| 1 | Expression and self-assembly of Heterocapsa circularisquama RNA virus-like particles synthesized in Pichia pastoris显示文摘Heterocapsa circularisquama RNA virus(HcRNAV) is the first single-stranded RNA virus to be characterized that infects dinoflagellates.The ability of HcRNAV coat protein(HcRNAV CP) to self-assemble into virus-like particles(VLPs) in vitro suggested that heterologous expression was possible,and that the VLPs might be ideal nanocontainers for the targeted delivery of genes and chemicals.In this paper,we report the expression of a codon-optimized HcRNAV 109 CP gene in Pichia pastoris and the production of self-assembled HcRNAV VLPs using large-scale fermentation.The HcRNAV 109 CP gene was synthesized according to the codon preference of P.pastoris and cloned into a pPICZA vector.The recombinant plasmid pPICZA-CPsyns was transformed into P.pastoris by electroporation.The resulting yeast colonies were screened by PCR and analyzed for protein expression by SDS polyacrylamide gel electrophoresis.After large-scale fermentation,the yield of HcRNAV CPsyns reached approximately 2.5 g L 1 within 4 d.The HcRNAV VLPs were purified using PEG precipitation followed by cesium chloride density gradient ultracentrifugation,and were subsequently analyzed using UV spectrophotometry and transmission electron microscopy.Fluorescence dye-labeled myoglobin was loaded into the cages of the HcRNAV VLPs and the encapsulation was confirmed by fluorescence spectroscopy.The results point to the possible utilization in pharmacology or nanotechnology of HcRNAV VLPs produced by P.pastoris fermentation. | WU YuanZheng KIM Wonduck KIM Si-Wou EOM Chi-Yong YANG HeTong SHIN Hyun-Jae | 2012 | Chinese Science Bulletin2012,57,25: | 2 |
| 2 | Influences of 1-methylcyclopropene-containing papers on the metabolisms of membrane lipids in Anxi persimmons during storage显示文摘Objectives:The aim of this work was to analyse the effects of 1-methylcyclopropene(1-MCP)treatment on the metabolisms of membrane lipids in postharvest Anxi persimmons during storage.Materials and methods:Anxi persimmon(Diospyros kaki L.f.cv.Anxi)fruits were treated by paper containing 1-MCP with a concentration of 1.35μl/l.The cellular membrane permeability was analysed by the electric conductivity meter.The activities of lipoxygenase(LOX),phospholipase(PLD)and lipase were determined by spectrophotometry.The component and relative amounts of membrane fatty acids were determined using gas chromatograph(GC).Results:The 1-MCP-treated Anxi persimmons manifested a lower electrolyte leakage rate,lower LOX,PLD and lipase activities,higher levels of unsaturated fatty acids(USFAs),higher ratio of USFAs to saturated fatty acids(SFAs)(U/S),higher index of USFAs(IUFA),but lower levels of SFAs.Conclusions:The degradation and the metabolisms of membrane lipids could be suppressed by 1-MCP treatment,which might be accountable for the delaying softening of postharvest Anxi persimmons during storage. | Hui Wang Guo Chen Lili Shi Hetong Lin Yihui Chen Yifen Lin Zhongqi Fan | 2020 | Food Quality and Safety2020,4,3: | 2 |

| 4 | A theoretical study of hydrogen-bonded molecular clusters of sulfuric acid and organic acids with amides显示文摘Amides,a series of significant atmospheric nitrogen-containing volatile organic compounds(VOCs),can participate in new particle formation(NPF)throught interacting with sulfuric acid(SA)and organic acids.In this study,we investigated the molecular interactions of formamide(FA),acetamide(AA),N-methylformamide(MF),propanamide(PA),Nmethylacetamide(MA),and N,N-dimethylformamide(DMF)with SA,acetic acid(HAC),propanoic acid(PAC),oxalic acid(OA),and malonic acid(MOA).Global minimum of clusters were obtained through the association of the artificial bee colony(ABC)algorithm and density functional theory(DFT)calculations.The conformational analysis,thermochemical analysis,frequency analysis,and topological analysis were conducted to determine the interactions of hydrogen-bonded molecular clusters.The heterodimers formed a hepta or octa membered ring through four different types of hydrogen bonds,and the strength of the bonds are ranked in the following order:SO-H···O>CO-H···O>N-H···O>C-H···O.We also evaluated the stability of the clusters and found that the stabilization effect of amides with SA is weaker than that of amines with SA but stronger than that of ammonia(NH 3)with SA in the dimer formation of nucleation process.Additionally,the nucleation capacity of SA with amides is greater than that of organic acids with amides. | Chenpeng Zuo Xianwei Zhao Hetong Wang Xiaohui Ma Siyuan Zheng Fei Xu Qingzhu Zhang | 2021 | Journal of Environmental Sciences2021,33,2: | 2 |

| 8 | Effectiveness of Jiedu granule(解毒颗粒) on gut microbiota in patients with advanced hepatocellular carcinoma:a randomized controlled trial显示文摘OBJECTIVE:To observe whether Jiedu granule(解毒颗粒,JDG)modulates the composition of the gut microbiota during advanced hepatocellular carcinoma(HCC).METHODS:A randomized controlled trial was conducted.Sixty-two advanced HCC participants were randomly allocated to receive JDG or placebo.The median overall survival(OS)times of patients and the variation of relative abundance of bacteria over time were used as main outcome measures.RESULTS:Patients who received JDG demonstrated significantly longer median OS times compared with the placebo group.Pyrosequencing of the V3 regions of 16S r RNA genes revealed a dose dependent deviation of gut microbiota in response to JDG treatment.Redundancy analysis identified Clostridium XI and Peptostreptococcaceae which related to the onset of liver cancer disappeared after 1-month and 2-month JDG treatment,while in control group,no significant changes of these two bacteria were found.The variation tendency of relative abundance of Bacteroides(essential in immunoblocking therapy of tumor)in JDG group was not obvious while in control group,it was decreased significantly with time.The relative abundance of Roseburia(correlated with the occurrence of liver cancer)was increased in JDG group and was decreased in control group over time.CONCLUSION:Changes in the gut microbiota may be associated with the efficiency of JDG on survival period of advanced HCC patients.Trial registration:Chinese Clinical TRIAL Registry Chi CTR-OOC-16008002. | FAN Yifu ZHAO Hetong ZHANG Yani YIN Zifei DU Juan LING Changquan | 2022 | Journal of Traditional Chinese Medicine2022,42,6: | 1 |

| 11 | Carbon dots enhanced gelatin/chitosan bio-nanocomposite packaging film for perishable foods显示文摘Carbon dots(CDs)with intriguing fluorescent property,good biocompatibility,high stability,easy interaction with substrates,are burgeoning carbon nanoparticles with large potential in various applications.Incorporating CDs into the polymer matrix is becoming a popular strategy to endow the complex with new functions.Herein,the green-synthesized CDs was integrated into the mixture of gelatin(derived from waste fish scale)and chitosan,and a multifunctional bio-nanocomposite(defined as Gelatin/Chitosan/CDs)film was developed,which showed the excellent antibacterial,antioxidant,pH-sensitivity,UV shielding,and blue-emission properties.The effects of different concentrations of CDs on the physical,mechanical,structural,and functional activity of bio-nanocomposite film were tested.Compared with the Gelatin/Chitosan film,the Gelatin/Chitosan/CDs film with an optimum addition of 20%CDs showed the enhanced antibacterial,antioxidant as well as UV shielding activities.More importantly,it was used as an effective packaging material for fish meat preservation,reducing the loss of nutritional quality consumption,extending the shelf life of food.Besides,the bio-nanocomposite films also possessed the anti-counterfeiting and pH-responsive properties due to the strong fluorescent emission of CDs,and had the great potential in developing the intelligent packaging materials.Our work shed new light on the new application of CDs and the synthesis of bio-nanocomposite film in food industry. | Bofei Fu Qiaoling Liu Minghuan Liu Xiaofang Chen Hetong Lin Zongping Zheng Junqiu Zhu Congjie Dai Xiaochen Dong Da-Peng Yang | 2022 | Chinese Chemical Letters2022,33,10: | 1 |
