1Characterization of the receptor-binding domain(RBD)of 2019 novel coronavirus:implication for development of RBD protein as a viral attachment inhibitor and vaccine显示文摘The outbreak of Coronavirus Disease 2019(COVID-19)has posed a serious threat to global public health,calling for the development of safe and effective prophylactics and therapeutics against infection of its causative agent,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),also known as 2019 novel coronavirus(2019-nCoV).The CoV spike(S)protein plays the most important roles in viral attachment,fusion and entry,and serves as a target for development of antibodies,entry inhibitors and vaccines.Here,we identified the receptor-binding domain(RBD)in SARS-CoV-2 S protein and found that the RBD protein bound strongly to human and bat angiotensin-converting enzyme 2(ACE2)receptors.SARS-CoV-2 RBD exhibited significantly higher binding affinity to ACE2 receptor than SARS-CoV RBD and could block the binding and,hence,attachment of SARS-CoV-2 RBD and SARS-CoV RBD to ACE2-expressing cells,thus inhibiting their infection to host cells.SARS-CoV RBD-specific antibodies could crossreact with SARS-CoV-2 RBD protein,and SARS-CoV RBD-induced antisera could cross-neutralize SARS-CoV-2,suggesting the potential to develop SARS-CoV RBD-based vaccines for prevention of SARS-CoV-2 and SARS-CoV infection.Wanbo Tai Lei He Xiujuan Zhang Jing Pu Denis Voronin Shibo Jiang Yusen Zhou Lanying Du 2020Cellular & Molecular Immunology2020,17,6:35
2Shear-wave splitting in the crust:Regional compressive stress from polarizations of fast shear-waves显示文摘When propagating through anisotropic rocks in the crust,shear-waves split into faster and slower components with almost orthogonal polarizations.For nearly vertical propagation the polarization of fast shear-wave(PFS)is parallel to both the strike of the cracks and the direction of maximum horizontal stress,therefore it is possible to use PFS to study stress in the crust.This study discusses several examples in which PFS is applied to deduce the compressive stress in North China,Longmenshan fault zone of east edge of Tibetan plateau and Yunnan zone of southeast edge of Tibetan plateau,also discusses temporal variations of PFS orientations of 1999 Xiuyan earthquake sequences of northeastern China.The results are consistent to those of other independent traditional stress measurements.There is a bridge between crustal PFS and the crustal principal compressive stress although there are many unclear disturbance sources.This study suggests the PFS results could be used to deduce regional and in situ principal compressive stress in the crust only if there are enough seismic stations and enough data.At least,PFS is a useful choice in the zone where there are a large number of dense seismic stations.Yuan Gao Yutao Shi Jing Wu Lingxue Tai 2012Earthquake Science2012,25,1:20
3Regulation of activin receptor-interacting protein 2 expression in mouse hepatoma Hepa1-6 cells and its relationship with collagen type Ⅳ显示文摘AIM: To investigate the regulation of activin receptor-interacting protein 2 (ARIP2) expression and its possible relationships with collagen type Ⅳ (collagen Ⅳ) in mouse hepatoma cell line Hepal-6 cells. METHODS: The ARIP2 mRNA expression kinetics in Hepal-6 cells was detected by RT-PCR,and its regulation factors were analyzed by treatment with signal transduction activators such as phorbol 12-myristate 13-acetate (PMA),forskolin and A23187. After pcDNA3-ARIP2 was transfected into Hepal-6 cells,the effects of ARIP2 overexpression on activin type Ⅱ receptor (ActRⅡ) and collagen Ⅳ expression were evaluated. RESULTS: The expression levels of ARIP2 mRNA in Hapel-6 cells were elevated in time-dependent manner 12 h after treatment with activin A and endotoxin LPS,but not changed evidently in the early stage of stimulation (2 or 4 h). The ARIP2 mRNA expression was increased after stimulated with signal transduction activators such as PMA and forskolin in Hepal-6 cells,whereas decreased after treatment with A23187 (25.3% ± 5.7% vs 48.1% ± 3.6%,P < 0.01). ARIP2 overexpression could remarkably suppress the expression of ActRⅡA mRNA in dose-dependent manner,but has no effect on ActRⅡB in Hepal-6 cells induced by activin A. Furthermore,we have found that overexpression of ARIP2 could inhibit collagen Ⅳ mRNA and protein expressions induced by activin A in Hapel-6 cells. CONCLUSION: These findings suggest that ARIP2 expression can be influenced by various factors. 4Liquid-Solid Phase Equilibria of Nb-Si-Ti Ternary Alloys显示文摘To determine the liquid-solid phase equilibria of the Nb-Si-Ti ternary system,Nb-Si-Ti alloys of different compositions are pre-pared. By means of scanning electron microscopy (SEM),X-ray diffraction (XRD) and electron probe microanalysis (EPMA),the phases in the alloys,such as Si-based solutions,Ti(Nb)Si,Ti(Nb)Si2,Nb(Ti)Si2,Ti(Nb)5Si4,Nb(Ti)5Si3,Ti(Nb)5Si3,Nb(Ti)3Si and Nb-based solutions are identified,and the phase evolution is analyzed. 5Application of Contact Graph Routing in Satellite Delay Tolerant Networks显示文摘Satellite networks have many inherent advantages over terrestrial networks and have become an important part of the global network infrastructure.Routing aimed at satellite networks has become a hot and challenging research topic.Satellite networks,which are special kind of Delay Tolerant Networks(DTN),can also adopt the routing solutions of DTN.Among the many routing proposals,Contact Graph Routing(CGR) is an excellent candidate,since it is designed particularly for use in highly deterministic space networks.The applicability of CGR in satellite networks is evaluated by utilizing the space oriented DTN gateway model based on OPNET(Optimized Network Engineering Tool).Link failures are solved with neighbor discovery mechanism and route recomputation.Earth observation scenario is used in the simulations to investigate CGR's performance.The results show that the CGR performances are better in terms of effectively utilizing satellite networks resources to calculate continuous route path and alternative route can be successfully calculated under link failures by utilizing fault tolerance scheme.ZHU Laitao LI Yong ZHANG Junxiang WU Jing TAI Xiao ZHOU Jianguo 2015空间科学学报2015,35,1:5
6Arterial relaxation is coupled to inhibition of mitochondrial fission in arterial smooth muscle cells:comparison of vasorelaxant effects of verapamil and phentolamine显示文摘Mitochondria are morphologically dynamic organelles which undergo fission and fusion processes. Our previous study found that arterial constriction was always accompanied by increased mitochondrial fission in smooth muscle cells, whereas inhibition of mitochondrial fission in smooth muscle cells was associated with arterial relaxation. Here, we used the typical vasorelaxants, verapamil and phentolamine, to further confirm the coupling between arterial constriction and mitochondrial fission in rat aorta. Results showed that phentolamine but not verapamil induced vasorelaxation in phenylephrine(PE)-induced rat thoracic aorta constriction. Verapamil, but not phentolamine, induced vasorelaxation in high K^+(KPSS)-induced rat thoracic aorta constriction. Pre-treatment with phentolamine prevented PEbut not KPSS-induced aorta constriction and pre-treatment with verapamil prevented both PE-and KPSSinduced aorta constriction. Transmission electron microscopy(TEM) results showed that verapamil but not phentolamine inhibited KPSS-induced excessive mitochondrial fission in aortic smooth muscle cells,and verapamil prevented both PE-and KPSS-induced excessive mitochondrial fission in aortic smoothmuscle cells. Verapamil inhibited KPSS-induced excessive mitochondrial fission in cultured vascular smooth muscle cells(A10). 8Flexible organic thin-film transistors based on poly(3-hexylthiophene) films for nitrogen dioxide detection显示文摘Flexible nitrogen dioxide(NO_2) gas sensors based on organic thin-film transistors(OTFTs) were developed in this work, in which conductive indium tin oxide(ITO) coated polyethylene naphthalene-2,6-dicarboxylate(PEN) was designed for the flexible substrates, and poly methyl methacrylate(PMMA) and poly(3-hexylthiophene)(P3HT) was spin-coated as the gate dielectric layer and spray-deposited as the active layer, respectively. The effects of PMMA concentrations and P3HT solution volume on performances of flexible OTFTs were systematically investigated. The results showed that the optimized flexible OTFT exhibited high field-effect mobility(9.51×10^(–3) cm^2/(V s) at a gate bias of –50 V) and excellent response characteristics,including high sensitivity(up to 0.169 ppm^(–1)), good repeatability and selectivity. The flexibility of the developed OTFT sensor was also investigated, and the results showed that the electrical and gas-sensing properties were affected by the bending cycles,thus further work should be done for improving the flexibility of the sensor. 9Lipomatous hemangiopericytoma of the stomach:A case report and a review of literature显示文摘Lipomatous hemangiopericytomas(LHPCs) are rare soft-tissue tumors that are histologically characterized by hemangiopericytomatous vasculature and the presence of mature adipocytes.We present the clinicopathological features of a case of gastric LHPC in a 56-year-old female,along with a literature review.Endoscopy and endoscopic ultrasound showed a submucosal tumor 0.8 cm across in the greatest dimension in the lesser curvature side of the gastric antrum.Grossly,the well-defined mass had a solid and tan-white cut surface admixed with myxoid regions and yellowish areas.Histological examination revealed a submucosal well-circumscribed lesion composed of cellular nodules with the classic appearance of an hemangiopericytoma admixed with clusters and lobules of mature adipocytes.The ill-defined tumor cells had weakly eosinophilic cytoplasm and contained spindled nuclei with occasional small nucleoli.Nuclei atypia and mitoses were absent,and no cellular atypia,necrosis or vascular invasion was observed.Immunohistochemistry showed that the tumor cells were diffusely positive for CD34,CD99,and vimentin and were focally reactive for bcl-2.This is the first known report of an LHPC in the stomach.The patient was followed for 12 mo without any evidence of metastasis or recurrence.Hong-Biao Jing Qing-Da Meng Yan-Hong Tai 2011World Journal of Gastroenterology2011,17,43:3

11Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. 12Changes on Characteristic Components During Shoots Development in Camellia Sinensis显示文摘Fresh leaves of tea cultivar Shuchazao were harvested from five different stages of shoot development including single-bud(SB),one leaf and one bud(BL1),two leaves and one bud(BL2),three leaves and one bud(BL3)and mature leaves(ML,including five leaves).The contents of tea-specific components,including caffeine,catechins and amino acids,in tea leaves were extracted and analyzed using the HPLC technique.The results showed that the content of caffeine in the buds in BL3 stage was generally the highest,while it did not change much in SB,BL1 and BL2 stages.The content of caffeine in the leaves at the same leaf-age was similar in five different development stages.The total contents of catechins in the first leaf were higher than that in the buds in all five development stages,and it was the highest in the first leaf at BL1 stage,but it decreased with the increase of the leaf-age.As far as the monomeric catechins were concerned,non-ester type catechins and ester type catechins can be detected in the buds or leaves in the five development stages.The majority of catechins were epigallocatechingallate(EGCG),which showed a similar variation tendency as that of total catechins.However,the contents of non-ester type catechins such as epicatechingallate(EGC),epicatechin(EC) and gallocatechin(GC) were higher in buds than in leaves,which were opposite to the content variation trends of ester type catechins in the course of shoot development.Theanine was the major amino acids in all development stages of tender shoots,and its content in the stage of single-bud was the highest.The content of theanine in buds was three or four times higher than in leaves,and the older the leaf-age was,the less theanine existed in the leaves.Jing Zhang Hua Yang Chao-Ling Wei Yu-Ling Tai Qi Chen Xiao-Chun Wan 2013茶叶2013,39,4:2
13Accurate and generalizable quantitative scoring of liver steatosis from ultrasound images via scalable deep learning显示文摘BACKGROUND Hepatic steatosis is a major cause of chronic liver disease.Two-dimensional(2D)ultrasound is the most widely used non-invasive tool for screening and monitoring,but associated diagnoses are highly subjective.AIM To develop a scalable deep learning(DL)algorithm for quantitative scoring of liver steatosis from 2D ultrasound images.METHODS Using multi-view ultrasound data from 3310 patients,19513 studies,and 228075 images from a retrospective cohort of patients received elastography,we trained a DL algorithm to diagnose steatosis stages(healthy,mild,moderate,or severe)from clinical ultrasound diagnoses.Performance was validated on two multiscanner unblinded and blinded(initially to DL developer)histology-proven cohorts(147 and 112 patients)with histopathology fatty cell percentage diagnoses and a subset with FibroScan diagnoses.We also quantified reliability across scanners and viewpoints.Results were evaluated using Bland-Altman and receiver operating characteristic(ROC)analysis.RESULTS The DL algorithm demonstrated repeatable measurements with a moderate number of images(three for each viewpoint)and high agreement across three premium ultrasound scanners.High diagnostic performance was observed across all viewpoints:Areas under the curve of the ROC to classify mild,moderate,and severe steatosis grades were 0.85,0.91,and 0.93,respectively.The DL algorithm outperformed or performed at least comparably to FibroScan control attenuation parameter(CAP)with statistically significant improvements for all levels on the unblinded histology-proven cohort and for“=severe”steatosis on the blinded histology-proven cohort.CONCLUSION The DL algorithm provides a reliable quantitative steatosis assessment across view and scanners on two multi-scanner cohorts.Diagnostic performance was high with comparable or better performance than the CAP.Bowen Li Dar-In Tai Ke Yan Yi-Cheng Chen Cheng-Jen Chen Shiu-Feng Huang Tse-Hwa Hsu Wan-Ting Yu Jing Xiao Lu Le Adam P Harrison 2022World Journal of Gastroenterology2022,28,22:2

16Adsorption behaviors of gas molecules on the surface of ZnO nanocrystals under UV irradiation显示文摘A UV-activated room temperature chemiresistive gas sensor based on ZnO nanocrystals film was fabricated. Its gas sensing properties under various conditions were also investigated in detail. The results shed new insight into the adsorption behaviors of gas molecules on the surface of ZnO nanocrystals under UV irradiation. The chemisorbed oxygen species(O^-2(ads)(hv)) induced by UV light govern the adsorption and desorption ways of other gas molecules on the surface of ZnO nanocrystals, which is dependent on the electron affinity of gas molecules. Gas molecules with higher electron affinity than oxygen molecules can be adsorbed on the surface by the competitive adsorption way, extracting electrons from the surface. Gas molecules with lower electron affinity than oxygen molecules are attracted by the adsorbed O^-2(ads)(hv)layer, releasing electrons to the surface. These processes can influence the gas sensing properties of the sensor.
