2ILC3-derived OX40L is essential for homeostasis of intestinal Tregs in immunodeficient mice显示文摘OX40L is one of the co-stimulatory molecules that can be expressed by splenic lymphoid tissue inducer(Lti)cells,a subset of group 3 innate lymphoid cells(ILC3s).OX40L expression in subsets of intestinal ILC3s and the molecular regulation of OX40L expression in ILC3s are unknown.Here,we showed intestinal ILC3s marked as an OX40L high population among all the intestinal leukocytes and were the dominant source of OX40L in Rag1–/–mice.All ILC3 subsets expressed OX40L,and NCR–ILC3s were the most abundant source of OX40L.The expression of OX40L in ILC3s could be upregulated during inflammation.In addition to tumor necrosis factor(TNF)-like cytokine 1A(TL1A),which has been known as a trigger for OX40L,we found that Poly(I:C)representing viral stimulus promoted OX40L expression in ILC3s via a cell-autonomous manner.Furthermore,we demonstrated that IL-7-STAT5 signaling sustained OX40L expression by ILC3s.Intestinal regulatory T cells(Tregs),most of which expressed OX40,had defective expansion in chimeric mice,in which ILC3s were specifically deficient for OX40L expression.Consistently,co-localization of Tregs and ILC3s was found in the cryptopatches of the intestine,which suggests the close interaction between ILC3s and Tregs.Our study has unveiled the crosstalk between Tregs and ILC3s in mucosal tissues through OX40–OX40L signaling,which is crucial for the homeostasis of intestinal Tregs.Tian Deng Caixia Suo Jiali Chang Rui Yang Jingyu Li Ting Cai Ju Qiu 2020Cellular & Molecular Immunology2020,17,2:3

6cFos-ANAB:A cFos-based Web Tool for Exploring Activated Neurons and Associated Behaviors显示文摘cFos is one of the most widely-studied genes in the field of neuroscience.Currently,there is no systematic database focusing on cFos in neuroscience.We developed a curated database-cFos-ANAB-a cFos-based web tool for exploring activated neurons and associated behaviors in rats and mice,comprising 398 brain nuclei and sub-nuclei,and five associated behaviors:pain,fear,feeding,aggression,and sexual behavior.Direct relationships among behaviors and nuclei(even cell types)under specific stimulating conditions were constructed based on cFos expression profiles extracted from original publications.Moreover,overlapping nuclei and sub-nuclei with potentially complex functions among different associated behaviors were emphasized,leading to results serving as important clues to the development of valid hypotheses for exploring as yet unknown circuits.Using the analysis function of cFos-ANAB,multi-layered pictures of networks and their relationships can quickly be explored depending on users’purposes.These features provide a useful tool and good reference for early exploration in neuroscience.The cFos-ANAB database is available at www.cfos-db.net.Fan Wang Wenjie Sun Lei Chang Kefang Sun Leying Hou Linna Qian Chaoyin Jin Jiandong Chen Jiali Pu Panmeng Ye Shuang Qiu Jianhong Luo Shumin Duan Baorong Zhang Zhihua Gao Xiaojun Hu 2021Neuroscience Bulletin2021,37,10:0
7The appearance of cytoplasmic cytochrome C precedes apoptosis during Drosophila salivary gland degradation显示文摘Apoptosis is an important process for organism development that functions to eliminate cell damage,maintain homeostasis,and remove obsolete tissues during morphogenesis.In mammals,apoptosis is accompanied by the release of cytochrome C(Cyt-c)from mitochondria to the cytoplasm.However,whether this process is conserved in the fruit fly,Drosophila melanogaster,remains controversial.In this study,we discovered that during the degradation of Drosophila salivary gland,the transcription of mitochondria apoptosis factors(MAPFs),Cyt-c,and death-associated APAF1-related killer(Dark)encoding genes are all upregulated antecedent to initiator and effector caspases encoding genes.The proteins Cyt-c and the active caspase 3 appear gradually in the cytoplasm during salivary gland degradation.Meanwhile,the Cyt-c protein colocates with mito-GFP,the marker indicating cytoplasmic mitochondria,and the change in mitochondrial membrane potential coincides with the appearance of Cyt-c in the cytoplasm.Moreover,impeding or promoting 20E-induced transcription factor E93 suppresses or enhances the staining of Cyt-c and the active caspase 3 in the cytoplasm of salivary gland,and accordingly decreases or increases the mitochondrial membrane potential,respectively.Our research provides evidence that cytoplasmic Cyt-c appears before apoptosis during Drosophila salivary gland degradation,shedding light on partial conserved mechanism in apoptosis between insects and mammals.Shihui Long Wenxin Cao Yongyu Qiu Ruohan Deng Jiali Liu Lidan Zhang Renke Dong Fengxin Liu Sheng Li Haigang Zhao Na Li Kang Li 2024Insect Science2024,31,1:0
8An iASPP-derived short peptide restores p53-mediated cell death in cancers with wild-type p53显示文摘Background:Inhibitor of apoptosis-stimulating protein of p53(iASPP)is an evolutionarily conserved p53 inhibitor.Mechanistically,iASPP can accelerate tumorigenesis by inhibiting the transactivation function of p53.Targeting the interaction between iASPP and p53 may be a potential therapy for restoring the activity of p53 in tumors.Methods:We constructed an iASPP-derived peptide,called A8,that was derived from the C-terminus of iASPP.Here,we transfected A8 into two wildtype(WT)p53 cell lines,U2OS and A549,and then determined the number of apoptotic cells.The mechanism by which A8 affected apoptosis was further examined by immunoprecipitation(IP),Dual-Luciferase reporter assays,and chromatin IP assays.Real-time polymerase chain reaction and western blots were also used to examine the expression levels of apoptosis-related factors.Results:Our data demonstrate that A8 can increase apoptosis rates in WT p53 cell lines.Functional analysis suggested that A8 restored the transcriptional function and DNA binding activities of p53 toward the Bax and PUMA gene promoters.Moreover,A8 reduced cell proliferation and inhibited tumor growth in xenograft nude mice.Conclusions:These data provide a new approach for restoring the tumor suppressor function of p53 in cancer cells that express WT p53 and therefore may serve as a novel cancer treatment strategy.Shi Qiu Wei Qi Wen Wu Qian Qiu Jiali Ma Yingjun Li Wenhui Fan Junli Li Yang Xu Hai Chen Jie Liu 2023iLABMED2023,1,2:0
9Multi-View & Transfer Learning for Epilepsy Recognition Based on EEG Signals显示文摘Epilepsy is a central nervous system disorder in which brain activity becomes abnormal.Electroencephalogram(EEG)signals,as recordings of brain activity,have been widely used for epilepsy recognition.To study epilep-tic EEG signals and develop artificial intelligence(AI)-assist recognition,a multi-view transfer learning(MVTL-LSR)algorithm based on least squares regression is proposed in this study.Compared with most existing multi-view transfer learning algorithms,MVTL-LSR has two merits:(1)Since traditional transfer learning algorithms leverage knowledge from different sources,which poses a significant risk to data privacy.Therefore,we develop a knowledge transfer mechanism that can protect the security of source domain data while guaranteeing performance.(2)When utilizing multi-view data,we embed view weighting and manifold regularization into the transfer framework to measure the views’strengths and weaknesses and improve generalization ability.In the experimental studies,12 different simulated multi-view&transfer scenarios are constructed from epileptic EEG signals licensed and provided by the Uni-versity of Bonn,Germany.Extensive experimental results show that MVTL-LSR outperforms baselines.The source code will be available on https://github.com/didid5/MVTL-LSR.Jiali Wang Bing Li Chengyu Qiu Xinyun Zhang Yuting Cheng Peihua Wang Ta Zhou Hong Ge Yuanpeng Zhang Jing Cai 2023Computers, Materials & Continua2023,,6:0
10Atomic insights into synergistic effect of pillared graphene by carbon nanotube on the mechanical properties of polymer nanocomposites显示文摘Molecular dynamics simulations have been performed to explore the underlying synergistic mechanism of pillared graphene or non-covalent connected graphene and carbon nanotubes(CNTs) on the mechanical properties of polyethylene(PE) nanocomposites. By constructing the pillared graphene model and CNTs/graphene model, the effect of the structure, arrangement and dispersion of hybrid fillers on the tensile mechanical properties of PE nanocomposites was studied. The results show that the pillared graphene/PE nanocomposites exhibit higher Young’s modulus, tensile strength and elongation at break than non-covalent connected CNTs/graphene/PE nanocomposites. The pull-out simulations show that pillared graphene by CNTs has both large interfacial load and long displacement due to the mixed modes of shear separation and normal separation. 12Modulation of the DNA repair system and ATR-p53 mediated apoptosis is relevant for tributyltin-induced genotoxic effects in human hepatoma G2 cells显示文摘The toxic effects of tributyltin(TBT) have been extensively documented in several types of cells, but the molecular mechanisms related to the genotoxic effects of TBT have still not been fully elucidated. Our study showed that exposure of human hepatoma G2 cells to 1–4 μmol/L TBT for 3 hr caused severe DNA damage in a concentration-dependent manner. Moreover, the expression levels of key DNA damage sensor genes such as the replication factor C, proliferating cell nuclear antigen and poly(ADP-ribose)polymerase-1 were inhabited in a concentration-dependent manner. We further demonstrated that TBT induced cell apoptosis via the p53-mediated pathway, which was most likely activated by the ataxia telangiectasia mutated and rad-3 related(ATR)protein kinase. The results also showed that cytochrome c, caspase-3, caspase-8,caspase-9, and the B-cell lymphoma 2 were involved in this process. Taken together, we demonstrated for the first time that the inhibition of the DNA repair system might be more responsible for TBT-induced genotoxic effects in cells.
