| 1 | Design,methodology,and baseline of whole city‑million scale children and adolescents myopia survey(CAMS)in Wenzhou,China显示文摘Background:Myopia is the most common visual impairment in children and adolescents worldwide.This study described an economical and effective population-based screening pipeline and performed the project of a million scale children and adolescents myopia survey(CAMS),which will shed light on the further study of myopia from the level of epidemiology and precision medicine.Methods:We developed a novel population-based screening pattern,an intelligent screening process and internetbased information transmission and analysis system to carry out the survey consisting of school children in Wenzhou,China.The examination items include unaided distance visual acuity,presenting distance visual acuity,and noncycloplegic autorefraction.Myopia and high myopia were defined as spherical equivalent(SE)≤−1.00 diopters(D)and SE≤−6.00 D,respectively.Next,the reports of the vision checking were automatically sent to parents and the related departments.The CAMS project will be done two to four times annually with the support of the government.An online eyesight status information management system(OESIMS)was developed to construct comprehensive and efficient electronic vision health records(EVHRs)for myopia information inquiry,risk pre-warning,and further study.Results:The CAMS completed the first-round of screening within 30 days for 99.41%of Wenzhou students from districts and counties,in June 2019.A total of 1,060,925 participants were eligible for CAMS and 1,054,251(99.37%participation rate)were selected through data quality control,which comprised 1305 schools,and 580,609,251,050 and 170,967 elementary,middle,and high school students.The mean age of participants was 12.21±3.32 years(6-20 years),the female-to-male ratio was 0.82.The prevalence of myopia in elementary,middle,and high school students was 38.16%,77.52%,and 84.00%,respectively,and the high myopia incidence was 0.95%,6.90%,and 12.98%.Conclusions:The CAMS standardized myopia screening model involves automating large-scale information collection,data transmission,data analysis and early warning,thereby supporting myopia prevention and control.The entire survey reduced 90%of staff,cost,and time consumption compared with previous surveys.This will provide new insights for decision support for public health intervention. | Liangde Xu Youyuan Zhuang Guosi Zhang Yunlong Ma Jian Yuan Changseng Tu MiaoMiao Li Wencan Wang Yaru Zhang Xiaoyan Lu Jing Li Xinting Liu Zhengbo Xue Meng Zhou Jie Sun Jinhua Bao Ming Li Fan Lu Hong Wang Jianzhong Su Jia Qu | 2021 | Eye and Vision2021,8,1: | 7 |
| 2 | Development and validation of a TaqMan^(TM) fluorescent quantitative real-time PCR assay for the rapid detection of Edwardsiella tarda显示文摘Edwardsiella tarda is one of the most important emerging pathogens in the global aquaculture industries. As such, an accurate diagnosis and quantitative analytical methods are urgently needed for this bacterium. In this study, primers and a TaqMan probe specific to the conservative sequences of the 16S rRNA gene of E. tarda were designed. The concentration of primers and TaqMan probe were optimized to 200 nmol/L and 120 nmol/L, respectively. The detection sensitivity of the FQ-PCR assay was determined to be as low as five copies of the target sequence per reaction using the pGEM-16S rDNA recombinant plasmid as a template, which was 100 times more sensitive than conventional PCR. A standard curve by plotting the threshold cycle values (y) against the common logarithmic copies (log_10 n_c as x; n_c is copy number) of pGEM-16S rDNA was generated. The results of intra-and inter-assay variability tests demonstrate that the established FQ-PCR method was highly reproducible. The assay was specific for E. tarda as it showed that there was no cross-reactivity to eight additional bacterial pathogen strains in aquaculture. | 5 | A Novel Vibriophage vB_VcaS_HC Containing Lysogeny-Related Gene Has Strong Lytic Ability against Pathogenic Bacteria显示文摘To avoid the negative effects of antibiotics,using phage to prevent animal disease becomes a promising method in aquaculture.Here,a lytic phage provisionally named vB_Vca S_HC that can infect the pathogen(i.e.,Vibrio campbellii 18)of prawn was isolated.The phage has an isometric head and a non-contractile tail.During phage infection,the induced host mortality in 5.5 h reached ca.96%,with a latent period of 1.5 h and a burst size of 172 PFU/cell.It has an 81,566 bp circular ds DNA genome containing 121 open reading frames(ORFs),and ca.71%of the ORFs are functionally unknown.Comparative genomic and phylogenetic analysis revealed that it is a novel phage belonging to Delepquintavirus,Siphoviridae,Caudovirales.In the phage genome,besides the ordinary genes related to structure assembly and DNA metabolism,there are 10 auxiliary metabolic genes.For the first time,the pyruvate phosphate dikinase(PPDK)gene was found in phages whose product is a key rate-limiting enzyme involving Embden-Meyerhof-Parnas(EMP)reaction.Interestingly,although the phage has a strong bactericidal activity and contains a potential lysogeny related gene,i.e.,the recombinase(Rec A)gene,we did not find the phage turned into a lysogenic state.Meanwhile,the phage genome does not contain any bacterial virulence gene or antimicrobial resistance gene.This study represents the first comprehensive characterization of a lytic V.campbellii phage and indicates that it is a promising candidate for the treatment of V.campbellii infections. | Chengcheng Li Zengmeng Wang Jiulong Zhao Long Wang Guosi Xie Jie Huang Yongyu Zhang | 2021 | Virologica Sinica2021,36,2: | 1 |
