| 2 | Multiplex genome editing using a dCas9-cytidine deaminase fusion in Streptomyces显示文摘CRISPR/Cas-mediated genome editing has greatly facilitated the study of gene function in Streptomyces. However, it could not be efficiently employed in streptomycetes with low homologous recombination(HR) ability. Here, a deaminase-assisted base editor d Cas9-CDA-ULstr was developed in Streptomyces, which comprises the nuclease-deficient Cas9(dCas9), the cytidine deaminase from Petromyzon marinus(PmCDA1), the uracil DNA glycosylase inhibitor(UGI) and the protein degradation tag(LVA tag). Using d Cas9-CDA-ULstr , we achieved single-, double-and triple-point mutations(cytosine-to-thymine substitutions)at target sites in Streptomyces coelicolor with efficiency up to 100%, 60% and 20%, respectively. This base editor was also demonstrated to be highly efficient for base editing in the industrial strain, Streptomyces rapamycinicus, which produces the immunosuppressive agent rapamycin. Compared with base editors derived from the cytidine deaminase rAPOBEC1, the PmCDA1-assisted base editor dCas9-CDA-ULstr could edit cytosines preceded by guanosines with high efficiency, which is a great advantage for editing Streptomyces genomes(with high GC content). Collectively, the base editor dCas9-CDA-ULstr could be employed for efficient multiplex genome editing in Streptomyces. | 3 | Statistical characteristics and mechanisms of mesoscale eddies in the North Indian Ocean显示文摘The statistical characteristics and mechanisms of mesoscale eddies in the North Indian Ocean are investigated by adopting multi-sensor satellite data from 1993 to 2019.In the Arabian Sea(AS),seasonal variation of eddy characteristics is remarkable,while the intraseasonal variability caused by planetary waves is crucial in the Bay of Bengal(BOB).Seasonal variation of the eddy kinetic energy(EKE)is distinct along the west boundary of AS,especially in the Somali Current region.In the BOB,larger EKE occurs at the northwest basin from March to May,to the east of Sri Lanka from June to September,and along the east coast of India from November to December.The wind stress work(WW)is further studied to figure out the direct influence of wind forcing on EKE.The WW exerts positive effects on EKE along the west boundary of AS and in the south of India/Sri Lanka during the two monsoon seasons.Besides,the WW also has impact on EKE along the east coast of India in November and December.Eventually,we investigate the characteristics and the driving mechanisms of long lifespan eddies.In the AS,long lifespan anti-cyclonic eddies(AEs)mainly generate in the Socotra,the West Indian Coastal Current and the East Arabian Current regions,while cyclonic eddies(CEs)are concentrated in the northwest region.In the BOB,long lifespan AEs mostly form near the west of Myanmar,while CEs are accumulated at the north and northwest basin.The instabilities caused by Rossby waves,coastal Kelvin waves,seasonal currents,together with wind stress forcing exert enormous efforts on the generation and evolution of these eddies. | Chunjian Sun Xidong Wang Anmin Zhang Lianxin Zhang Caixia Shao Guosong Wang | 2022 | Acta Oceanologica Sinica2022,41,5: | 1 |

| 5 | Telomere-to-telomere pear (Pyrus pyrifolia) reference genome reveals segmental and whole genome duplication driving genome evolution显示文摘Previously released pear genomes contain a plethora of gaps and unanchored genetic regions.Here,we report a telomere-to-telomere(T2T)gap-free genome for the red-skinned pear,‘Yunhong No.1’(YH1;Pyrus pyrifolia),which is mainly cultivated in Yunnan Province(southwest China),the pear’s primary region of origin.The YH1 genome is 501.20 Mb long with a contig N50 length of 29.26 Mb.All 17 chromosomes were assembled to the T2T level with 34 characterized telomeres.The 17 centromeres were predicted and mainly consist of centromeric-specific monomers(CEN198)and long terminal repeat(LTR)Gypsy elements(≥74.73%).By filling all unclosed gaps,the integrity of YH1 is markedly improved over previous P.pyrifolia genomes(‘Cuiguan’and‘Nijisseiki’).A total of 1531 segmental duplication(SD)driven duplicated genes were identified and enriched in stress response pathways.Intrachromosomal SDs drove the expansion of disease resistance genes,suggesting the potential of SDs in adaptive pear evolution.A large proportion of duplicated gene pairs exhibit dosage effects or sub-/neo-functionalization,whichmay affect agronomic traits like stone cell content,sugar content,and fruit skin russet.Furthermore,as core regulators of anthocyanin biosynthesis,we found that MYB10 and MYB114 underwent various gene duplication events.Multiple copies of MYB10 and MYB114 displayed obvious dosage effects,indicating role differentiation in the formation of red-skinned pear fruit.In summary,the T2T gap-free pear genome provides invaluable resources for genome evolution and functional genomics. | Manyi Sun Chenjie Yao Qun Shu Yingyun He Guosong Chen Guangyan Yang Shaozhuo Xu Yueyuan Liu Zhaolong Xue Jun Wu | 2023 | Horticulture Research2023,10,11: | 0 |

| 8 | Achieving gradient heterogeneous structure in Mg alloy for excellent strength-ductility synergy显示文摘Most metals including Mg alloys have a longstanding dilemma of strength-ductility trade-off,which is hindering their wider applications.In this study,we propose a gradient heterogeneous grain(GHG)structure for evading this trade-off dilemma and ultrasonic severe surface rolling is attempted to construct this novel structure in ZE41 Mg alloy.Here,the GHG structure combine the benefits of gradient structure and heterogeneous grain structure and introduce large microstructural heterogeneities.Compared to the coarse-grain and heterogeneous-grain structured alloys,the GHG structured one exhibits dramatical enhancement in strength,ductility,and strain hardening capability.To the best of our knowledge,its strength becomes much higher than that of common ZE41 Mg alloys at no reduction in ductility.These unique mechanical properties stem from not only the individual contribution of the heterogeneous structure components including the fine/ultrafine grains and deformed coarse grains but also their synergistic effect via hetero-deformation induced strengthening and hardening effects.In summary,our study provides a feasible way to develop new Mg alloys with high strength and good ductility. | Jing Han Jiapeng Sun Yuanming Song Bingqian Xu Zhenquan Yang Songsong Xu Ying Han Guosong Wu Jiyun Zhao | 2023 | Journal of Magnesium and Alloys2023,11,7: | 0 |
