1An isoflavone derivative potently inhibits the angiogenesis and progression of triple-negative breast cancer by targeting the MTA2/SerRS/VEGFA pathway显示文摘Objective:Angiogenesis plays a vital role in tumor growth and metastasis.Here,we aimed to find novel efficient antiangiogenic molecules targeting vascular endothelial growth factor A(VEGFA)at the transcriptional level to treat triple-negative breast cancer(TNBC).Methods:We used a cell-based seryl tRNA synthetase(SerRS)promoter-driven dual-luciferase reporter system to screen an in-house library of 384 naturally occurring small molecules and their derivatives to find candidate molecules that could upregulate the expression of SerRS,a potent transcriptional repressor of VEGFA.The levels of SerRS and VEGFA were examined by quantitative RT-PCR(qRT-PCR),western blotting,and/or ELISAs in TNBC cells after candidate molecule administration.Zebrafish,the Matrigel plug angiogenesis assay in mice,the TNBC allograft,and xenograft mouse models were used to evaluate thein vivoanti-angiogenic and anti-cancer activities.Furthermore,the potential direct targets of the candidates were identified by proteomics and biochemical studies.Results:We found the most active compound was 3-(4-methoxyphenyl)quinolin-4(1H)-one(MEQ),an isoflavone derivative.In TNBC cells,MEQ treatment resulted in increased SerRS mRNA(P<0.001)and protein levels and downregulated VEGFA production.Both the vascular development of zebrafish and Matrigel plug angiogenesis in mice were inhibited by MEQ.MEQ also suppressed the angiogenesis in TNBC allografts and xenografts in mice,resulting in inhibited tumor growth and prolonged overall survival(P<0.05).Finally,we found that MEQ regulated SerRS transcription by interacting with MTA2(Metastasis Associated 1 Family Member 2).Conclusions:Our findings suggested that the MTA2/SerRS/VEGFA axis is a drug-treatable anti-angiogenic target,and MEQ is a promising anti-tumor molecule that merits further investigation for clinical applications.Xiaotong Zhang Gengyi Zou Xiyang Li Lun Wang Tianyu Xie Jin Zhao Longlong Wang Shunchang Jiao Rong Xiang Haoyu Ye Yi Shi 2020Cancer Biology & Medicine2020,17,3:6
2Trace Elements in Sphalerite from the Dadongla Zn-Pb Deposit, Western Hunan-Eastern Guizhou Zn-Pb Metallogenic Belt, South China显示文摘The western Hunan-eastern Guizhou Zn-Pb metallogenic belt is one of the important Zn-Pb mineralization regions in China.The Dadongla deposit,located in the northeast of Guizhou Province,is one of the typical Zn-Pb deposits in the region and has estimated resources more than 12 million metric tons(Mt)with an average grade of 4.11 wt%Zn+Pb.Its orebodies are hosted in the lower Cambrian Aoxi Formation dolomite,occurring as bedded,para-bedded in shape,and in conformity with the wall rock.The ore mineral assemblage is simple,dominated by sphalerite with minor pyrite and galena,and the gangue minerals are composed of dolomite,calcite with minor bitumen and barite.In view of the lack of geological and geochemical researches,the genesis of Zn-Pb ore is still unclear.Laser ablation-inductively coupled plasma mass spectrometry(LA-ICPMS)spot and mapping analyses were used to obtain sphalerite trace element chemistry in the Dadongla Zn-Pb deposit in Guizhou,China,aiming to constrain its ore genesis.The results show that sphalerite is characterized by the enrichment of Cd,Fe,Ge and Hg,corresponding with that of typical MVT deposits.Four zones were identified in the sphalerite crystal from Dadongla from the center to margin according to the color bands.in which the zone in the center,representing the early ore-stage sphalerite,is characterized by enrichment of Cd relatively,while the zone forming at late ore-stage is enriched in Ge and Hg relatively.The finding was controlled by differential leached metals content in ore-forming fluid from its source rock.Notably,critical element Ge trends to be enriched at the late ore-stage and follows a substitution of 2 Zn^2+(?)Ge^4++□(vacancy).Moreover,the calculated ore-forming temperature ranges from 79.9℃to 177.6℃by the empirical formula,which is similar to that of typical Mississippi Valley-type(MVT)deposits.Compared with the features of trace elements in sphalerite from different types of deposits,together with the geology,the Dadongla deposit belongs to an MVT Zn-Pb deposit.HU Yusi YE Lin WEI Chen LI Zhenli HUANG Zhilong WANG Haoyu 2020Acta Geologica Sinica(English Edition)2020,94,6:3

4Enhancing thermal conductivity of silicone rubber composites by insitu constructing SiC networks:A finite-element study based on first principles calculation显示文摘Polymer composites as thermal interface materials have been widely used in modern electronic equipment.In this work,we report a novel method to prepare highly through-plane thermally conductive silicone rubber(SR)composites with vertically aligned silicon carbide fibers(VA-SiCFs)entangled by SiC nanowires(SiCNWs)networks.First,a series of carbon fibers(CFs)skeletons were fabricated in sequence of coating poor thermally conductive polyacrylonitrile-based CFs with polydopamine,icetemplated assembly,and freeze-drying processes.Furthermore,VA-SiCFs networks,i.e.,long-range continuous SiCFs-SiCNWs networks,based on the prepared CFs skeletons,were in-situ obtained via template-assisted chemical vapor deposition method.The thermal conductivity enhancement mechanism of VA-SiCFs networks on its SR composites was also intensively studied by finite element simulation,based on the first principles investigation of SiC,and Foygel’s theory.The in-situ grown VA-SiCFs networks possess high intrinsic thermal conductivity without the thermal interface between fillers,acting as the high-efficiency through-plane long-range continuous thermal conduction path,in which the SiCNWs were the in-plane“thermal spreader”.The VA-SiCFs/SR composites reached a high through-plane thermal conductivity,2.13 W/(m·K),at the filler loading of 15 vol.%,which is 868.2%,and 249.2%higher than that of pure SR sample,and random-CFs@polydopamine(PDA)/SR composites at the same content,respectively.The VA-SiCFs/SR composites also exhibited good electrical insulation performance and excellent dimensional stability,which guaranteed the stable interfacial heat transfer of high-power density electronic devices.Dongliang Ding Shiyu Zhang Haoyu Liang Xu Wang Ya Wu Yuanming Ye Zhenguo Liu Qiuyu Zhang Guangzhao Qin Yanhui Chen 2023Nano Research2023,16,1:0
5Design and Fabrication of Reflective Phase Shifter for Two-Dimensional Terahertz Beam-Scanning Reflectarray显示文摘A reflective phase shifter is proposed to realize the two-dimensional beam-scanning reflectarray.The reflectarray is composed of double-dipole resonant elements in which the phase shift is implemented by applying a driving voltage to the liquid crystal(LC). A 30 ×30 phase shifter sample is fabricated on a quartz substrate with a 480 μm thickness and a 4 cm × 4 cm area.According to the experimental results, 0 to 360° phase shift can be achieved within the bandwidth of 5 GHz. In order to accomplish two-dimensional beam-scanning and reduce the negative impact of driving lines on the reflectarray, a new LC driving method is developed. To verify the accuracy of the proposed approach, three samples of different driving line numbers are designed using the above-mentioned phase shifter, which all have achieved a0-360° phase shift in the bandwidth of 4 GHz. 6Revisiting the gravity laws of inter-city mobility in megacity regions显示文摘Inter-city mobility is one of the most important issues in the UN Sustainable Development Goals,as it is essential to access the regional labour market,goods and services,and to constrain the spread of infectious diseases.Although the gravity model has been proved to be an effective model to describe mobility among settlements,knowledge is still insufficient in regions where dozens of megacities interact closely and over 100 million people reside.In addition,the existing knowledge is limited to overall population mobility,while the difference in inter-city travel with different purposes is unexplored on such a large geographic scale.We revisited the gravity laws of inter-city mobility using the 2.12 billion trip chains recorded by 40.48 million mobile phone users’trajectories in the Jing-Jin-Ji Region,which contains China’s capital Beijing.Firstly,unlike previous studies,we found that non-commuting rather than commuting is the dominant type of inter-city mobility(89.3%).Noncommuting travellers have a travel distance 42.3%longer than commuting travellers.Secondly,we developed more accurate gravity models for the spatial distribution of inter-city commuting and non-commuting travel.We also found that inter-city mobility has a hierarchical structure,as the distribution of inter-city travel volume follows Zipf’s law.In particular,the hierarchy of non-commuting travel volume among the cities is more in line with an ideal Zipf distribution than commuting travel.Our findings contribute to new knowledge on basic inter-city mobility laws,and they have significant applications for regional policies on human mobility.Pengjun ZHAO Haoyu HU Liangen ZENG Jun CHEN Xinyue YE 2023Science China Earth Sciences2023,66,2:0
7Purification and characterization of a cold-active myrosinase from marine Pseudomonas oleovorans SuMy07显示文摘Myrosinase(EC 3.2.1.147),also known asβ-thioglucoside glucohydrolase,was first discovered in mustard seeds.Myrosinase hydrolyzes glucosinolate to produce D-glucose and an unstable aglycon,which upon rearrangement yields metabolites such as isothiocyanate,sulfate,acetonitrile,and thiocyanate.Isothiocyanate has a wide range of biological functions[1].Previous studies have shown that isothiocyanate has insect-repellant and antibacterial effects[2].Isothiocyanate also has anti-inflammatory and antioxidant properties,making it a potential anticancer compound[3,4].Sulforaphane is one of the most representative isothiocyanate substances against numerous cancers.Myrosinase extraction is technically challenging,although myrosinase metabolites have a variety of uses.At present,myrosinase is primarily extracted from white mustard seeds after a time-consuming and expensive preparation.Therefore,the search for myrosinase that can hydrolyze glucosinolates to produce isothiocyanate efficiently and economically has become an urgent task.Microbes can secrete myrosinase with rapid growth,lack of seasonal variation,simple intensive cultivation,and simple product extraction.Zhifa Huang Nannan Liu Yaowei Fang Xiaoyue Hou Guang Yang Jing Lu Haoyu Mi Qinwen Ye Rongjun Zhu Shu Liu 2023Acta Biochimica et Biophysica Sinica2023,55,4:0
8Targeting TRPV1-mediated autophagy attenuates nitrogen mustard-induced dermal toxicity显示文摘Nitrogen mustard(NM)causes severe vesicating skin injury,which lacks effective targeted therapies.The major limitation is that the specific mechanism of NM-induced skin injury is not well understood.Recently,autophagy has been found to play important roles in physical and chemical exposure-caused cutaneous injuries.However,whether autophagy contributes to NM-induced dermal toxicity is unclear.Herein,we initially confrmed that NM dose-dependently caused cell death and induced autophagy in keratinocytes.Suppression of autophagy by 3-methyladenine,chloroquine,and baflomycin A1 or ATG5 siRNA attenuated NM-induced keratinocyte cell death.Furthermore,NM increased transient receptor potential vanilloid 1(TRPV1)expression,intracellular Ca2+content,and the activities of Car+/calmodulin-dependent kinase kinaseβ(CaMKKB),AMP-activated protein kinase(AMPK),unc-51-like kinase 1(ULK1),and mammalian target of rapamycin(mTOR).NM-induced autophagy in keratinocytes was abolished by treatment with inhibitors of TRPV1(capsazepine),CaMKKβ(STO-609),AMPK(compound C),and ULK1(SBI-0206965)as well as TRPV1,CaMKKB,and AMPK siRNA transfection.In addition,an mTOR inhibitor(rapamycin)had no signifcant effect on NM-stimulated autophagy or cell death of keratinocytes.Finally,the results of the in vivo experiment in NM-treated skin tissues were consistent with the findings of the in vitro experiment.In conclusion,NM-caused dermal toxicity by overactivating autophagy partially through the activation of TRPV1-Ca2+-CaMKKB-AMPK-ULK1 signaling pathway.These results suggest that blocking TRPV1-dependent autophagy could be a potential treatment strategy for NM-caused cutaneous injury.Mingliang Chen Xunhu Dong Haoyue Deng Feng Ye Yuanpeng Zhao Jin Cheng Guorong Dan Jiqing Zhao Yan Sai Xiuwu Bian Zhongmin Zou 2021Signal Transduction and Targeted Therapy2021,6,2:0
9Fabricating a PVDF skin for PEO-based SPE to stabilize the interface both at cathode and anode for Li-ion batteries显示文摘Poly(ethylene oxide)(PEO)-based solid polymer electrolyte is always the most promising candidate for preparing thinner, lighter and safer lithium-ion batteries. However, the lithium dendrites growth of lithium anode and the high-voltage oxidation of cathode are easy to cause the PEO-based battery failure.The way to deal with the different challenges on both sides of the anode and cathode is pursued all the time. In this study, we reported a new strategy to construct the PVDF/PEO/PVDF three-layer structure for solid polymer electrolyte(marked as PVDF@PEO) using PVDF as the functional “skin”. The PVDF@PEO electrolyte can effectively prevent from the lithium dendrites, and shows a stable cycling life over1000 h in the Li/PVDF@PEO/Li cell. In addition, the PVDF@PEO electrolyte exhibits higher oxidation resistance and can be matched with high-voltage LiCoO_(2) cathode. The Li/PVDF@PEO/LiCoO_(2) cell delivered a specific capacity of about 150 m Ah g^(-1) over 150 cycles and maintained good cycling stability. 10Preliminary Study on Water Demand Law of 1-0 Rooted Cuttings of Populus szechuanica显示文摘In order to provide theoretical basis and technical support for the afforestation and artificial water supply of P.szechuanica in arid areas,the characteristics of water consumption of P.szechuanica were explored,and the law of water demand of P.szechuanica was grasped.In this paper,potted seedlings of 1-0 rooted cuttings of P.szechuanica were taken as research objects,and change situation of water consumption under different water control gradients was measured regularly by using weighing method,further analyzing dynamic change of water consumption of P.szechuanica and revealing water demand law of 1-0 rooted cuttings of P.szechuanica.The results showed that total change of water consumption of 1-0 rooted cuttings of P.szechuanica had'slow-fast-slow-fast'double-peak trend in the growth period of the current year,and corresponded with univariate linear relation(R^(2)=0.7137),with significant difference.In whole growth period,water consumption in August was the highest,which was 2.7 times of that in June and July and 1.5 times of that after September.In different water control treatments,the dynamic changes of daily and monthly water consumption were significantly different.In seven water control treatments,monthly water consumption was between(6315.95±1690.70)and(10105.28±3065.30)g/month,and mean was(8211.07±2308.23)g/month.With intensification of water control treatment,water consumption increased,but there was no seedling death due to water shortage.P.szechuanica has great plasticity in water demand,and can survive in both arid and humid environments.Meanwhile,it is revealed that P.szechuanica is the most widely distributed tree species in the region.Dabuqiong Haoyu WANG Huanhuan XIE Zhen XING Yanhui YE 2021Meteorological and Environmental Research2021,12,4:0
11Crustal velocity structure of the Northwest Sub-basin of the South China Sea based on seismic data reprocessing显示文摘The deep crustal structure of the Northwest Sub-basin(NWSB)of the South China Sea(SCS)is of great importance for understanding the tectonic nature of the continent-ocean transition(COT)and magmatism in this oceanic basin.The 2-D wide-angle reflection/refraction seismic profile OBS2006-2 is almost parallel to the extinct spreading ridge(ESR)of the NWSB.In addition to the original data,we added the data of two reprocessed OBS stations,and carried out seismic phase re-picking and travel-time imaging to obtain the crustal velocity structure along this profile.Resolution tests demonstrate that the newly acquired velocity structure is more reliable than the prior interpretation.The depth of the Moho(23.5–11.8 km)and crustal thickness(20.5–6.5 km)systematically changes from continental crust of the Xisha Block to the oceanic crust within the NWSB.The COT zone has a width of^20 km and the depth of the Moho decreases from 15.0 to 11.0 km,corresponding to a^4 km decrease in crustal thickness(6–10 km).A high velocity layer(HVL,7.2–7.4 km s–1)exists at the bottom of the crust at the location where the sharp lateral transition of the continental crust to the oceanic crust occurs.Age dating shows that the Doublepeak Seamount was formed at^23 Ma,after the cessation of the NWSB seafloor spreading(~32–25 Ma).The crust beneath the Double-peak Seamount is oceanic with a thickness of 9 km.We infer that this oceanic crust was formed by magmatic upwelling and decompression melting along a pre-existing zone of weakness.Qiang WANG Minghui ZHAO Haoyu ZHANG Jiazheng ZHANG Enyuan HE Ye YUAN Xuelin QIU 2020Science China Earth Sciences2020,63,11:0
