1Sodium oligomannate therapeutically remodels gut microbiota and suppresses gut bacterial amino acids-shaped neuroinflammation to inhibit Alzheimer's disease progression显示文摘Recently,increasing evidence has suggested the association between gut dysbiosis and Alzheimer's disease(AD)progression,yet the role of gut microbiota in AD pathogenesis remains obscure.Herein,we provide a potential mechanistic link between gut microbiota dysbiosis and neuroinflammation in AD progression.Using AD mouse models,we discovered that,during AD progression,the alteration of gut microbiota composition leads to the peripheral accumulation of phenylalanine and isoleucine,which stimulates the differentiation and proliferation of pro-inflammatory T helper 1(Thl)cells.The brain-infiltrated peripheral Th1 immune cells are associated with the Ml microglia aaivation,contributing to AD-associated neuroinflammation.Importantly,the elevation of phenylalanine and isoleucine concentrations and the increase of Th1 cell frequency in the blood were also observed in two small independent cohorts of patients with mild cognitive impairment(MCI)due to AD.Furthermore,GV-971,a sodium oligomannate that has demonstrated solid and consistent cognition improvement in a phase 3 clinical trial in China,suppresses gut dysbiosis and the associated phenylalanine/isoleucine accumulation,harnesses neuroinflammation and reverses the cognition impairment.Together,our findings highlight the role of gut dysbiosis-promoted neuroinflammation in AD progression and suggest a novel strategy for AD therapy by remodelling the gut microbiota.Xinyi Wang Guangqiang Sun Teng Feng Jing Zhang Xun Huang Tao Wang Zuoquan Xie Xingkun Chu Jun Yang Huan Wang Shuaishuai Chang Yanxue Gong Lingfei Ruan Guanqun Zhang Siyuan Yan Wen Lian Chen Du Dabing Yang Qingli Zhang Feifei Lin Jia Liu Haiyan Zhang Changrong Ge Shifu Xiao Jian Ding Meiyu Geng 2019Cell Research2019,29,10:192
2Contrasting trends of PM2.5 and surface-ozone concentrations in China from 2013 to 2017显示文摘Although much attention has been paid to investigating and controlling air pollution in China,the trends of air-pollutant concentrations on a national scale have remained unclear.Here,we quantitatively investigated the variation of air pollutants in China using long-term comprehensive data sets from 2013 to 2017,during which Chinese government made major efforts to reduce anthropogenic emission in polluted regions.Our results show a significant decreasing trend in the PM2.5 concentration in heavily polluted regions of eastern China,with an annual decrease of〜7%compared with measurements in 2013.The measured decreased concentrations of SO2>NO2 and CO(a proxy for anthropogenic volatile organic compounds)could explain a large fraction of the decreased PM2.5 concentrations in different regions.As a consequence;the heavily polluted days decreased significantly in corresponding regions.Concentrations of organic aerosol;nitrate;sulfate,ammonium and chloride measured in urban Beijing revealed a remarkable reduction from 2013 to 2017;connecting the decreases in aerosol precursors with corresponding chemical components closely.However;surface-ozone concentrations showed increasing trends in most urban stations from 2013 to 2017;which indicates stronger photochemical pollution.The boundary-layer height in capital c让ies of eastern China showed no significant trends over the Beijing-Tianjin-Hebei,Yangtze River Delta and Pearl River Delta regions from 2013 to 2017;which confirmed the reduction in anthropogenic emissions.Our results demonstrated that the Chinese government was successful in the reduction of particulate matter in urban areas from 2013 to 2017,although the ozone concentration has increased significantly;suggesting a more complex mechanism of improving Chinese air quality in the future.Yonghong Wang Wenkang Gao Shuai Wang Tao Song Zhengyu Gong Dongsheng Ji Lili Wang Zirui Liu Guiqian Tang Yanfeng Huo Shili Tian Jiayun Li Mingge Li Yuan Yang Biwu Chu Tuukka Petaja Veli-Matti Kerminen Hong He Jiming Hao Markku Kulmala Yuesi Wang Yuanhang Zhang 2020National Science Review2020,7,8:51

4Root microbiota shift in rice correlates with resident time in the field and developmental stage显示文摘Land plants in natural soil form intimate relationships with the diverse root bacterial microbiota. A growing body of evidence shows that these microbes are important for plant growth and health. Root microbiota composition has been widely studied in several model plants and crops; however, little is known about how root microbiota vary throughout the plant's life cycle under field conditions. We performed longitudinal dense sampling in field trials to track the time-series shift of the root microbiota from two representative rice cultivars in two separate locations in China. We found that the rice root microbiota varied dramatically during the vegetative stages and stabilized from the beginning of the reproductive stage, after which the root microbiota underwent relatively minor changes until rice ripening. Notably, both rice genotype and geographical location influenced the patterns of root microbiota shift that occurred during plant growth. The relative abundance of Deltaproteobacteria in roots significantly increased overtime throughout the entire life cycle of rice, while that of Betaproteobacteria, Firmicutes, and Gammaproteobacteria decreased. By a machine learning approach, we identified biomarker taxa and established a model to correlate root microbiota with rice resident time in the field(e.g., Nitrospira accumulated from 5 weeks/tillering in field-grown rice). 6Relationship between cell adhesion molecules expression and the biological behavior of gastric carcinoma显示文摘AIM: To evaluate the relationship between the expression of cell adhesion molecules (CAMs) and the biological behavior of gastric carcinoma. METHODS: Expression of syndecan-1, E-cadherin and integrin β3 were evaluated by immunohistochemical study in a total of 118 gastric carcinomas and 20 non- tumor gastric mucosas. RESULTS: The expressions of syndecan-1 and E-cadherin were significantly lower in gastric carcinoma compared to non-tumor gastric mucosa, and the low expression rates were positively correlated to the tumor invasion depth, vessel invasion, lymph node metastasis and distant metastasis (P < 0.01 in all cases). However, the expression of integrin β3 was significantly higher in gastric carcinoma compared to non-tumor gastric mucosa, and the high expression rates were positively correlated to the tumor invasion depth, vessel invasion, lymph node metastasis and distant metastasis (P < 0.01 in all cases). In addition, the three protein expressions were correlated to the tumor growth pattern (P < 0.01, P < 0.01, and P < 0.05 respectively), but not correlated to tumor differentiation (P > 0.05, P > 0.05 and P > 0.05 respectively). Positive correlation was observed between the expressions of syndecan-1 and E-cadherin, but they which were negatively correlated to the expression of integrin β3 (P < 0.01 in all cases). Univariate analysis demonstrated that the mean survival time and 5-year survival rate were lower in the cases with low expressions of syndecan-1 and E-cadherin and high expression of integrin β3 (P < 0.01, in all cases). COX multivariate analysis showed that the expression level of syndecan-1 could be an independent prognostic index of gastric carcinoma (P < 0.01), whereas E-cadherin and integrin β3 could not be independent indexes (P > 0.05, P > 0.05 respectively). CONCLUSION: The low expression of syndecan-1 and E-cadherin and the high expression of integrin β3 are significantly correlated with the invasion and metastasis of gastric carcinoma, and they are highly correlated with each other. 7Effects of continuously regenerating diesel particulate filters on regulated emissions and number-size distribution of particles emitted from a diesel engine显示文摘The effects of continuously regenerating diesel particulate filter (CRDPF) systems on regulated gaseous emissions, and number-size distribution and mass of particles emanated from a diesel engine have been investigated in this study. Two CRDPF units (CRDPF-1 and CRDPF-2) with different specifications were separately retrofitted to the engine running with European steady-state cycle (ESC). An electrical low pressure impactor (ELPI) was used for particle number-size distribution measurement and mass estimation. The conversion/reduction rate (R CR ) of hydrocarbons (HC) and carbon monoxide (CO) across CRDPF-1 was 83% and 96.3%, respectively. Similarly, the R CR of HC and CO and across CRDPF-2 was 91.8% and 99.1%, respectively. The number concentration of particles and their concentration peaks; nuclei mode, accumulation mode and total particles; and particle mass were highly reduced with the CRDPF units. The nuclei mode particles at downstream of CRDPF-1 and CRDPF-2 decreased by 99.9% to 100% and 97.8% to 99.8% respectively; and the particle mass reduced by 73% to 92.2% and 35.3% to 72.4%, respectively, depending on the engine conditions. 8Feasibility of endoscopic treatment and predictors of lymph node metastasis in early gastric cancer显示文摘BACKGROUND Endoscopic submucosal dissection (ESD) has been routinely performed in applicable early gastric cancer (EGC) patients as an alternative to conventional surgical operations that involve lymph node dissection. The indications for ESD have been recently expanded to include larger, ulcerated, and undifferentiated mucosal lesions, and differentiated lesions with slight submucosal invasion. The risk of lymph node metastasis (LNM) is the most important consideration when deciding on a treatment strategy for EGC. Despite the advantages over surgical procedures, lymph nodes cannot be removed by ESD. In addition, whether patients who meet the expanded indications for ESD can be managed safely remains controversial. AIM To determine whether the ESD indications are applicable to Chinese patients and to investigate the predictors of LNM in EGC. METHODS We retrospectively analyzed 12552 patients who underwent surgery for gastric cancer between June 2007 and December 2018 at the Affiliated Hospital of Qingdao University. A total of 1262 (10.1%) EGC patients were eligible for inclusion in this study. Data on the patients’ clinical, endoscopic, and histopathological characteristics were collected. The absolute and expanded indications for ESD were validated by regrouping the enrolled patients and determining the positive LNM results in each subgroup. Predictors of LNM in patients were evaluated by univariate and multivariate analyses. RESULTS LNM was observed in 182 (14.4%) patients. No LNM was detected in the patients who met the absolute indications (0/90). LNM occurred in 4/311 (1.3%) patients who met the expanded indications. According to univariate analysis, LNM was significantly associated with positive tumor marker status, medium (20-30 mm) and large (>30 mm) lesion sizes, excavated macroscopic-type tumors, ulcer presence, submucosal invasion (SM1 and SM2), poor differentiation, lymphovascular invasion (LVI), perineural invasion, and diffuse and mixed Lauren’s types. Multivariate analysis demonstrated SM1 invasion (odds ration [OR]= 2.285, P = 0.03), SM2 invasion (OR = 3.230, P < 0.001), LVI (OR = 15.702, P < 0.001), mucinous adenocarcinoma (OR = 2.823, P = 0.015), and large lesion size (OR = 1.900, P = 0.006) to be independent risk factors. CONCLUSION The absolute indications for ESD are reasonable, and the feasibility of expanding the indications for ESD requires further investigation. 9Preparation, characterization and salt-resistance of a coal based super absorbent composite显示文摘Potassium humate was extracted from brown coal. A novel super absorbent composite, poly (acrylic acid-co-acryla-mide)/potassium humate (PAA-AM/KHA), was prepared by graft polymerization of acrylic acid, acrylamide and coal based potas-sium humate using N, N’-methylenebisacrylamide as a crosslinker and potassium peroxydisulfate as an initiator. The effects of reaction temperature, degree of neutralization of the poly (acrylic acid) and the amounts of crosslinker, initiator and potassium hu-mate were investigated. Salt resistance tests were also carried out. The composite prepared under optimal conditions had a potas-sium humate content of 10% and exhibited a water absorption of 770 g/g in distilled water, and 349, 286 and 41 g/g in 0.5 mol/L KCl, MgCl2 and AlCl3 solutions respectively. The results indicate that the salt resistance of PAA-AM/KHA was superior to that of poly (acrylic acid-co-acrylamide) because of the collaborative effect of functional groups of the coal based potassium humate. The PAA-AM/KHA micro powder was characterized by IR spectroscopy and the micrographic surface was characterized by scanning electron microscopy. Introduction of potassium humate into the poly (acrylic acid-co-acrylamide) structure creates a composite more suitable for use as a water-managing material in the renewal of arid and desert environments. 10Treatment of hepatic cysts by B-ultrasound-guided radiofrequency ablation显示文摘BACKGROUND:The traditional therapy for hepatic cysts has limited success because of recrudescence. Radiofrequency ablation(RFA)has become popular because of its advantages including little damage,therapeutic effect and reduced suffering.This report describes the effects and reliability of RFA in the treatment of 29 patients with hepatic cysts. METHODS:B-ultrasound-guided RFA was used to treat hepatic mono-cyst or multi-cysts of 29 patients(63 tumors). Ablative efficiency and complications were assessed by imaging and clinical symptoms. RESULTS:The tumors were abated completely in 34 cysts with a diameter<5 cm and no recurrence was seen after 3 months.In 21 cysts with a diameter of 5-10 cm,tumor volume was decreased by over 70%,then reduction and fiberosis were found.In 8 cysts with a diameter greater than 10 cm,tumor volume was decreased by more than 60%,and in 2 cysts it was increased more slightly than that at 1 month after RFA.In subsequent follow-up(6 and 12 months after RFA),tumors<10 cm in diameter were fully ablated.No significant discomfort and complications were found in any patient. 11Genetic Analysis and Primary Mapping of pms4, a Photoperiod-Sensitive Genic Male Sterility Gene in Rice (Oryza sativa)显示文摘To understand the genetic characteristics of a new photoperiod-sensitive genic male sterile line Mian 9S, some reciprocal crosses were made between Mian 9S and six indica rice materials, Yangdao 6, Luhui 602, Shuihui 527, Mianhui 725, Fuhui 838 and Yixiang 1B. Genetic analysis results suggested that the photoperiod-sensitive genic male sterility (PGMS) of Mian 9S was controlled by a single recessive nuclear gene. Thus, the F2 population derived from the cross of Yangdao 6/Mian 9S was used to map the PGMS gene in Mian 9S. By using SSR markers, the PGMS gene of Mian 9S was mapped on one side of the markers, RM6659 and RM1305, on rice chromosome 4, with the genetic distances of 3.0 cM and 3.5 cM, respectively. The gene was a novel PGMS gene and designated tentatively as pms4. 13Spatial and temporal variation of fish assemblages in a subtropical small stream of the Huangshan Mountain显示文摘Spatial and temporal variation of fish assemblages were investigated seasonally from May 2007 to February 2008across 11 study sites in a subtropical small stream, the Puxi Stream, of the Huangshan Mountain. Along the longitudinal gradientfrom headwater to downstream, fish species richness and abundance increased gradually, but then decreased significantly at thelower reaches. The highest species richness and abundance were observed in August and the lowest in February. Based on analysisof similarities (ANOSIM), fish assemblages were significantly different in spatial variation but not in temporal variation. Althoughdifferences were observed both among sites and among stream orders, the lower R value in order-variation suggestedstream order was not the optimal factor explaining the spatial variation of fish assemblages. In addition, dam construction did notsignificantly alter fish assemblages in the sites adjacent to and immediately downstream to dams. Using cluster analysis andnon-metric Multi Dimensional Scaling analysis (NMS), assemblages were separated into three groups at a Bray-Curtis similarityvalue of 42%: the upper, middle and lower groups. Following analysis of similarity percentages of species contributions (SIMPER),shifts in occurrence or abundance of S. curriculus, Z. platypus, R. bitterling and A.fasciatus contributed most to the differencesamongst the three groups. Standard Deviation Redundancy Analysis (RDA) suggested that habitat structure (such as elevation,substrate, and flow velocity) contributed to the spatial and temporal pattern of fish assemblages in the Puxi Stream. In conclusion,the fish assemblages in Puxi Stream presented significant spatial but not temporal variation. Human disturbance has perhapsinduced the decrease in species diversity in the lower reaches. 14Confirmation of pelitic granulite in the Altai orogen and its geological significance显示文摘The existence of pelitic granulite in the Altai orogen was confirmed for the first time by detailed petrographic research and P-T pseudosection modeling. The pelitic granulite has the assemblage of garnet + cordierite + K-feldspar + biotite + sillimanite + plagioclase + quartz with some samples containing the paragenesis of cordierite + spinel. Peak conditions of the pelitic granulite determined from the P-T pseudosection involved P = 0.5―0.6 GPa, T = 780―800℃, belonging to medium-to low-pressure type. SHRIMP U-Pb dating of zircon presented a metamorphic age of 292.8 ± 2.3 Ma. 15Osteoarthritis:pathogenic signaling pathways and therapeutic targets显示文摘Osteoarthritis(OA)is a chronic degenerative joint disorder that leads to disability and affects more than 500 million population worldwide.OA was believed to be caused by the wearing and tearing of articular cartilage,but it is now more commonly referred to as a chronic whole-joint disorder that is initiated with biochemical and cellular alterations in the synovial joint tissues,which leads to the histological and structural changes of the joint and ends up with the whole tissue dysfunction.Currently,there is no cure for OA,partly due to a lack of comprehensive understanding of the pathological mechanism of the initiation and progression of the disease.Therefore,a better understanding of pathological signaling pathways and key molecules involved in OA pathogenesis is crucial for therapeutic target design and drug development.In this review,we first summarize the epidemiology of OA,including its prevalence,incidence and burdens,and OA risk factors.We then focus on the roles and regulation of the pathological signaling pathways,such as Wnt/β-catenin,NF-κB,focal adhesion,HIFs,TGFβ/ΒΜP and FGF signaling pathways,and key regulators AMPK,mTOR,and RUNX2 in the onset and development of OA.In addition,the roles of factors associated with OA,including MMPs,ADAMTS/ADAMs,and PRG4,are discussed in detail.Finally,we provide updates on the current clinical therapies and clinical trials of biological treatments and drugs for OA.Research advances in basic knowledge of articular cartilage biology and OA pathogenesis will have a significant impact and translational value in developing OA therapeutic strategies.Qing Yao Xiaohao Wu Chu Tao Weiyuan Gong Mingjue Chen Minghao Qu Yiming Zhong Tailin He Sheng Chen Guozhi Xiao 2023Signal Transduction and Targeted Therapy2023,8,3:11

17Human organoids in basic research and clinical applications显示文摘Organoids are three-dimensional(3D)miniature structures cultured in vitro produced from either human pluripotent stem cells(hPSCs)or adult stem cells(AdSCs)derived from healthy individuals or patients that recapitulate the cellular heterogeneity,structure,and functions of human organs.The advent of human 3D organoid systems is now possible to allow remarkably detailed observation of stem cell morphogens,maintenance and differentiation resemble primary tissues,enhancing the potential to study both human physiology and developmental stage.As they are similar to their original organs and carry human genetic information,organoids derived from patient hold great promise for biomedical research and preclinical drug testing and is currently used for personalized,regenerative medicine,gene repair and transplantation therapy.In recent decades,researchers have succeeded in generating various types of organoids mimicking in vivo organs.Herein,we provide an update on current in vitro differentiation technologies of brain,retinal,kidney,liver,lung,gastrointestinal,cardiac,vascularized and multi-lineage organoids,discuss the differences between PSC-and AdSC-derived organoids,summarize the potential applications of stem cell-derived organoids systems in the laboratory and clinic,and outline the current challenges for the application of organoids,which would deepen the understanding of mechanisms of human development and enhance further utility of organoids in basic research and clinical studies.Xiao-Yan Tang Shanshan Wu Da Wang Chu Chu Yuan Hong Mengdan Tao Hao Hu Min Xu Xing Guo Yan Liu 2022Signal Transduction and Targeted Therapy2022,7,6:10
18Unsuspected Gallbladder Cancer During or After Laparoscopic Cholecystectomy显示文摘Objective To summarize the clinical features and outcomes of unsuspected gallbladder carcinoma (UGC) detected during or after laparoscopic cholecystectomy.Methods Medical records of 8005 patients,who underwent laparoscopic cholecystectomy in Peking Union Medical College Hospital between June 1993 and June 2011,were reviewed.Patients that pathologically diagnosed as UGC were retrospectively studied in terms of clinical features,preoperative and postoperative diagnosis,surviving period,and complications.Results In the 8005 patients who received laparoscopic cholecystectomy,36 (0.45%) were diagnosed as UGC during (25 patients) or after (11 patients) laparoscopic cholecystectomy.The gallbladder cancer was staged as T1 in 16 patients,T2 in 11 patients,and T3 in 9 patients.The 1-,3-,and 5-year survival rates of all the patients were 88.9% (32/36),63.9% (23/36),and 58.3% (21/36).The 5-year survival rates in T1 stage,T2 stage,and T3 stage patients were 100%,75.0%,and 0.0%,respectively.Conclusions The survival rate of UGC is associated with tumor stage,not with operation approaches.Laparoscopic cholecystectomy is appropriate for T1 patients.Xie-qun Xu Wei Liu Bing-lu Li Tao Hong Chao-ji Zheng Chu Wang Yu-pei Zhao 2013Chinese Medical Sciences Journal2013,28,2:10
19Efficient adiabatic silicon-on-insulator waveguide taper显示文摘A silicon-on-insulator-based adiabatic waveguide taper with a high coupling efficiency and small footprint is presented.The taper was designed to reduce the incidence of mode conversion to higher-order and radiation modes inside the waveguide.In connecting a 0.5-μm-wide output waveguide and a 12-μm-wide input waveguide of a grating coupler,a compact 120-μm-long taper was demonstrated,achieving a transmission of 98.3%.Previously,this transmission level could only be achieved using a conventional linear taper with a length of more than 300μm.Yunfei Fu Tong Ye Weijie Tang Tao Chu 2014Photonics Research2014,2,3:10
20Microalgae:A potential alternative to health supplementation for humans显示文摘Microalgae has been consumed in human diet for thousands of years.It is an under-exploited crop for production of dietary foods.Microalgae cultivation does not compete with land and resources required for traditional crops and has a superior yield compared to terrestrial crops.Its high protein content has exhibited a huge potential to meet the dietary requirements of growing population.Apart from being a source of protein,presence of various bio-active components in microalgae provide an added health benefit.This review describes various microalgal sources of proteins and other bio-active components.One of the heavily studied group of bio-active components are pigments due to their anticarcenogenic,antioxidative and antihypertensive properties.Compared to various plant and floral species,microalgae contain higher amounts of pigments.Microalgal derived proteins have complete Essential Amino Acids(EAA)profiles and their protein content is higher than conventional sources such as meat,poultry and dairy products.However,microalgal based functional foods have not flooded the market.The lack of awareness coupled with scarce incentives for producers result in under-exploitation of microalgal potential.Application of microalgal derived components as dietary and nutraceutical supplements is discussed comprehensively.Apurav Krishna Koyande Kit Wayne Chew Krishnamoorthy Rambabu Yang Tao Dinh-Toi Chu Pau-Loke Show 2019Food Science and Human Wellness2019,8,1:10
