2Contribution of atmospheric nitrogen deposition to diffuse pollution in a typical hilly red soil catchment in southern China显示文摘Atmospheric nitrogen(N) deposition is currently high and meanwhile diffuse N pollution is also serious in China. The correlation between N deposition and riverine N export and the contribution of N deposition to riverine N export were investigated in a typical hilly red soil catchment in southern China over a two-year period. N deposition was as high as 26.1 to55.8 kg N/(ha·yr) across different land uses in the studied catchment, while the riverine N exports ranged from 7.2 to 9.6 kg N/(ha·yr) in the forest sub-catchment and 27.4 to30.3 kg N/(ha·yr) in the agricultural sub-catchment. The correlations between both wet N deposition and riverine N export and precipitation were highly positive, and so were the correlations between NH4+-N or NO3--N wet deposition and riverine NH4+-N or NO3--N exports except for NH4+-N in the agricultural sub-catchment, indicating that N deposition contributed to riverine N export. The monthly export coefficients of atmospheric deposited N from land to river in the forest sub-catchment(with a mean of 14%) presented a significant positive correlation with precipitation, while the monthly contributions of atmospheric deposition to riverine N export(with a mean of 18.7% in the agricultural sub-catchment and a mean of 21.0% in the whole catchment) were significantly and negatively correlated with precipitation. 3Soil phosphorus availability and rice phosphorus uptake in paddy fields under various agronomic practices显示文摘Agronomic practices affect soil phosphorus(P) availability, P uptake by plants, and subsequently the efficiency of P use. A field experiment was carried out to investigate the effects of various agronomic practices(straw incorporation, paddy water management, nitrogen(N) fertilizer dose, manure application,and biochar addition) on soil P availability(e.g., soil total P(STP), soil available P(SAP), soil microbial biomass P(SMBP), and rice P uptake as well as P use efficiency(PUE)) over four cropping seasons in a rice-rice cropping system, in subtropical central China. Compared to the non-straw treatment(control,using full dose of chemical N fertilizer), straw incorporation increased SAP and SMBP by 9.3%–18.5% and 15.5%–35.4%, respectively;substituting half the chemical N fertilizer dose with pig manure and the biochar application increased STP, SAP, and SMBP by 10.5%–48.3%, 30.2%–236.0%, and 19.8%–72.4%,respectively, mainly owing to increased soil P and organic carbon inputs;adding a half dose of N and no N input(reduced N treatments) increased STP and SAP by 2.6%–7.5% and 19.8%–33.7%, respectively, due to decreased soil P outputs. Thus, soil P availability was greatly affected by soil P input and use. The continuous flooding water regime without straw addition significantly decreased SMBP by 11.4% compared to corresponding treatments under a mid-season drainage water regime. Total P uptake by rice grains and straws at the harvest stage increased under straw incorporation and under pig manure application, but decreased under the reduced N treatments and under biochar application at a rate of 48 t ha-1, compared to the control. Rice P uptake was significantly positively correlated with rice biomass, and both were positively correlated with N fertilizer application rates, SAP, SMBP, and STP. Phosphorus use efficiency generally increased under straw incorporation but decreased under the reduced N treatments and under the manure application(with excessive P input), compared to the control. These results showed that straw incorporation can be used to increase soil P availability and PUE while decreasing the use of chemical P fertilizers. 5ZmOST1 mediates abscisic acid regulation of guard cell ion channels and drought stress responses显示文摘The phytohormone abscisic acid(ABA) is an important mediator in the drought response, participating in, among other processes, stomatal movements. In Arabidopsis thaliana, the serine/threonine protein kinase,OST1, regulates this response, but the function of its maize homolog has yet to be established. Here, we isolated ZmOST1 and show that its encoded protein indeed acts to regulate guard cell movement. ZmOST1 was ubiquitously expressed throughout the plant, being highly expressed in guard cells, and inducible both by exogenous ABA and water stress. Transient expression of a ZmOST1-GFP fusion protein, in maize mesophyll protoplasts, indicated its subcellular localization in the cytoplasm and nucleus. A Zmost1 loss-of-function mutant exhibited reduced sensitivity to ABA-activated slow anion channels in maize guard cells, and reduced drought tolerance. Constitutive expression of ZmOST1, in an A. thaliana ost1-1 mutant rescued the phenotype with respect both to the sensitivity of guard cell slow anion currents to ABA treatment and stomatal closure. 13Pelagic Manganese Nodules--A New Type of Oncolites显示文摘Pelagic manganese nodules from the East Pacific Ocean have been studied using polarizing,scanning electron and transmission electron microscopes. It has been revealed that the manganesenodules are composed of cores and stromatolite coatings. The structures and textures of thesenodules are peculiar to oncolite. Consequently, the pelagic manganese nodules are manganeseoncolites. Based on the stromatolites in the coatings, the manganese oncolites from the East Paci-fic Ocean can be divided into two types. One is smooth on surface and dense inside. Its coatingsare composed mainly of Minima. The other has a knobby surface and is loose internally. Itscoatings consist mainly of Admirabilis. The TEM investigation has also revealed that the con-structors of the manganese oncolites are ultra-microbes. 14Bacterial Metabolism-Initiated Nanocatalytic Tumor Immunotherapy显示文摘The low immunogenicity of tumors remains one of the major limitations of cancer immunotherapy.Herein,we report a bacterial metabolisminitiated and photothermal-enhanced nanocatalytic therapy strategy to completely eradicate primary tumor by triggering highly effective antitumor immune responses.Briefly,a microbiotic nanomedicine,designated as Cu_(2)O@ΔSt,has been constructed by conjugating PEGylated Cu_(2)O nanoparticles on the surface of an engineered Salmonella typhimurium strain(ΔSt).Owing to the natural hypoxia tropism ofΔSt,Cu_(2)O@ΔSt could selectively colonize hypoxic solid tumors,thus minimizing the adverse effects of the bacteria on normal tis-sues.Upon bacterial metabolism within the tumor,Cu_(2)O@ΔSt generates H_(2)S gas and other acidic substances in the tumor microenvironment(TME),which will in situ trigger the sulfidation of Cu_(2)O to form CuS facilitating tumor-specific photothermal therapy(PTT)under local NIR laser irradiation on the one hand.Meanwhile,the dissolved Cu+ions from Cu_(2)O into the acidified TME enables the nanocatalytic tumor therapy by catalyzing the Fenton-like reaction of decom-posing endogenous H_(2)O_(2) into cytotoxic hydroxyl radicals(·OH)on the other hand.Such a bacterial metabolism-triggered PTT-enhanced nanocatalytic treatment could effectively destroy tumor cells and induce a massive release of tumor antigens and damage-associated molecular patterns,thereby sensitizing tumors to checkpoint blockade(ICB)therapy.The combined nanocatalytic and ICB therapy results in the much-inhibited growth of distant and metastatic tumors,and more importantly,induces a powerful immunological memory effect after the primary tumor ablation.Wencheng Wu Yinying Pu Shuang Gao Yucui Shen Min Zhou Heliang Yao Jianlin Shi 2022Nano-Micro Letters2022,14,12:1

20Quantitative analysis of glutamate compounds in the swine brain following central analgesics nasal spray using proton magnetic resonance spectroscopy and linear combination model显示文摘<正>BACKGROUND:In localized brain proton magnetic resonance spectroscopy(~1H-MRS),metabolite levels are often expressed as ratios,rather than absolute concentrations.Frequently,the denominator is creatine,which is assumed to be stable in normal,as well as many pathological, states.However,in vivo creatine levels do not remain constant.Therefore,absolute metabolite measurements,which provide the precise concentrations of certain chemical compounds,are superior to metabolite ratios for determining pathological and evolutional changes. OBJECTIVE:To investigate the feasibility of quantification analysis of brain metabolite changes caused by central analgesics nasal spray using the ~1H-MRS and linear combination model(LCModel) methods. DESIGN,TIME AND SETTING:This neuroimaging,observational,animal study was performed at the Laboratory of the Department of Medical Imaging,Second Affiliated Hospital,Medical College, Shantou University,China from July to December 2007. MATERIALS:Butorphanol tartrate nasal spray,as a mixed agonist-antagonist opioid analgesic,was purchased from Shanghai Hengrui Pharmacy,China.A General Electric Signa 1.5T System (General Electric Medical Systems,Milwaukee,Wl,USA) and LCModel software(Stephen Provencher,Oakville,Ontario,Canada) were used in this study. METHODS:MRS images were acquired in ten healthy swine aged 2 weeks using single-voxel point-resolved spectroscopic sequence.A region of interest(2 cm×2 cm×2 cm) was placed in the image centers of maximum brain parenchyma.Repeated MRS scanning was performed 15-20 minutes after intranasal administration of 1 mg of butorphanol tartrate.Three settings of repetition time/echo time were selected before and after nasal spray administration 3000 ms/30 ms, 1500 ms/30 ms,and 3000 ms/50 ms.Metabolite concentrations were estimated by LCModel software. MAIN OUTCOME MEASURES:~1H-MRS spectra was obtained using various repetition time/echo time settings.Concentrations of glutamate compounds(glutamate+glutamine),N-acetyl aspartate, and choline were detected in swine brain prior to and following nasal spray treatment. RESULTS:The glutamate compounds curve was consistent with original spectra,when a repetition time/echo time of 3 000 ms/30 ms was adopted.Concentrations of glutamate compounds,N-acetyl aspartate,and choline decreased following administration.The most significant reduction was observed in glutamate compound concentrations from(9.28±0.54) mmol/kg to(7.28±0.54) mmol/kg (P<0.05).
