1Microbial treatment in chronic constipation显示文摘Chronic functional constipation is a kind of common intestinal disease that occurs in children, adults and elderly people. This disease not only causes great influence to physiological function, but also results in varying degrees of psychological barriers. At present, constipation treatments continue to rely on traditional methods such as purgative therapy and surgery. However, these approaches can disrupt intestinal function. Recent research between intestinal diseases and gut microbiota has gradually revealed a connection between constipation and intestinal flora disturbance, providing a theoretical basis for microbial treatment in chronic constipation. Microbial treatment mainly includes probiotic preparations such as probiotics, prebiotics, synbiotics and fecal microbiota transplantation(FMT). Due to its safety, convenience and curative effect, probiotic preparations have been widely accepted, especially gradually developed FMT with higher curative effects. Microbial treatment improves clinical symptoms, promotes the recovery of intestinal flora, and has no complications during the treatment process. 2Dysbiosis of gut microbiota was closely associated with psoriasis显示文摘Psoriasis is an autoimmune disease and gut microbiota participate in the establishment of intestinal immunity. This study was performed to identify the fecal microbial composition of psoriasis patients, and investigated the influence of subgroup(type and severity) on the fecal microbial composition, and to define the key microbiota in the pathogenesis of psoriasis. Fecal samples from 35 psoriasis patients and 27 healthy controls were sequenced by 16 S rRNA and then analyzed by informatics methods. We found that the microbiota of the psoriasis group differed from that of the heathy group. The relative abundances of Firmicutes and Bacteroidetes were inverted at the phylum level, and 16 kinds of phylotype at the genus level were found with significant difference. No microbial diversity and composition alteration were observed among the four types of psoriasis. The microbiota of psoriasis patients in the severe state differs from those of psoriasis patients with more mild conditions and also the healthy controls. The veillonella in fecal microbiota showed a positive relationship with h-CRP in blood. This research proved that psoriasis patients have a significant disturbed microbiota profiles. 3Ketogenic diet alleviates colitis by reduction of colonic group 3 innate lymphoid cells through altering gut microbiome显示文摘Accumulating evidence suggests that ketogenic diets(KDs)mediate the rise of circulating ketone bodies and exert a potential antiinflammatory effect;however,the consequences of this unique diet on colitis remain unknown.We performed a series of systematic studies using a dextran sulfate sodium(DSS)animal model of inflammatory colitis.Animals were fed with a KD,low-carbohydrate diet(LCD),or normal diet(ND).Germ-free mice were utilized in validation experiments.Colon tissues were analyzed by transcriptome sequencing,RT2 profiler PCR array,histopathology,and immunofluorescence.Serum samples were analyzed by metabolic assay kit.Fecal samples were analyzed by 16S rRNA gene sequencing,liquid chromatography-mass spectrometry and gas chromatography-mass spectrometry.We observed that KD alleviated colitis by altering the gut microbiota and metabolites in a manner distinct from LCD.Quantitative diet experiments confirmed the unique impact of KD relative to LCD with a reproducible increase in Akkermansia,whereas the opposite was observed for Escherichia/Shigella.After colitis induction,the KD protected intestinal barrier function,and reduced the production of R0Ryt+CD3_group 3 innate lymphoid cells(ILC3s)and related inflammatory cytokines(IL-17a,IL-18,IL-22,Ccl4).Finally,fecal microbiota transplantation into germ-free mice revealed that the KD-mediated colitis inhibition and ILC3 regulation were dependent on the modification of gut microbiota.Taken together,our study presents a global view of microbiome-metabolomics changes that occur during KD colitis treatment,and identifies the regulation of gut microbiome and ILC3s as novel targets involving in IBD dietary therapy.Cheng Kong Xuebing Yan Yongqiang Liu Linsheng Huang Yefei Zhu Jide He Renyuan Gao Matthew F.Kalady Ajay Goel Huanlong Qin Yanlei Ma 2021Signal Transduction and Targeted Therapy2021,6,5:8
4Alterations in intestinal microbiota of colorectal cancer patients receiving radical surgery combined with adjuvant CapeOx therapy显示文摘An intricate relationship exists and interactions occur between gut microbiota and colorectal cancer(CRC).Radical surgery combined with adjuvant chemotherapy(AC) serves as the mainstream therapeutic scheme for most CRC patients.The current research was conducted to assess the effect of surgery or chemotherapy on gut microbiota.Forty-three CRC patients who received radical surgery and AC were enrolled.Fecal samples were collected preoperatively,postoperatively,and after the first to fifth cycles of postoperative chemotherapy.The microbial community of each sample was analyzed using high throughput 16S rRNA amplicon sequencing.Compared with preoperative samples,fecal samples collected postoperatively exhibited a significant decrease of obligate anaerobes,tumor-related bacteria,and butyric acid-producing bacteria.However,a significant increase of some conditional pathogens was observed.In addition,the AC regimen(CapeOx) was found to alter intestinal microbiota dramatically.In particular,several changes were observed after chemotherapy including an increase of pathogenic bacteria,the 'rebound effect' of chemotherapy-adapted bacteria,the shift of lactate-utilizing microbiota from Veillonella to Butyricimonas and Butyricicoccus,as well as the decrease of probiotics.Both radical surgery and CapeOx chemotherapy exert a non-negligible effect on the gut microbiota of CRC patients.Microbiota-based intervention may be beneficial for patients during postoperative clinical management.Cheng Kong Renyuan Gao Xuebing Yan Linsheng Huang Jide He Hao Li Jie You Huanlong Qin 2019Science China(Life Sciences)2019,62,9:7
5Large eddy simulation of unsteady transitional flow on the low-pressure turbine blade显示文摘The aim of this paper is to predict the phenomenon of laminar separation,transition and reattachment in a low-pressure turbine(LPT).Self-developed large eddy simulation program of compressible N-S equations was used to describe the flow structures of T106A LPT blade profile at Reynolds number of 1.1X105 based on the exit isentropic velocity and chord length.The computational results show the distributions of time-averaged wall-static pressure coefficient and mean skin-friction coefficient on the blade surface.The locations of laminar separation and reattachment points occur around 87%and 98%axial chord,which agree well with experiment data.The two-dimensional shear layer is gradually unstable along the downstream half of the suction side as a result of the spanwise fluctuation and the roll up of shear layer via Kelvin-Helmholtz(KH)instability.Three-dimensional motions appear near 84%axial chord which later triggers spanwise vortexes and streamwise vortexes,leading to transition to turbulence in the separation bubble.Through introducing the concept of dissipation function,the high loss mainly comes from the places where strong shear layer and intense fluctuation exist.Furthermore,the separation region is only an accumulation center of the low-energy fluid rather than an area of loss source.WANG YunFei CHEN Fu LIU HuaPing CHEN HuanLong 2014Science China(Technological Sciences)2014,57,9:4

7Fusobacterium nucleatum enhances the efficacy of PD-L1 blockade in colorectal cancer显示文摘Given that only a subset of patients with colorectal cancer(CRC)benefit from immune checkpoint therapy,efforts are ongoing to identify markers that predict immunotherapeutic response.Increasing evidence suggests that microbes influence the efficacy of cancer therapies.Fusobacterium nucleatum induces different immune responses in CRC with different microsatellite-instability(MSI)statuses.Here,we investigated the effect of F.nucleatum on anti-PD-L1 therapy in CRC.We found that high F.nucleatum levels correlate with improved therapeutic responses to PD-1 blockade in patients with CRC.Additionally,F.nucleatum enhanced the antitumor effects of PD-L1 blockade on CRC in mice and prolonged survival.Combining F.nucleatum supplementation with immunotherapy rescued the therapeutic effects of PD-L1 blockade.Furthermore,F.nucleatum induced PD-L1 expression by activating STING signaling and increased the accumulation of interferon-gamma(IFN-γ)^(+)CD8^(+)tumor-infiltrating lymphocytes(TILs)during treatment with PD-L1 blockade,thereby augmenting tumor sensitivity to PD-L1 blockade.Finally,patient-derived organoid models demonstrated that increased F.nucleatum levels correlated with an improved therapeutic response to PD-L1 blockade.These findings suggest that F.nucleatum may modulate immune checkpoint therapy for CRC.Yaohui Gao Dexi Bi Ruting Xie Man Li Jing Guo Hu Liu Xianling Guo Juemin Fang Tingting Ding Huiyuan Zhu Yuan Cao Meichun Xing Jiayi Zheng Qing Xu Qian Xu Qing Wei Huanlong Qin 2021Signal Transduction and Targeted Therapy2021,6,12:3

10Optimization of Hydrolysis Conditions for the Isolation of Angiotensin-I Converting Enzyme(ACE) Inhibitory Peptides from Rhopilema hispidum显示文摘To obtain the maximum angiotensin-I converting enzyme(ACE) inhibitory activity, the protein hydrolysis conditions of the jellyfish Rhopilema hispidum were optimized using response surface methodology(RSM). Trypsin was selected to produce R. hispidum protein hydrolysates(RPH) with ACE inhibitory activity. The optimal parameters for producing protein hydrolysates with the highest ACE inhibitory activity were as follows: hydrolysis time 5 h, hydrolysis temperature 50℃, and the enzyme-to-substrate ratio 6%. Under these conditions, the ACE inhibitory rate of RPH could reach 64.28% ± 5.72%. In addition, RPH contained high levels of Gly, Glu, Pro, Ala, Asp and Arg, with a molecular weight distribution range of 0.32–6.84 kDa. The following three novel ACE inhibitory peptides were isolated and identified: Ile-Gly-Glu-Thr-Gly-Pro, Gly-Ala-Thr-Gly-Pro-Ala-Gly-Tyr-Val and Gly-AlaPhe-Gly-Pro-Gly-Gly-Leu-Val-Gly-Arg-Pro. The IC_(50) values of the ACE inhibitory activity of these three purified peptides were 19.07, 27.42 and 31.26 μmol L^(-1), respectively. 12Ketogenic diet for human diseases:the underlying mechanisms and potential for clinical implementations显示文摘The ketogenic diet(KD)is a high-fat,adequate-protein,and very-low-carbohydrate diet regimen that mimics the metabolism of the fasting state to induce the production of ketone bodies.The KD has long been established as a remarkably successful dietary approach for the treatment of intractable epilepsy and has increasingly garnered research attention rapidly in the past decade,subject to emerging evidence of the promising therapeutic potential of the KD for various diseases,besides epilepsy,from obesity to malignancies.In this review,we summarize the experimental and/or clinical evidence of the efficacy and safety of the KD in different diseases,and discuss the possible mechanisms of action based on recent advances in understanding the influence of the KD at the cellular and molecular levels.We emphasize that the KD may function through multiple mechanisms,which remain to be further elucidated.The challenges and future directions for the clinical implementation of the KD in the treatment of a spectrum of diseases have been discussed.We suggest that,with encouraging evidence of therapeutic effects and increasing insights into the mechanisms of action,randomized controlled trials should be conducted to elucidate a foundation for the clinical use of the KD.Huiyuan Zhu Dexi Bi Youhua Zhang Cheng Kong Jiahao Du Xiawei Wu Qing Wei Huanlong Qin 2022Signal Transduction and Targeted Therapy2022,7,2:2

17Structural Networks Constraints on Alteration and Mineralization Processes in the Jiaojia Gold Deposit, Jiaodong Peninsula, China显示文摘Structural network studies could give appropriate opportunities to understanding structural/hydrothermal events,transportation of ore-forming fluids and water/rock interaction process.Four structural deformation/hydrothermal events have been identified in the Jiaojia fault zone according to microtexture and deformation of quartz and feldspars.Plagioclase experienced ductile deformation period with bended polysynthetic twin stripes(>450℃)in the early stage,followed by K-feldspar alteration period with ductile-brittle deformation and subgrain rotation recrystallization of quartz(380-450℃).Then,sericitization period occurred extensive ductile-brittle deformation(350-420℃)and extensive subgrain rotation recrystallization with a little bulging recrystallization in quartz.In the last,gold precipitation-related pyrite-sericite-quartz alteration was dominated by brittle deformation(300-380℃)and total bulging recrystallization of quartz.From the K-feldspar alteration zone and sericitization zone to pyrite-sericitequartz alteration zone,fractal dimension values of dynamically recrystallized quartz grains increase from 1.07 and 1.24 to 1.32,the calculated paleo strain rate values of dynamically recrystallized quartz range from 10^-10^.7(380℃)-10^-9.6(450℃)and 10^-9.3(350℃)-10^-8.2(420℃)to 10^-9.5(300℃)-10^-8.0(380℃),and the paleo differential stress values increase from 36.9 and 39.3,to 121.3 MPa.The increase of fractal dimension values and decrease of grain size from pyrite-sericite-quartz alteration zone and sericitization zone to K-feldspar alteration zone decreased average water/rock ratio values,which could lead to different acidity and redox conditions of ore-forming fluids and mineralization differences.Two kinds of orecontrolling fractures have been distinguished which include the gentle dip types(18°-50°)with NW(315°-355°)and SW(180°-235°)dip hosting No.Ⅰorebodies and the steep dip types(74°-90°)with NE(45°-85°)and SE(95°-165°)dip hosting No.Ⅲorebodies.These faults/fractures crosscut altered Linglong granite of footwall of the Jiaojia fault zone as rhombohedrons that promoted the connection between fractures in the K-feldspar alteration zone and fluid flow passages near the main fault face.Research results indicate No.Ⅰand No.Ⅲorebodies should be derived from the same mineralization event and belong to different orebody types in different mineralization sites under the same structural networks.Huanlong Hu Shuangliang Liu Hong-Rui Fan Kuifeng Yang Yabin Zuo Yachun Cai 2020Journal of Earth Science2020,31,3:1
