| 1 | Antigenically dominant proteins within the human liver mitochondrial proteome identified by monoclonal antibodies显示文摘Analysis of the mitochondrial proteome would provide valuable insight into the function of this important organelle, which plays key roles in energy metabolism, apoptosis, free radical production, thermogenesis, and calcium signaling. It could also increase our understanding about the mechanisms that promote mitochondrial disease. To identify proteins that are antigenically dominant in human liver mitochondria, we generated >240 hybridoma cell lines from native mitochondrial proteins after cell fusion, screening, and cloning. Antibodies that recognized mitochondrial proteins were identified by screening human liver cDNA expression libraries. In this study, we identified 6 major antigens that were recognized by at least 2 different monoclonal antibodies (mAbs). The proteins that were antigenically dominant were: acetyl-Coenzyme A acyltransferase 2 (mitochondrial 3-oxoacyl-Coenzyme A thiolase), aldehyde dehydrogenase 1 family member A1, carbamoyl phosphate synthetase 1, dihydrolipoamide S-acetyltransferase (E2 component of pyruvate dehydrogenase complex), enoyl coenzyme A hydratase 1, and hydroxysteroid (11-beta) dehydrogenase 1. We also determined the subcellular localizations of these enzymes within the mitochondria using immunohistocytochemistry. We believe that these well-characterized antibodies will provide a valuable resource for the Human Liver Proteome Project (HLPP), and will make studies aimed at investigating liver mitochondrial function far easier to perform in future. | 3 | Recent progress in Co-based metal-organic framework derivatives for advanced batteries显示文摘To date,Co-based metal-organic frameworks(Co-MOFs)have drawn much attention owing to their advantages of easy preparation,high porosity and adjustable structure.Because of these enticing properties,numerous efforts have been devoted to their applications in energy storage and conversion.However,poor conductivity has become one of the biggest obstacles for large-scale use of pristine Co-MOFs.Subsequently,many attempts have been carried out to develop various Co-MOF derived materials as electrodes for rechargeable batteries in order to address the above-mentioned shortcoming and to enhance the electrical conductivity with improved stability during cycling.Moreover,in addition to improvement of Li-ion batteries in practical utilization,seeking for other rechargeable batteries is another urgent task due to the high cost and limited sources of metallic Li.Herein,by following the recent research progress,this review provides an overview of applications of Co-MOF derived materials in various rechargeable batteries including lithium-ion batteries,sodium-ion batteries,lithium-sulfur batteries,zinc air batteries and other rechargeable batteries,where they have been utilized as cathodes,anodes,separators and electrocatalysts.Accordingly,we categorize and compare the morphology driven electrochemical performance of various Co-MOF derivatives including porous carbon,cobalt oxides,cobalt chalcogenides,cobalt phosphides and corresponding composites.Finally,current challenges for large-scale production and commercialization of Co-MOF derived materials as well as some reasonable solutions have been discussed at the end. | Jianen Zhou Qingyun Yang Qiongyi Xie Hong Ou Xiaoming Lin Akif Zeb Lei Hu Yongbo Wu Guozheng Ma | 2022 | Journal of Materials Science & Technology2022,,1: | 2 |

| 12 | Natural epitope variants of the hepatitis C virus impair cytotoxic T lymphocyte activity显示文摘AIM:To understand how interactions between hepatitis C virus(HCV) and the host's immune system might lead to viral persistence or effective elimination of HCV.METHODS:Nucleotides 3519-3935 of the non-structural 3(NS3) region were amplified by using reverse transcription polymerase chain reaction(PCR).PCR products of the HCV NS3 regions were integrated into a PCR T7TOPO TA vector and then sequenced in both directions using an automated DNA sequencer.Relative major histocompatibility complex binding levels of wild-type and variant peptides were performed by fluorescence polarization-based peptide competition assays.Peptides with wild type and variant sequences of NS3 were synthesized locally using F-moc chemistry and purified by high-performance liquid chromatography.Specific cytotoxic T lymphocytes(CTLs) clones toward HCV NS3 wild-type peptides were generated through limiting dilution cloning.The CTL clones specifically recognizing HCV NS3 wild-type peptides were tested by tetramer staining and flow cytometry.Cytolytic activity of CTL clones was measured using target cells labeled with the fluorescence enhancing ligand,DELFIA EuTDA.RESULTS:The pattern of natural variants within three human leukocyte antigen(HLA)-A2-restricted NS3 epitopes has been examined in one patient with chronic HCV infection at 12,28 and 63 mo post-infection.Results obtained may provide convincing evidence of immune selection pressure for all epitopes investigated.Statistical analysis of the extensive sequence variation found within these NS3 epitopes favors a Darwinian selection model of variant viruses.Mutations within the epitopes coincided with the decline of CTL responses,and peptide-binding studies suggested a signif icant impact of the mutation on T cell recognition rather than peptide presentation by HLA molecules.While most variants were either not recognized or elicited low responses,such could antagonize CTL responses to target cells pulsed with wild-type peptides.CONCLUSION:Cross-recognition of CTL epitopes from wild-type and naturally-occurring HCV variants may lead to impaired immune responses and ultimately contribute to viral persistence. | Shuping Wang Rico Buchli Jennifer Schiller Jianen Gao Rodney S VanGundy William H Hildebrand David D Eckels | 2010 | World Journal of Gastroenterology2010,16,16: | 1 |
