Inhibitive effect of triptolide on invasiveness of human fibrosarcoma cells by downregulating matrix metalloproteinase-9 expression显示文摘Objective:To explore the molecular mechanisms of antitumor properties of triptolide,a bioactive component isolated from the Chinese herb Tripterygium wolfordii Hook F.Methods:Human fibrosarcoma HT-1080 cells were treated with different doses of triptolide for 72 h.Then the expression and activity of matrix metalloproteinase(MMP)-2 and -9 were measured and the invasiveness of triptolide-treated HT-1080 cells was compared with that of anti-MMP-9- treated HT-1080 cells.Results:18 nmol/L triptolide inhibited the gene expression and activity of MMP-9,but not those of MMP-2,in HT-1080 cells. In addition, both 18 nmol/L triptolide and 3μg/mL anti-MMP-9 significantly reduced the invasive potential of HT-1080 cells,by about 50%and 35%, respectively, compared with the control.Whereas there was no significant difference between the effect of 18 nmol/L triptolide and that of anti-MMP-9 on invasive potential of HT-1080 cells. Conclusions:These data suggest that triptolide inhibits tumor cell invasion partly by reducing MMP-9 gene expression and activity. | Shengbo Yang Can Gu Guiying Zhang Jian Kang Haiquan Wen QianJin Lu Jinhua Huang | 2011 | Asian Pacific Journal of Tropical Medicine2011,4,6: | 3

Modular Extremely Large-Scale Array Communication:Near-Field Modelling and Performance Analysis显示文摘This paper investigates the wireless communication with a novel architecture of antenna arrays, termed modular extremely large-scale array(XLarray),where array elements of an extremely large number/size are regularly mounted on a shared platform with both horizontally and vertically interlaced modules. Each module consists of a moderate/flexible number of array elements with the inter-element distance typically in the order of the signal wavelength, while different modules are separated by the relatively large inter-module distance for convenience of practical deployment.By accurately modelling the signal amplitudes and phases,as well as projected apertures across all modular elements,we analyse the near-field signal-to-noise ratio(SNR)performance for modular XL-array communications.Based on the non-uniform spherical wave(NUSW)modelling, the closed-form SNR expression is derived in terms of key system parameters, such as the overall modular array size, distances of adjacent modules along all dimensions, and the user's three-dimensional(3D)location.In addition, with the number of modules in different dimensions increasing infinitely, the asymptotic SNR scaling laws are revealed.Furthermore,we show that our proposed near-field modelling and performance analysis include the results for existing array architectures/modelling as special cases,e.g.,the collocated XL-array architecture, the uniform plane wave(UPW)based far-field modelling, and the modular extremely large-scale uniform linear array(XL-ULA)of onedimension.Extensive simulation results are presented to validate our findings. | Xinrui Li Haiquan Lu Yong Zeng Shi Jin Rui Zhang | 2023 | China Communications2023,20,4: | 0
Rational Design of a Near-infrared Fluorescent Material with High Solid-state Efficiency,Aggregation-induced Emission and Live Cell Imaging Property显示文摘Near-infrared(NIR)fluorescent materials with high photoluminescent quantum yields(PLQYs)have wide application prospects. Therefore,we design and synthesize a D-A type NIR organic molecule,TPATHCNE,in which triphenylamine and thiophene are utilized as the donors and fumaronitrile is applied as the acceptor. We systematically investigate its molecular structure and photophysical property.TPATHCNE shows high T_(g) of 110℃ and T_(d) of 385℃ and displays an aggregation-induced emission(AIE)property.A narrow optical bandgap of 1.65 eV is obtained. The non-doped film of TPATHCNE exhibits a high PLQY of 40.3%with an emission peak at 732 nm,which is among the best values of NIR emitters. When TPATHCNE is applied in organic light-emitting diode(OLED),the electroluminescent peak is located at 716 nm with a maximum external quantum efficiency of 0.83%.With the potential in cell imaging, the polystyrene maleic anhydride(PMSA)modified TPATHCNE nanoparticles(NPs)emit strong fluorescence when labeling HeLa cancer cells, suggesting that TPATHCNE can be used as a fluorescent carrier for specific staining or drug delivery for cellular imaging.TPATHCNE NPs fabricated by bovine serum protein(BSA)are cultivated with mononuclear yeast cells, and the intense intracellular red fluorescence indicates that it can be adopted as a specific stain for imaging. | SHANG Anqi ZHAO Lele LI Zhenhua CHENG Zhuang JIN Haixu FENG Zijun CHEN Zhijun ZHANG Haiquan LU Ping | 2022 | Chemical Research in Chinese Universities2022,38,6: | 0
Chemotherapy induces breast cancer stem cell enrichment through repression of glutathione S-transferase Mu显示文摘Triple-negative breast cancer(TNBC)has aggressive characteristics as exemplified by the high risk of recurrence, metastasis, and patient mortality.1 Cytotoxic chemotherapy may reduce tumor burden initially, but leave behind chemo-resistant breast cancer stem cells(BCSCs)that lead to tumor recurrence and metastasis.2 Chemotherapy induces BCsC enrichment through the activation of glutathione biosynthesis pathways, which increases intracellular glutathione levels and specifies the BCSC phenotype through complicated downstream signaling pathways.3 Glutathione S-transferases(GSTs),a superfamily(which contains seven cytosolic classes:Alpha,Kappa,Mu,Omega,Pi,Theta, and Zeta, and one microsomal class MGSTs)of phase Il enzymes that catalyze the conjugation of glutathione with electrophilic compounds, play a critical role in detoxification and chemotherapy resistance of cancer cells. However, the role of GSTs in the regulation of BCSCs is largely elusive. | Jing He Yiran Yu Yilin He Jie He Guangyu Ji Haiquan Lu | 2024 | Genes & Diseases2024,11,2: | 0
Targeting hypoxia-induced CBS expression inhibits breast cancer stem cells through the induction of ferroptosis显示文摘Triple-negative breast cancer(TNBC)has a poor prognosis because of its aggressive characteristics and lack of targeted therapies.1 Recently,it has been reported that TNBC is sensitiveto ferroptosis,an iron-dependent type of programmed cell death, making it a potential target for the treatment of TNBC.2 Breast cancer stem cells(BCSCs),a small population of cancer cells that possess the infinite proliferative potential and tumor-initiating properties, play important roles in drug resistance.3 The responses of BCsCs to ferroptosis-inducing agents are controversial and remain elusive. | Guangyao Wei Jia Liu Jie Lan Guangyu Ji Huize Xia Zhiqun Zhao Zhaoxue Yu Rong Sun Chuanzhao Zhang Haiquan Lu | 2023 | Genes & Diseases2023,10,6: | 0
