1miRNA studies in in vitro and in vivo activated hepatic stellate cells显示文摘AIM: To understand which and how different miRNAs are implicated in the process of hepatic stellate cell (HSC) activation. METHODS: We used microarrays to examine the differential expression of miRNAs during in vitro activation of primary HSCs (pHSCs). The transcriptome changes upon stable transfection of rno-miR-146a into an HSC cell line were studied using cDNA microarrays. Selected differentially regulated miRNAs were investigated by quantitative real-time polymerase chain reaction during in vivo HSC activation. The effect of miRNA mimics and inhibitor on the in vitro activation of pHSCs was also evaluated.RESULTS: We found that 16 miRNAs were upregulated and 26 were downregulated significantly in 10-d in vitro activated pHSCs in comparison to quiescent pHSCs. Overexpression of rno-miR-146a was characterized by marked upregulation of tissue inhibitor of metalloproteinase-3, which is implicated in the regulation of tumor necrosis factor-α activity. Differences in the regulation of selected miRNAs were observed comparing in vitro and in vivo HSC activation. Treatment with miR-26a and 29a mimics, and miR-214 inhibitor during in vitro activation of pHSCs induced significant downregulation of collagen type Ⅰ transcription. CONCLUSION: Our results emphasize the different regulation of miRNAs in in vitro and in vivo activated pHSCs. 3Inferring the Tree of Life of the order Cypriniformes, the earth's most diverse clade of freshwater fishes: Implications of varied taxon and character sampling显示文摘The phylogenetic relationships of species are fundamental to any biological investigation, including all evolutionary studies. Accurate inferences of sister group relationships provide the researcher with an historical framework within which the attributes or geographic origin of species (or supraspecific groups) evolved. Taken out of this phylogenetic context, interpretations of evolutionary processes or origins, geographic distributions, or speciation rates and mechanisms, are subject to nothing less than a biological experiment without controls. Cypriniformes is the most diverse clade of freshwater fishes with estimates of diversity of nearly 3,500 species. These fishes display an amazing array of morphological, ecological, behavioral, and geographic diversity and offer a tremendous opportunity to enhance our understanding of the biotic and abiotic factors associated with diversification and adaptation to environments. Given the nearly global distribution of these fishes, they serve as an important model group for a plethora of biological investigations, including indicator species for future cli-matic changes. The occurrence of the zebrafish, Danio rerio, in this order makes this clade a critical component in understanding and predicting the relationship between mutagenesis and phenotypic expressions in vertebrates, including humans. With the tremendous diversity in Cypriniformes, our understanding of their phylogenetic relationships has not proceeded at an acceptable rate, despite a plethora of morphological and more recent mo-lecular studies. Most studies are pre-Hennigian in origin or include relatively small numbers of taxa. Given that analyses of small numbers of taxa for molecular characters can be compromised by peculiarities of long-branch attraction and nodal-density effect, it is critical that significant progress in our understanding of the relationships of these important fishes occurs with increasing sampling of species to mitigate these potential problems. The recent Cypriniformes Tree of Life initiative is an effort to achieve this goal with morphological and molecular (mitochondrial and nuclear) data. In this early synthesis of our understanding of the phylogenetic relationships of these fishes, all types of data have contributed historically to improving our understanding, but not all analyses are complementary in taxon sampling, thus precluding direct understanding of the impact of taxon sampling on achieving accurate phylogenetic inferences. However, recent molecular studies do provide some insight and in some instances taxon sampling can be implicated as a variable that can influence sister group relationships. Other instances may also exist but without inclusion of more taxa for both mitochondrial and nuclear genes, one cannot distinguish between inferences being dictated by taxon sampling or the origins of the molecular data.Richard L. 4Study on cosmogenic activation in germanium detectors for future tonne-scale CDEX experiment显示文摘A study on cosmogenic activation in germanium was carried out to evaluate the cosmogenic background level of natural and ^(70)Ge depleted germanium detectors. The production rates of long-lived radionuclides were calculated with Geant4 and CRY.Results were validated by comparing the simulated and experimental spectra of CDEX-1B detector. Based on the validated codes, the cosmogenic background level was predicted for further tonne-scale CDEX experiment. The suppression of cosmogenic background level could be achieved by underground germanium crystal growth and high-purity germanium detector fabrication to reach the sensitivity requirement for direct detection of dark matter. 7First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. 8First results on ^(76)Ge neutrinoless double beta decay from CDEX-1 experiment显示文摘We report the first results on ^(76)Ge neutrinoless double beta decay from stage one of the China dark-matter experiment(CDEX).A p-type point-contact high-purity germanium detector with a mass of 994 g has been installed to detect neutrinoless double beta decay events, as well as to directly detect dark matter particles. An exposure of 304 kg d has been analyzed over a wide spectral band from 500 keV to 3 MeV. The average event rate obtained was about 0.012 counts per keV per kg per day over the 2.039 MeV energy range. The half-life of ^(76)Ge neutrinoless double beta decay derived based on this result is T^(0ν)1/2>6.4×10^(22) yr(90% C.L.). 9Targeted Ptpn11 deletion in mice reveals the essential role of SHP2 in osteoblast differentiation and skeletal homeostasis显示文摘The maturation and function of osteoblasts(OBs)rely heavily on the reversible phosphorylation of signaling proteins.To date,most of the work in OBs has focused on phosphorylation by tyrosyl kinases,but little has been revealed about dephosphorylation by protein tyrosine phosphatases(PTPases).SHP2(encoded by PTPN11)is a ubiquitously expressed PTPase.PTPN11 mutations are associated with both bone and cartilage manifestations in patients with Noonan syndrome(NS)and metachondromatosis(MC),although the underlying mechanisms remain elusive.Here,we report that SHP2 deletion in bone gamma-carboxyglutamate protein-expressing(Bglap+)bone cells leads to massive osteopenia in both trabecular and cortical bones due to the failure of bone cell maturation and enhanced osteoclast activity,and its deletion in Bglap+chondrocytes results in the onset of enchondroma and osteochondroma in aged mice with increased tubular bone length.Mechanistically,SHP2 was found to be required for osteoblastic differentiation by promoting RUNX2/OSTERIX signaling and for the suppression of osteoclastogenesis by inhibiting STAT3-mediated RANKL production by osteoblasts and osteocytes.These findings are likely to explain the compromised skeletal system in NS and MC patients and to inform the development of novel therapeutics to combat skeletal disorders.Lijun Wang Huiliang Yang Jiahui Huang Shaopeng Pei Liyun Wang Jian Q.Feng Dian Jing Hu Zhao Henry M.Kronenberg Douglas C.Moore Wentian Yang 2021Bone Research2021,9,1:3

11Growth arrest-specific gene 2 suppresses hepatocarcinogenesis by intervention of cell cycle and p53-dependent apoptosis显示文摘BACKGROUND Growth arrest-specific gene 2(GAS2)plays a role in modulating in reversible growth arrest cell cycle,apoptosis,and cell survival.GAS2 protein is universally expressed in most normal tissues,particularly in the liver,but is depleted in some tumor tissues.However,the functional mechanisms of GAS2 in hepatocellular carcinoma(HCC)are not fully defined.AIM To investigate the function and mechanism of GAS2 in HCC.METHODS GAS2 expression in clinic liver and HCC specimens was analyzed by real-time PCR and western blotting.Cell proliferation was analyzed by counting,MTS,and colony formation assays.Cell cycle analysis was performed by flow cytometry.Cell apoptosis was investigated by Annexin V apoptosis assay and western blotting.RESULTS GAS2 protein expression was lower in HCC than in normal tissues.Overexpression of GAS2 inhibited the proliferation of HCC cells with wide-type p53,while knockdown of GAS2 promoted the proliferation of hepatocytes(P<0.05).Furthermore,GAS2 overexpression impeded the G1-to-S cell cycle transition and arrested more G1 cells,particularly the elevation of sub G1(P<0.01).Apoptosis induced by GAS2 was dependent on p53,which was increased by etoposide addition.The expression of p53 and apoptosis markers was further enhanced when GAS2 was upregulated,but became diminished upon downregulation of GAS2.In the clinic specimen,GAS2 was downregulated in more than 60%of HCCs.The average fold changes of GAS2 expression in tumor tissues were significantly lower than those in paired non-tumor tissues(P<0.05).CONCLUSION GAS2 plays a vital role in HCC cell proliferation and apoptosis,possibly by regulating the cell cycle and p53-dependent apoptosis pathway.Ran-Xu Zhu Alfred Sze Lok Cheng Henry Lik Yuen Chan Dong-Ye Yang Wai-Kay Seto 2019World Journal of Gastroenterology2019,25,32:2
12Hydro-mechanical analysis of rainfall-induced fines migration process within unsaturated soils显示文摘Seepage-induced fines migration under rainfall infiltration is a main cause leading to shallow failures in loose colluvial slopes. To describe the full process of fines migration within unsaturated soils during rainfall infiltration and the associated hydromechanical behaviors, a seepage-erosion-deformation coupled formulation is proposed in this paper. The governing equations proposed are implemented into a finite element code and used to investigate the influences of skeleton deformation on the rainfall infiltration process through unsaturated soil columns.The numerical results were presented in detail for a better understanding of the rainfall-induced fines migration process within unsaturated soils. Further,the obtained results are integrated into an infinite slope model for slope stability analysis. The results show that, the skeleton deformation will affect the rainfall infiltration rate and hence the timing of slope failures; meanwhile their influences are more evident if the fines deposition process is taken into account.Moreover, the slope stability could be reduced gradually due to the soil strength loss along with loss of fine particles. 20Nano-yttrium-containing precipitates of T6 heat-treated A356.2 alloy when trace yttrium(Y less than 0.100 wt%) added显示文摘To investigate the effect of yttrium(Y) on microstructure refinement and mechanical properties of aluminum alloy A356.2,the different trace contents of Y(0 wt%,0.025 wt%,0.050 wt%,0.075 wt%,or 0.100 wt%)were introduced into the liquid alloy.The alloys were fabricated in a preheated permanent mold,and subsequently treated by a T6 heat treatment.The results of tensile testing indicate that the yield strength(YS),the ultimate tensile strength(UTS) and the elongation(El) of the A356.2 alloy are improved by the Y additions.The YS dependence on grain size for the test alloys follows the Hall-Petch equation,which gives YS=-354.1+2875.2 d^(-1/2) with a correlation of R^(2)=0.83.As 0.050 wt%Y is added,the optimum values of the YS,UTS and El are achieved after T6 heat treatment.The secondary phases were identified by X-ray diffraction(XRD) which mainly consisted as Si,Mg2 Si and Al3 Y.The scanning electron microscope(SEM) and energy-dispersive spectrometer(EDS) analyses reveal the presence of the nano-sized Al3 Y particles on the surface of the Si phase.The A356.2 alloy with the Y addition is strengthened by the dendritic refinement,and the presence of the micron-and nano-sized Al3 Y precipitates.Xin-Ping Hu Qing Wang Henry Hu Rui-Xin Li Yang Zhao Zhi-Ming Wang Bing-Rong Zhang 2021Rare Metals2021,40,11:1
