1Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual.Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang 2020Protein & Cell2020,11,4:44
2Fuzzy logic based coordinated control of battery energy storage system and dispatchable distributed generation for microgrid显示文摘Microgrid is a good option to integrate renewableenergy sources (RES) into power systems. Inorder to deal with the intermittent characteristics of therenewable energy based distributed generation (DG) units,a fuzzy-logic based coordinated control strategy of a batteryenergy storage system (BESS) and dispatchable DGunits is proposed for the microgrid management system(MMS). 3First observation of tropospheric nitrogen dioxide from the Environmental Trace Gases Monitoring Instrument onboard the GaoFen-5 satellite显示文摘The Environmental Trace Gases Monitoring Instrument(EMI)is the first Chinese satellite-borne UV–Vis spectrometer aiming to measure the distribution of atmospheric trace gases on a global scale.The EMI instrument onboard the GaoFen-5 satellite was launched on 9 May 2018.In this paper,we present the tropospheric nitrogen dioxide(NO2)vertical column density(VCD)retrieval algorithm dedicated to EMI measurement.We report the first successful retrieval of tropospheric NO_(2) VCD from the EMI instrument.Our retrieval improved the original EMI NO_(2) prototype algorithm by modifying the settings of the spectral fit and air mass factor calculations to account for the on-orbit instrumental performance changes.The retrieved EMI NO_(2) VCDs generally show good spatiotemporal agreement with the satellite-borne Ozone Monitoring Instrument and TROPOspheric Monitoring Instrument(correlation coefficient R of ~0.9,bias<50%).A comparison with ground-based MAX-DOAS(Multi-Axis Differential Optical Absorption Spectroscopy)observations also shows good correlation with an R of 0.82.The results indicate that the EMI NO_(2) retrieval algorithm derives reliable and precise results,and this algorithm can feasibly produce stable operational products that can contribute to global air pollution monitoring.Chengxin Zhang Cheng Liu Ka Lok Chan Qihou Hu Haoran Liu Bo Li Chengzhi Xing Wei Tan Haijin Zhou Fuqi Si Jianguo Liu 2020Light(Science & Applications)2020,9,1:11
4Ni Flower/MXene-Melamine Foam Derived 3D Magnetic/Conductive Networks for Ultra-Efficient Microwave Absorption and Infrared Stealth显示文摘The development of multifunctional and efficient electromagnetic wave absorbing materials is a challenging research hotspot.Here,the magnetized Ni flower/MXene hybrids are successfully assembled on the surface of melamine foam(MF)through electrostatic self-assembly and dip-coating adsorption process,realizing the integration of microwave absorption,infrared stealth,and flame retardant.Remarkably,the Ni/MXene-MF achieves a minimum reflection loss(RLmin)of−62.7 dB with a corresponding effective absorption bandwidth(EAB)of 6.24 GHz at 2 mm and an EAB of 6.88 GHz at 1.8 mm.Strong electromagnetic wave absorption is attributed to the three-dimensional magnetic/conductive networks,which provided excellent impedance matching,dielectric loss,magnetic loss,interface polarization,and multiple attenuations.In addition,the Ni/MXene-MF endows low density,excellent heat insulation,infrared stealth,and flame-retardant functions.This work provided a new development strategy for the design of multifunctional and efficient electromagnetic wave absorbing materials.Haoran Cheng Yamin Pan Xin Wang Chuntai Liu Changyu Shen Dirk W.Schubert Zhanhu Guo Xianhu Liu 2022Nano-Micro Letters2022,14,4:7
5Effects of Exogenous 5-Aminolevulinic Acid and 24-Epibrassinolide on Cd Accumulation in Rice from Cd-Contaminated Soil显示文摘High grain-Cd-accumulating rice variety Yongyou 9 was planted in Cd-contaminated farmland in Taizhou City, Zhejiang Province, China to study the effects of 5-aminolevulinic acid(ALA) and24-epibrassinolide(EBR) on Cd accumulation in brown rice. Results showed that the exogenous ALA and EBR had no significant effects on agronomic traits, soil pH and total Cd content in soil, but had some effects on the available Cd content in soil, and significantly influenced the Cd accumulation in the different parts of rice. Results also showed that 100 mg/L exogenous ALA significantly reduced the Cd accumulation in brown rice to blow the food safety standard(0.2 mg/kg), and also significantly reduced the Cd contents in the roots and culm of rice. However, 200 mg/L exogenous ALA treatment increased the Cd content in brown rice remarkably. In addition, 0.15 mg/L EBR treatment increased Cd accumulation in roots, culm, leaves and brown rice notably, whereas 0.30 mg/L exogenous EBR treatment reduced the Cd accumulation in brown rice properly, but it was not significant. 6Wafer-scale patterned growth of type-II Dirac semimetal platinum ditelluride for sensitive room-temperature terahertz photodetection显示文摘As the lastly unexplored electromagnetic wave,terahertz(THz)radiation has been exploited in a plenty of contexts such as fundamental research,military and civil fields.Most recently,representative two-dimensional(2D)topological semimetal,platinum ditelluride(PtTe_(2))has attracted considerable research interest in THz detection due to its unique physical properties.However,to achieve practical applications,the low-cost,large-scale,controllable synthesis and efficient patterning of 2D materials are key requirements,which remain a challenge for PtTe_(2)and its photodetectors(PDs).Herein,a facile approach is developed to obtain waferscale(2-inches)patterned PtTe_(2)arrays using one-step tellurium-vapor transformation method and micro-Nano technology.PtTe_(2)PD arrays are fabricated with the as-grown PtTe_(2)arrays evenly distributed on a 2-inch wafer,exhibiting high conductivity(~2.7×105 S m^(-1))and good electrical consistency.Driven by the Dirac fermions,PtTe_(2)PDs achieve a broadband(0.02-0.3 THz)response with a fast response speed(~4.7μs),a high sensitivity(~47 pW Hz^(-1/2))and high-resolution transmission THz-imaging capability,which displays the potential of large-area THz array imaging.These results are one step towards the practical applications of integrated PD arrays based on 2D materials.Zhuo Dong Wenzhi Yu Libo Zhang Liu Yang Luyi Huang Yan Zhang Zeqian Ren Haoran Mu Cheng Chen Junrong Zhang Jie Li Lin Wang Kai Zhang 2023InfoMat2023,5,5:1
7Vertical distribution and temporal evolution of formaldehyde and glyoxal derived from MAX-DOAS observations:The indicative role of VOC sources显示文摘Formaldehyde(HCHO)and glyoxal(CHOCHO)are important oxidization intermediates of most volatile organic compounds(VOCs),but their vertical evolution in urban areas is not well understood.Vertical profiles of HCHO,CHOCHO,and nitrogen dioxide(NO_(2))were retrieved from ground-based Multi-Axis Differential Optical Absorption Spectroscopy(MAXDOAS)observations in Hefei,China.HCHO and CHOCHO vertical profiles prefer to occur at higher altitudes compared to NO_(2),which might be caused by the photochemistry-oxidation of longer-lived VOCs at higher altitudes.Monthly means of HCHO concentrations were higher in summer,while enhanced amounts of NO_(2)were mainly observed in winter.CHOCHO exhibited a hump-like seasonal variation,with higher monthly-averaged values not only occurred in warm months(July-August)but also in cold months(November-December).Peak values mainly occurred during noon for HCHO but emerged in the morning for CHOCHO and NO_(2),suggesting that HCHO is stronger link to photochemistry than CHOCHO.We further use the glyoxal to formaldehyde ratio(GFR)to investigate the VOC sources at different altitudes.The lowest GFR value is almost found in the altitude from 0.2 to 0.4 km,and then rises rapidly as the altitude increases.The GFR results indicate that the largest contributor of the precursor VOC is biogenic VOCs at lower altitudes,while at higher altitudes is anthropogenic VOCs.Our findings provide a lot more insight into VOC sources at vertical direction,but more verification is recommended to be done in the future.Qianqian Hong Cheng Liu Qihou Hu Yanli Zhang Chengzhi Xing Jinping Ou Wei Tan Haoran Liu Xiaoqing Huang Zhenfeng Wu 2022Journal of Environmental Sciences2022,34,12:1
8Programmed death ligand-1 regulates angiogenesis and metastasis by participating in the c-JUN/VEGFR2 signaling axis in ovarian cancer显示文摘Background:Although programmed cell death-ligand 1(PD-L1)plays a wellknown function in immune checkpoint response by interacting with programmed cell death-1(PD-1),the cell-intrinsic role of PD-L1 in tumors is still unclear.Here,we explored the molecular regulatory mechanism of PD-L1 in the progression and metastasis of ovarian cancer.Methods:Immunohistochemistry of benign tissues and ovarian cancer samples was performed,followed by migration,invasion,and angiogenesis assays in PDL1-knockdown ovarian cancer cells.Immunoprecipitation,mass spectrometry,and chromatin immunoprecipitation were conducted along with zebrafish and mouse experiments to explore the specific functions and mechanisms of PD-L1 in ovarian cancer.Results:Our results showed that PD-L1 induced angiogenesis,which further promoted cell migration and invasion in vitro and in vivo of ovarian cancer.Mechanistically,PD-L1 was identified to directly interact with vascular endothelial growth factor receptor-2(VEGFR2)and then activated the FAK/AKT pathway,which further induced angiogenesis and tumor progression,leading to poor prognosis of ovarian cancer patients.Meanwhile,PD-L1 was found to be regulated by the oncogenic transcription factor c-JUN at the transcriptional level,which enhanced the expression of PD-L1 in ovarian cancer.Furthermore,we demonstrated that PD-L1 inhibitor durvalumab,combined with the antiangiogenic drug,apatinib,could enhance the effect of anti-angiogenesis and the inhibition of cell migration and invasion.Conclusion:Our results demonstrated that PD-L1 promoted the angiogenesis and metastasis of ovarian cancer by participating in the c-JUN/VEGFR2 signaling axis,suggesting that the combination of PD-L1 inhibitor and antiangiogenic drugs may be considered as a potential therapeutic approach for ovarian cancer patients.Yufei Yang Lingfang Xia Yong Wu Hongyu Zhou Xin Chen Haoran Li Midie Xu Zihao Qi Ziliang Wang Huizhen Sun Xi Cheng 2021Cancer Communications2021,41,6:1
9Cost-benefit Analysis of Land Development Projects Based on Fuzzy Comprehensive Evaluation Model显示文摘Ecological and environmental issues have always been the focus of government work. The economic losses caused by air pollution,water pollution,noise pollution and solid waste pollution caused by land use development projects are estimated,and the total population economic losses are incorporated into project costing. Using the analytic hierarchy process,four benefit indicators and five cost indicators are selected for constructing a fuzzy comprehensive evaluation model to evaluate the cost-benefit of projects of different scales. The projects of different scales are ranked according to the level of comprehensive scores. The results show that large-scale projects are cost-effective and stable over the long term,and there are differences in the main sources of pollution-caused economic losses for projects of different scales. 11Cloning and characterization of the PtVIP1 gene in Populus显示文摘The VirE2-interaction protein 1(VIP1)serves as a regulator of mitogen-activated protein kinase 3(MPK3)-mediated stress gene modulation under biotic stress,which in turn activates the MPK3 pathway in Arabidopsis.The mode of action of the VIP1 protein in Populus in response to biotic stress remains unknown.In this study,we cloned the full-length cDNA of the PtVIP1 gene from Populus trichocarpa(accession number of GenBank:KY793105).The VIP1 protein harboured a conserved bZIP(basic leucine zipper)domain located in the C-terminus.The VIP1 subcellular localization assay indicated that the VIP1 protein was present in the cytoplasm and nucleus under normal conditions,and that an increase in the amount of the protein in the nucleus occurred after treatment with flg22,the elicitor-active epitope of flagellin which triggers the innate immune response in plants.Transgenic Populus plants overexpressing VIP1 genes(PtVIP1 of Populus;or AtVIP1 of Arabidopsis,as positive control)were generated to investigate the role of VIP1 in vivo.The expression of poplar pathogenesis-related protein 1(PR1)genes was upregulated in transgenic-PtVIP1 or AtVIP1 poplar plants.The transgenic poplar plants overexpressing PtVIP1 or AtVIP1 also showed enhanced resistance to Brenneria salicis infection.These results suggest that the VIP1 protein accumulates in the nucleus in response to biotic stress,and that the pathogen resistance of transgenic VIP1 poplar may be associated with the induced expression of PR1 genes in response to pathogen challenge.Haoran Wang Weitao Zhang Mingxiu Wang Qiang Cheng 2019Journal of Forestry Research2019,30,6:1

13Solution processable transition metal dichalcogenides-based hybrids for photodetection显示文摘Transition metal dichalcogenides(TMDs), as one of the most promising two-dimensional(2D) materials, have attracted considerable attention for use in photodetection applications over the past few years due to their distinct properties, such as atomic-scale thickness, tunable direct bandgaps, and decent carrier mobilities at room temperature. Compared with pure 2D TMDs, the construction of hybrids consisting of TMDs and other low-dimensional materials can further improve the performance of photodetectors including their spectral range, responsivity and detectivity, which significantly boosts interest in the development of TMDs-based photodetectors. On the other hand, solution-phase synthesis methods provide a facile strategy for the scalable production of TMD hybrids, opening an exciting avenue to develop low-cost devices. 15Ground-Based Hyperspectral Stereoscopic Remote Sensing Network: A Promising Strategy to Learn Coordinated Control of O_(3) and PM_(2.5) over China显示文摘With the coming of the“14th Five-Year Plan,”the coordinated control of particulate matter with an aerodynamic diameter no greater than 2.5 lm(PM_(2.5))and O_(3) has become a major issue of air pollution prevention and control in China.The stereoscopic monitoring of regional PM_(2.5) and O_(3) and their precursors is crucial to achieve coordinated control.However,current monitoring networks are currently inadequate for monitoring the vertical profiles of both PM_(2.5) and O_(3) simultaneously and support air quality control.The University of Science and Technology of China(USTC)has established a nationwide ground-based hyperspectral stereoscopic remote sensing network based on multi-axis differential optical absorption spectroscopy(MAX-DOAS)since 2015.This monitoring network provides a significant opportunity for the regional coordinated control of PM_(2.5) and O_(3) in China.One-year vertical profiles of aerosol,NO_(2) and HCHO monitored from four MAX-DOAS stations installed in four megacities(Beijing,Shanghai,Shenzhen,and Chongqing)were used to characterize their vertical distribution differences in four key regions,Jing–Jin–Ji(JJJ),Yangtze River Delta(YRD),Pearl River Delta(PRD),and Sichuan Basin(SB),respectively.The normalized and yearly averaged aerosol vertical profiles below 400 m in JJJ and PRD exhibit a box shape and a Gaussian shape,respectively,and both show exponential shapes in YRD and SB.The NO_(2) vertical profiles in four regions all exhibit exponential shapes because of vehicle emissions.The shape of the HCHO vertical profile in JJJ and PRD was Gaussian,whereas an exponential shape was shown in YRD and SB.Moreover,a regional transport event occurred at an altitude of 600–1000 m was monitored in the southwest–northeast pathway of the North China Plain(NCP)by five MAX-DOAS stations(Shijiazhuang(SJZ),Wangdu(WD),Nancheng(NC),Chinese Academy of Meteorological Sciences(CAMS),and University of Chinese Academy of Sciences(UCAS))belonging to the above network.The aerosol optical depths(AOD)in these five stations decreased in the order of SJZ>WD>NC>CAMS>UCAS.The short-distance regional transport of NO2 in the 700–900 m layer was monitored between WD and NC.As an important precursor of secondary aerosol,the peak of NO_(2) air mass in WD and NC all occurred 1 h earlier than that of aerosol.This was also observed for the short-distance regional transport of HCHO in the 700–900 m layer between NC and CAMS,which potentially affected the O_(3) concentration in Beijing.Finally,CAMS was selected as a typical site to determine the O_(3)–NO_(x)–volatile organic compounds(VOCs)sensitivities in vertical space.We found the production of O_(3) changed from predominantly VOCs-limited conditions to mainly mixed VOCs–NO_(x)-limited condition from the 0–100 m layer to the 200–300 m layer.In addition,the downward transport of O_(3) could contribute to the increase of ground surface O_(3) concentration.This ground-based hyperspectral stereoscopic remote sensing network provide a promising strategy to support management of PM_(2.5) and O_(3) and their precursors and conduct attribution of sources.Cheng Liu Chengzhi Xing Qihou Hu Qihua Li Haoran Liu Qianqian Hong Wei Tan Xiangguang Ji Hua Lin Chuan Lu Jinan Lin Hanyang Liu Shaocong Wei Jian Chen Kunpeng Yang Shuntian Wang Ting Liu Yujia Chen 2022Engineering2022,,12:0
16A novel gene-activated matrix composed of PEI/plasmid-BMP2 complexes and hydroxyapatite/chitosan-microspheres promotes bone regeneration显示文摘The incorporation of pro-osteogenic growth factors into bone graft materials to enhance bone regeneration is a key research area within the field of bone tissue engineering and regenerative medicine.However,growth factors directly incorporated in protein form are easily degraded,and have a limited active half-life,which cannot exert long-term and stable osteoinductive and oteoconductive effects.The combination of gene therapy and tissue engineering through gene-activated matrix(GAM)may provide a good alternative solution to overcome such limitations.Scaffold materials can be combined together with plasmid DNA and a chemical-based transfection agent to form GAM,through which transfected cells could secrete growth factors in a sustained manner over a longer time duration;thereby enabling bone graft materials to act as a repository of therapeutic genes,while providing structural support and a scaffold matrix for new bone tissue ingrowth.In this study,we prepared hydroxyapatite/chitosan-microspheres(HA/CS-MS)with microfabrication technology and emulsification method,and loaded the polyethylene imine/bone morphogenetic protein 2 plasmid(PEI/pBMP2)complexes with high transfection capacity(transfection efficiency up to 54.79%±4.95%),thus forming a novel GAM system with superior bone regeneration capacity—PEI/pBMP2-HA/CS-MS.The in vitro experiments in this study demonstrated that our GAM had excellent biocompatibility(with cell viability over 95%),and that the as-fabricated microsphere material possessed a nano-network fibrous structure similar to natural extracellular matrix(ECM),together with a higher surface area that can provide more cell adhesion sites.The sizes of the prepared microspheres were mainly distributed in the 160–180μm range,while the maximal loading rate of PEI-pBMP2 complexes was 59.79%±1.85%.As a loaded complexes system,the GAM can release plasmids in a slow controlled manner,effectively transfecting surrounding target cells(release effect for up to 21 days),while cells adherent to the material can also take up plasmids,resulting in sustained secretion of the target protein,thereby effectively promoting bone regeneration.In vivo data from micro-computed tomography(micro-CT)and histological staining showed that the use of the composite materials effectively enhanced bone regeneration in defect areas.These findings thus demonstrated that the novel GAM system had excellent osteoinductivity with significant clinical potential.Ruyuan Ding Yajun Liu Dawei Cheng Gang Yang Wenjing Wu Haoran Du Xin Jin Yihan Chen Yuanyin Wang Boon Chin HENG Qing Yang Jianguang XU 2022Nano Research2022,15,7:0
17A review of interaction mechanisms and microscopic simulation methods for CO_(2)-water-rock system显示文摘This work systematically reviews the complex mechanisms of CO_(2)-water-rock interactions,microscopic simulations of reactive transport(dissolution,precipitation and precipitate migration)in porous media,and microscopic simulations of CO_(2)-water-rock system.The work points out the key issues in current research and provides suggestions for future research.After injection of CO_(2) into underground reservoirs,not only conventional pressure-driven flow and mass transfer processes occur,but also special physicochemical phenomena like dissolution,precipitation,and precipitate migration.The coupling of these processes causes complex changes in permeability and porosity parameters of the porous media.Pore-scale microscopic flow simulations can provide detailed information within the three-dimensional pore and throat space and explicitly observe changes in the fluid-solid interfaces of porous media during reactions.At present,the research has limitations in the decoupling of complex mechanisms,characterization of differential multi-mineral reactions,precipitation generation mechanisms and characterization(crystal nucleation and mineral detachment),simulation methods for precipitation-fluid interaction,and coupling mechanisms of multiple physicochemical processes.In future studies,it is essential to innovate experimental methods to decouple“dissolution-precipitation-precipitate migration”processes,improve the accuracy of experimental testing of minerals geochemical reaction-related parameters,build reliable characterization of various precipitation types,establish precipitation-fluid interaction simulation methods,coordinate the boundary conditions of different physicochemical processes,and,finally,achieve coupled flow simulation of“dissolution-precipitation-precipitate migration”within CO_(2)-water-rock systems.ZHANG Liehui ZHANG Tao ZHAO Yulong HU Haoran WEN Shaomu WU Jianfa CAO Cheng WANG Yongchao FAN Yunting 2024Petroleum Exploration and Development2024,51,1:0
18Identification of potential sex-related genes in Siniperca chuatsi显示文摘Mandarin fish(Siniperca chuatsi)is an economically important freshwater fish cultured in China.In this species,females grow faster than males.However,due to the lack of available genomic and transcriptome information,the mechanisms of sex differentiation remain poorly understood.In this study,Illumina high-throughput sequencing technology was used to sequence four cDNA libraries;the tissues examined included hypothalamus,pituitary gland,ovary,and testis.A total of 134124 high-quality unigenes were obtained on average length of 1361 bp and N50 of 3312 bp.A search of all-unigene sequences against NR,NT,SwissProt,KOG,KEGG,GO,and InterPro databases resulted in 59688(44.50%),76329(56.91%),50432(37.60%),45741(34.10%),48760(36.35%),5241(3.91%),and 46099(34.37%)annotations,respectively.In a comparison of ovarian and testicular libraries,15289 ovary-biased genes and 10035 testis-biased genes were identified,including a series of genes related to sex determination and differentiation,such as cyp19a1a,foxl2,sox9,dmrt1,amh,and others.In addition,49495 SSRs and 85899 SNPs were detected in transcriptome data.Quantitative real-time PCR results of 15 sex-related functional genes indicated that RNA-seq data was reliable.This study will contribute to a better understanding of the molecular mechanisms of sex differentiation and development in Mandarin fish.Qiaoying ZHU Chong HAN Cheng PENG Xingni ZHOU Chongwei WANG Linqiang HAN Shuisheng LI Guifeng LI Haoran LIN Yong ZHANG 2021Journal of Oceanology and Limnology2021,39,4:0
19eIF-Three to Tango: emerging functions of translation initiation factor eIF3 in protein synthesis and disease显示文摘Studies over the past three years have substantially expanded the involvements of eukaryotic initiation factor 3 (eIF3) in messenger RNA (mRNA) translation. It now appears that this multi-subunit complex is involved in every possible form of mRNA translation, controlling every step of protein synthesis from initiation to elongation, termination, and quality control in positive as well as negative fashion. Through the study of eIF3, we are beginning to appreciate protein synthesis as a highly integrated process coordinating protein production with protein folding, subcellular targeting, and degradation. At the same time, eIF3 subunits appear to have specific functions that probably vary between different tissues and individual cells. Considering the broad functions of eIF3 in protein homeostasis, it comes as little surprise that eIF3 is increasingly implicated in major human diseases and first attempts at therapeutically targeting eIF3 have been undertaken. 20A digital microfluidic system with 3D microstructures for single-cell culture显示文摘Despite the precise controllability of droplet samples in digital microfluidic(DMF)systems,their capability in isolating single cells for long-time culture is still limited:typically,only a few cells can be captured on an electrode.Although fabricating small-sized hydrophilic micropatches on an electrode aids single-cell capture,the actuation voltage for droplet transportation has to be significantly raised,resulting in a shorter lifetime for the DMF chip and a larger risk of damaging the cells.In this work,a DMF system with 3D microstructures engineered on-chip is proposed to form semi-closed micro-wells for efficient single-cell isolation and long-time culture.Our optimum results showed that approximately 20%of the micro-wells over a 30×30 array were occupied by isolated single cells.In addition,low-evaporation-temperature oil and surfactant aided the system in achieving a low droplet actuation voltage of 36V,which was 4 times lower than the typical 150 V,minimizing the potential damage to the cells in the droplets and to the DMF chip.To exemplify the technological advances,drug sensitivity tests were run in our DMF system to investigate the cell response of breast cancer cells(MDA-MB-231)and breast normal cells(MCF-10A)to a widely used chemotherapeutic drug,Cisplatin(Cis).The results on-chip were consistent with those screened in conventional 96-well plates.This novel,simple and robust single-cell trapping method has great potential in biological research at the single cell level.Jiao Zhai Haoran Li Ada Hang-Heng Wong Cheng Dong Shuhong Yi Yanwei Jia Pui-In Mak Chu-Xia Deng Rui PMartins 2020Microsystems & Nanoengineering2020,6,1:0
