| 1 | Anti-SARS-CoV-2 activities in vitro of Shuanghuanglian preparations and bioactive ingredients显示文摘Human infection with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)causes coronavirus disease 2019(COVID-19)and there is no cure currently.The 3CL protease(3CLpro)is a highly conserved protease which is indispensable for CoVs replication,and is a promising target for development of broad-spectrum antiviral drugs.In this study we investigated the anti-SARS-CoV-2 potential of Shuanghuanglian preparation,a Chinese traditional patent medicine with a long history for treating respiratory tract infection in China.We showed that either the oral liquid of Shuanghuanglian,the lyophilized powder of Shuanghuanglian for injection or their bioactive components dose-dependently inhibited SARS-CoV-23CLpro as well as the replication of SARS-CoV-2 in Vero E6 cells.Baicalin and baicalein,two ingredients of Shuanghuanglian,were characterized as the first noncovalent,nonpeptidomimetic inhibitors of SARS-CoV-23CLpro and exhibited potent antiviral activities in a cell-based system.Remarkably,the binding mode of baicalein with SARS-CoV-23CLpro determined by X-ray protein crystallography was distinctly different from those of known 3CLpro inhibitors.Baicalein was productively ensconced in the core of the substrate-binding pocket by interacting with two catalytic residues,the crucial S1/S2 subsites and the oxyanion loop,acting as a“shield”in front of the catalytic dyad to effectively prevent substrate access to the catalytic dyad within the active site.Overall,this study provides an example for exploring the in vitro potency of Chinese traditional patent medicines and effectively identifying bioactive ingredients toward a specific target,and gains evidence supporting the in vivo studies of Shuanghuanglian oral liquid as well as two natural products for COVID-19 treatment. | Hai-xia Su Sheng Yao Wen-feng Zhao Min-jun Li Jia Liu Wei-juan Shang Hang Xie Chang-qiang Ke Hang-chen Hu Mei-na Gao Kun-qian Yu Hong Liu Jing-shan Shen Wei Tang Lei-ke Zhang Geng-fu Xiao Li Ni Dao-wen Wang Jian-ping Zuo Hua-liang Jiang Fang Bai Yan Wu Yang Ye Ye-chun Xu | 2020 | Acta Pharmacologica Sinica2020,41,9: | 32 |

| 3 | Isolation and characterization of dengue virus serotype 2 from the large dengue outbreak in Guangdong, China in 2014显示文摘Dengue has been well recognized as a global public health threat,but only sporadic epidemics and imported cases were reported in recent decades in China.Since July 2014,an unexpected large dengue outbreak has occurred in Guangdong province,China,resulting in more than 40000 patients including six deaths.To clarify and characterize the causative agent of this outbreak,the acute phase serum from a patient diagnosed with severe dengue was subjected to virus isolation and high-throughput sequencing(HTS).Traditional real-time RT-PCR and HTS with Ion Torrent PGM detected the presence of dengue virus serotype 2(DENV-2).A clinical DENV-2 isolate GZ05/2014 was obtained by culturing the patient serum in mosquito C6/36 cells.The complete genome of GZ05/2014 was determined and deposited in Gen Bank under the access number KP012546.Phylogenetic analysis based on the complete envelope gene showed that the newly DENV-2 isolate belonged to Cosmopolitan genotype and clustered closely with other Guangdong strains isolated in the past decade.No amino acid mutations that are obviously known to increase virulence or replication were identified throughout the genome of GZ05/2014.The high homology of Guangdong DENV-2 strains indicated the possibility of establishment of local DENV-2 circulation in Guangdong,China.These results help clarify the origin of this epidemic and predict the future status of dengue in China. | ZHAO Hui ZHAO Ling Zhai JIANG Tao LI Xiao Feng FAN Hang HONG Wen Xin ZHANG Yu ZHU Qin YE Qing TONG Yi Gang CAO Wu Chun ZHANG Fu Chun QIN Cheng Feng | 2014 | Science China(Life Sciences)2014,57,12: | 21 |
| 4 | Variants of the arachidonate 5-lipoxygenase-activating protein (ALOX5AP) gene and risk of ischemic stroke in Han Chinese of eastern China显示文摘Variants of the arachidonate 5-lipoxygenase-activating protein (ALOX5AP) gene have been suggested to play an important role in the pathogenesis of atherosclerosis and ischemic stroke.This study was aimed to explore the association of ALOX5AP variants with ischemic stroke risk in Han Chinese of eastern China.A total of 690 ischemic stroke cases and 767 controls were recruited.The subjects were further subtyped according to the Trial of Org 10172 in Acute Stroke Treatment (TOAST) criteria.On the basis of that,two polymorphisms of the ALOX5AP gene (rs10507391 and rs12429692) were determined by TaqMan genotyping assay.In addition,plasma leukotriene B4 (LTB4) levels were analyzed in these subjects.There was no evidence of association between the two variants of ALOX5AP and the risk of ischemic stroke or its TOAST-subtypes.Haplotype analysis and stratification analysis according to sex,age,body mass index,hypertension,and diabetes also showed negative association.Analysis of LTB4 levels in a subset of cases and controls revealed that LTB4 levels were significantly higher in ischemic stroke cases than in the controls (70.06±14.75 ng/L vs 57.34±10.93 ng/L;P=0.000) and carriers of the T allele of the rs10507391 variant were associated with higher plasma LTB4 levels (P=0.000).The present study suggests there is no association of the two polymorphisms in the ALOX5AP gene with ischemic stroke risk in Han Chinese of eastern China. | Gannan Wang Yao Wang Hao Sun Weijuan Cao Jing Zhang Hang Xiao Jinsong Zhang | 2011 | The Journal of Biomedical Research2011,25,5: | 12 |
| 5 | Strengths, weaknesses, opportunities and threats (SWOT) analysis of reinitiation into methamphetamine abusers: qualitative findings from an exploration of methamphetamine abusers in Shanghai, China显示文摘Background Despite high relapse rate among methamphetamine (MA) abusers, there still have been little empirical data to date detecting the risk factors related to craving and relapse from the perspective of MA abusers. Therefore, the aim of this study was to use qualitative research methods exploring the strengths, weaknesses, opportunities and threat factors that could facilitate or impede individuals' abstinence in a real-life context from the perspectives of MA abusers. Aim To use qualitative research methods exploring the strengths, weaknesses, opportunities and threat factors relating to individuals' abstinence in a real-life context. Method Semistructured interviews were conducted with 32 MA abusers recruited in Shanghai guided by open-ended questions on narrating the real-life catalysts and inhibitors related to craving, initiation, relapse and abstinence. All data were transcribed verbatim and analysed using a strengths, weaknesses, opportunities and threats (SWOT) analysis. Results The results of the SWOT analysis revealed that contextual factors including peer influence, prevalence and availability of MA, familiar venue of MA use, discrimination, sexual behaviours, alcohol, emotional states and their attitudes towards smoking MA were important factors that contribute to reinitiation and relapse. Surveillance systems, antidrug social workers, vocational skills trainings, moving to another city and family responsibility might serve as counter measures targeti叩 those mentioned weaknesses and threats above. Conclusion This SWOT analysis highlights the complex nature of relapse. | 7 | A telomere-to-telomere gap-free reference genome of watermelon and its mutation library provide important resources for gene discovery and breeding显示文摘Watermelon,Citrullus lanatus,is the world's third largest fruit crop.Reference genomes with gaps and a narrow genetic base hinder functional genomics and genetic improvement of watermelon.Here,we report the assembly of a telomere-to-telomere gap-free genome of the elite watermelon inbred line G42 by incorporating high-coverage and accurate long-read sequencing data with multiple assembly strategies.All 11 chromosomes have been assembled into single-contig pseudomolecules without gaps,representing the highest completeness and assembly quality to date.The G42 reference genome is 369321829 bp in length and contains 24205 predicted protein-coding genes,with all 22 telomeres and 11 centromeres characterized.Furthermore,we established a pollen-EMS mutagenesis protocol and obtained over 200000M1 seeds from G42.In a sampling pool,48 monogenic phenotypic mutations,selected from 223M1and 78 M2 mutants with morphological changes,were confirmed.The average mutation density was 1 SNP/1.69Mband1 indel/4.55 Mb per M1 plant and 1SNP/1.08Mb and 1 indel/6.25 Mb per M2 plant.Taking advantage of the gap-free G42 genome,8039 mutations from 32 plants sampled from M1 and M2 families were identified with 100%accuracy,whereas only 25% of the randomly selected mutations identified using the 97103v2 reference genome could be confirmed.Using this library and the gap-free genome,two genes responsible for elongated fruit shape and male sterility(CiMs1)were identified,both caused by a single basechange from G to A.The validated gap-free genome and its EMS mutation library provide invaluable resources for functional genomics and genetic improvement of watermelon. | Yun Deng Shoucheng Liu Yilin Zhang Jingsheng Tan Xiaopeng Li Xiao Chu Binghua Xu Yao Tian Yudong Sun Bosheng Li YunbiXu Xing Wang Deng Hang He Xingping Zhang | 2022 | Molecular Plant2022,15,8: | 9 |
| 8 | Association between Plasma Metal Levels and Diabetes Risk: a Case-control Study in China显示文摘Objective Many metals, some of which have been classified as environmental endocrine disruptors, are used extensively in everyday consumer products and are ubiquitous in our living environment. In the present study, we aimed to explore the associations between the prevalence risk of type 2 diabetes and plasma levels of 20 trace elements as well as those of heavy metals in a Han Chinese population. Methods We conducted a case-control study to investigate the associations between plasma concentrations of 20 metals and diabetes in Jiangsu province. A total of 122 newly diagnosed cases of type 2 diabetes and 429 matched controls were recruited from community physical examinations in Suzhou City of Jiangsu Province. Plasma metal levels were measured by inductively-coupled plasma mass spectrometry. Results After adjusting for confounders, plasma vanadium, chromium, manganese, copper, zinc, arsenic, selenium, strontium, palladium, cadmium, cesium, and barium were associated with diabetes risk(P < 0.05). The adjusted OR increased with increasing concentration of vanadium, manganese, copper, zinc, and cesium. Conclusion Many metals, including manganese, copper, zinc, arsenic, selenium, and cadmium in plasma, are associated with the morbidity of diabetes. | 9 | Aerobic granules cultivated and operated in continuous-flow bioreactor under particle-size selective pressure显示文摘A novel method based on the selective pressure of particle size(particle-size cultivation method, PSCM) was developed for the cultivation and operation of aerobic granular sludge in a continuous-flow reactor, and compared with the conventional method based on the selective pressure of settling velocity(settling-velocity cultivation method, SVCM). Results indicated that aerobic granules could be cultivated in continuous operation mode by this developed method within 14 days. Although in the granulation process, under particle-size selective pressure, mixed liquor suspended solids(MLSS) in the reactor fluctuated greatly and filamentous bacteria dominated the sludge system during the initial operation days, no obvious difference in profile was found between the aerobic granules cultivated by PSCM and SVCM. Moreover, aerobic granules cultivated by PSCM presented larger diameter, lower water content and higher specific rates of nitrification, denitrification and phosphorus removal, but lower settling velocity. Under long term operation of more than 30 days, aerobic granules in the continuous-flow reactor could remain stable and obtain good chemical oxygen demand(COD), NH4+-N, total nitrogen(TN) and total phosphorus(TP) removal. | 13 | CFD study on double-to single-loop flow pattern transition and its influence on macro mixing efficiency in fully baffled tank stirred by a Rushton turbine显示文摘For a fully baffled tank stirred by a Rushton turbine (RT), the flow pattern will change from double- to single-loop as the off bottom clearance (C) of the RT decreases from one third of the tank diameter. Such a flow pattern transition as well as its influence on the macro mixing efficiency was investigated via CFD simulation. The transient sliding mesh approach coupled with the standard k-s turbulence model could correctly and efficiently reproduce the reported critical C range where the flow pattern changes. Simulation results indicated that such a critical C range varied hardly with the impeller rotation speed but decreased significantly with increasing impeller diameter. Small RTs are preferable to generating the single-loop flow pattern. A mechanism of the flow pattern transition was further proposed to explain these phe no mena. The discharge stream from the RT deviates down wards from the horizontal direction for small C values;if it meets the tank wall first, the double-loop will form;if it hits the tank bottom first, the single-loop will form. With the flow pattern transition, the mixing time decreased by about 35% at the same power input (P), indicating that the single-loop flow pattern was more efficient than the double-loop to enhance the macro mixing in the tank. A comparison was further made between the single-loop RT and pitched blade turbine (PBT, 45°) from macro mixing perspective. | 14 | Petrogenesis and tectonic implications of Indosinian granitoids from Western Qinling Orogen,China:Products of magma-mixing and fractionation显示文摘The Western Qinling Orogen(WQO) is characterized by voluminous distribution of Indosinian granitoids,the formation of which provides an important window to unravel the geochemical and geodynamic evolution and associated metallogeny.Here we investigate a group of intrusions termed 'Five Golden Flowers' based on petrological,geochemical,zircon U-Pb geochronological and Lu-Hf isotopic studies on the granitoids and their mafic microgranular enclaves(MMEs).Our results show that these intrusions are genetically divided into two types,namely,magma-mixing and highly fractionated.The Jiaochangba,Lujing,Zhongchuan,and Luchuba granitoids are biotite monzogranites(220±0.8 Ma to 217±2.6 Ma) with abundant coeval MMEs(220±.1 Ma to 217±2.7 Ma).The rocks contain moderate to high SiO2,high MgO,Rb,Sr,Ba,and Th contents,but low TiO2,P2 O5,and Sc values,A/CNK of <1.1,and a range of εHf(t) values of-11.7 to +2.23 with corresponding TDM2values of 1967-1228 Ma.The MMEs possess K-feldspar megacrysts,abundant acicular apatites,and show lopsided textures.They have lower SiO2,Al2 O3,and Th contents,but higher MgO,TiO2,and Sc,with εHf(t) values of-18.0 to +3.18 and TDM1 of 849-720 Ma.The data indicate that the MMEs were derived from a magma sourced from the enriched lithospheric mantle.We suggest that these host granitoids were produced by partial melting of latePaleoproterozoic to early-Mesoproterozoic lower crust with the involvement of Neoproterozoic SCLM-derived mafic magmas.The Baijiazhuang pluton is dominantly composed of leucogranite(muscovite granite and twomica monzogranite,216±1.5 Ma) without MMEs.The rocks are peraluminous with high A/CNK(1.06-1.27).Compared with the other four granitoids,the Baijiazhuang leucogranite shows higher SiO2 content,markedly lower concentrations of TiO2,MgO,Al2 O3,CaO,and Fe2 O3T,and lower LREE/HREE and(La/Yb)N values.These leucogranites are also rich in Rb,Th,and U,and display marked depletions in Ba,Sr,Ti,and Eu,indicating that they experienced significant fractionation.Zircon εHf(t) values(-10.2 to-3.27) and TDM2(1868-1424 Ma),as well as the Nb/Ta and K2 O/Na2 O values are similar to the other four granitoids,indicating that they are likely to have been derived from a similar source;with sediments playing only a minor role in the magma generation.The low contents of Yb and Y suggest that their partial melting was controlled by garnets and micrographic texture of K-feldspar reflects high-temperature melting through undercooling.Based on the above features,we infer that the Baijiazhuang leucogranite likely represents the product of high degree fractionation of the I-type biotite monzogranite magma which generated the other four granitoids at relatively high temperatures,within magma chambers at mid-crust depths.We propose that the granitoid suite was formed in the transitional setting from synto post-collision during the collisional orogeny between the SCB and NCB,following break-off of the subducted South China Block lithosphere during 220-216 Ma. | Xiao Xiong Laimin Zhu Guowei Zhang M.Santosh Hang Jiang Jun Zheng Anlin Guo Lele Ding | 2020 | Geoscience Frontiers2020,11,4: | 6 |

| 17 | Zircon U-Pb Geochronology and Geochemical Characteristics of the Volcanic Host Rocks from the Tongyu VHMS Copper Deposit in the Western North Qinling Orogen and Their Geological Significance显示文摘Precise in situ zircon U-Pb dating and Lu–Hf isotopic measurement using an LA-ICP-MS system, whole-rock major and trace element geochemistry and Sr–Nd isotope geochemistry were conducted on the volcanic host rocks of the Tongyu copper deposit on the basis of further understanding of its geological characteristics. Three zircon samples from the volcanic host rocks yielded 206Pb/238 U weighted average ages ranging from 436±4 Ma to 440±5 Ma, which are statistically indistinguishable and coeval with the ca. 440 Ma northward subduction event of the Paleo-Qinling oceanic slab. The volcanic host rocks were products of magmatic differentiation that evolved from basalt to andesite to dacite to rhyolite, forming an integrated tholeiitic island arc volcanic rock suite. The primitive mantle-normalized trace element patterns for most samples show characteristics of island arc volcanic rocks, such as relative enrichment of LILE(e.g. Th, U, Pb and La) and depletion of HFSE(e.g. Nb, Ta, Ti, Zr and Hf). Discrimination diagrams of Ta/Yb vs Th/Yb, Ta vs Th, Yb vs Th/Ta, Ta/Hf vs Th/Hf, Hf/3 vs Th vs Nb/16, La vs La/Nb and Nb vs Nb/Th all suggest that both the volcanic host rocks from the Tongyu copper deposit and the volcanic rocks from the regional Xieyuguan Group were formed in an island arc environment related to subduction of an oceanic slab. Values of ISr(0.703457 to 0.708218) and εNd(t)(-2 to 5.8) indicate that the source materials of volcanic rocks from the Tongyu copper deposit and the Xieyuguan Group originated from the metasomatised mantle wedge with possible crustal material assimilation. Most of the volcanic rock samples show good agreement with the values of typical island arc volcanic rocks in the ISr-εNd(t) diagram. The involvement of crustal-derived material in the magma of the volcanic rocks from the Tongyu copper deposit was also reflected in the zircon εHf(t) values, which range from-3.08 to 10.7, and the existence of inherited ancient xenocrystic zircon cores(2616±39 Ma and 1297±22 Ma). The mineralization of the Tongyu copper deposit shows syn-volcanic characteristics such as layered orebodies interbedded with the volcanic rock strata, thus, the zircon U-Pb age of the volcanic host rocks can approximately represent the mineralization age of the Tongyu copper deposit. Both the Meigou pluton and the volcanic host rocks were formed during the ca. 440 Ma northward subduction of the Paleo-Qinling Ocean when high oxygen fugacity aqueous hydrothermal fluid released by dehydration of the slab and the overlying sediments fluxed into the mantle wedge, triggered partial melting of the mantle wedge, and activated and extracted Cu and other ore-forming elements. | 19 | Pathogen development and host responses to Plasmopara viticola in resistant and susceptible grapevines:an ultrastructural study显示文摘The downy mildew disease in grapevines is caused by Plasmopara viticola.This disease poses a serious threat wherever viticulture is practiced.Wild Vitis species showing resistance to P.viticola offer a promising pathway to develop new grapevine cultivars resistant to P.viticola which will allow reduced use of environmentally unfriendly fungicides.Here,transmission and scanning microscopy was used to compare the resistance responses to downy mildew of three resistant genotypes of V.davidii var.cyanocarpa,V.piasesezkii and V.pseudoreticulata and the suceptible V.vinifera cultivar‘Pinot Noir’.Following inoculation with sporangia of P.viticola isolate‘YL’on V.vinifera cv.‘Pinot Noir’,the infection was characterized by a rapid spread of intercellular hyphae,a high frequency of haustorium formation within the host’s mesophyll cells,the production of sporangia and by the absence of host-cell necrosis.In contrast zoospores were collapsed in the resistant V.pseudoreticulata‘Baihe-35-1’,or secretions appeared arround stomata at the beginning of the infection period in V.davidii var.cyanocarpa‘Langao-5’and V.piasezkii‘Liuba-8’.The main characteristics of the resistance responses were the rapid depositions of callose and the appearance of empty hyphae and the plasmolysis of penetrated tissue.Moreover,collapsed haustoria were observed in V.davidii var.cyanocarpa‘Langao-5’at 5 days post inoculation(dpi)and in V.piasezkii‘Liuba-8’at 7 dpi.Lastly,necrosis extended beyond the zone of restricted colonization in all three resistant genotypes.Sporangia were absent in V.piasezkii‘Liuba-8’and greatly decreased in V.davidii var.cyanocarpa‘Langao-5’and in V.pseudoreticulata‘Baihe-35-1’compared with in V.vinifera cv.‘Pinot Noir’.Overall,these results provide insights into the cellular biological basis of the incompatible interactions between the pathogen and the host.They indicate a number of several resistant Chinese wild species that could be used in developing new cultivars having good levels of downy mildew resistance. | Xiao Yin Rui-Qi Liu Hang Su Li Su Yu-Rui Guo Zi-Jia Wang Wei Du Mei-Jie Li Xi Zhang Yue-Jin Wang Guo-Tian Liu Yan Xu | 2017 | Horticulture Research2017,4,1: | 5 |
