1Deubiquitinase JOSD2 stabilizes YAP/TAZ to promote cholangiocarcinoma progression显示文摘Cholangiocarcinoma(CCA)has emerged as an intractable cancer with scanty therapeutic regimens.The aberrant activation of Yes-associated protein(YAP)and transcriptional co-activator with PDZ-binding motif(TAZ)are reported to be common in CCA patients.However,the underpinning mechanism remains poorly understood.Deubiquitinase(DUB)is regarded as a main orchestrator in maintaining protein homeostasis.Here,we identified Josephin domain-containing protein 2(JOSD2)as an essential DUB of YAP/TAZ that sustained the protein level through cleavage of polyubiquitin chains in a deubiquitinase activity-dependent manner.The depletion of JOSD2 promoted YAP/TAZ proteasomal degradation and significantly impeded CCA proliferation in vitro and in vivo.Further analysis has highlighted the positive correlation between JOSD2 and YAP abundance in CCA patient samples.Collectively,this study uncovers the regulatory effects of JOSD2 on YAP/TAZ protein stabilities and profiles its contribution in CCA malignant progression,which may provide a potential intervention target for YAP/TAZ-related CCA patients.Meijia Qian Fangjie Yan Weihua Wang Jiamin Du Tao Yuan Ruilin Wu Chenxi Zhao Jiao Wang Jiabin Lu Bo Zhang Nengming Lin Xin Dong Xiaoyang Dai Xiaowu Dong Bo Yang Hong Zhu Qiaojun He 2021Acta Pharmaceutica Sinica B2021,11,12:5
2Cesium-rubidium mineralization in Himalayan leucogranites显示文摘This paper presents a study of a newly discovered pollucite-lepidolite-albite granite(PLAG)in the Himalayan leucogranite belt,which marks the first occurrence of pollucite,a major cesium silicate mineral,in the Himalayan orogenic belt(China).The rock appears at the northern part of the Gyirong pluton,coexisting with the tourmaline-bearing two-mica granite(TMG).Primary rare-metal minerals include lepidolite(Li),spodumene(Li),pollucite(Cs),cassiterite(Sn),and microlite(Ta).Micas,mainly lithian muscovite to lepidolite,contain 4.07 wt.%Li_2O and 0.76 wt.%Rb_2O on average.The average Li_2O content of the spodumene is 7.95 wt.%.Pollucite not only has an average Cs_2O content of 34 wt.%,but also has an elevated Rb_2O content of about 0.16 wt.%.Notably,this granite attains industrial grades for rare metals,specifically with Li_2O,Rb_2O,and Cs_2O contents of 0.49–1.19 wt.%,0.12–0.24 wt.%,and 0.69–2.33 wt.%,respectively.Dating results of magmatic accessory cassiterite and monazite indicated that the PLAG was formed at 19–18 Ma,slightly later than the TMG(22–20 Ma)of the Gyirong pluton.Thus,these two types of granites may form within the same magmatic system considering their pulsating intrusive contact,formation ages,and whole-rock and mineral chemical compositions.Furthermore,the abundant presence of pollucite suggests that the PLAG experienced high degrees of magmatic fractionation.In comparison to the Pusila spodumene pegmatite in the Himalaya and the Yashan topaz-lepidolite granite in Jiangxi,South China,the Gyirong PLAG exhibits different whole-rock and mineral compositions,resulting from differences in source materials and fractionation processes.Notably,the difference in fluorine(F)content may determine the degree of rare-metal element enrichment.The discovery of Gyirong PLAG highlights multiple stages and types of rare-metal mineralization in the Himalayan leucogranite belt,which is controlled by the South Tibetan Detachment System.The Cs-bearing geyserite deposit exposed along the Yarlung-Zangbo River,together with Himalayan leucogranites,constitutes two systems of rare-metal elements migration and enrichment.These two systems reflect the interaction among Earth systems across time and space,emphasizing how the Himalayan orogeny controls mineralization.As a result,the Himalayan leucogranite belt has considerable prospecting potential for cesium and rubidium resources and may be a crucial area for future exploration and resource utilization.Fangyang HU Xiaochi LIU Shaoxiong HE Jiamin WANG Fuyuan WU 2023Science China Earth Sciences2023,66,12:3
3Targeting slug-mediated non-canonical activation of c-Met to overcome chemo-resistance in metastatic ovarian cancer cells显示文摘Metastasis-associated drug resistance accounts for high mortality in ovarian cancer and remains to be a major barrier for effective treatment. In this study, SKOV3/T4, a metastatic subpopulation of ovarian cancer SKOV3 cells, was enriched to explore potential interventions against metastaticassociated drug resistance. Quantitative genomic and functional analyses were performed and found that slug was significantly increased in the SKOV3/T4 subpopulation and contributed to the high resistance of SKOV3/T4. Further studies showed that slug activated c-Met in a ligand-independent manner due to elevated levels of fibronectin and provoked integrin α V function, which was confirmed by the significant correlation of slug and p-Met levels in 121 ovarian cancer patient samples. Intriguingly,c-Met inhibitor(s) exhibited greatly enhanced anti-cancer effects in slug-positive ovarian cancer models both in vitro and in vivo. Additionally, IHC analyses revealed that slug levels were highly correlated with reduced survival of ovarian cancer patients. 4Characteristics of oral methicillin-resistant Staphylococcus epidermidis isolated from dental plaque显示文摘The oral microbial community is widely regarded as a latent reservoir of antibiotic resistance genes. This study assessed the molecular epidemiology, susceptibility profile, and resistance mechanisms of 35 methicillin-resistant Staphylococcus epidermidis(MRSE) strains isolated from the dental plaque of a healthy human population. Broth microdilution minimum inhibitory concentrations(MICs) revealed that all the isolates were nonsusceptible to oxacillin and penicillin G. Most of them were also resistant to trimethoprim(65.7%) and erythromycin(54.3%). The resistance to multiple antibiotics was found to be largely due to the acquisition of plasmid-borne genes. The mec A and dfr A genes were found in all the isolates, mostly dfr G(80%), aac A-aph D(20%), aad D(28.6%), aph A3(22.9%), msr A(5.7%), and the erm C gene(14.3%). Classical mutational mechanisms found in these isolates were mainly efflux pumps such as qac A(31.4%), qac C(25.7%), tet K(17.1%), and nor A(8.6%). Multilocus sequence type analysis revealed that sequence type 59(ST59) strains comprised 71.43% of the typed isolates, and the e BURST algorithm clustered STs into the clonal complex 2-II(CC2-II). The staphyloccoccal cassette chromosome mec(SCCmec) type results showed that 25(71.43%) were assigned to type IV. Moreover, 88.66% of the isolates were found to harbor six or more biofilm-associated genes. The aap, atl E, embp, sdr F, and IS256 genes were detected in all 35 isolates. 5Stoichiometric effect on electrical and near-infrared photodetection properties of full-composition-range GaAs1−xSbx nanowires显示文摘As one of the most important narrow bandgap ternary semiconductors, GaAs1−xSbx nanowires (NWs) have attracted extensive attention recently, due to the superior hole mobility and the tunable bandgap, which covers the whole near-infrared (NIR) region, for technological applications in next-generation high-performance electronics and NIR photodetection. However, it is still a challenge to the synthesis of high-quality GaAs1−xSbx NWs across the entire range of composition, resulting in the lack of correlation investigation among stoichiometry, microstructure, electronics, and NIR photodetection. Here, we demonstrate the success growth of high-quality GaAs1−xSbx NWs with full composition range by adopting a simple and low-cost surfactant-assisted solid source chemical vapor deposition method. All of the as-prepared NWs are uniform, smooth, and straight, without any phase segregation in all stoichiometric compositions. The lattice constants of each NW composition have been well correlated with the chemical stoichiometry and confirmed by high-resolution transmission electron microscopy, X-ray diffraction, and Raman spectrum. Moreover, with the increase of Sb concentration, the hole mobility of the as-fabricated field-effect-transistors and the responsivity and detectivity of the as-fabricated NIR photodetectors increase accordingly. 6Unsupervised content-preserving transformation for optical microscopy显示文摘The development of deep learning and open access to a substantial collection of imaging data together provide a potential solution for computational image transformation,which is gradually changing the landscape of optical imaging and biomedical research.However,current implementations of deep learning usually operate in a supervised manner,and their reliance on laborious and error-prone data annotation procedures remains a barrier to more general applicability.Here,we propose an unsupervised image transformation to facilitate the utilization of deep learning for optical microscopy,even in some cases in which supervised models cannot be applied.Through the introduction of a saliency constraint,the unsupervised model,named Unsupervised content-preserving Transformation for Optical Microscopy(UTOM);can learn the mapping between two image domains without requiring paired training data while avoiding distortions of the image content.UTOM shows promising performance in a wide range of biomedical image transformation tasks,including in silico histological staining,fluorescence image restoration,and virtual fluorescence labeling.Quantitative evaluations reveal that UTOM achieves stable and high-fidelity image transformations across different imaging conditions and modalities.We anticipate that our framework will encourage a paradigm shift in training neural networks and enable more applications of artificial intelligence in biomedical imaging.Xinyang Li Guoxun Zhang Hui Qiao Feng Bao Yue Deng Jiamin Wu Yangfan He Jingping Yun Xing Lin Hao Xie Haoqian Wang Qionghai Dai 2021Light(Science & Applications)2021,10,3:1

11Endoplasmic reticulum stress is involved in retinal injury induced by repeated transient spikes of intraocular pressure显示文摘Clinically,a large proportion of glaucoma patients undergo repeated intraocular pressure(IOP)spike(Spike IOP)attacks during their sleep,which may facilitate retinopathy.In this study,we established a mouse model of repeated transient Spike IOP to investigate the direct damage to the retina following Spike IOP attacks,and elucidated the underlying molecular mechanism.We analyzed the changes in the number of retinal ganglion cells(RGCs)via immunofluorescence.Thereafter,we detected retinal cell apoptosis via terminal deoxynucleotidyl transferase deoxyuridine triphosphate(d UTP)nick-end labeling(TUNEL)staining,and performed RNA sequencing(RNA-seq)to reveal the underlying molecular mechanism.Finally,we validated the expression of key molecules in the endoplasmic reticulum(ER)stress pathway using quantitative real-time polymerase chain reaction(q RT-PCR)and western blot analysis.Results revealed a time-dependent RGC loss in Spike IOP,evidenced by a reduction in the number of Brn3 a-positive RGCs in experimental eyes following a 7-d continuous treatment with Spike IOP.In addition,TUNEL staining indicated that apoptosis of retinal cells started in the outer nuclear layer(ONL),and then spread to the ganglion cell layer(GCL)with time.RNA-seq analysis revealed that ER stress might be involved in Spike IOP-induced retinal injury.This result was corroborated by western blot,which revealed upregulation of ER stress-related proteins including binding immunoglobulin protein/glucose-regulated protein 78(Bi P/GRP78),phosphorylated inositolrequiring enzyme 1(p-IRE1),unspliced X-box-binding protein 1(XBP1-u),spliced X-box-binding protein 1(XBP1-s),phosphorylated c-Jun N-terminal kinase(p-JNK),C/EBP-homologous protein(CHOP),and B-cell lymphoma 2(Bcl-2)-associated X protein(Bax).These findings indicate that repeated IOP transients are detrimental to the retina,while ER stress plays an important role in retinal cell apoptosis in this situation.Notably,repeated Spike IOP among glaucoma patients is a crucial factor for progressive retinopathy.Xue YANG Xiaowei YU Zhenni ZHAO Yuqing HE Jiamin ZHANG Xiaoqian SU Nannan SUN Zhigang FAN 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,9:0
12Multifunctional polyeugenol-based nanoparticles with antioxidant and antibacterial properties显示文摘Over the past few decades,extensive scientific has been dedicated to polymer synthesis employing renewable resources.In this study,we devised and synthesized multifunctional polyeugenol-based nanoparticles,exhibiting remarkable antioxidant and antibacterial properties.The grafting of eugenol onto the nanoparticle surface was achieved via a thiol-ene chemical reaction with a grafting rate of 3.5%.To comprehend the properties of the synthesized nanoparticles,we employed Fourier transform infrared spectroscopy,X-ray photoelectron spectroscopy and pyrolysis-gas chromatography mass spectrometry.Subsequent scanning electron microscopic and transmission electron microscopic analyses revealed the presence of a crosslinked structure within the polyeugenol-based nanoparticles,as well as a heteroge-neous microsphere structure on the surface.Due to the inherent crosslinking structure,the polyeugenol-based nanoparticles demonstrated robust the resistance to solvents,as ascertained through thermog-ravimetric analysis and solvent resistance tests.Notably,nitrogen adsorption/desorption studies confirmed the adsorption capacity of the polyeugenol-based nanoparticles,rendering them potentially suitable for drug transport applications.Moreover,the assays for assessing cytocompatibility and reactive oxygen species scavenging activities demonstrated better performance of the eugenol-based nano-particles compared to eugenol.Furthermore,the polyeugenol-based nanoparticles exhibited certain bactericidal activities against Escherichia coli,Staphylococcus aureus,and Fusobacterium nucleatum.Consequently,these observations indicated the nontoxic nature and expansive application prospects of the polyeugenol-based nanoparticles in the domains of medicine and food preservation.This work presented a pioneering concept for the development of antioxidant and antibacterial multifunctional polymer materials derived from eugenol.Qiaoqiao Lan Xuying Zhang Jiaming Liang Wei Wang Leili Lv Jiamin He Zefeng Wang Chunsheng Qu 2024Particuology2024,87,4:0
13A deep learning based fine-grained classification algorithm for grading of visual impairment in cataract patients显示文摘Recent advancements in artificial intelligence(AI)have shown promising potential for the automated screening and grading of cataracts.However,the different types of visual impairment caused by cataracts exhibit similar phenotypes,posing significant challenges for accurately assessing the severity of visual impairment.To address this issue,we propose a dense convolution combined with attention mechanism and multi-level classifier(DAMC_Net)for visual impairment grading.First,the double-attention mechanism is utilized to enable the DAMC_Net to focus on lesions-related regions.Then,a hierarchical multi-level classifier is constructed to enhance the recognition ability in distinguishing the severities of visual impairment,while maintaining a better screening rate for normal samples.In addition,a cost-sensitive method is applied to address the problem of higher false-negative rate caused by the imbalanced dataset.Experimental results demonstrated that the DAMC_Net outperformed ResNet50 and dense convolutional network 121(DenseNet121)models,with sensitivity improvements of 6.0%and 3.4%on the category of mild visual impairment caused by cataracts(MVICC),and 2.1%and 4.3%on the category of moderate to severe visual impairment caused by cataracts(MSVICC),respectively.The comparable performance on two external test datasets was achieved,further verifying the effectiveness and generalizability of the DAMC_Net.JIANG Jiewei ZHANG Yi XIE He YANG Jingshi GONG Jiamin LI Zhongwen 2024Optoelectronics Letters2024,20,1:0

16Generation and evolution of bright-dark pulses in a thulium/holmium co-doped fiber laser based on nonlinear polarization rotation显示文摘We report an experimental phenomenon of the splitting and merging of the bright-dark pulses(BDPs)in a mode-locked thulium/holmium co-doped fiber laser(THDFL)based on nonlinear polarization rotation(NPR).By adding 100-m-long highly nonlinear fiber(HNLF)into a simple ring cavity,the BDPs can be generated.The time interval between the bright and dark pulses increases linearly with the pump power and approximately equals to the inverse of the modulation frequency of the radio frequency(RF)spectrum.Apart from that,the bright and dark pulses are shown not to be orthogonally polarized.The obtained results are valuable for the evolutionary mechanism of the BDPs in passively mode-locked fiber lasers(PMLFLs).WANG Xiaofa HE Yiping PAN Jiamin 2023Optoelectronics Letters2023,19,12:0

18Observation of an acoustic non-Hermitian topological Anderson insulator显示文摘The interaction of band topology and disorder can give rise to intriguing phenomena.One paradigmatic example is the topological Anderson insulator,whose nontrivial topology is induced in a trivial system by disorders.In this study,we investigate the efect of purely non-Hermitian disorders on topological systems using a one-dimensional acoustic lattice with coupled resonators.Specifically,we construct a theoretical framework to describe the non-Hermitian topological Anderson insulator phase solely driven by disordered loss modulation.Then,the complete evolution of non-Hermitian disorder-induced topological phase transitions,from an initial trivial phase to a topological Anderson phase and finally to a trivial Anderson phase,is revealed experimentally using both bulk and edge spectra.Interestingly,topological modes induced by non-Hermitian disorders to be immune to both weak Hermitian and non-Hermitian disorders.These findings pave the way for future research on disordered non-Hermitian systems for novel wave manipulation.Zhongming Gu He Gao Haoran Xue Di Wang Jiamin Guo Zhongqing Su Baile Zhang Jie Zhu 2023Science China(Physics,Mechanics & Astronomy)2023,66,9:0
19Regulating nitrogen vacancies within graphitic carbon nitride to boost photocatalytic hydrogen peroxide production显示文摘Introducing nitrogen vacancies is an effective method to improve the catalytic performance of g-C_(3)N_(4)-based photocatalysts,whereas understanding how nitrogen vacancies types affect the catalytic performance remains unclear.Herein,two different types of nitrogen vacancies were successfully introduced into g-C_(3)N_(4)by pyrolysis of melamine under argon and ammonia atmosphere with subsequent HNO3 oxidation.The pyrolysis atmosphere is found to have a significant influence on the introduced nitrogen vacancies type,where tertiary nitrogen groups(N_(3)C)and sp2-hybridized nitrogen atoms(N_(2)C)were the preferred sites for the formation of nitrogen vacancies under ammonia and argon pyrolysis,respectively.Moreover,nitrogen vacancies from N3C are experimentally and theoretically demonstrated to facilitate the narrowed band gap and the improved oxygen absorption capability.As expected,the optimal catalyst exhibits high H_(2)O_(2)yield of 451.8µM,which is 3.8 times higher than the pristine g-C_(3)N_(4)(119.0µM)after 4 h and good stability after10 photocatalytic runs.Yanlin Zhu Xuetao Liu Heng Liu Guangling He Jiamin Xiao Haijiao Xie Yanyan Sun Lei Han 2022SusMat2022,2,5:0
20Josephin domain containing 2(JOSD2)promotes lung cancer by inhibiting LKB1(Liver kinase B1)activity显示文摘Non-small cell lung cancer(NSCLC)ranks as one of the leading causes of cancer-related deaths worldwide.Despite the prominence and effectiveness of kinase-target therapies in NSCLC treatment,these drugs are suitable for and beneficial to a mere~30%of NSCLC patients.Consequently,the need for novel strategies addressing NSCLC remains pressing.Deubiquitinases(DUBs),a group of diverse enzymes with well-defined catalytic sites that are frequently overactivated in cancers and associated with tumorigenesis and regarded as promising therapeutic targets.Nevertheless,the mechanisms by which DUBs promote NSCLC remain poorly understood.Through a global analysis of the 97 DUBs’contribution to NSCLC survival possibilities using The Cancer Genome Atlas(TCGA)database,we found that high expression of Josephin Domain-containing protein 2(JOSD2)predicted the poor prognosis of patients.Depletion of JOSD2 significantly impeded NSCLC growth in both cell/patient-derived xenografts in vivo.Mechanically,we found that JOSD2 restricts the kinase activity of LKB1,an important tumor suppressor generally inactivated in NSCLC,by removing K6-linked polyubiquitination,an action vital for maintaining the integrity of the LKB1-STRAD-MO25 complex.Notably,we identified the first small-molecule inhibitor of JOSD2,and observed that its pharmacological inhibition significantly arrested NSCLC proliferation in vitro/in vivo.Our findings highlight the vital role of JOSD2 in hindering LKB1 activity,underscoring the therapeutic potential of targeting JOSD2 in NSCLC,especially in those with inactivated LKB1,and presenting its inhibitors as a promising strategy for NSCLC treatment.Tao Yuan Chenming Zeng Jiawei Liu Chenxi Zhao Fujing Ge Yuekang Li Meijia Qian Jiamin Du Weihua Wang Yonghao Li Yue Liu Xiaoyang Dai Jianya Zhou Xueqin Chen Shenglin Ma Hong Zhu Qiaojun He Bo Yang 2024Signal Transduction and Targeted Therapy2024,9,2:0
