| 1 | DEVELOPMENT OF STEP-POOL SEQUENCE AND ITS EFFECTS IN RESISTANCE AND STREAM BED STABILITY显示文摘Experiments were conducted and field investigations were performed to study the development of step-pool sequence and its effects on resistance to the flow and stream bed stability. Step-pool sequence develops in incised channels as a result of streambed erosion, which is compared with sand dunes and armor layer of the role in resistance and streambed protection. The tight interlocking of particles in steps gives them an inherent stability which only extreme floods are likely to disturb. That stability suggests that step-pools are a valid equilibrium form, especially when coupled with their apparent regularity form and their role in satisfying the extreme condition of resistance maximization. The development degree of step-pools, SP, is proportional to the streambed slope. If the incoming sediment load is equal to or more than the sediment-carrying capacity of the flow, there is no bed erosion and thence there are no step-pools. Ifthe flow depth increases and is over the step-height the resistance caused by the step-pool sequence will be greatly reduced. The rate of energy dissipation by step-pools is a function of SP. The higher is SP, the larger is the rate of energy dissipation. The step-pool sequence increases the resistance and flow depth, reduces the shear stress of the flow and protects the streambed from erosion. | 2 | MicroRNA in vivo precipitation identifies miR-151-3p as a computational unpredictable miRNA to target Stat3 and inhibits innate IL-6 production显示文摘MicroRNAs(miRNAs)function as important regulators in the immune response and inflammation.Several approaches have been reported to computationally predict miRNAs and their potential targets.However,there are still many miRNA–target interactions that are unpredictable by using the current computational algorithms.We established a miRNA in vivo precipitation method(miRIP)to identify unpredictable miRNAs with definite targets in these cells.Because Stat3 is a well-known transcription factor involved in innate immunity and inflammation,we utilized the miRIP method to identify miRNAs that bind Stat3 mRNA in macrophages.Among the captured miRNAs,miR-151-3p was confirmed to interact with Stat3 mRNA 3′-UTR and downregulate the Stat3 protein levels.LPS stimulation decreased miR-151-3p expression,thereby increasing IL-6 production.Therefore,we found that miR-151-3p inhibited LPS-induced IL-6 production by targeting Stat3.These data further confirmed miRIP as an efficient method to identify unpredictable miRNAs and explore miRNAs-mediated regulation in innate immunity and inflammation. | Xiang Liu Xiaoping Su Sheng Xu Huamin Wang Dan Han Jiangxue Li Mingyan Huang Xuetao Cao | 2018 | Cellular & Molecular Immunology2018,15,2: | 8 |
| 3 | Investigation and experimental validation of curcumin-related mechanisms against hepatocellular carcinoma based on network pharmacology显示文摘Objective:To determine the potential molecular mechanisms underlying the therapeutic effect of curcumin on hepatocellular carcinoma(HCC)by network pharmacology and experimental in vitro validation.Methods:The predictive targets of curcumin or HCC were collected from several databases.the identified overlapping targets were crossed with Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses using the Database for Annotation,Visualization,and Integrated Discovery(DAVID)platform.Two of the candidate pathways were selected to conduct an experimental verification.The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide tetrazolium(MTT)assay was used to determine the effect of curcumin on the viability of Hep G2 and LO2 cells.The apoptosis and autophagy of Hep G2 cells were respectively detected by flow cytometry and transmission electron microscopy.Besides,western blot and real-time polymerase chain reaction(PCR)were employed to verify the p53 apoptotic pathway and adenosine 5’-monophosphate(AMP)-activated protein kinase(AMPK)autophagy pathway.Hep G2 cells were pretreated with pifithrin-α(PFT-α)and GSK690693 for further investigation.Results:The 167 pathways analyzed by KEGG included apoptosis,autophagy,p53,and AMPK pathways.The GO enrichment analysis demonstrated that curcumin was involved in cellular response to drug,regulation of apoptotic pathway,and so on.The in vitro experiments also confirmed that curcumin can inhibit the growth of Hep G2 cells by promoting the apoptosis of p53 pathway and autophagy through the AMPK pathway.Furthermore,the protein and messenger RNA(m RNA)of the two pathways were downregulated in the inhibitor-pretreated group compared with the experimental group.The damage-regulated autophagy modulator(DRAM)in the PFT-α-pretreated group was downregulated,and p62 in the GSK690693-pretreated group was upregulated.Conclusions:Curcumin can treat HCC through the p53 apoptotic pathway and the AMPK/Unc-51-like kinase 1(ULK1)autophagy pathway,in which the mutual transformation of autophagy and apoptosis may occur through DRAM and p62. | Yang CHEN Qian LI Sisi REN Ting CHEN Bingtao ZHAI Jiangxue CHENG Xiaoyan SHI Liang SONG Yu FAN Dongyan GUO | 2022 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,8: | 3 |
| 4 | Synthetic lethal short hairpin RNA screening reveals that ring finger protein 183 confers resistance to trametinib in colorectal cancer cells显示文摘Background: The mitogen-activated extracellular signal-regulated kinase 1/2(MEK1/2) inhibitor trametinib has shown promising therapeutic effects on melanoma, but its efficacy on colorectal cancer(CRC) is limited. Synthetic lethality arises with a combination of two or more separate gene mutations that causes cell death, whereas individual mutations keep cells alive. This study aimed to identify the genes responsible for resistance to trametinib in CRC cells,using a synthetic lethal short hairpin RNA(shRNA) screening approach.Methods: We infected HT29 cells with a pooled lentiviral shRNA library and applied next-generation sequencing to identify shRNAs with reduced abundance after 8-day treatment of 20 nmol/L trametinib. HCT116 and HT29 cells were used in validation studies. Stable ring finger protein 183(RNF183)-overexpressing cell lines were generated by pcDNA4-myc/his-RNF183 transfection. Stable RNF 183-knockdown cell lines were generated by infection of lentiviruses that express RNF183 shRNA, and small interference RNA(siRNA) was used to knock down RNF183 transiently.Quantitative real-time PCR was used to determine the mRNA expression. Western blotting, immunohistochemical analysis, and enzyme-linked immunosorbent assay(ELISA) were used to evaluate the protein abundance. MTT assay,colony formation assay, and subcutaneous xenograft tumor growth model were used to evaluate cell proliferation.Results: In the primary screening, we found that the abundance of RNF183 shRNA was markedly reduced after treatment with trametinib. Trametinib induced the expression of RNF183, which conferred resistance to drug-induced cell growth repression and apoptotic and non-apoptotic cell deaths. Moreover, interleukin-8(IL-8) was a downstream gene of RNF183 and was required for the function of RNF183 in facilitating cell growth. Additionally, elevated RNF183 expression partly reduced the inhibitory effect of trametinib on IL-8 expression. | 8 | Dry wear behaviors of wear resistant composite coatings produced by laser cladding显示文摘Using different proportional mixtures of Ni-coated MoS2, TiC and pure Ni powders, new typical wear resistant and selflubricant coatings were formed on low carbon steel by laser cladding process. The microstructures and phase composition of the composite coatings were studied by SEM and XRD. The typical microstructure of the composite coating is composed of multisulfide phases including binary element sulfide and ternary element sulfide, γ-Ni, TiC and Mo2C. Wear tests were carried out using an FALEX-6 type pin-on-disc machine. The friction coefficient and mass loss of three kinds of MoS2/TiC/Ni laser clad coatings are lower than those of quenched 45 steel, and the worn surfaces of the laser cladding coatings are very smooth. Because of high hardness combined with low friction, the laser cladding composite coating with a mixture of 70% Ni-coated MoS2, 20%TiC and 10%pure Ni powder presents better wear behaviors than the composite coating with other powder blends.
