3Precise nanomedicine for intelligent therapy of cancer显示文摘Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. 5Angular response of ‘pin-hole' imaging structure measured by collimated β source显示文摘The pitch-angle distribution of energetic particles is important for space physics studies on magnetic storm and particle acceleration.A‘pin-hole’imaging structure is built with the‘pin-hole’technique and a position sensitive detector,which can be used to measure the pitch angle distribution of energetic particles.To calibrate the angular response of the‘pin-hole’imaging structure,special experiment facilities are needed,such as the particle accelerator with special design.The features of this kind of particle accelerator are:1)The energy range of the outgoing particles should be mid-energy particles(tens keV to several hundred keV);2)the particle flux should be consistent in time-scale;3)the directions of the outgoing particles should be the same and 4)the particle number within the spot should be low enough.In this paper,a method to calibrate the angular response of the‘pin-hole’imaging structure by the90Sr/90Y β source with a collimator is introduced and simulated by Geant4 software.The result of the calibration with the collimated β source is in accord with the Geant4 simulations,which verifies the validity of this method.ZOU Hong LUO Lin LI ChenFang JIA XiangHong XU Feng CHEN HongFei CHEN Jiang SHI WeiHong YU XiangQian ZOU JiQing 2013Science China(Technological Sciences)2013,56,11:10
6Progress on Porous Ceramic Membrane Reactors for Heterogeneous Catalysis over Ultrafine and Nano-sized Catalysts显示文摘Heterogeneous catalysts with ultrafine or nano particle size have currently attracted considerable attentions in the chemical and petrochemical production processes, but their large-scale applications remain challenging because of difficulties associated with their efficient separation from the reaction slurry. A porous ceramic membrane reactor has emerged as a promising method to solve the problem concerning catalysts separation in situ from the reaction mixture and make the production process continuous in heterogeneous catalysis. This article presents a review of the present progress on porous ceramic membrane reactors for heterogeneous catalysis, which covers classification of configurations of porous ceramic membrane reactor, major considerations and some important industrial applications. A special emphasis is paid to major considerations in term of application-oriented ceramic membrane design, optimization of ceramic membrane reactor performance and membrane fouling mechanism. 7Improvement of pristinamycin I(PI)production in Streptomyces pristinaespiralis by metabolic engineering approaches显示文摘Pristinamycin,produced by Streptomyces pristinaespiralis,which is a streptogramin-like antibiotic consisting of two chemically unrelated components:pristinamycin I(PI)and pristinamycin II(PII),shows potent activity against many antibiotic-resistant pathogens.However,so far pristinamycin production titers are still quite low,particularly those of PI.In this study,we constructed a PI single component producing strain by deleting the PII biosynthetic genes(snaE1 and snaE2).Then,two metabolic engineering approaches,including deletion of the repressor gene papR3 and chromosomal integration of an extra copy of the PI biosynthetic gene cluster(BGC),were employed to improve PI production.The final engineered strain DPIIDpapR3/PI produced a maximum PI level of 132 mg/L,with an approximately 2.4-fold higher than that of the parental strain S.pristinaespiralis HCCB10218.Considering that the PI biosynthetic genes are clustered in two main regions in the 210 kb“supercluster”containing the PI and PII biosynthetic genes as well as a cryptic polyketide BGC,these two regions were cloned separately and then were successfully assembled into the PI BGC by the transformation-associated recombination(TAR)system.Collectively,the metabolic engineering approaches employed is very efficient for strain improvement in order to enhance PI titer.Jiali Meng Rongrong Feng Guosong Zheng Mei Ge Yvonne Mast Wolfgang Wohlleben Jufang Gao Weihong Jiang Yinhua Lu 2017Synthetic and Systems Biotechnology2017,2,2:7
8Metabolic engineering of Streptomyces coelicolor for enhanced prodigiosins (RED) production显示文摘Bacterial prodigiosins are red-colored secondary metabolites with multiple activities,such as anticancer,antimalarial and immunosuppressive,which hold great potential for medical applications.In this study,dramatically enhanced prodigiosins(RED) production in Streptomyces coelicolor was achieved by combinatorial metabolic engineering,including inactivation of the repressor gene ohkA,deletion of the actinorhodin(ACT) and calcium-dependent antibiotic(CDA) biosynthetic gene clusters(BGCs) and multi-copy chromosomal integration of the RED BGC.The results showed that ohkA deletion led to a 1-fold increase of RED production over the wild-type strain M145.Then,the ACT and CDA BGCs were deleted successively based on the AohkA mutant(SBJ101).To achieve multi-copy RED BGC integration,artificial ΦC31 attB site(s) were inserted simultaneously at the position where the ACT and CDA BGCs were deleted.The resulting strains SBJ102(with a single deletion of the ACT BGC and insertion of one artificial attB site) and SBJ103(with the deletion of both BGCs and insertion of two artificial attB sites) produced 1.9-and 6-fold higher RED titers than M145,respectively.Finally,the entire RED BGC was introduced into mutants from SBJ101 to SBJ103,generating three mutants(from SBJ104 to SBJ106) with chromosomal integration of one to three copies of the RED BGC.The highest RED yield was from SBJ106,which produced a maximum level of 96.8 mg g^(-1) cell dry weight,showing a 12-fold increase relative to M145.Collectively,the metabolic engineering strategies employed in this study are very efficient for the construction of high prodigiosin-producing strains.Panpan Liu Hong Zhu Guosong Zheng Weihong Jiang Yinhua Lu 2017Science China(Life Sciences)2017,60,9:7
9Detection and characterization of diverse alpha-and betacoronaviruses from bats in China显示文摘Bats have been implicated as important reservoir hosts of alpha- and betacoronaviruses. In this study, diverse coronaviruses(CoVs) were detected in 50 of 951(positive rate 5.3%) intestinal specimens of eight bat species collected in four provinces and the Tibet Autonomous Region of China by pan-coronavirus RT-PCR screening. Based on 400-nt RNA-dependent RNA polymerase(Rd RP) sequence analysis, eight belonged to genus Alphacoronavirus and 42 to Betacoronavirus.Among the 50 positive specimens, thirteen gave rise to CoV full-length Rd RP gene amplification for further sequence comparison, of which three divergent sequences(two from a unreported province) were subjected to full genome sequencing. Two complete genomes of betacoronaviruses(JTMC15 and JPDB144) and one nearly-complete genome of alphacoronavirus(JTAC2) were sequenced and their genomic organization predicted. The present study has identified additional numbers of genetically diverse bat-borne coronaviruses with a wide distribution in China. 10Nanoparticle monolayer-based flexible strain gauge with ultrafast dynamic response for acoustic vibration detection显示文摘The relatively poor dynamic response of current flexible strain gauges has prevented their wide adoption in portable electronics. In this work, we present a greatly improved flexible strain gauge, where one strip of Au nanoparticle (NP) monolayer assembled on a polyethylene terephthalate film is utilized as the active unit. The proposed flexible gauge is capable of responding to applied stimuli without detectable hysteresis via electron tunneling between adjacent nanoparticles within the Au NP monolayer. Based on experimental quantification of the time and frequency domain dependence of the electrical resistance of the proposed strain gauge, acoustic vibrations in the frequency range of 1 to 20,000 Hz could be reliably detected. 11Freeform OST-HMD system with large exit pupil diameter and vision correction capability显示文摘Compactness and light weight,large exit pupil diameter and distance,small distortion for virtual image,and see-through light paths are pivotal factors to achieve a better,wearable experience of optical see-through headmounted displays(OST-HMDs).In addition,light efficiency of the virtual image light path is an important factor for heat dissipation in HMD devices.This paper presents a new type of OST-HMD optical system that includes three wedge-shaped freeform prisms and two symmetric lenses.Based on a 0.71 in.microdisplay,an OST-HMD prototype with a diagonal field of view(FOV)of 45.3°,an F-number(F/#)of 1.8,an exit pupil size of 12 mm×8 mm,and an eye relief of 18 mm is demonstrated.The maximum value of distortion of the final system is 0.6%and 0.4%for virtual image and see-through light path,respectively.The overall dimension of the optical system per eye is no larger than 30 mm(width)×40 mm(height)×14 mm(thickness),and the weight of the optical module including lenses,holder,and microdisplay is 12.8 g.The light efficiency of the virtual image light path is up to 50%higher than those of other OST-HMD optical solutions.DEWEN CHENG JIAXI DUAN HAILONG CHEN HE WANG DANYANG LI QIWEI WANG QICHAO HOU TONG YANG WEIHONG HOU DONGHUA WANG XIAOYU CHI BIN JIANG YONGTIAN WANG 2022Photonics Research2022,10,1:5
12Imaging energetic electron spectrometer onboard a Chinese navigation satellite in the inclined GEO orbit显示文摘The energetic electron measurement is one of the most important issues to understand dynamics in space physics and the applications for space weather. In this study, the principle and functional components of the imaging energetic electron spectrometer(IES) onboard a Chinese navigation satellite in the inclined GEO orbit(IGSO) was introduced. The IES instrument is developed by the team in Peking University(BeiDa), thus it is named as BD-IES. Based on the pin-hole technique, the instrument can measure 50–600 keV electrons incident from 9 directions over a range of 180° in polar angle. With pulse height analysis(PHA), the spectrum can be determined for each direction. The energy and angular calibrations were performed, which show the good energy and angular characteristics of BD-IES. Monte Carlo simulations show that the anti-proton design of BDIES can effectively decrease the proton contamination on the electron measurements in the inclined GEO orbit. 14Regional relationship between the Jiang-Huai Meiyu and the equatorial surface-subsurface temperature anomalies显示文摘The Jiang-Huai Meiyu rainy season can be distinguished into the Jiangnan Meiyu spell and the Huaihe Meiyu spell. The Jiangnan Meiyu spell appears on the last ten days in June and the Huaihe Meiyu spell lasts from early July to middle July. An inter-decadal transition was observed in 1998 respectively from the anomalies of Jiangnan Meiyu rainfall,the sea surface temperature (SST),and the subsurface tem-perature in the equatorial Pacific. Since the beginning of the 21st century,opposite trends and biennial oscillations of the Meiyu rainfall are observed in the Jiangnan and Huaihe basins. Before the strong La Nia of 1999―2000,the positive SST anomalies usually occurred in the eastern equatorial Pacific. Since the beginning of the 21st century,a precursory warming signal of SST anomaly comes from the subsurface temperature which is centrally exposed near the dateline in the central equatorial Pacific. The above-normal Meiyu rainfall in 2003,2005 and 2007 over the Huaihe basin followed the prior winter-spring positive SST anomaly near the dateline. 15Optimal operation of cold–heat–electricity multi-energy collaborative system based on price demand response显示文摘In a multi-energy collaboration system, cooling, heating, electricity, and other energy components are coupled to complement each other. Through multi-energy coordination and cooperation, they can significantly improve their individual operating efficiency and overall economic benefits. Demand response, as a multi-energy supply and demand balance method, can further improve system flexibility and economy. Therefore, a multi-energy cooperative system optimization model has been proposed, which is driven by price-based demand response to determine the impact of power-demand response on the optimal operating mode of a multi-energy cooperative system. The main components of the multi-energy collaborative system have been analyzed. The multi-energy coupling characteristics have been identified based on the energy hub model. Using market elasticity as a basis, a price-based demand response model has been built. The model has been optimized to minimize daily operating cost of the multi-energy collaborative system. 16Decoupled Delay and Bandwidth Centralized Queue-Based QoS Scheme in OpenFlow Networks显示文摘There are an increasing of scenarios that require the independent bandwidth and delay demands. For instance, in a data center, the interactive message would not occupy much bandwidth, but it requires the rigorous demands for the delay. However, the existing QoS approaches are mainly bandwidth based, which are inappropriate for these scenarios. Hence, we propose the decoupled scheme in the OpenFlow networks to provide the centralized differential bandwidth and delay control. We leverage the mature HTB to manage the bandwidth. And we design the Queue Delay Management Scheme (QDMS) for queuing delay arrangement, as well as the Comprehensive Parameters based Dijkstra Route algorithm (CPDR) for the propagation delay control. 17Phenol hydrogenation to cyclohexanone over palladium nanoparticles loaded on charming activated carbon adjusted by facile heat treatment显示文摘Selective phenol hydrogenation is a green approach to produce cyclohexanone.It still remains a big challenge to prepare efficient supports of the catalysts for the phenol hydrogenation via a simple and cost-effective approach.Herein,a facile approach was developed,i.e.,direct calcination of activated carbon(AC)under argon at high temperature,to improve its structure and surface properties.The modified AC materials were supported with Pd nanoparticles(NPs)to fabricate the Pd/C catalysts.The as-prepared Pd/C600 catalyst exhibits superior catalytic performance in the phenol hydrogenation,and its turnover frequency(TOF)value is 199.2 h^-1,1.31 times to that of Pd/C-raw.The Pd/C600 catalyst presents both better hydrophobicity and more structural defects,contributing to the improved dispersibility in the reaction solution(phenol-cyclohexane),the better Pd dispersion and the smaller Pd size,which result in the enhancement of the catalytic performance.Furthermore,the as-prepared Pd/C600 catalyst shows a good recyclability.Chunhua Zhang Guangxin Yang Hong Jiang Yefei Liu Rizhi Chen Weihong Xing 2020Chinese Journal of Chemical Engineering2020,28,10:2
18Overexpression of the diguanylate cyclase CdgD blocks developmental transitions and antibiotic biosynthesis in Streptomyces coelicolor显示文摘Cyclic dimeric GMP(c-di-GMP)has emerged as the nucleotide second messenger regulating both development and antibiotic production in high-GC,Gram-positive streptomycetes.Here,a diguanylate cyclase(DGC),CdgD,encoded by SCO5345 from the model strain Streptomyces coelicolor,was functionally identified and characterized to be involved in c-di-GMP synthesis through genetic and biochemical analysis.cdgD overexpression resulted in significantly reduced production of actinorhodin and undecylprodigiosin,as well as completely blocked sporulation or aerial mycelium formation on two different solid media.In the cdgD-overexpression strain,intracellular c-di-GMP levels were 13-27-fold higher than those in the wild-type strain.In vitro enzymatic assay demonstrated that CdgD acts as a DGC,which could efficiently catalyze the synthesis of c-di-GMP from two GTP molecules.Heterologous overproduction of cdgD in two industrial Streptomyces strains could similarly impair developmental transitions as well as antibiotic biosynthesis.Collectively,our results combined with previously reported data clearly demonstrated that c-di-GMP-mediated signalling pathway plays a central and universal role in the life cycle as well as secondary metabolism in streptomycetes.Xiaocao Liu Guosong Zheng Gang Wang Weihong Jiang Lei Li Yinhua Lu 2019Science China(Life Sciences)2019,62,11:2
19Multiplex genome editing using a dCas9-cytidine deaminase fusion in Streptomyces显示文摘CRISPR/Cas-mediated genome editing has greatly facilitated the study of gene function in Streptomyces. However, it could not be efficiently employed in streptomycetes with low homologous recombination(HR) ability. Here, a deaminase-assisted base editor d Cas9-CDA-ULstr was developed in Streptomyces, which comprises the nuclease-deficient Cas9(dCas9), the cytidine deaminase from Petromyzon marinus(PmCDA1), the uracil DNA glycosylase inhibitor(UGI) and the protein degradation tag(LVA tag). Using d Cas9-CDA-ULstr , we achieved single-, double-and triple-point mutations(cytosine-to-thymine substitutions)at target sites in Streptomyces coelicolor with efficiency up to 100%, 60% and 20%, respectively. This base editor was also demonstrated to be highly efficient for base editing in the industrial strain, Streptomyces rapamycinicus, which produces the immunosuppressive agent rapamycin. Compared with base editors derived from the cytidine deaminase rAPOBEC1, the PmCDA1-assisted base editor dCas9-CDA-ULstr could edit cytosines preceded by guanosines with high efficiency, which is a great advantage for editing Streptomyces genomes(with high GC content). Collectively, the base editor dCas9-CDA-ULstr could be employed for efficient multiplex genome editing in Streptomyces.
