1Molecular Characterization of a Leaf Senescence-Related Transcription Factor BrWRKY75 of Chinese Flowering Cabbage显示文摘WRKY is a plant-specific transcription factor(TF) involved in the regulation of many biological processes; however, its role in leaf senescence of leafy vegetables remains unknown. In the present work, a WRKY TF, termed Br WRKY75 was isolated from Chinese flowering cabbage [Brassica rapa L. ssp. chinensis(L.) Mokino var. utilis Tsen et Lee]. Analysis of deduced amino acid sequence and the phylogenetic tree showed that Br WRKY75 has high homology with WRKY75 from Brassica oleracea and Arabidopsis thaliana, and belongs to the II c sub-group. Sub-cellular localization and transcriptional activity analysis revealed that Br WRKY75 is a nuclear protein with transcriptional repression activity, and was up-regulated during leaf senescence. Electrophoretic mobility shift assay confirmed that Br WRKY75 directly bound to the W-box(TTGAC) cis-element. 2A NAC transcription factor, NOR-like1, is a new positive regulator of tomato fruit ripening显示文摘Ripening of the model fruit tomato(Solanum lycopersicum)is controlled by a transcription factor network including NAC(NAM,ATAF1/2,and CUC2)domain proteins such as No-ripening(NOR),SlNAC1,and SlNAC4,but very little is known about the NAC targets or how they regulate ripening.Here,we conducted a systematic search of fruit-expressed NAC genes and showed that silencing NOR-like1(Solyc07g063420)using virus-induced gene silencing(VIGS)inhibited specific aspects of ripening.Ripening initiation was delayed by 14 days when NOR-like1 function was inactivated by CRISPR/Cas9 and fruits showed obviously reduced ethylene production,retarded softening and chlorophyll loss,and reduced lycopene accumulation.RNA-sequencing profiling and gene promoter analysis suggested that genes involved in ethylene biosynthesis(SlACS2,SlACS4),color formation(SlGgpps2,SlSGR1),and cell wall metabolism(SlPG2a,SlPL,SlCEL2,and SlEXP1)are direct targets of NOR-like1.Electrophoretic mobility shift assays(EMSA),chromatin immunoprecipitation-quantitative PCR(ChIP-qPCR),and dual-luciferase reporter assay(DLR)confirmed that NOR-like1 bound to the promoters of these genes both in vitro and in vivo,and activated their expression.Our findings demonstrate that NOR-like1 is a new positive regulator of tomato fruit ripening,with an important role in the transcriptional regulatory network.Ying Gao Wei Wei Xiaodan Zhao Xiaoli Tan Zhongqi Fan Yiping Zhang Yuan Jing Lanhuan Meng Benzhong Zhu Hongliang Zhu Jianye Chen Cai-Zhong Jiang Donald Grierson Yunbo Luo Da-Qi Fu 2018Horticulture Research2018,5,1:9
3Adsorption of rubidium ion from aqueous solution by surface ion imprinted materials显示文摘A new type of rubidium ion-imprinted polymer has been synthesized by the surface-imprinting technique using methacrylic acid as the functional monomer,the rubidium ion as the template,methanol as the solvent,and silica as a carrier.Ethylene glycol dimethacrylate and 2,2-azobisisobutyronitrile were used as acrosslinker and an initiator,respectively.In addition,based on the macrocyclic effect of crown ethers,the 18-crown-6 ligand was introduced as a ligand to fix the template ions better.Scanning electron microscopy,zeta-potential analysis,Fourier transform infrared spectroscopy,thermogravimetric analysis,and X-ray photoelectron spectroscopy were performed to characterize the ion-imprinted polymer.The effects of the preparation and adsorption conditions on the adsorption performance of the rubidium ion-imprinted polymer were investigated.The results indicated that the rubidium ion-imprinted polymer has high selectivity and faster kinetics than other adsorbents,with an equilibrium adsorption capacity of 200.19 mg·g^(-1)at 298 K within 25 min.The sorption isotherm was well described by the Freundlich isotherm model,while the adsorption kinetics fitted the pseudo-second-order kinetic model.Consecutive adsorption-desorption experiments showed that the ion-imprinted polymer had good chemical stability and reusability.Zhongqi Ren Jie Wang Hewei Zhang Fan Zhang Shichao Tian Zhiyong Zhou 2023Chinese Journal of Chemical Engineering2023,54,2:3
4A Banana PHD-Type Transcription Factor MaPHD1 Represses a Cell Wall-Degradation Gene MaXTH6 during Fruit Ripening显示文摘Plant homeobox domain(PHD)-type transcription factors(TFs) are involved in a variety of biological processes. However, its involvement in commercially important fruit ripening process remains largely unclear. In the present work, the characterization of a PHD-type TF termed MaPHD1 from banana fruit is reported. Multiple alignments of the deduced amino acid sequence revealed that Ma PHD1 showed a high homology with Arabidopsis thaliana Alfin1-like proteins belonging to plant-specific sub-family of PHD finger proteins. MaPHD1 was found localized in the nucleus and exhibited trans-repression ability. It was down-regulated by ethylene and ripening. Electrophoretic Mobility Shift Assay(EMSA) and transient expression analysis demonstrated that Ma PHD1 directly bound to the G-rich motifs in the promoter of Ma XTH6, which is associated with cell wall degradation, and subsequently repressed its expression. These findings suggest that MaPHD1 may be negatively associated with banana fruit ripening, at least in part, by the direct suppression of Ma XTH6. 5Influences of 1-methylcyclopropene-containing papers on the metabolisms of membrane lipids in Anxi persimmons during storage显示文摘Objectives:The aim of this work was to analyse the effects of 1-methylcyclopropene(1-MCP)treatment on the metabolisms of membrane lipids in postharvest Anxi persimmons during storage.Materials and methods:Anxi persimmon(Diospyros kaki L.f.cv.Anxi)fruits were treated by paper containing 1-MCP with a concentration of 1.35μl/l.The cellular membrane permeability was analysed by the electric conductivity meter.The activities of lipoxygenase(LOX),phospholipase(PLD)and lipase were determined by spectrophotometry.The component and relative amounts of membrane fatty acids were determined using gas chromatograph(GC).Results:The 1-MCP-treated Anxi persimmons manifested a lower electrolyte leakage rate,lower LOX,PLD and lipase activities,higher levels of unsaturated fatty acids(USFAs),higher ratio of USFAs to saturated fatty acids(SFAs)(U/S),higher index of USFAs(IUFA),but lower levels of SFAs.Conclusions:The degradation and the metabolisms of membrane lipids could be suppressed by 1-MCP treatment,which might be accountable for the delaying softening of postharvest Anxi persimmons during storage.Hui Wang Guo Chen Lili Shi Hetong Lin Yihui Chen Yifen Lin Zhongqi Fan 2020Food Quality and Safety2020,4,3:2
6Comprehensive treatment of latex wastewater and resource utilization of concentrated liquid显示文摘Latex wastewater is a kind of refractory organic wastewater containing high concentrations of organics and ammonia nitrogen.In this work,the combined process of forward osmosis(FO)and reverse osmosis(RO)was designed to treat the latex wastewater in the whole process,achieving the water recovery rate of 99%and basically no waste discharge after the catalytic oxidation process.The turbidity of the latex wastewater was decreased to below 1 NTU by microfiltration pretreatment,and then using MgCl_2 worked as the draw solution for FO process to treat the latex wastewater.Different operation conditions including adding acid or scale inhibitor as the pretreatment methods were conducted to improve the treatment performance of the combined process.After the treatment of the whole process,the concentration of COD was less than 20 mg·L^(-1),the concentration of NH_3-N was less than 10 mg·L^(-1),and the concentration of TP was less than 0.5 mg·L^(-1)for the treated latex wastewater.The water quality met standards of industrial water reuse after the complete analysis of the treated latex wastewater,meanwhile,useful substances of L-Quebrachitol(L-Q)were successfully extracted from the concentrated solution.Therefore,the combined process of FO and RO could realize the efficient treatment and reuse of latex wastewater,which provided with some important guidance on the industrial application.Shichao Tian Yuming Tu Rujie Li Yufan Du Zhiyong Zhou Fan Zhang Zhongqi Ren 2023Chinese Journal of Chemical Engineering2023,57,5:0
7Enrichment of nervonic acid in Acer truncatum Bunge oil by combination of two-stage molecular distillation,one-stage urea complexation and five-stage solvent crystallization显示文摘Nervonic acid is the world’s first and only potent substance that can repair damaged nerve fibers and promote nerve cell regeneration with high nutritional value.The wide variety of fatty acids in plant oils and fats with similar structures makes the large-scale separation and purification of high-purity nervonic acid very difficult.A new combined process of molecular distillation,urea inclusion and solvent crystallization was established to prepare high-purity nervonic acid with the mixed fatty acids obtained after saponification and acidification of Acer truncatum Bunge oil as raw materials.First,according to the difference in the mean free path of fatty acids,molecular distillation was used to separate and remove C16 saturated fatty acid of palmitic acid and four C18-C20 fatty acids of stearic,oleic,linoleic,and linolenic acids.The content of C16-C20 fatty acids decreased from 72.92% to 19.22% after two-stage molecular distillation processes,in which the contents of saturated fatty acid of palmitic acid decreased to about 0.5%.Then,according to the difference in carbon chain length and saturation of fatty acid,the contents of C22-C24 saturated fatty acids of tetracosanoic and docosanoic acids decreased to 0.21% and 0.07% by urea inclusion with urea/free fatty acid preparation by saponification(SPOMFs)ratio as 0.6.In addition,all saturated fatty acids were basically separated.Finally,according to the difference in the solubility of fatty acids in solvents,the C18-C20 unsaturated fatty acids of oleic,linoleic,and linolenic acids and C22 unsaturated fatty acid of erucic acid were removed by solvent crystallization.The content of C18-C20 unsaturated fatty acids decreased to less than 5% with pentanol as the solvent after the first stage solvent crystallization.The content of erucic acid decreased to 3.47% with anhydrous ethanol as the solvent after the second to fifth stage solvent crystallization.The combined process of molecular distillation,urea inclusion and low temperature crystallization innovatively adopted an efficient,simple and easy-toindustrial solvent crystallization method to separate erucic and nervonic acids,obtaining nervonic acid with purity of 96.53% and final yield of 47.99%.Yingxi Gao Jiayi Shi Jie Wang Fan Zhang Shichao Tian Zhiyong Zhou Zhongqi Ren 2023Chinese Journal of Chemical Engineering2023,59,7:0
8VabHLH137 promotes proanthocyanidin and anthocyanin biosynthesis and enhances resistance to Colletotrichum gloeosporioides in grapevine显示文摘Proanthocyanidins(PAs)and anthocyanins are involved in the response of plants to various environmental stresses.However,the mechanism behind defense-induced PA biosynthetic regulation is still not completely elucidated,also in grapevine.This study performed a transcriptome sequencing analysis of grape berries infected with Colletotrichum gloeosporioides to highlight the induction of the VabHLH137 factor from the basic helix-loop-helix(bHLH)XII subfamily by the fungus,which appeared to be significantly co-expressedwith PA-related genes.The functional analysis of VabHLH137 overexpression and knockdownin transgenic grape calli showed that it positively regulated PA and anthocyanin biosynthesis.Moreover,VabHLH137 overexpression in the grape calli significantly increased resistance to C.gloeosporioides.A yeast one-hybrid and electrophoretic mobility shift assay revealed that VabHLH137 directly bound to the VaLAR2 promoter,enhancing its activity and interacting with VaMYBPAR,a transcriptional activator of PA biosynthesis.Furthermore,transient experiments showed that although the VabHLH137+VaMYBPAR complex activated VaLAR2 expression,it failed to further enhance VaLAR2 expression compared to VaMYBPAR alone.The findings indicated that VabHLH137 enhanced PA biosynthesis by activating of VaLAR2 expression,providing new insight into the transcriptional regulation of defense-induced PA biosynthesis in grapevine.Dan Yu Wei Wei Zhongqi Fan Jianye Chen Yilin You Weidong Huang Jicheng Zhan 2023Horticulture Research2023,10,2:0
9Carnosine concentration and expression profiles of carnosine related genes in Mytilus after beta-alanine injection显示文摘Carnosine and its analogues are histidine-containing dipeptides(HCDs)playing diverse functions in vertebrates.However,the distribution and the metabolism of carnosine in invertebrates are still unknown.In this study,Mytilus coruscus,a shellfish with important economic value in China,was selected for the investigation of HCD content and the expression profiling of carnosine-related genes in various mussel tissues.Quantification of HCD by amino acids analyzer revealed a low concentration of anserine in muscular tissues in Mytilus,indicating the presence of HCD even in an invertebrate.mRNA expression of five carnosine metabolic-related genes was profiled in various tissues,and the results highlighted the relative higher expression level of these genes in muscular tissues.Considering the fact that beta-alanine supplementation can increase the HCD content in vertebrates,a beta-alanine injection was performed and the changes of HCD concentration and the mRNA expression of carnosine related genes were investigated in five mussel tissues.The results revealed the increase of HCD concentration,as well as the up-regulated expression level of related genes,in tested tissues of beta-alanine injected mussels.Transcriptomic analysis for the whole soft tissue of mussel before and after beta-alanine injection were performed,and 3569 differential expression genes(DEGs)were identified in the beta-alanine injected group when compared to their expression levels in the control.These data indicated the complex eff ects of betaalanine on M.coruscus metabolism,and those DEGs enriched in pathways of cancers,muscle contraction,and tyrosine metabolism highlighted the possible functions of beta-alanine in cell proliferation,sports,and melanogenesis,respectively.Chen PAN Zhi LIAO Jianyu HE Zhongqi GU Chunyue WANG Meihua FAN Isabella BUTTINO Baoying GUO Xiaojun YAN Xiaolin ZHANG 2022Journal of Oceanology and Limnology2022,40,3:0

11Highly selective and green recovery of lithium ions from lithium iron phosphate powders with ozone显示文摘Since lithium iron phosphate cathode material does not contain high-value metals other than lithium,it is therefore necessary to strike a balance between recovery efficiency and economic benefits in the recycling of waste lithium iron phosphate cathode materials.Here,we describe a selective recovery process that can achieve economically efficient recovery and an acceptable lithium leaching yield.Adjusting the acid concentration and amount of oxidant enables selective recovery of lithium ions.Iron is retained in the leaching residue as iron phosphate,which is easy to recycle.The effects of factors such as acid concentration,acid dosage,amount of oxidant,and reaction temperature on the leaching of lithium and iron are comprehensively explored,and the mechanism of selective leaching is clarified.This process greatly reduces the cost of processing equipment and chemicals.This increases the potential industrial use of this process and enables the green and efficient recycling of waste lithium iron phosphate cathode materials in the future.Ruiqi Li Kang Li Wei Wang Fan Zhang Shichao Tian Zhongqi Ren Zhiyong Zhou 2023Frontiers of Chemical Science and Engineering2023,17,6:0
