sheng 2012Biomedical and Environmental Sciences2012,25,2:16
3PARylation regulates stress granule dynamics, phase separation, and neurotoxicity of disease-related RNA-binding proteins显示文摘Mutations in RNA-binding proteins (RBPs) localized in ribonucleoprotein (RNP) granules, such as hnRNP A1 and TDP-43, promote aberrant protein aggregation, which is a pathological hallmark of various neurodegenerative diseases, such as amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Protein posttranslational modifications (PTMs) are known to 佗gulate RNP granules. In this study, we investigate the function of poly(ADP-ribosyl)ation (PARylation), an important PTM involved in DNA damage repair and cell death, in RNP granule-related neurodegeneration. We reveal that PARylation levels are a major regulator of the assembly-disassembly dynamics of RNP granules containing disease-related RBPs, hnRNP A1 and TDP-43. We find that hnRNP A1 can both be PARylated and bind to PARylated proteins or poly(ADP-ribose)(PAR). We further uncover that PARylation of hnRNP A1 at K298 controls its nucleocytoplasmic transport, whereas PAR-binding via the PAR-binding motif (PBM) of hnRNP Al regulates its association with stress granules. Moreover, we reveal that PAR not only dramatically enhances the liquid-liquid phase separation of hnRNP A1, but also promotes the co-phase separation of hnRNP A1 and TDP-43 in vitro and their interaction in vivo. Finally, both genetic and pharmacological inhibition of PARP mitigates hnRNP Al- and TDP-43-mediated neurotoxicity in cell and Drosophila models of ALS. 5Spraying printing of liquid metal electronics on various clothes to compose wearable functional device显示文摘Liquid metal printing is emerging as an important tool for making wearable electronics. However, very limited academic efforts were made to fulfill such an increasing need. This paper is dedicated to present relatively complete theoretical and experimental characterizations for liquid metal spraying printing towards developing wearable electronic textile. The practical conditions of liquid metal droplets in the spraying printing process such as the jet velocity, the size distribution of droplets and their evenness degree, the morphology of droplets and their unrolling areas after impacting the substrate are quantified. The dominating factors, including the oxidation of liquid metal and the pressure force on cloth substrate during the impacting process, which ensure liquid metal firmly adhere to the cloth, are clarified. Further, various clothes are comparatively investigated to test their capabilities in printing liquid metal conductors, where the resistance difference can be over thousand-fold. In addition to interpreting the basic mechanisms and performances of the spraying printing, two programmable flexible circuits with specifically designed functions such as blinking LED lighting and wireless infrared temperature measurement via current manufacture technology were also demonstrated and evaluated for their washable ability. 6Shear wave elastography results correlate with liver fibrosis histology and liver function reserve显示文摘AIM: To evaluate the correlation of shear wave elastography(SWE) results with liver fibrosis histology and quantitative function reserve.METHODS: Weekly subcutaneous injection of 60% carbon tetrachloride(1.5 m L/kg) was given to 12 canines for 24 wk to induce experimental liver fibrosis, with olive oil given to 2 control canines. At 24 wk, liver condition was evaluated using clinical biochemistry assays, SWE imaging, lidocaine metabolite monoethylglycine-xylidide(MEGX) test, and histologic fibrosis grading. Clinical biochemistry assays were performed at the institutional central laboratory for routine liver function evaluation. Liver stiffness was measured in triplicate from three different intercostal spaces and expressed as mean liver stiffness modulus(LSM). Plasma concentrations of lidocaine and its metabolite MEGX were determined using high-performance liquid chromatography repeated in duplicate. Liver biopsy samples were fixed in 10% formaldehyde, and liver fibrosis was graded using the modified histological activity index Knodell score(F0-F4). Correlations among histologic grading, LSM, and MEGX measures were analyzed with the Pearson linear correlation coefficient.RESULTS: At 24 wk liver fibrosis histologic grading was as follows: F0, n = 2(control); F1, n = 0; F2, n = 3; F3, n = 7; and F4, n = 2. SWE LSM was positively correlated with histologic grading(r = 0.835, P < 0.001). Specifically, the F4 group had a significantly higher elastic modulus than the F3, F2, and F0 groups(P = 0.002, P = 0.003, and P = 0.006, respectively), and the F3 group also had a significantly higher modulus than the control F0 group(P = 0.039). LSM was negatively associated with plasma MEGX concentrations at 30 min(r =-0.642; P = 0.013) and 60 min(r =-0.651; P = 0.012), time to ? of the maximum concentration(r =-0.538; P = 0.047), and the area under the curve(r =-0.636; P = 0.014). 12Role of EGFL7/EGFR-signaling pathway in migration and invasion of growth hormone-producing pituitary adenomas显示文摘Currently, the primary therapeutic strategy for most growth hormone-producing pituitary adenomas(GHPA) is surgery. Due to the invasiveness of GHPA, high recurrence has limited the benefit of complete adenoma removal surgery. Epidermal growth factor-like domain 7(EGFL7) is a secreted factor implicated in tumor angiogenesis, growth, invasiveness and metastasis in GHPA. Herein, we observed that the expression level of EGFL7 and p-EGFR in invasive GHPAwas much higher than that of non-invasive GHPA. The overexpression of EGFL7 was positively correlated with activation of EGFR(p-EGFR). Noticeably, EGFL7 knockdown significantly inhibited activation of EGFR signaling cascades, including p-ERGR, p-AKT and p-ERK. Further studies showed that EGFL7 knockdown or pharmacological inhibition of EGFR-pathway, using EGFR inhibitor Tyrphostin AG-1478, significantly suppressed migration and invasion of GH3 and GT1-1 cells. 13In vivo degradation and endothelialization of an iron bioresorbable scaffold显示文摘Detection of in vivo biodegradation is critical for development of next-generation medical devices such as bioresorbable stents or scaffolds(BRSs).In particular,it is urgent to establish a nondestructive approach to examine in vivo degradation of a new-generation coronary stent for interventional treatment based on mammal experiments;otherwise it is not available to semi-quantitatively monitor biodegradation in any clinical trial.Herein,we put forward a semi-quantitative approach to measure degradation of a sirolimus-eluting iron bioresorbable scaffold(IBS)based on optical coherence tomography(OCT)images;this approach was confirmed to be consistent with the present weight-loss measurements,which is,however,a destructive approach.The IBS was fabricated by a metal-polymer composite technique with a polylactide coating on an iron stent.The efficacy as a coronary stent of this new bioresorbable scaffold was compared with that of a permanent metal stent with the name of trade mark Xience,which has been widely used in clinic.The endothelial coverage on IBS was found to be greater than on Xience after implantation in a rabbit model;and our well-designed ultrathin stent exhibited less individual variation.We further examined degradation of the IBSs in both minipig coronary artery and rabbit abdominal aorta models.The present result indicated much faster iron degradation of IBS in the rabbit model than in the porcine model.The semi-quantitative approach to detect biodegradation of IBS and the finding of the species difference might be stimulating for fundamental investigation of biodegradable implants and clinical translation of the next-generation coronary stents.Wenjiao Lin Hongjie Zhang Wanqian Zhang Haiping Qi Gui Zhang Jie Qian Xin Li Li Qin Haifeng Li Xiang Wang Hong Qiu Xiaoli Shi Wei Zheng Deyuan Zhang Runlin Gao Jiandong Ding 2021Bioactive Materials2021,6,4:3
14Chidamide combined with cyclophosphamide,doxorubicin,vincristine and prednisone in previously untreated patients with peripheral T-cell lymphoma显示文摘Objective:Chidamide is an oral histone deacetylase subtype-selective inhibitor approved for relapsed or refractory peripheral T-cell lymphoma(PTCL).This phase 1 b study evaluated the safety,pharmacokinetics,and preliminary efficacy of chidamide in combination with cyclophosphamide,doxorubicin,vincristine and prednisone(CHOP)for treatment-na?ve PTCL patients.Methods:This study was an open-label,multicenter trial composed of dose escalation and dose expansion.Patients received CHOP for six 21-d cycles and chidamide on d 1,4,8 and 11 in each cycle.Four dose levels of chidamide(20,25,30 and 35 mg)were evaluated.The primary objective was to evaluate the safety and tolerability of the combination regimen.Results:A total of 30 patients were evaluated in this study:15 in the dose-escalation part and 15 in the doseexpansion part.In the dose-escalation study,three patients were enrolled in the 35 mg chidamide cohort.One had dose-limiting toxicity with grade 3 vascular access complications,and one had grade 2 neutropenia with a sustained temperature>38°C.Dose escalation was stopped at this chidamide dose level.The most common(≥10%)grade 3 or 4 adverse events(AEs)were leukopenia(90.0%),neutropenia(83.3%),vomiting(13.3%),thrombocytopenia(10.0%)and febrile neutropenia(10.0%).No significant changes in chidamide pharmacokinetic properties were observed before and after combination treatment.The objective response rate for the 28 patients evaluable for preliminary efficacy was 89.3%(25/28),with 16(57.1%)achieving complete response or unconfirmed complete response.The estimated median progression-free survival was 14.0 months.In summary,we chose chidamide 30 mg as the recommended dose for phase 2.Conclusions:The addition of chidamide to standard CHOP chemotherapy was tolerable with promising preliminary efficacy in previously untreated PTCL patients,which supports further clinical studies with this combination regimen for the frontline treatment of PTCL.Lin Gui Junning Cao Dongmei Ji Huilai Zhang Qian Fan Jun Zhu Yuqin Song Shiyu Jiang Zhiqiang Ning Jia Yu Yuankai Shi 2021Chinese Journal of Cancer Research2021,33,5:3
15Cis-2-dodecenoic Acid Mediates Its Synergistic Effect with Triazoles by Interfering with Efflux Pumps in Fluconazole-resistant Candida albicans显示文摘Objective To evaluate the synergy of the Burkholderia signaling molecule cis-2-dodecenoic acid(BDSF) and fluconazole(FLU) or itraconazole(ITRA) against two azole-resistant C. albicans clinical isolates in vitro and in vivo. Methods Minimum inhibitory concentrations(MICs) of antibiotics against two azole-resistant C. albicans were measured by the checkerboard technique, E-test, and time-kill assay. In vivo antifungal synergy testing was performed on mice. Analysis of the relative gene expression levels of the strains was conducted by quantitative reverse-transcription polymerase chain reaction(qR T-PCR). Results BDSF showed highly synergistic effects in combination with FLU or ITRA with a fractional inhibitory concentration index of ≤ 0.08. BDSF was not cytotoxic to normal human foreskin fibroblast cells at concentrations of up to 300 μg/mL. The qR T-PCR results showed that the combination of BDSF and FLU/ITRA significantly inhibits the expression of the efflux pump genes CDR1 and MDR1 via suppression of the transcription factors TAC1 and MRR1, respectively, when compared with FLU or ITRA alone. No dramatic difference in the mR NA expression levels of ERG1, ERG11, and UPC2 was found, which indicates that the drug combinations do not significantly interfere with UPC2-mediated ergosterol levels. In vivo experiments revealed that combination therapy can be an effective therapeutic approach to treat candidiasis. 16The apoptosis of HEL cells induced by hydroxyurea显示文摘Hydroxyurea has been used to synchronize cultured cells to S-phase and used to treat patients with sicklecell anemia. Recently, we found that hydroxyurea can induce the apoptosis of HEL (human erythroleukemia) cells.The induced HEL cells showed ultrastructurally chromatin condensation with regular crescents at the nuclear edges and apoptotic bodies. However, the cells of K562, another human erythroleukemia cell line, did not show such morphological changes. Under fluoroscope, the HEL cells after induction often displayed a clear reduction in nuclear diameter and nuclear chromatin cleavage and condensation and the presence of nuclear ring and apoptotic bodies. Analysis with flow cytometry showed that the percentage of apoptotic cells is about 30-40% after HEL cells were induced by hydroxyurea for 3 days. 17Clinical application of free/total PSA ratio in the diagnosis of prostate cancer in men over 50 years of age with total PSA levels of 2.0-25.0 ng ml^(−1) in Western China显示文摘The goal of this study was to investigate the clinical application of free/total prostate-specific antigen(F/T PSA)ratio,considering the new broad serum total PSA(T-PSA)“gray zone”of 2.0–25.0 ng ml^(−1)in differential diagnosis of prostate cancer(PCa)and benign prostate diseases(BPD)in men over 50 years in Western China.A total of 1655 patients were included,528 with PCa and 1127 with BPD.Serum T-PSA,free PSA(F-PSA),and F/T PSA ratio were analyzed.Receiver operating characteristic curves were used to assess the efficiency of PSA and F/T PSA ratio.There were 47.4%of cancer patients with T-PSA of 2.0–25.0 ng ml^(−1).When T-PSA was 2.0–4.0 ng ml^(−1),4.0–10.0 ng ml^(−1),and 10.0–25.0 ng ml^(−1),the area under the curve(AUC)of F/T PSA ratio was 0.749,0.769,and 0.761,respectively.The best AUC of F/T PSA ratio was 0.811 when T-PSA was 2.0–25.0 ng ml^(−1),with a specificity of 0.732,a sensitivity of 0.788,and an optimal cutoff value of 15.5%.The AUC of F/T PSA ratio in different age groups(50–59 years,60–69 years,70–79 years,and≥80 years)was 0.767,0.806,0.815,and 0.833,respectively,and the best sensitivity(0.857)and specificity(0.802)were observed in patients over 80 years.The T-PSA trend was in accordance with the Gleason score,tumor node metastasis(TNM)stage,and American Joint Committee on Cancer prognosis group.Therefore,the F/T PSA ratio can facilitate the differential diagnosis of PCa and BPD in the broad T-PSA“gray zone”.Serum T-PSA can be a Gleason score and prognostic indicator.Xue-Dan Gao Qiang Miao Jun-Long Zhang Jian-Zhao Zhai Xue-Mei Gui Yi-Han Cai Qian Niu Bei Cai 2022Asian Journal of Andrology2022,24,2:2
18HIV-1 Vif suppresses antiviral immunity by targeting STING显示文摘HIV-1 infection-induced cGAS–STING–TBK1–IRF3 signaling activates innate immunity to produce type I interferon(IFN).The HIV-1 nonstructural protein viral infectivity factor(Vif)is essential in HIV-1 replication,as it degrades the host restriction factor APOBEC3G.However,whether and how it regulates the host immune response remains to be determined.In this study,we found that Vif inhibited the production of type I IFN to promote immune evasion.HIV-1 infection induced the activation of the host tyrosine kinase FRK,which subsequently phosphorylated the immunoreceptor tyrosine-based inhibitory motif(ITIM)of Vif and enhanced the interaction between Vif and the cellular tyrosine phosphatase SHP-1 to inhibit type I IFN.Mechanistically,the association of Vif with SHP-1 facilitated SHP-1 recruitment to STING and inhibited the K63-linked ubiquitination of STING at Lys337 by dephosphorylating STING at Tyr162.However,the FRK inhibitor D-65495 counteracted the phosphorylation of Vif to block the immune evasion of HIV-1 and antagonize infection.These findings reveal a previously unknown mechanism through which HIV-1 evades antiviral immunity via the ITIM-containing protein to inhibit the posttranslational modification of STING.These results provide a molecular basis for the development of new therapeutic strategies to treat HIV-1 infection.Yu Wang Gui Qian Lingyan Zhu Zhuo Zhao Yinan Liu Wendong Han Xiaokai Zhang Yihua Zhang Tingrong Xiong Hao Zeng Xianghui Yu Xiaofang Yu Xiaoyan Zhang Jianqing Xu Quanming Zou Dapeng Yan 2022Cellular & Molecular Immunology2022,19,1:2
19Similarities and differences in clinical and pathologic features of inflammatory bowel disease-associated colorectal cancer in China and Canada显示文摘Background:Colorectal cancer(CRC)has become one of the major life-threatening complications in patients with inflammatory bowel disease(IBD),which includes ulcerative colitis(UC)and Crohn's disease(CD).This study aimed to explore the clinicalpathologic similarities and differences in the IBD-associ吐ed CRC(IBD-CRC)between patients in China and Canada.Methods:Data of 78 patients with IBD-CRC retrospectively retrieved from two representative medical institutions in Beijing(China)and Calgary(Canada)over the same past 13 years,including 25(22 UC-associated and three CD-associated)from Beijing group and 53(32 UC-associated and 21 CD-associated)from Calgary group,were compared with regards to their clinical and pathologic characteristics.Results:Several known features of IBD-CRC were seen in both groups,including long duration and large extent of colitis,active inflammation background,multifocal lesions,and advanced tumor-node-metastasis stage.Beijing group showed a significantly higher percentage of UC(88.0%vs.60.4%,P=0.018),younger age at diagnosis of CRC(48.6±12.8 years vs.61.6±14.7 years,P<0.001),lower ratio of mucinous adenocarcinoma(7.1%us.42.4%,P=0.001)compared with Calgary group.None of the Beijing group had concurrent primary sclerosing cholangitis,while 5.7%of Calgary group did.Surveillance colonoscopy favored the detection rate of precancerous lesions(41.4%vs.17.0%,P=0.002).Conclusions:As compared with patients from the Calgary group,the IBD-CRC patients in Beijing group were younger,less CDassociated and had less mucinous features,otherwise they were similar in many common features.Ji Li Wei-Xun Zhou Shuang Liu Wei-Yang Zheng Ya-Nan Wang Jing-Nan Li Jose Gp Ferraz Jia-Ming Qian Xian-Yong Gui 2019Chinese Medical Journal2019,,22:2
