| 1 | Micro-droplet Digital Polymerase Chain Reaction and Real-Time Quantitative Polymerase Chain Reaction Technologies Provide Highly Sensitive and Accurate Detection of Zika Virus显示文摘The establishment of highly sensitive diagnostic methods is critical in the early diagnosis and control of Zika virus(ZIKV)and in preventing serious neurological complications of ZIKV infection. In this study, we established micro-droplet digital polymerase chain reaction(ddPCR) and real-time quantitative PCR(RT-qPCR) protocols for the detection of ZIKV based on the amplification of the NS5 gene. For the ZIKV standard plasmid, the RT-qPCR results showed that the cycle threshold(Ct) value was linear from 10~1 to 10~8 copy/l L, with a standard curve R^2 of 0.999 and amplification efficiency of 92.203%;however, a concentration as low as 1 copy/l L could not be detected. In comparison with RT-qPCR, the dd PCR method resulted in a linear range of 10~1–10~4 copy/l L and was able to detect concentrations as low as 1 copy/l L. Thus, for detecting ZIKV from clinical samples, RT-qPCR is a better choice for high-concentration samples(above 10~1 copy/l L),while ddPCR has excellent accuracy and sensitivity for low-concentration samples. | 2 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 3 | Short-term and imminent geomagnetic anomalies of the Wenchuan M_S8.0 earthquake and exploration on earthquake forecast显示文摘The diurnal variation of the geomagnetic vertical component is exhibited mainly by changes of phase and amplitude before strong earthquakes.Based on data recorded by the network of geomagnetic observatories in China for many years, the anomalous features of the appearance time of the minima of diurnal variations(i.e, low-point time) of the geomagnetic vertical components and the variation of their spatial distribution(i.e, phenomena of low-point displacement) have been studied before the Wenchuan MS8.0 earthquake.The strong aftershocks after two months' quiescence of M6 aftershocks of the MS8.0 event were forecasted based on these studies.There are good correlativities between these geomagnetic anoma-lies and occurrences of earthquakes.It has been found that most earthquakes occur near the boundary line of sudden changes of the low-point time and generally within four days before or after the 27th or 41st day counting from the day of the appearance of the anomaly.In addition, the imminent anomalies in diurnal-variation amplitudes near the epicentral areas have also been studied before the Wenchuan earthquake. | Wuxing Wang Jianhai Ding Surong Yu Yongxian Zhang | 2009 | Earthquake Science2009，22，2： | 2 |
| 4 | Genesis of S-type Granites in the Pengshan Sn-polymetallic Ore Field, Northern Jiangxi Province and its Implications显示文摘The Pengshan Sn-polymetallic ore field is located in the southeastern part of the Yangtze block,spanning the southeast edge of the MLYDZ and the northern edge of the mid-segment of the Jiangnan Uplift,and on one side of the MLYDZ.The studies of LA-ICP-MS zircon U-Pb chronology and petrogeochemistry for Early Cretaceous acid granites from the Pengshan ore field were carried out in this paper.We report zircon U-Pb geochronology and whole-rock geochemistry for acid granites in the Pengshan ore field.The zircon U-Pb ages of the muscovite-granite,biotite adamellite and granite-porphyry are 127.6±1.7 Ma,126.9±1.6 Ma and 126.6±2.0 Ma,respectively.The granites in Pengshan are characterized by a high silicon content and are rich in alkali.They belong to high-potassium,calc-alkaline,peraluminous granite.The rocks have a relatively high Rb/Ba ratio,and the data points for muscovite-granite and biotite adamellite all fall within the clay-rich sources region,near the pelite-derived end-member,showing that the Pengshan muscovite-granite and biotite adamellite mainly originated from the partial melting of metapelites with high maturity.The transformation of the compressional and extensional tectonics in this region approximately 128 Ma obviously lags behind that in the mid-segment of the Jiangnan Uplift(135 Ma),but occurred earlier than the MLYDZ(126 Ma).The Pengshan ore field extends from the mid-segment of the Jiangnan Uplift to the MLYDZ.Although the tectonic stress field is constrained by the combination of the two secondary tectonic units,the time of tectonic system transformation is closer to the MLYDZ because the spatial orientation of the area is enclosed in the MLYDZ.Relevant geophysical and drilling data confirm the rationality of Pengshan-Ao'xia as a multi-center vertical zoning ore field,and show the scientificity of the prospecting idea of abutting joint between the north-west of Pengshan area and the south-east of Ao'xia area. | YIN Qingqing XIANG Xinkui YU Zhendong YANG Xiufeng WANG Tianchen ZHONG Bo TAN Rong LIAO Jianhai ZHU Yuansong | 2020 | Acta Geologica Sinica(English Edition)2020，94，6： | 0 |

| 6 | simple and effective aerosol pathogen disinfection test for a flowing air disinfector显示文摘Aerosol transmission is an important disease transmission route and has been especially pertinent to hospital and biosafety laboratories during the SARS-CoV-2 pandemic.The thermal resistance of airborne SARS-CoV-2 is lower than that of Bacillus subtilis spores,which are often used to test the effectiveness of SARS-CoV-2 and other pathogen disinfection methods.Herein,we propose a new method to test the disinfection ability of a flowing air disinfector(a digital electromagnetic induction air heater)using B.subtilis spores.The study provides an alternative air disinfection test method.The new test system combined an aerosol generator and a respiratory filter designed in-house and could effectively recover spores on the filter membrane at the air outlet after passing through the flowing air disinfector.The total number of bacterial spores used in the test was within the range of 5×10^(5)–5×10^(6)colony-forming units(CFUs)specified in the technical standard for disinfection.The calculation was based on the calculation method in Air Disinfection Effect Appraisal Test in Technical Standard for Disinfection(2002 Edition).At an air speed of 3.5 m/s,we used a digital electromagnetic induction air heater to disinfect flowing air containing 4.100×10^(6)CFUs of B.subtilis spores and determined that the minimum disinfection temperature was 350℃for a killing rate of 99.99%.At 400℃,additional experiments using higher spore concentrations(4.700×10^(6)±1.871×10^(5)CFU)and a higher airspeed(4 m/s)showed that the killing rate remained>99.99%.B.subtilis spores,as a biological indicator for testing the efficiency of dry-heat sterilization,were killed by the high temperatures used in this system.The proposed method used to test the flowing air disinfector is simple,stable,and effective.This study provides a reference for the development of test systems that can assess the disinfection ability of flowing air disinfectors. | Xuling Liu Zhiran Qin Linqing Wang Xiaoting Xie Yifang Fu Jianhai Yu Zuxin Liang Xiaoen He Jingshu Li Hong Dai Jinxiu Yao Qinghua Wu Weiwei Xiao Li Zhu Chengsong Wan Bao Zhang Wei Zhao | 2023 | Journal of Biosafety and Biosecurity2023，5，1： | 0 |
