1Contrasting trends of PM2.5 and surface-ozone concentrations in China from 2013 to 2017显示文摘Although much attention has been paid to investigating and controlling air pollution in China,the trends of air-pollutant concentrations on a national scale have remained unclear.Here,we quantitatively investigated the variation of air pollutants in China using long-term comprehensive data sets from 2013 to 2017,during which Chinese government made major efforts to reduce anthropogenic emission in polluted regions.Our results show a significant decreasing trend in the PM2.5 concentration in heavily polluted regions of eastern China,with an annual decrease of〜7%compared with measurements in 2013.The measured decreased concentrations of SO2>NO2 and CO(a proxy for anthropogenic volatile organic compounds)could explain a large fraction of the decreased PM2.5 concentrations in different regions.As a consequence;the heavily polluted days decreased significantly in corresponding regions.Concentrations of organic aerosol;nitrate;sulfate,ammonium and chloride measured in urban Beijing revealed a remarkable reduction from 2013 to 2017;connecting the decreases in aerosol precursors with corresponding chemical components closely.However;surface-ozone concentrations showed increasing trends in most urban stations from 2013 to 2017;which indicates stronger photochemical pollution.The boundary-layer height in capital c让ies of eastern China showed no significant trends over the Beijing-Tianjin-Hebei,Yangtze River Delta and Pearl River Delta regions from 2013 to 2017;which confirmed the reduction in anthropogenic emissions.Our results demonstrated that the Chinese government was successful in the reduction of particulate matter in urban areas from 2013 to 2017,although the ozone concentration has increased significantly;suggesting a more complex mechanism of improving Chinese air quality in the future.Yonghong Wang Wenkang Gao Shuai Wang Tao Song Zhengyu Gong Dongsheng Ji Lili Wang Zirui Liu Guiqian Tang Yanfeng Huo Shili Tian Jiayun Li Mingge Li Yuan Yang Biwu Chu Tuukka Petaja Veli-Matti Kerminen Hong He Jiming Hao Markku Kulmala Yuesi Wang Yuanhang Zhang 2020National Science Review2020,7,8:51

5Ginsenoside Rgl protects against ischemic/reperfusioninduced neuronal injury through miR-144/Nrf2/ARE pathway显示文摘Ginsenoside Rg1 (Rg1),a saponin extracted from Panax ginseng,has been well documented to be effective against ischemic/ reperfusion (I/R)neuronal injury.However,the underlying mechanisms remain obscure.In the present study,we investigated the roles of Nrf2 and miR-144 in the protective effects of Rgl against I/R-induced neuronal injury.In OGD/R-treated PC12 cells,Rgl (0.01-1 μmol/L)dose-dependently attenuated the cell injury accompanied by prolonging nuclear accumulation of Nrf2,enhancing the transcriptional activity of Nrf2,as well as promoting the expression of ARE-target genes.The activation of the Nrf2/ARE pathway by Rgl was independent of disassociation with Keapl,but resulted from post-translational regulations.Knockdown of Nrf2 abolished all the protective changes of Rgl in OG-D/R-treated PC12 cells.Furthermore,Rgl treatment significantly decreased the expression of miR-144,which downregulated Nrf2 production by targeting its 3'-untranlated region after OGD/R.Knockdown of Nrf2 had no effect on the expression of miR-144,suggesting that miR-144 was an upstream regulator of Nrf2.We revealed that there was a direct binding between Nrf2 and miR-144 in PC12 cells.Application of anti-miR-144 occluded the activation of the Nrf2/ ARE pathway by Rgl in OGD/R-treated PC12 cells.In tMCAO rats,administration of Rgl (20 mg/kg).significantly alleviated ischemic injury,and activated Nff2/ARE pathway.The protective effects of Rgl were abolished by injecting of AAV-HIF-miR-144-shRNA into the predicted ischemic penumbra.In conclusion,our results demonstrate that Rgl alleviates oxidative,stress after I/R through inhibiting miR-144 activity and subsequently promoting the Nrf2/ARE pathway at the post-translational-level.Shi-feng Chu Zhao Zhang Xin Zhou Wen-bin He Chen Chen Piao Luo Dan-dan Liu Qi-di Ai Hai-fan Gong Zhen-zhen Wang Hong-shuo Sun Zhong-ping Feng Nai-hong Chen 2019Acta Pharmacologica Sinica2019,40,1:30

8Targeted puncture of left branch of intrahepatic portal vein in transjugular intrahepatic portosystemic shunt to reduce hepatic encephalopathy显示文摘BACKGROUND Transjugular intrahepatic portosystemic shunt(TIPS) is currently used for the treatment of complications of portal hypertension. The incidence of hepatic encephalopathy(HE) remains a problem in TIPS placement. It has been reported that the right branch mainly receives superior mesenteric venous blood while the left branch mainly receives blood from the splenic vein. We hypothesized that targeted puncture of the left portal vein would divert the non-nutritive blood from the splenic vein into the TIPS shunt; therefore, targeted puncture of the left branch of the intrahepatic portal vein during TIPS may reduce the risk of HE.AIM To evaluate the influence of targeted puncture of left branch of portal vein in TIPS on HE.METHODS A retrospective analysis of 1244 patients with portal-hypertension-related complications of refractory ascites or variceal bleeding who underwent TIPS from January 2000 to January 2013 was performed. Patients were divided into group A(targeting left branch of portal vein, n = 937) and group B(targeting right branch of portal vein, n = 307). TIPS-related HE and clinical outcomes were analyzed.RESULTS The symptoms of ascites and variceal bleeding disappeared within a short time.By the endpoint of follow-up, recurrent bleeding and ascites did not differ significantly between groups A and B(P = 0.278, P = 0.561, respectively).Incidence of HE differed significantly between groups A and B at 1 mo(14.94% vs36.80%, χ~2 = 4.839, P = 0.028), 3 mo(12.48% vs 34.20%, χ~2 = 5.054, P = 0.025), 6 mo(10.03% vs 32.24%, χ~2 = 6.560, P = 0.010), 9 mo(9.17% vs 31.27%, χ~2 = 5.357, P =0.021), and 12 mo(8.21% vs 28.01, χ~2 = 3.848, P = 0.051). There were no significant differences between groups A and B at 3 years(6.61% vs 7.16%, χ~2 = 1.204, P =0.272) and 5 years(5.01% vs 6.18%, χ~2 = 0.072, P = 0.562). 9Delayed sowing can increase lodging resistance while maintaining grain yield and nitrogen use efficiency in winter wheat显示文摘Lodging resistance of winter wheat(Trnticum aestivum L.) can be increased by late sowing.However, whether grain yield and nitrogen use efficiency(NUE) can be maintained with delayed sowing remains unknown. During the 2013-2014 and 2014-2015 growing seasons, two winter wheat cultivars were sown on three dates(early sowing on October 1, normal so,wing on October8, and late sowing on October 15) to investigate the responses of lodging resistance, grain yield,and NUE to sowing date. No significant differences in lodging resistance, grain yield, or NUE between early and normal sowing were observed. Averaging over the two cultivars and years,postponing the sowing date significantly increased lodging resistance by 53.6% and 49.6%compared with that following early and normal sowing, respectively. Lodging resistance was improved mainly through a reduction in the culm height at the center of gravity and an increase in the tensile strength of the base internode. Late sowing resulted in similar grain yield as well as kernel weight and number of kernels per square meter, compared to early and normal sowing.Averaging over the two cultivars and years, delayed sowing resulted in a reduction in nitrogen uptake efficiency(UPE) by 11.0% and 9.9% compared to early and normal sowing, respectively,owing to reduced root length density and dry matter accumulation before anthesis. An average increase in nitrogen utilization efficiency(UTE) of 12.9% and 11.2% compared to early and normal sowing, respectively, was observed with late sowing owing to a reduction in the grain nitrogen concentration. The increase in UTE offset the reduction in UPE, resulting in equal NUEs among all sowing dates. 10Significant concurrent decrease in PM_(2.5) and NO_(2) concentrations in China during COVID-19 epidemic显示文摘The strict control measures and social lockdowns initiated to combat COVID-19 epidemic have had a notable impact on air pollutant concentrations.According to observation data obtained from the China National Environmental Monitoring Center,compared to levels in 2019,the average concentration of NO_(2) in early 2020 during COVID-19 epidemic has decreased by 53%,50%,and 30%in Wuhan city,Hubei Province(Wuhan excluded),and China(Hubei excluded),respectively.Simultaneously,PM_(2.5) concentration has decreased by 35%,29%,and 19%in Wuhan,Hubei(Wuhan excluded),and China(Hubei excluded),respectively.Less significant declines have also been found for SO_(2) and CO concentrations.We also analyzed the temporal variation and spatial distribution of air pollutant concentrations in China during COVID-19 epidemic.The decreases in PM_(2.5) and NO_(2) concentrations showed relatively consistent temporal variation and spatial distribution.These results support control of NO x to further reduce PM_(2.5) pollution in China.The concurrent decrease in NOx and PM_(2.5) concentrations resulted in an increase of O3 concentrations across China during COVID-19 epidemic,indicating that coordinated control of other pollutants is needed.Biiuu Chu Shuping Zhang Jun Liu Qingxin Ma Hong He 2021Journal of Environmental Sciences2021,33,1:21

13Risk factors for progression to acute-on-chronic liver failure during severe acute exacerbation of chronic hepatitis B virus infection显示文摘BACKGROUND Acute exacerbation in patients with chronic hepatitis B virus(HBV) infection results in different severities of liver injury. The risk factors related to progression to hepatic decompensation(HD) and acute-on-chronic liver failure(ACLF) in patients with severe acute exacerbation(SAE) of chronic HBV infection remain unknown.AIM To identify risk factors related to progression to HD and ACLF in compensated patients with SAE of chronic HBV infection.METHODS The baseline characteristics of 164 patients with SAE of chronic HBV infection were retrospectively reviewed. Independent risk factors associated with progression to HD and ACLF were identified. The predictive values of our previously established prediction model in patients with acute exacerbation(AE model) and the model for end-stage liver disease(MELD) score in predicting the development of ACLF were evaluated.RESULTS Among 164 patients with SAE, 83(50.6%) had compensated liver cirrhosis(LC),43 had progression to HD without ACLF, and 29 had progression to ACLF within 28 d after admission. Independent risk factors associated with progression to HD were LC and low alanine aminotransferase. Independent risk factors for progression to ACLF were LC, high MELD score, high aspartate aminotransferase(AST) levels, and low prothrombin activity(PTA). The area under the receiver operating characteristic of the AE model [0.844, 95%confidence interval(CI): 0.779-0.896] was significantly higher than that of MELD score(0.690, 95%CI: 0.613-0.760, P < 0.05) in predicting the development of ACLF.CONCLUSION In patients with SAE of chronic HBV infection, LC is an independent risk factor for progression to both HD and ACLF. High MELD score, high AST, and low PTA are associated with progression to ACLF. 14Genome-wide characterization and analysis of b HLH transcription factors in Panax ginseng显示文摘Ginseng(Panax ginseng C.A. Meyer) is one of the best-selling herbal medicines, with ginsenosides as its main pharmacologically active constituents. Although extensive chemical and pharmaceutical studies of these compounds have been performed, genome-wide studies of the basic helix-loop-helix(b HLH) transcription factors of ginseng are still limited. The b HLH transcription factor family is one of the largest transcription factor families found in eukaryotic organisms, and these proteins are involved in a myriad of regulatory processes. In our study, 169 bHLH transcription factor genes were identified in the genome of P. ginseng, and phylogenetic analysis indicated that these PGb HLHs could be classified into 24 subfamilies. A total of 21 RNA-seq data sets, including two sequencing libraries for jasmonate(JA)-responsive and 19 reported libraries for organ-specific expression analyses were constructed. Through a combination of gene-specific expression patterns and chemical contents,6 PGbHLH genes from 4 subfamilies were revealed to be potentially involved in the regulation of ginsenoside biosynthesis. These 6 PGbHLHs, which had distinct target genes, were further divided into two groups depending on the absence of MYC-N structure. 15Qingxuan Jiangya Decoction(清眩降压汤) Prevents Blood Pressure Elevation and Ameliorates Vascular Structural Remodeling via Modulating TGF-β 1/Smad Pathway in Spontaneously Hypertensive Rats显示文摘Objective:To elevate the effects of Qingxuan Jiangya Decoction(清眩降压汤,QXJYD)on hypertension and vascular structural remodeling(VSR)in spontaneously hypertensive rats(SHRs),and investigate the underlying mechanisms.Methods:SHRs(n=8)were given intra-gastric administration with 60 mg/kg of QXJYD or saline,daily for 8 weeks,while rats in SHR-control(n=8)and WKY(n=8)groups were received equal volumes of saline solution.Systolic blood pressures(SBP),diastolic blood pressures(DBP)and mean blood pressures(MBP)were measured once a week.The levels of angiotensinⅡ(AngⅡ),endothelin 1(ET-1)and plasma renin activity(PRA)were tested by enzyme-linked immunosorbent assay(ELISA)and radioimmunoassay,respectively.The effect of QXJYD on VSR was determined by examining the media thickness and the ex vivo contractility of thoracic aortic.The proliferation and fibrosis of vascular smooth muscle cells(VSMCs)were examined via immunohistochemical(IHC)staining for proliferating cell nuclear antigen(PCNA),collagen Ⅰ and collagen Ⅲ,respectively.The mRNA and protein expressions of transforming growth factor β1(TGF-β1),Smad3 and phosphorylation of Smad3 in thoracic aorta tissues were determined by real-time polymerase chain reaction(PCR)and Western blot assay,respectively.Results:QXJYD treatment led to a significant decrease of the elevation of blood pressure in SHRs and reduced the levels of AngⅡ,ET-1 and PRA in the serum(P<0.05).In addition,QXJYD treatment remarkably ameliorated VSR and vascular function in SHRs.Moreover,QXJYD inhibited VSMC proliferation and fibrosis by suppressing the expression of PCNA,collagen Ⅰ and collagen Ⅲ in thoracic aortic.Furthermore,QXJYD inhibited the expression of TGF-β1,Smad3 and the phosphorylation of Smad3,respectively(P<0.05).Conclusion:QXJYD reversed VSR by inhibiting VSMC proliferation and collagen deposition via regulation of TGF-β1/Smad signaling pathway,which may,in part,illuminate its anti-hypertensive activities.HE Fei CHU Jian-feng CHEN Hong-wei LIN Wei LIN Shan CHEN You-qin PENG Jun CHEN Ke-ji 2020Chinese Journal of Integrative Medicine2020,26,3:12

17Effect of initial stent position on patency of transjugular intrahepatic portosystemic shunt显示文摘AIM To evaluate the effect of initial stent position on transjugular intrahepatic portosystemic shunt(TIPS).METHODS We studied 425 patients from January 2004 to January 2015 with refractory ascites or variceal bleeding who required TIPS placement. Patients were randomly divided into group A(stent in hepatic vein, n = 57), group B(stent extended to junction of hepatic vein and inferior vena cava, n = 136), group C(stent in left branch of portal vein, n = 83) and group D(stent in main portal vein, n = 149). Primary unassisted patency was compared using Kaplan-Meier analysis, and incidence of recurrence of bleeding, ascites and hepatic encephalopathy(HE) were analyzed.RESULTS The mean primary unassisted patency rate in group B tended to be higher than in group A at 3, 6 and 12 mo(P = 0.001, 0.000 and 0.005), and in group D it tended to be lower than in group C at 3, 6 and 12 mo(P = 0.012, 0.000 and 0.028). The median shunt primary patency time for group A was shorter than for group B(5.2 mo vs 9.1 mo, 95%CI: 4.3-5.6, P = 0.013, logrank test), while for group C it was longer than for group D(8.3 mo vs 6.9 mo, 95%CI: 6.3-7.6, P = 0.025, log-rank test). Recurrence of bleeding and ascites in group A was higher than in group B at 3 mo(P = 0.014 and 0.020), 6 mo(P = 0.014 and 0.019) and 12 mo(P = 0.024 and 0.034. Recurrence in group D was higher than in group C at 3 mo(P = 0.035 and 0.035), 6 mo(P = 0.038 and 0.022) and 12 mo(P = 0.017 and 0.009). 18Spatial and temporal variation of fish assemblages in a subtropical small stream of the Huangshan Mountain显示文摘Spatial and temporal variation of fish assemblages were investigated seasonally from May 2007 to February 2008across 11 study sites in a subtropical small stream, the Puxi Stream, of the Huangshan Mountain. Along the longitudinal gradientfrom headwater to downstream, fish species richness and abundance increased gradually, but then decreased significantly at thelower reaches. The highest species richness and abundance were observed in August and the lowest in February. Based on analysisof similarities (ANOSIM), fish assemblages were significantly different in spatial variation but not in temporal variation. Althoughdifferences were observed both among sites and among stream orders, the lower R value in order-variation suggestedstream order was not the optimal factor explaining the spatial variation of fish assemblages. In addition, dam construction did notsignificantly alter fish assemblages in the sites adjacent to and immediately downstream to dams. Using cluster analysis andnon-metric Multi Dimensional Scaling analysis (NMS), assemblages were separated into three groups at a Bray-Curtis similarityvalue of 42%: the upper, middle and lower groups. Following analysis of similarity percentages of species contributions (SIMPER),shifts in occurrence or abundance of S. curriculus, Z. platypus, R. bitterling and A.fasciatus contributed most to the differencesamongst the three groups. Standard Deviation Redundancy Analysis (RDA) suggested that habitat structure (such as elevation,substrate, and flow velocity) contributed to the spatial and temporal pattern of fish assemblages in the Puxi Stream. In conclusion,the fish assemblages in Puxi Stream presented significant spatial but not temporal variation. Human disturbance has perhapsinduced the decrease in species diversity in the lower reaches. 19Osteoarthritis:pathogenic signaling pathways and therapeutic targets显示文摘Osteoarthritis(OA)is a chronic degenerative joint disorder that leads to disability and affects more than 500 million population worldwide.OA was believed to be caused by the wearing and tearing of articular cartilage,but it is now more commonly referred to as a chronic whole-joint disorder that is initiated with biochemical and cellular alterations in the synovial joint tissues,which leads to the histological and structural changes of the joint and ends up with the whole tissue dysfunction.Currently,there is no cure for OA,partly due to a lack of comprehensive understanding of the pathological mechanism of the initiation and progression of the disease.Therefore,a better understanding of pathological signaling pathways and key molecules involved in OA pathogenesis is crucial for therapeutic target design and drug development.In this review,we first summarize the epidemiology of OA,including its prevalence,incidence and burdens,and OA risk factors.We then focus on the roles and regulation of the pathological signaling pathways,such as Wnt/β-catenin,NF-κB,focal adhesion,HIFs,TGFβ/ΒΜP and FGF signaling pathways,and key regulators AMPK,mTOR,and RUNX2 in the onset and development of OA.In addition,the roles of factors associated with OA,including MMPs,ADAMTS/ADAMs,and PRG4,are discussed in detail.Finally,we provide updates on the current clinical therapies and clinical trials of biological treatments and drugs for OA.Research advances in basic knowledge of articular cartilage biology and OA pathogenesis will have a significant impact and translational value in developing OA therapeutic strategies.Qing Yao Xiaohao Wu Chu Tao Weiyuan Gong Mingjue Chen Minghao Qu Yiming Zhong Tailin He Sheng Chen Guozhi Xiao 2023Signal Transduction and Targeted Therapy2023,8,3:11
20Safety and efficacy of transjugular intrahepatic portosystemic shunt combined with palliative treatment in patients with hepatocellular carcinoma显示文摘BACKGROUND There is a close relationship between cirrhosis and hepatocellular carcinoma (HCC). Transjugular intrahepatic portosystemic shunt (TIPS) has good clinical effect in treating the complication of portal hypertension. However, because of the risk of postoperative liver failure, severe complications, and low survival rate for HCC, TIPS is contraindicated in patients with portal hypertension and liver cancer. We studied a large cohort of patients with cirrhosis and HCC who underwent TIPS for recurrent variceal bleeding and/or ascites. AIM To assess the safety, efficacy, and survival rate in patients with HCC who underwent TIPS. METHODS Group A comprised 217 patients with HCC and portal hypertension who underwent the TIPS procedure between 1999 and 2014. After TIPS deployment, these patients received palliative treatment for HCC. Group B comprised a cohort of 136 HCC patients with portal hypertension who did not undergo TIPS placement. Group B received palliative treatment for HCC plus medical therapy for portal hypertension. The clinical outcomes and survival rate were assessed. RESULTS In Group A, the primary technical success rate was 97.69% for TIPS placement,and no severe procedure-related complications of TIPS placement were reported. The control of variceal bleeding (VB) within 1 mo did not differ significantly between the groups (P = 0.261). Absorption of refractory ascites within 1 mo, recurrence of VB, and recurrence of refractory ascites differed significantly between the groups (P = 0.017, 0.023, and 0.009, respectively). By comparison, the rate of hepatic encephalopathy in Group B was lower than that in Group A (P = 0.036). The 1-, 2-, 3-, 4-, and 5-year survival rates were significantly different between Groups A and B (χ^2 = 12.227, P = 0.018;χ^2 = 12.457, P = 0.014;χ^2 = 26.490, P = 0.013;χ^2 = 21.956, P = 0.009, and χ^2 = 24.596, P = 0.006, respectively). The mean survival time was 43.7 mo in Group A and 31.8 mo in Group B. Median survival time was 50.0 mo in Group A and 33.0 mo in Group B. Mean and median survival differed significantly between the two groups (P = 0.000,χ^2 = 35.605, log-rank test). The mortality rate from VB in Group A was higher than that in Group B (P = 0.006), but the rates of hepatic tumor, hepatic failure, and multiorgan failure did not differ significantly between the two groups (P = 0.173, 0.246 and 0.257, respectively).
