| 2 | Detection of tris-(2, 3-dibromopropyl) isocyanurate as a neuronal toxicant in environmental samples using neuronal toxicity-directed analysis显示文摘Neuronal toxic pollutants in environment possess hazards to human health. It is essential to determine the causative neuronal toxicants in environmental samples. In the present study, viability of primary cultured cerebellar granule neurons (CGNs), combined with sample extraction, chemical fractionation and identification, was applied for screening acid-resistant neuronal toxic substances in environmental samples. River sediments and agricultural soils along the river near a brominated flame retardant (BFR) manufacturing plant in South China were collected to screen the key neuronal toxicants. The results indicated that the manufacturing plant was a source of neuronal toxicity risks. In the sediment and soil near the plant, one of the causative toxicants was identified as tris-(2,3-dibromopropyl) isocyanurate (TBC) using HPLC-MS/MS. | 3 | Alterations of endogenous metabolites in urine of rats exposed to decabromodiphenyl ether using metabonomic approaches显示文摘There is large usage of polybrominated diphenyl ethers(PBDEs) especially for decabromodiphenyl ether(BDE-209, Deca-BDE) in controlling the risks of fire. The toxicological effects of PBDEs are worth being concerned about. Female SD rats were daily gavaged with BDE-209 ether at the dose of 100 mg/kg for 20 days. Histological observation was performed for the screening of the target organs for BDE-209 exposure. The distribution and metabolism of PBDEs in the exposed main organs were evidenced by HRGC-HRMS. Alterations of the endogenous metabolite concentrations in urine were investigated using metabonomic approaches based on1H NMR spectrum. Histopathological changes including serious edema in kidney, hepatocellular spotty necrosis and perivasculitis in liver indicated that BDE-209 caused potential influences on endogenous metabolism in the exposed liver and the kidney. BDE-209 was found to be highly accumulated in lipid, ovary, kidney and liver after 20 days' exposure. Occurrence of other lower brominated PBDEs in the rats demonstrated that reductive debromination process happened in vivo. Hydroxylated and methoxylated–BDEs, as metabolism products, were also detected in the rat tissues. A total of 12 different endogenous metabolites showed obvious alterations in urine from the exposed rats, indicating the disturbance of the corresponding internal biochemical processes induced by BDE-209 exposure. | 4 | Chlorination pattern effect on thermodynamic parameters and environmental degradability for C_(10)-SCCPs: Quantum chemical calculation based on virtual combinational library显示文摘Short-chain chlorinated paraffins(SCCPs) are still controversial candidates for inclusion in the Stockholm Convention.The inherent mixture nature of SCCPs makes it rather difficult to explore their environmental behaviors.A virtual molecule library of 42,720 C_(10)-SCCP congeners covering the full structure spectrum was constructed.We explored the structural effects on the thermodynamic parameters and environmental degradability of C_(10)-SCCPs through semi-empirical quantum chemical calculations.The thermodynamic properties were acquired using the AM1 method,and frontier molecular orbital analysis was carried out to obtain the E_(HOMO),E_(LUMO) and E_(LUMO)-E_(HOMO) for degradability exploration at the same level.The influence of the chlorination degree(N_(Cl) on the relative stability and environmental degradation was elucidated.A novel structural descriptor,μ,was proposed to measure the dispersion of the chlorine atoms within a molecule.There were significant correlations between thermodynamic values and N_(Cl),while the reported N_(Cl)-dependent pollution profile of C_(10)-SCCPs in environmental samples was basically consistent with the predicted order of formation stability of C_(10)-SCCP congeners.In addition,isomers with largeμ showed higher relative stability than those with small μ.This could be further verified by the relationship between μ and the reactivity of nucleophilic substitution and · OH attack respectively.The C_(10)-SCCP congeners with less Cl substitution and lower dispersion degree are susceptible to environmental degradation via nucleophilic substitution and hydroxyl radical attack,while direct photolysis of C_(10)-SCCP congeners cannot readily occur due to the large E_(LUMO)-E_(HOMO) values.The chlorination effect and the conclusions were further checked with appropriate density functional theory(DFT) calculations. | Yuzhen Sun Wenxiao Pan Yuan Lin Jianjie Fu Aiqian Zhang | 2016 | Journal of Environmental Sciences2016,28,1: | 2 |
| 5 | Experimental Study on Analgesia and Hemostasis of Paris polyphylla,Scutellaria baicalensis and Their Compatibility显示文摘[Objectives]The analgesic and hemostatic effects of Paris polyphylla,Scutellaria baicalensis,and their compatibility were studied.[Methods](i)In the experiment on the writhing of mice induced by acetic acid,60 Kunming SPF mice were randomly selected,and were randomly divided into 6 groups according to the male-to-female ratio of 1∶1,including blank control group,P.polyphylla group,S.baicalensis group,and three P.polyphylla and S.baicalensis compatibility groups(with the ratio of 1∶2,1∶1 and 1∶2).There were 10 mice in each group.In the experimental groups,the mice were given 1 g/mL Chinese medicine extract according to a dose of 20 mL/kg.In the control group,the mice were given 0.9%normal saline in equal volumes.Gavage was performed one time every 24 h and lasted for 14 d.1 h after the gavage on the 14th day,they were given 0.5%glacial acetic acid solution via intraperitoneal injection.(ii)In the hot plate experiment,60 Kunming SPF mice were selected,and the ratio of male to female,grouping,administration and gavage were the same as those of the glacial acetic acid-induced writhing experiment.Gavage was lasted for 14 d.1 h after the gavage on the 14th day,the mice were placed on a hot plate apparatus at(55±0.5)℃,and the time of licking hindfoot was measured.(iii)The hemostatic effect was explored through three experimental methods of tail hemostasis,femoral artery hemostasis and liver hemostasis.60 Kunming SPF mice were taken in each of the three experiments,and the male-female ratio and grouping were the same as above.The powder was covered on the surface of the wound sites,and the bleeding was observed.The hemostatic time was recorded,and the hemostatic time was recorded as 3 min if it exceeded 3 min.[Results](i)In the acetic acid-induced writhing experiment,compared with the blank control group,the pain of mice in the experimental groups was inhibited.Among them,P.polyphylla group,S.baicalensis group,compatibility 1∶2 group,compatibility 1∶1 group,and compatibility 2∶1 group had significant effects(P<0.05),and the inhibition rate of writhing was 20.43%,28.32%,52.30%,32.79%,and 39.02%,respectively.The analgesic effect of the compatibility 1∶2 group was the most obvious.(ii)In the hot plate experiment,compared with the blank control group,the experimental groups had analgesic effect.Among them,P.polyphylla group,S.baicalensis group,compatibility 1∶2 group,compatibility 1∶1 group,and compatibility 2∶1 group had significant effects(P<0.05),and the increase rate of pain threshold was 29.17%,47.83%,61.54%,and 50.61%,and 53.83%,respectively.The analgesic effect of the compatibility 2∶1 group was the most significant.(iii)In the hemostatic experiment,there was a significant difference in hemostatic time between the compatibility 1∶2 group and the blank control group,P.polyphylla group,and S.baicalensis group(P<0.05).The hemostatic effect of the compatibility 1∶2 group was the best.[Conclusions]P.polyphylla,S.baicalensis,and its compatibility had good analgesic and hemostatic effects,and the best compatibility ratio was 1∶2. | Shibo ZHAO Suoyi HUANG Zhenjing SI Lanlan GAN Shusheng HUANG Ye FU Jianjie LAN Xueying YU Yuekong CEN | 2020 | Medicinal Plant2020,11,2: | 2 |
| 6 | Mercury in alpine fish from four rivers in the Tibetan Plateau显示文摘As a global pollutant, high levels of mercury(Hg) have been found in remote ecosystem due to the long range atmospheric transport. In this study, a total of 60 fish samples were collected from four rivers across the Tibetan Plateau to study the accumulation of Hg in remote and high-altitude aquatic environment. The total Hg(THg) and methylmercury(MeHg) in fish muscles ranged from 11 to 2097 ng/g dry weight(dw)(average: 819 ng/g dw)and from 14 to 1960 ng/g dw(average: 756 ng/g dw), respectively. Significantly positive linear relationships were observed between the THg(r = 0.591, p < 0.01, n = 36) and MeHg concentrations(r = 0.473, p < 0.01, n = 36) with the trophic level of fish from Lhasa River,suggesting trophic transfer and biomagnification of Hg in this aquatic ecosystem. Moreover,the THg levels in fish had significantly positive correlations with the length(r = 0.316,p < 0.05, n = 60) and weight(r = 0.271, p < 0.05, n = 60) of fish. The high levels of Hg were attributed to the slow growth and long lifespan of the fish under this sterile and cold environment. Consequently, concern has arisen regarding their potential effects on the environment and human health. However, the current understanding on the stability of Ag NPs is limited. Therefore, in this study the influence of environmental and biological factors, such as ionic strength, valence, biological molecules, pH and temperature, on the properties of Ag NPs was investigated. The absorption spectra, size, zeta potential, and release of silver ions were evaluated. Ionic strength, valence, Dulbecco’s Modified Eagle’s Medium (DMEM), without fetal bovine serum (FBS), reduced the stability of the Ag NPs. However, in DMEM with 10% FBS, the Ag NPs maintained their stability and nanoscale characteristics. | 16 | Inhibition of thrombin by functionalized C_(60)nanoparticles revealed via in vitro assays and in silico studies显示文摘The studies on the human toxicity of nanoparticles(NPs) are far behind the rapid development of engineered functionalized NPs. Fullerene has been widely used as drug carrier skeleton due to its reported low risk. However, different from other kinds of NPs, fullerene-based NPs(C_(60) NPs) have been found to have an anticoagulation effect, although the potential target is still unknown. In the study, both experimental and computational methods were adopted to gain mechanistic insight into the modulation of thrombin activity by nine kinds of C_(60) NPs with diverse surface chemistry properties. In vitro enzyme activity assays showed that all tested surface-modified C_(60) NPs exhibited thrombin inhibition ability. Kinetic studies coupled with competitive testing using 3 known inhibitors indicated that six of the C_(60) NPs, of greater hydrophobicity and hydrogen bond(HB) donor acidity or acceptor basicity, acted as competitive inhibitors of thrombin by directly interacting with the active site of thrombin. A simple quantitative nanostructure-activity relationship model relating the surface substituent properties to the inhibition potential was then established for the six competitive inhibitors.Molecular docking analysis revealed that the intermolecular HB interactions were important for the specific binding of C_(60) NPs to the active site canyon, while the additional stability provided by the surface groups through van der Waals interaction also play a key role in the thrombin binding affinity of the NPs. | 20 | Trophic transfer of mercury and methylmercury in an aquatic ecosystem impacted by municipal sewage effluents in Beijing, China显示文摘Gaobeidian Lake, located in Beijing, China, serves as a recipient lake for efluents from a large municipal sewage treatment plant (MSTP). In order to evaluate the effects of discharging MSTP efluent on the mercury contamination of the local aquatic ecosystem, sediment cores, water, plankton, fish, and turtle samples were collected from Gaobeidian Lake for mercury speciation analysis. High concentrations of total mercury (T-Hg) were detected in sediment cores (5.24–17.0 μg/g dry weight (dw), average: 10.1 μg/g). The ratio of methylmercury (MeHg) to T-Hg was less than 0.3% in sediments and ranged from 35% to 76% in biota samples. The highest level of T-Hg and MeHg were found in aquatic bryophyte and crucian carp (3673 and 437 ng/g dw, respectively).
