1MicroRNA in vivo precipitation identifies miR-151-3p as a computational unpredictable miRNA to target Stat3 and inhibits innate IL-6 production显示文摘MicroRNAs(miRNAs)function as important regulators in the immune response and inflammation.Several approaches have been reported to computationally predict miRNAs and their potential targets.However,there are still many miRNA–target interactions that are unpredictable by using the current computational algorithms.We established a miRNA in vivo precipitation method(miRIP)to identify unpredictable miRNAs with definite targets in these cells.Because Stat3 is a well-known transcription factor involved in innate immunity and inflammation,we utilized the miRIP method to identify miRNAs that bind Stat3 mRNA in macrophages.Among the captured miRNAs,miR-151-3p was confirmed to interact with Stat3 mRNA 3′-UTR and downregulate the Stat3 protein levels.LPS stimulation decreased miR-151-3p expression,thereby increasing IL-6 production.Therefore,we found that miR-151-3p inhibited LPS-induced IL-6 production by targeting Stat3.These data further confirmed miRIP as an efficient method to identify unpredictable miRNAs and explore miRNAs-mediated regulation in innate immunity and inflammation.Xiang Liu Xiaoping Su Sheng Xu Huamin Wang Dan Han Jiangxue Li Mingyan Huang Xuetao Cao 2018Cellular & Molecular Immunology2018,15,2:8
2Investigation and experimental validation of curcumin-related mechanisms against hepatocellular carcinoma based on network pharmacology显示文摘Objective:To determine the potential molecular mechanisms underlying the therapeutic effect of curcumin on hepatocellular carcinoma(HCC)by network pharmacology and experimental in vitro validation.Methods:The predictive targets of curcumin or HCC were collected from several databases.the identified overlapping targets were crossed with Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses using the Database for Annotation,Visualization,and Integrated Discovery(DAVID)platform.Two of the candidate pathways were selected to conduct an experimental verification.The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide tetrazolium(MTT)assay was used to determine the effect of curcumin on the viability of Hep G2 and LO2 cells.The apoptosis and autophagy of Hep G2 cells were respectively detected by flow cytometry and transmission electron microscopy.Besides,western blot and real-time polymerase chain reaction(PCR)were employed to verify the p53 apoptotic pathway and adenosine 5’-monophosphate(AMP)-activated protein kinase(AMPK)autophagy pathway.Hep G2 cells were pretreated with pifithrin-α(PFT-α)and GSK690693 for further investigation.Results:The 167 pathways analyzed by KEGG included apoptosis,autophagy,p53,and AMPK pathways.The GO enrichment analysis demonstrated that curcumin was involved in cellular response to drug,regulation of apoptotic pathway,and so on.The in vitro experiments also confirmed that curcumin can inhibit the growth of Hep G2 cells by promoting the apoptosis of p53 pathway and autophagy through the AMPK pathway.Furthermore,the protein and messenger RNA(m RNA)of the two pathways were downregulated in the inhibitor-pretreated group compared with the experimental group.The damage-regulated autophagy modulator(DRAM)in the PFT-α-pretreated group was downregulated,and p62 in the GSK690693-pretreated group was upregulated.Conclusions:Curcumin can treat HCC through the p53 apoptotic pathway and the AMPK/Unc-51-like kinase 1(ULK1)autophagy pathway,in which the mutual transformation of autophagy and apoptosis may occur through DRAM and p62.Yang CHEN Qian LI Sisi REN Ting CHEN Bingtao ZHAI Jiangxue CHENG Xiaoyan SHI Liang SONG Yu FAN Dongyan GUO 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,8:3
3Mobility Support for Content Source in Content-Centric Networking显示文摘Content-centric networking(CCN) is rising to solve the problems suffered by traditional networks,especially in terms of content delivery.One of the critical issues for basic CCN is supporting mobility.Handoff of Mobile Content Source(MCS) will result in a large scale routing update,which generates huge amount of routing overhead and leads to service interruption.Most of the existing CCN mobility mechanisms are transplanted from the IP mobility solutions,which are unnaturally integrated with CCN.Different from previous works,a mobility strategy from the perspective of CCN architecture is proposed to support the handoff of MCS in this paper.Especially,we define the critical network routers that can limit the routing update scale effectively when MCS handoff is conducted.Based on the defined critical network routers,the proposed scheme can provide lower routing update overhead,faster routing convergence and shorter service interruption time.Finally,series of simulations are conducted to validate the effectiveness of our proposed scheme.HUANG Tao LI Jiangxu LIU Jiang ZHANG Yunyong LIU Yunjie 2015China Communications2015,12,3:1

5Surface structure change properties:Auto-soft bionic fibrous membrane in reducing postoperative adhesion显示文摘Peritoneal adhesion is the most common adverse effect following abdominal surgery or inflammation.The occurrence in clinical trials has been successfully reduced using barriers.However,the shortcomings of frequently used adhesion barriers,such as rapid degradation rate of gel barrier and inadequate operation ability of solid barrier,cannot be ignored.In this study,a fibrous membrane with an ECM-like structure was prepared.The adhesion properties were reduced significantly by changing the surface structure.The fibrous membrane caused less inflammatory response and much less peripheral adhesion and intestinal obstruction compared to the casting film and the commercial film with smooth surface,though with the same components.Because of the auto-soft bionic structure and similarity in the mechanical modulus of the tissues,the fibrous membrane was more flexible when it adhered to the tissues,showed excellent effectiveness and biocompatibility.In addition to the rat and miniature pig models,a randomized,placebo-controlled,and multicenter clinical pilot study with 150 patients confirmed that because of its flexibility,biodegradability,and similarity to mechanical modulus and structure with tissues involved,the fibrous membrane served as a favorable implant for preventing post-operation adhesion.Shanshan Xu Chenhong Wang Ruoqing Mao Xiaoyu Liang Heran Wang Zhenyu Lin Jiangxue Li Shilin Li Jipeng Jiang Tongshuo Zhang Yongfu Ma Yang Liu Charles C.Han Ying Liu 2022Bioactive Materials2022,7,6:1

8Segregation of Re at theγ/γ'boundary of Ni-based single crystal superalloys revealed by first-principles calculations based Monte-Carlo simulations显示文摘Nickel-based single crystal superalloys have been widely used in aero-engines and gas turbine engines.To improve the creep resistance,rhenium is often added to the alloys.However,it is not yet fully under-stood how the added Re elements distribute in the alloys and how the microstructure evolves with the addition of Re.Here,we performed extensive first-principles calculations based Monte-Carlo simulated annealing of Ni-Al-Re ternary alloys with different Re concentrations ranging from 0.5 at.%to 6.0 at.%.The results demonstrate that with the decreasing temperature,most of Re atoms stay in theγphase,while a few of Re atoms stay in theγ'phase and tend to occupy the Al positions.At low temperatures,the Re atoms segregate at theγ/γ'boundary,in good agreement with experiment.We find that the disorder-order transition temperature of the Ni-Al-Re ternary alloys increases with the Re concentration due to the Re-enhanced Al-Al ordering tendency.In addition,we observe that at low temperatures the Re segregation at theγ/γ'boundary promotes the formation of Ni 4 Re-or Ni 8 Re-like local structures as the Re concentration is over 2 at.%.The formation of a large amount of these local structures consumes the Re atoms in solid solutions,and thus from the solid-solution strengthening point of view,this would have a negative influence on the creep resistance of the superalloys.This work provides important atomistic insights on the Re distribution and its effects on the stability of superalloys.Heyu Zhu Jiantao Wang Lei Wang Yongpeng Shi Mingfeng Liu Jiangxu Li Yun Chen Yingche Ma Peitao Liu Xing-Qiu Chen 2023Journal of Materials Science & Technology2023,,12:0
9A Hybrid Biofuel and Triboelectric Nanogenerator for Bioenergy Harvesting显示文摘Various types of energy exist everywhere around us,and these energies can be harvested from multiple sources to power micro-/nanoelectronic system and even personal electronic products.In this work,we proposed a hybrid energy-harvesting system(HEHS)for potential in vivo applications.The HEHS consisted of a triboelectric nanogenerator and a glucose fuel cell for simultaneously harvesting biomechanical energy and biochemical energy in simulated body fluid.These two energy-harvesting units can work individually as a single power source or work simultaneously as an integrated system.This design strengthened the flexibility of harvesting multiple energies and enhanced corresponding electric output.Compared with any individual device,the integrated HEHS outputs a superimposed current and has a faster charging rate.Using the harvested energy,HEHS can power a calculator or a green light-emitting diode pattern.Considering the widely existed biomechanical energy and glucose molecules in the body,the developed HEHS can be a promising candidate for building in vivo self-powered healthcare monitoring system.Hu Li Xiao Zhang Luming Zhao Dongjie Jiang Lingling Xu Zhuo Liu Yuxiang Wu Kuan Hu Ming‑Rong Zhang Jiangxue Wang Yubo Fan Zhou Li 2020Nano-Micro Letters2020,12,4:0

12Apoferritin nanoparticle based dual-antigen influenza conjugate vaccine with potential cross-protective efficacy against heterosubtypic influenza virus显示文摘Ferritin nanoparticles with self-assembling properties have been widely explored as vaccine carrier by displaying foreign antigens through genetic fusion strategy.In the present work,an apoferritin(AFt)nanoparticle was tested as influenza vaccine carrier by chemically conjugating a matrix protein 2 ectodomain(M2e)antigen peptide or/and the full-length hemagglutinin(HA)antigen on the outer surface of the AFt,with heterobifunctional sSMCC or SM(PEG)_(24) containing PEG chain as linkers.To each AFt nanoparticle,about 30-32 M2e or 1.8 HA antigen could be coupled.The AFt-(PEG)24-M2e,in which the M2e was coupled through SM(PEG)_(24) containing PEG chain,conferred higher protective efficacy in immunized mice than AFt-M2e did,but was less effective than AFt-(PEG)_(24)-HA.When both M2e and HA were coupled,the synthesized dual-antigen vaccine candidate AFt-(PEG)_(24)-M2e/HA elicited high level of M2e and HA antigen-specific antibodies and conferred 100%protection against lethal infection of homologous PR8 HI N1 virus strain and 70%protection against a heterologous A/FM/1/47(FM1,H1N1)strain,which was more effective than the M2e or HA single antigen vaccine candidates.The potential cross-protective effect of the dual-antigen vaccine was further demonstrated by significant specific hemagglutination inhibition(HAI)titers in serum of the immunized mice against three other heterologous viral strains including A/Singapore/GPl908/2015(IVR-180)H1N1,A/Anhui/1/2005 H5N1,and A/Hong Kong H3N2.Yanan Sheng Jiangxue Wei Zhengjun Li Zhiguo Su Guanghui Ma Songping Zhang 2022Particuology2022,20,5:0
13Electrospinning of a sandwich-structured membrane with sustained release capability and long-term anti-inflammatory effects for dental pulp regeneration显示文摘Current electro spun membranes used for pulp capping still lack the sustained-release capability and long-term anti-inflammatory effects that are favorable for dental pulp regeneration.In this work,a single-layered poly(lac tic acid)(PLA)electro spun membrane loaded with amorphous calcium phosphate(ACP)and aspirin(PLA/ACP/Aspirin membrane,i.e.,PA A membrane)is sandwiched between two poly(lactic-co-glycolic acid)(PLGA)electro spun membranes as a novel sandwich-structured PLGA and PA A composite electro spun membrane(PLGA-PAA membrane)to resolve the need for sustained-release design and anti-inflammatory effects.Contact angle measurements indicate that the PLGA-PAA membrane is more hydrophilic than the PAA membrane.An in vitro release study reveals that PLGA membranes coated on PAA membrane could slightly slow down ion release,while signiificantly prolonging aspirin release.We also co-cultured membranes with dental pulp stem cells(DPSCs)and human monocytic THP-1 cells to evaluate their osteogenic ability and anti-inflammatory effects,respectively.Compared with the PAA membrane,the PLGA-PAA membrane promotes cell adhesion,proliferation,and osteogenic differentiation.A prolonged anti-inflammatory effect of up to 18 days is also observed in the PLGA-PAA group.The results suggest a promising strategy for fabricating an electro spun membrane system with controlled release capabilities and long-term anti-inflammatory effects for use as pulp-capping material for regeneration of the dentin-pulp complex.Fenghe Yang Jiangxue Wang Xiaoyu Li Zhenzhen Jia Qiang Wang Dazhi Yu Jinyu Li Xufeng Niu 2022Bio-Design and Manufacturing2022,5,2:0
