1Quasi-real-time simulation of rotating drum using discrete element method with parallel GPU computing显示文摘Real-time simulation of industrial equipment is a huge challenge nowadays. The high performance and fine-grained parallel computing provided by graphics processing units (GPUs) bring us closer to our goals. In this article, an industrial-scale rotating drum is simulated using simplified discrete element method (DEM) without consideration of the tangential components of contact force and particle rotation. A single GPU is used first to simulate a small model system with about 8000 particles in realtime, and the simulation is then scaled up to industrial scale using more than 200 GPUs in a 1D domain-decomposition parallelization mode. The overall speed is about 1/11 of the real-time. 3Bifurcation analysis on full annular rub of a nonlinear rotor system显示文摘In this paper, analytical and numerical studies are carried out on the full annular rub motions of a nonlinear Jeffcott rotor. Transition sets of the synchronous full annular rub are given with the help of averaging method and constraint bifurcation theory to discuss the effects of system parameters on jump phenomena. Routh-Hurwitz criteria are employed to analyze the stability of synchronous full annular rub solution and determine the boundaries of static and Hopf bifurcations. 4A modified HLA-A^(*)0201-restricted CTL epitope from human oncoprotein (hPEBP4) induces more efficient antitumor responses显示文摘We previously identified human phosphatidylethanolamine-binding protein 4(hPEBP4)as an antiapoptotic protein with increased expression levels in breast,ovarian and prostate cancer cells,but low expression levels in normal tissues,which makes hPEBP4 an attractive target for immunotherapy.Here,we developed hPEBP4-derived immunogenic peptides for inducing antigen-specific cytotoxic T lymphocytes(CTLs)targeting breast cancer.A panel of hPEBP4-derived peptides predicted by peptide-MHC-binding algorithms was evaluated to characterize their HLAA2.1 affinity and immunogenicity.We identified a novel immunogenic peptide,P40–48(TLFCQGLEV),that was capable of eliciting specific CTL responses in HLA-A2.1/K^(b) transgenic mice,as well as in peripheral blood lymphocytes from breast cancer patients.Furthermore,amino-acid substitutions in the P40–48 sequence improved its immunogenicity against hPEBP4,a self-antigen,thus circumventing tolerance.We designed peptide analogs by preferred auxiliary HLA-A^(*)0201 anchor residue replacement,which induced CTLs that were crossreactive to the native peptide.Several analogs were able to stably bind to HLA-A^(*)0201 and elicit specific CTL responses better than the native sequence.Importantly,adoptive transfer of CTLs induced by vaccination with two analogs more effectively inhibited tumor growth than the native peptide.These data indicate that peptide analogs with high immunogenicity represent promising candidates for peptide-mediated therapeutic cancer vaccines.Weihong Sun Junyi Shi Jian Wu Junchu Zhang Huabiao Chen Yuanyuan Li Shuxun Liu Yanfeng Wu Zhigang Tian Xuetao Cao Nan Li 2018Cellular & Molecular Immunology2018,15,8:4
5Numerical investigation of granular flow similarity in rotating drums显示文摘The theory of flow similarity has not been well established for granular flows,in contrast to the case for conventional fluids,owing to a lack of reliable and general constitutive laws for their continuum description.A rigorous investigation of the similarity of velocity fields in different granular systems would be valuable to theoretical studies.However,experimental measurements face technological and physical problems.Numerical simulations that employ the discrete element method(DEM) may be an alternative to experiments by providing similar results,where quantitative analysis could be implemented with virtually no limitation.In this study,the similarity of velocity fields is investigated for the rolling regime of rotating drums by conducting simulations based on the DEM and using graphics processing units.For a constant Froude number,it is found that the particle-to-drum size ratio plays a dominant role in the determination of the velocity field,while the velocity field is much more sensitive to some material properties than to others.The implications of these findings are discussed in terms of establishing theoretical similarity laws for granular flows.Huabiao Qi Ji Xu Guangzheng Zhou Feiguo Chen Wei Ge Jinghai Li 2015Particuology2015,13,5:4
6In vitro inhibition of HIV-1 replication in autologous CD4*T cells indicates viral containment by multifactorial mechanisms显示文摘HIV-1-specific cytotoxic T lymphocytes(CTLs) and neutralizing antibodies(NAbs) are present during chronic infection, but the relative contributions of these effector mechanisms to viral containment remain unclear. Here, using an in vitro model involving autologous CD4+ T cells,primary HIV-1 isolates, HIV-1-specific CTLs, and neutralizing monoclonal antibodies, we show that b12, a potent and broadly neutralizing monoclonal antibody to HIV-1, was able to block viral infection when preincubated with virus prior to infection, but was much less effective than CTLs at limiting virus replication when added to infected cell cultures. However, the same neutralizing antibody was able to contain viruses by antibody-dependent cell-mediated virus inhibition in vitro,which was mediated by natural killer cells(NKs) and dependent on an Fc-Fc receptor interaction.Meanwhile, bulk CTLs from HIV-1 controllers were more effective in suppression of virus replication than those from progressors. These findings indicate that control of HIV-1 replication in activated CD4^+ T cells is ineffectively mediated by neutralizing antibodies alone, but that both CTLs and antibody-dependent NK-mediated immune mechanisms contribute to viral containment.
