| 1 | Highly fractionated granites:Recognition and research显示文摘Granite is one of the most important components of the continental crust on our Earth; it thus has been an enduring studied subject in geology. According to present knowledge, granite shows a great deal of heterogeneity in terms of its texture,structure, mineral species and geochemical compositions at different scales from small dike to large batholith. However, the reasons for these variations are not well understood although numerous interpretations have been proposed. The key point of this debate is whether granitic magma can be effectively differentiated through fractional crystallization, and, if so, what kind of crystallization occurred during the magmatic evolution. Although granitic magma has high viscosity because of its elevated SiO2 content, we agree that fractional crystallization is effectively processed during its evolution based on the evidence from field investigation,mineral species and its chemical variations, and geochemical compositions. These data indicate that crystal settling by gravitation is not the only mechanism dominating granitic differentiation. On the contrary, flow segregation or dynamic sorting may be more important. Accordingly, granite can be divided into unfractionated, fractionated(including weakly fractionated and highly fractionated) and cumulated types, according to the differentiation degree. Highly fractionated granitic magmas are generally high in primary temperature or high with various volatiles during the later stage, which make the fractional crystallization much easier than the common granitic melts. In addition, effective magmatic differentiation can be also expected when the magma emplaced along a large scale of extensional structure. Highly fractionated granitic magma is easily contaminated by country rocks due to its relatively prolonged crystallization time. Thus, granites do not always reflect the characteristics of the source areas and the physical and chemical conditions of the primary magma. | 2 | Targeting slug-mediated non-canonical activation of c-Met to overcome chemo-resistance in metastatic ovarian cancer cells显示文摘Metastasis-associated drug resistance accounts for high mortality in ovarian cancer and remains to be a major barrier for effective treatment. In this study, SKOV3/T4, a metastatic subpopulation of ovarian cancer SKOV3 cells, was enriched to explore potential interventions against metastaticassociated drug resistance. Quantitative genomic and functional analyses were performed and found that slug was significantly increased in the SKOV3/T4 subpopulation and contributed to the high resistance of SKOV3/T4. Further studies showed that slug activated c-Met in a ligand-independent manner due to elevated levels of fibronectin and provoked integrin α V function, which was confirmed by the significant correlation of slug and p-Met levels in 121 ovarian cancer patient samples. Intriguingly,c-Met inhibitor(s) exhibited greatly enhanced anti-cancer effects in slug-positive ovarian cancer models both in vitro and in vivo. Additionally, IHC analyses revealed that slug levels were highly correlated with reduced survival of ovarian cancer patients. | 3 | The application of proton spectrometers at the SG-III facility for ICF implosion areal density diagnostics显示文摘Charged particle diagnostics is one of the required techniques for implosion areal density diagnostics at the SG-III facility.Several proton spectrometers are under development, and some preliminary areal density diagnostics have been carried out. The response of the key detector, CR39, to charged particles was investigated in detail. A new track profile simulation code based on a semi-empirical model was developed. The energy response of the CR39 detector was calibrated with the accelerator protons and alphas from a241 Am source. A proton spectrometer based on the filtered CR39 detector was developed, and D–D primary proton measurements were implemented. A step range filter spectrometer was developed,and preliminary areal density diagnostics was carried out. A wedged range filter spectrometer array made of Si with a higher resolution was designed and developed at the SG-III facility. A particle response simulation code by the Monte Carlo method and a spectra unfolding code were developed. | 8 | Endogenous peroxynitrite activated fluorescent probe for revealing anti-tuberculosis drug induced hepatotoxicity显示文摘Py^(+)razinamide (PZA), isoniazid (INH) and rifampicin (RFP) are all commonly used anti-tuberculosis drugs in clinical practice, and long-term medication may cause severe liver damage and toxicity. The level of peroxynitrite (ONOO^(-)) generated in liver has long been regarded as a biomarker for the prediction and measurement of drug-induced liver injury (DILI). In this article, we constructed a BODIPY-based fluorescent probe (BDP-Py^(+)) that enabled quickly and sensitively detect and image ONOO^(-) in vivo. Utilizing this probe, we demonstrated the change of ONOO^(-) content in cells and mice model of DILI induced by acetaminophen (APAP), and for the first time revealed the mechanism of liver injury induced by antituberculosis drug PZA. Moreover, BDP-Py^(+) could be applied to screen out and evaluate the hepatotoxicity of different anti-tuberculosis drugs. Comparing with the existing serum enzymes detection and H&E staining, the probe could achieve early diagnosis of DILI before solid lesions in liver via monitoring the up-regulation of ONOO^(-) levels.
