| 5 | The Medium Energy X-ray telescope (ME) onboard the Insight-HXMT astronomy satellite显示文摘The Medium Energy X-ray telescope(ME) is one of the three main telescopes on board the Insight hard X-ray modulation telescope(Insight-HXMT) astronomy satellite. ME contains 1728 pixels of Si-PIN detectors sensitive in 5-30 ke V with a total geometrical area of 952 cm^2. The application specific integrated circuit(ASIC) chip, VA32TA6, is used to achieve low power consumption and low readout noise. The collimators define three kinds of field of views(FOVs) for the telescope, 1°×4°, 4°×4°,and blocked ones. Combination of such FOVs can be used to estimate the in-orbit X-ray and particle background components.The energy resolution of ME is ~3 ke V at 17.8 ke V(FWHM) and the time resolution is 255 μs. | 8 | A comparison study of the simulation accuracy between WRF and MM5 in simulating local atmospheric circulations over Greater Beijing显示文摘Several multi-scale numerical simulation experiments were carried out using the mesoscale modeling systems MM5V3.7 and WRFV2.2 for Greater Beijing to estimate the accuracy of WRF and MM5 in simulating the characteristics and variations of mesoscale local circulation in the atmospheric boundary layer of this area. We simulated the horizontal distribution and diurnal variations of temperature and wind fields near the ground and compared them with Automatic Weather System (AWS) data collected from 19 AWS stations in Beijing. Correlation and error analyses were also made. The modeling and statistical results showed that both WRF and MM5 model the temperature field near the ground significantly better than they model the wind field. The temperature field simulated by MM5 is more coincident than that of WRF with the AWS observation records, while WRF does better than MM5 in simulating the wind field, especially under the condition of gusty wind. Neither WRF nor MM5 can capture the fine structure of urban architectural complexity, which is the main error in the wind field simulation. Both models underestimate the land surface temperature at night and overestimate the temperature during the day. | 16 | Genomic Epidemiology of Carbapenemaseproducing Klebsiella pneumoniae in China显示文摘The rapid spread of carbapenemase-producing Klebsiella pneumoniae(cpKP)poses serious threats to public health;however,the underlying genetic basis for its dissemination is still unknown.We conducted a comprehensive genomic epidemiology analysis on 420 cpKP isolates collected from 70 hospitals in 24 provinces/autonomous regions/municipalities of China during 2009–2017 by short-/long-read sequencing.The results showed that most cpKP isolates were categorized into clonal group 258(CG258),in which ST11 was the dominant clone.Phylogenetic analysis revealed three major clades including the top one of Clade 3 for CG258 cpKP isolates.Additionally,carbapenemase gene analysis indicated that blaKPC was dominant in the cpKP isolates,and most blaKPC genes were located in five major incompatibility(Inc)groups of blaKPC-harboring plasmids.Importantly,three advantageous combinations of host–blaKPC-carrying plasmid(Clade 3.1+3.2–IncFIIpHN7A8,Clade 3.1+3.2–IncFIIpHN7A8:IncR,and Clade 3.3–IncFIIpHN7A8:IncpA1763-KPC)were identified to confer cpKP isolates the advantages in both genotypes(strong correlation/coevolution)and phenotypes(resistance/growth/competition)to facilitate the nationwide spread of ST11/CG258 cpKP.Intriguingly,Bayesian skyline analysis illustrated that the three advantageous combinations might be directly associated with the strong population expansion during 2007–2008 and subsequent maintenance of the population of ST11/CG258 cpKP after 2008.We then examined drug resistance profiles of these cpKP isolates and proposed combination treatment regimens for CG258/non-CG258 cpKP infections.Thus,the findings of our systematical analysis shed light on the molecular epidemiology and genetic basis for the dissemination of ST11/CG258 cpKP in China,and much emphasis should be given to the close monitoring of advantageous cpKP–plasmid combinations. | Cuidan Li Xiaoyuan Jiang Tingting Yang Yingjiao Ju Zhe Yin Liya Yue Guannan Ma Xuebing Wang Ying Jing Xinhua Luo Shuangshuang Li Xue Yang Fei Chen Dongsheng Zhou | 2022 | Genomics, Proteomics & Bioinformatics2022,20,6: | 1 |
| 17 | Icariin ameliorates memory deficits through regulating brain insulin signaling and glucose transporters in 3×Tg-AD mice显示文摘Icariin,a major prenylated flavonoid found in Epimedium spp.,is a bioactive constituent of Herba Epimedii and has been shown to exert neuroprotective effects in experimental models of Alzheimer’s disease.In this study,we investigated the neuroprotective mechanism of icariin in an APP/PS1/Tau triple-transgenic mouse model of Alzheimer’s disease.We performed behavioral tests,pathological examination,and western blot assay,and found that memory deficits of the model mice were obviously improved,neuronal and synaptic damage in the cerebral cortex was substantially mitigated,and amyloid-βaccumulation and tau hyperphosphorylation were considerably reduced after 5 months of intragastric administration of icariin at a dose of 60 mg/kg body weight per day.Furthermore,deficits of proteins in the insulin signaling pathway and their phosphorylation levels were significantly reversed,including the insulin receptor,insulin receptor substrate 1,phosphatidylinositol-3-kinase,protein kinase B,and glycogen synthase kinase 3β,and the levels of glucose transporter 1 and 3 were markedly increased.These findings suggest that icariin can improve learning and memory impairments in the mouse model of Alzheimer’s disease by regulating brain insulin signaling and glucose transporters,which lays the foundation for potential clinical application of icariin in the prevention and treatment of Alzheimer’s disease. | Fei Yan Ju Liu Mei-Xiang Chen Ying Zhang Sheng-Jiao Wei Hai Jin Jing Nie Xiao-Long Fu Jing-Shan Shi Shao-Yu Zhou Feng Jin | 2023 | Neural Regeneration Research2023,18,1: | 1 |
| 18 | Validation of the eighth edition of the AJCC staging system for patients with pancreatic adenocarcinoma initially receiving chemoradiotherapy and proposal of modifications显示文摘Objective:To validate the eighth edition of the AJCC staging system in patients with pancreatic adenocarcinoma receiving only stereotactic body radiation therapy and chemotherapy,and to propose modifications to improve prognostic accuracy.Methods:Patients with pathologically confirmed pancreatic adenocarcinoma without metastasis who were undergoing only chemoradiotherapy were included and staged according to the seventh and eighth editions of the AJCC staging system.Meanwhile,another group of stage T4 patients from the above enrollment with only portal vein involvement with or without tumor thrombi(PV±PVTT)were retrieved for survival comparisons.Modifications were proposed according to the survival comparisons.A cohort from the SEER database was used for external validation of the modified staging system.Results:A total of 683 patients were included.Patients with N2 or N1 but different T stages had significantly different survival outcomes according to the eighth edition.The survival of patients with(PV±PVTT)was comparable to that of patients with T4 tumors.The concordance index of the seventh and eighth editions,and the modified staging system was 0.744(95%CI:0.718—0.769),0.750(95%CI:0.725—0.775),and 0.788(95%CI:0.762-0.813),respectively.For external validation,the concordance index was 0.744(95%CI:0.718-0.770),0.750(95%CI:0.724-0.776),and 0.788(95%CI:0.762-0.814),respectively.Conclusions:The modified staging system is suggested to have the m ost accurate prognostic value.Hence,PV土PVTT should be added to the definition of T4 tumors regardless of tumor size.Patients with N2 or N1 in different T stages could be regrouped into different substages.Additionally,stage III should be subclassified into IIIA(T3N 2 and T4N 0)and IIIB(T4N 1-2). | Xiaofei Zhu Di Chen Yangsen Cao Xianzhi Zhao Xiaoping Ju Yuxin Shen Fei Cao Shuiwang Qing Fang Fang Zhen Jia Huojun Zhang | 2020 | Cancer Biology & Medicine2020,17,2: | 1 |
