| 1 | Atorvastatin protects against cerebral ischemia/ reperfusion injury through anti-inflammatory and antioxidant effects显示文摘In addition to its lipid-lowering effect, atorvastatin exerts anti-inflammatory and antioxidant effects as well. In this study, we hypothesized that atorvastatin could protect against cerebral ischemia/reperfusion injury. The middle cerebral artery ischemia/reperfusion model was established, and atorvastatin, 6.5 mg/kg, was administered by gavage. We found that, after cerebral ischemia/ reperfusion injury, levels of the inflammation-related factors E-selectin and myeloperoxidase were upregulated, the oxidative stress-related marker malondialdehyde was increased, and superoxide dismutase activity was decreased in the ischemic cerebral cortex. Atorvastatin pretreatment significantly inhibited these changes. | 2 | Neutrophil extracellular traps mediate the crosstalk between glioma progression and the tumor microenvironment via the HMGB1/RAGE/IL-8 axis显示文摘Objective:Neutrophil extracellular traps(NETs)produced by tumor-infiltrating neutrophils(TINs)are associated with poor prognosis in patients with several types of cancer.However,the mechanisms underlying the involvement of NETs in glioma progression remain largely unknown.This study aimed to elucidate the roles of NETs in biological processes that drive the crosstalk between glioma progression and the tumor microenvironment.Methods:Neutrophil infiltration and NETs formation were investigated in glioma tissue through immunohistochemistry,and their relationships with clinicopathological features and outcomes were statistically evaluated.The effects of NETs on glioma cell progression were studied in a co-culture system.In vivo and in vitro experiments validated the reactive oxygen species activity and cytokine production of TINs,as well as the ERK signaling pathway activation and the metastasis of gliomas.Results:Neutrophil infiltration and NETs formation were induced in high-grade glioma compared with low-grade glioma.NETs induced by TINs were determined to be an oncogenic marker of high-grade gliomas and to be involved in cell proliferation and invasion.NETs overproduction promoted glioma cell proliferation,migration,and invasion.Furthermore,HMGB1 was found to bind to RAGE and activate the NF-κB signaling pathway in vitro.In addition,NETs stimulated the NF-κB signaling pathway,thus promoting IL-8 secretion in glioblastoma.Subsequently,IL-8 recruited neutrophils which in turn mediated NETs formation via the PI3 K/AKT/ROS axis in TINs.Conclusions:Our results suggest that NETs produced by TINs mediate the crosstalk between glioma progression and the tumor microenvironment by regulating the HMGB1/RAGE/IL-8 axis.Targeting NETs formation or IL-8 secretion may be an effective approach to inhibit glioma progression. | Caijun Zha Xiangqi Meng Lulu Li Shan Mi Da Qian Ziwei Li Pengfei Wu Shaoshan Hu Shihong Zhao Jinquan Cai Yanhong Liu | 2020 | Cancer Biology & Medicine2020,17,1: | 19 |
| 3 | Prognostic value of tertiary lymphoid structure and tumour infiltrating lymphocytes in oral squamous cell carcinoma显示文摘Tertiary lymphoid structures(TLS)are ectopic lymphoid structures in cancers that are largely associated with favourable prognosis.However,the prognostic value of TLSs in oral squamous cell carcinoma(OSCC)is largely unknown,and the association between tumour infiltrating lymphocytes(TILs)and TLSs has been rarely explored in OSCC.In this study,associated markers of TLS,including peripheral node address(PNAd)in high endothelial venules,CD20 in B cells and CD3 in T cells,were examined in 168 OSCC patients,and survival analysis was performed between TLS-positive and TLS-negative cohorts.We detected the presence of TILs by staining CD8+cytotoxic T cells and CD57+NK cells as well.TLSs appeared as highly organized structures in 45(26.8%)cases.TLSpositive patients had a better 5-year overall survival(OS)rate(88.9%vs.56.1%,P<0.001)and relapse-free survival(RFS)rate(88.9%vs.63.4%,P=0.002).Moreover,the presence of TLS was an independent prognostic factor for both the 5-year OS rate(hazard ratio[HR]=3.784;95%confidence interval[CI],1.498–9.562)and RFS rate(HR=3.296;95%CI,1.279–8.490)in multivariate analysis.Furthermore,a higher density of CD8+T cells and CD57+NK cells was found in TLS-positive sections than in TLS-negative counterparts(P<0.001),and their combination provided a higher predictive accuracy(AUC=0.730;95%CI,0.654–0.805).In conclusion,our results suggest that TLS is an independent positive prognostic factor for OSCC patients.These findings provide a theoretical basis for the future diagnostic and therapeutic value of TLSs in OSCC treatment. | Qunxing Li Xiangqi Liu Dikan Wang Yanqiong Wang Huanzi Lu Shuqiong Wen Juan Fang Bin Cheng Zhi Wang | 2020 | International Journal of Oral Science2020,12,3: | 12 |

| 5 | Transcriptomic and genetic approaches reveal an essential role of the NAC transcription factor SlNAP1 in the growth and defense response of tomato显示文摘With global climate change,plants are frequently being exposed to various stresses,such as pathogen attack,drought,and extreme temperatures.Transcription factors(TFs)play crucial roles in numerous plant biological processes;however,the functions of many tomato(Solanum lycopersicum L.)TFs that regulate plant responses to multiple stresses are largely unknown.Here,using an RNA-seq approach,we identified SlNAP1,a NAC TF-encoding gene,which was strongly induced by various stresses.By generating SlNAP1 transgenic lines and evaluating their responses to biotic and abiotic stresses in tomato,we found that SlNAP1-overexpressing plants showed significantly enhanced defense against two widespread bacterial diseases,leaf speck disease,caused by Pseudomonas syringae pv.tomato(Pst)DC3000,and root-borne bacterial wilt disease,caused by Ralstonia solanacearum.In addition,SlNAP1 overexpression dramatically improved drought tolerance in tomato.Although the SlNAP1-overexpressing plants were shorter than the wild-type plants during the early vegetative stage,eventually,their fruit yield increased by 10.7%.Analysis of different hormone contents revealed a reduced level of physiologically active gibberellins(GAs)and an increased level of salicylic acid(SA)and abscisic acid(ABA)in the SlNAP1-overexpressing plants.Moreover,EMSAs and ChIP-qPCR assays showed that SlNAP1 directly activated the transcription of multiple genes involved in GA deactivation and both SA and ABA biosynthesis.Our findings reveal that SlNAP1 is a positive regulator of the tomato defense response against multiple stresses and thus may be a potential breeding target for improving crop yield and stress resistance. | Jiao Wang Chenfei Zheng Xiangqi Shao Zhangjian Hu Jianxin Li Ping Wang Anran Wang Jingquan Yu Kai Shi | 2020 | Horticulture Research2020,7,1: | 9 |
| 6 | Photodegradation of 17α-ethynylestradiol in dissolved humic substances solution:Kinetics,mechanism and estrogenicity variation显示文摘17α-Ethynylestradiol(EE2) in natural waters may cause adverse effects on organisms due to its high estrogenic potency. Laboratory studies were performed to study the effects of a local humic acid(LHA), fulvic acid(LFA) and Aldrich humic acid(AHA) on the photochemical behavior and estrogenic potency of EE2. Here photolytic experiments demonstrated that pure aqueous EE2 could undergo direct and self-sensitized photodegradation at a global rate of 0.0068 hr^(-1).Photodegradation rate of EE2 in 5.0 mg/L dissolved humic substances(DHS) was determined to be 0.0274, 0.0296 and 0.0254 hr^(-1) for LHA, LFA and AHA, respectively. Reactive oxygen species(ROS) and triplet dissolved humic substances(~3DHS*) scavenging experiments indicated that the promotion effect of DHS on EE2 photodegradation was mainly aroused by the reactions of HOU(35%–50%),~1O_2(<10%) andDHS*(22%–34%). However, the photodegradation of EE2 could also be inhibited when DHS exceeded the threshold of 10 mg/L. Three hydroxylation products of EE2 were identified using GC–MS and their formation pathways were also proposed. In vitro estrogenicity tests showed that EE2 was transformed into chemicals without estrogenic potency. It usually incubates from atomic/molecular level and progressively aggravates along with lithiation. Understanding the internal failure is of great importance for developing solutions of failure prevention and advanced anode materials. In this research, different internal failure processes of anode materials for lithium batteries are discussed. | 14 | Neck observation versus elective neck dissection in management of clinical T1/2N0 oral squamous cell carcinoma: a retrospective study of 232 patients显示文摘Objective: The management of early-stage(cT1/2N0) oral squamous cell carcinoma(OSCC) remains a controversial issue. The aim of this study was to compare the clinical outcomes of neck observation(OBS) and elective neck dissection(END) in treating patients with cT1/2N0 OSCC.Methods: A total of 232 patients with cT1/2N0 OSCC were included in this retrospective study. Of these patients, 181 were treated with END and 51 with OBS. The survival curves of 5-year overall survival(OS), diseasespecific survival(DSS), and recurrence-free survival(RFS) rates were plotted using the Kaplan-Meier method for each group, and compared using the Log-rank test.Results: There was no significant difference in 5-year OS and DSS rates between END and OBS groups(OS:89.0% vs. 88.2%, P=0.906; DSS: 92.3% vs. 92.2%, P=0.998). However, the END group had a higher 5-year RFS rate than the OBS group(90.1% vs. 76.5%, P=0.009). Patients with occult metastases in OBS group(7/51) had similar 5-year OS rate(57.1% vs. 64.1%, P=0.839) and DSS rate(71.4% vs. 74.4%, P=0.982) to those in END group(39/181). In the regional recurrence patients, the 5-year OS rate(57.1% vs. 11.1%, P=0.011) and DSS rate(71.4% vs. 22.2%, P=0.022) in OBS group(7/51) were higher than those in END group(9/181).Conclusions: The results indicated that OBS policy could obtain the same 5-year OS and DSS as END. It could transfer useful alien genes into wheat varieties, enhancing genetic diversity for investigation of genetics and breeding. In the present study, two new procedures, at genome level and single chromosome level, were established.Compared with the classical chromosome engineering, the chromosome engineering of pollen wheat has the following main characters: ( ⅰ ) diversity of products, ( ⅱ ) rapid stability and (ⅲ) high efficiency of selection. | 16 | Development, identification and utilization of introgression lines using Chinese endemic and synthetic wheat as donors显示文摘Chromosome segmental introgression lines(ILs)are an effective way to utilize germplasm resources in crops.To improve agronomic traits of wheat cultivar(Triticum aestivum) Shi 4185, four sets of ILs were developed. The donors were Chinese endemic subspecies accessions Yunnan wheat(T. aestivum ssp. yunnanense) YN3, Tibetan semiwild wheat(T. aestivum ssp. tibetanum) XZ-ZM19450, and Xinjiang wheat(T. aestivum ssp. petropavlovskyi) XJ5, and synthetic wheat HC-XM1620 derived from a cross between T. durum acc. D67.2/P66.270 with Aegilops tauschii acc. 218.Totals of 356, 366, 445 and 457 simple sequence repeat(SSR)markers were polymorphic between Shi 4185 and YN3, XZZM19450, XJ5 and HC-XM1620, respectively. In total, 991 ILs were identified, including 300 derived from YN3, covering 95%of the genome of Shi 4185, 218 from XZ-ZM19450(79%), 279 from XJ5(97%), and 194 from HC-ZX1620(84%). The sizes and locations of each introgression were determined from a consensus SSR linkage map. Using the ILs, 11 putative Resear quantitative trait loci(QTLs) were identified for plant height(PH), spike length(SL) and grain number per spike(GNS).Comparative analyses of 24 elite ILs with the parents revealed that the four donor parents could be important resources to improve wheat SL and GNS.
