1Correlation of Gut Microbiome Between ASD Children and Mothers and Potential Biomarkers for Risk Assessment显示文摘Variation of maternal gut microbiota may increase the risk of autism spectrum disorders(ASDs) in offspring. Animal studies have indicated that maternal gut microbiota is related to neurodevelopmental abnormalities in mouse offspring, while it is unclear whether there is a correlation between gut microbiota of ASD children and their mothers. We examined the relationships between gut microbiome profiles of ASD children and those of their mothers, and evaluated the clinical discriminatory power of discovered bacterial biomarkers. Gut microbiome was profiled and evaluated by 16S ribosomal RNA gene sequencing in stool samples of 59 mother–child pairs of ASD children and 30 matched mother–child pairs of healthy children. Significant differences were observed in the gut microbiome composition between ASD and healthy children in our Chinese cohort. Several unique bacterial biomarkers, such as Alcaligenaceae and Acinetobacter, were identified. Mothers of ASD children had more Proteobacteria, Alphaproteobacteria, Moraxellaceae, and Acinetobacter than mothers of healthy children. There was a clear correlation between gut microbiome profiles of children and their mothers; however, children with ASD still had unique bacterial biomarkers, such as Alcaligenaceae, Enterobacteriaceae, and Clostridium. Candidate biomarkers discovered in this study had remarkable discriminatory power. 3Nasal delivery of broadly neutralizing antibodies protects mice from lethal challenge with SARS-CoV-2 delta and omicron variants显示文摘Multiple new variants of severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)have constantly emerged,as the delta and omicron variants,which have developed resistance to currently gained neutralizing antibodies.This highlights a critical need to discover new therapeutic agents to overcome the variants mutations.Despite the availability of vaccines against coronavirus disease 2019(COVID-19),the use of broadly neutralizing antibodies has been considered as an alternative way for the prevention or treatment of SARS-Co V-2 variants infection.Here,we show that the nasal delivery of two previously characterized broadly neutralizing antibodies(F61 and H121)protected K18-h ACE2 mice against lethal challenge with SARS-Co V-2 variants.The broadly protective efficacy of the F61 or F61/F121 cocktail antibodies was evaluated by lethal challenge with the wild strain(WIV04)and multiple variants,including beta(B.1.351),delta(B.1.617.2),and omicron(B.1.1.529)at 200or 1000 TCID_(50),and the minimum antibody administration doses(5-1.25 mg/kg body weight)were also evaluated with delta and omicron challenge.Fully prophylactic protections were found in all challenged groups with both F61 and F61/H121 combination at the administration dose of 20 mg/kg body weight,and corresponding mice lung viral RNA showed negative,with almost all alveolar septa and cavities remaining normal.Furthermore,low-dose antibody treatment induced significant prophylactic protection against lethal challenge with delta and omicron variants,whereas the F61/H121 combination showed excellent results against omicron infection.Our findings indicated the potential use of broadly neutralizing monoclonal antibodies as prophylactic and therapeutic agent for protection of current emerged SARS-Co V-2 variants infection.Jia Lu Qiangling Yin Rongjuan Pei Qiu Zhang Yuanyuan Qu Yongbing Pan Lina Sun Ding Gao Cuiqin Liang Jingwen Yang Wei Wu Jiandong Li Zongqiang Cui Zejun Wang Xinguo Li Dexin Li Shiwen Wang Kai Duan Wuxiang Guan Mifang Liang Xiaoming Yang 2022Virologica Sinica2022,37,2:4

5Effects of silver nanoparticles with different dosing regimens and exposure media on artificial ecosystem显示文摘Due to the wide use of silver nanoparticles(AgNPs) in various fields, it is crucial to explore the potential negative impacts on the aquatic environment of AgNPs entering into the environment in different ways. In this study, comparative experiments were conducted to investigate the toxicological impacts of polyvinylpyrrolidone-coated silver nanoparticles(PVP-AgNPs) with two kinds of dosing regimens, continuous and one-time pulsed dosing, in different exposure media(deionized water and XiangJiang River water). There were a number of quite different experimental results(including 100% mortality of zebrafish,decline in the activity of enzymes, and lowest number and length of adventitious roots) in the one-time pulsed dosing regimen at high PVP-AgNP concentration exposure(HOE)compared to the three other treatments. Meanwhile, we determined that the concentration of leached silver ions from PVP-AgNPs was too low to play a role in zebrafish death. Those results showed that HOE led to a range of dramatic ecosystem impacts which were more destructive than those of other treatments. Moreover, compared with the continuous dosing regimen, despite the fact that higher toxicity was observed for HOE, there was little difference in the removal of total silver from the aquatic environment for the different dosing regimens. No obvious differences in ecological impacts were observed between different water columns under low concentration exposure. 7Distinct mononuclear diploid cardiac subpopulation with minimal cell-cell communications persists in embryonic and adult mammalian heart显示文摘A small proportion of mononuclear diploid cardiomyocytes(MNDCMs),with regeneration potential,could persist in adult mammalian heart.However,the heterogeneity of MNDCMs and changes during development remains to be illuminated.To this end,12645 cardiac cells were generated from embryonic day 17.5 and postnatal days 2 and 8 mice by single-cell RNA sequencing.Three cardiac developmental paths were identified:two switching to cardiomyocytes(CM)maturation with close CM–fibroblast(FB)communications and one maintaining MNDCM status with least CM–FB communications.Proliferative MNDCMs having interactions with macrophages and non-proliferative MNDCMs(non-pMNDCMs)with minimal cell–cell communications were identified in the third path.The non-pMNDCMs possessed distinct properties:the lowest mitochondrial metabolisms,the highest glycolysis,and high expression of Myl4 and Tnni1.Single-nucleus RNA sequencing and immunohistochemical staining further proved that the Myl4^(+)Tnni1+MNDCMs persisted in embryonic and adult hearts.These MNDCMs were mapped to the heart by integrating the spatial and single-cell transcriptomic data.In conclusion,a novel non-pMNDCM subpopulation with minimal cell–cell communications was unveiled,highlighting the importance of microenvironment contribution to CM fate during maturation.These findings could improve the understanding of MNDCM heterogeneity and cardiac development,thus providing new clues for approaches to effective cardiac regeneration.Miaomiao Zhu Huamin Liang Zhe Zhang Hao Jiang Jingwen Pu Xiaoyi Hang Qian Zhou Jiacheng Xiang Ximiao He 2023Frontiers of Medicine2023,17,5:1
8'Three-in-One' Multi-Scale Structural Design of Carbon Fiber-Based Composites for Personal Electromagnetic Protection and Thermal Management显示文摘Wearable devices with efficient thermal management and electromagnetic interference(EMI) shielding are highly desirable for improving human comfort and safety. Herein, a multifunctional wearable carbon fibers(CF) @ polyaniline(PANI)/silver nanowires(Ag NWs) composites with a “branch-trunk” interlocked micro/nanostructure were achieved through 'three-in-one' multi-scale design. The reasonable assembly of the three kinds of one-dimensional(1D) materials can fully exert their excellent properties i.e., the superior flexibility of CF, the robustness of PANI, and the splendid conductivity of Ag NWs. Consequently, the constructed flexible composite demonstrates enhanced mechanical properties with a tensile stress of 1.2 MPa, which was almost 6 times that of the original material. This is mainly attributed to the fact that the PNAI(branch) was firmly attached to the CF(trunk) through polydopamine(PDA), forming a robust interlocked structure. Meanwhile, the composite possesses excellent thermal insulation and heat preservation capacity owing to the synergistically low thermal conductivity and emissivity. More importantly, the conductive path of the composite established by the three 1D materials greatly improved its EMI shielding property and Joule heating performance at low applied voltage. There are hydrogen bonds in the Complex. 19Reduced expression of semaphorin 3A in osteoclasts causes lymphatic expansion in a Gorham-Stout disease(GSD)mouse model显示文摘Gorham-Stout disease(GSD)is a sporadic chronic disease characterized by progressive bone dissolution,absorption,and disappearance along with lymphatic vessel infiltration in bone-marrow cavities.Although the osteolytic mechanism of GSD has been widely studied,the cause of lymphatic hyperplasia in GSD is rarely investigated.In this study,by comparing the RNA expression profile of osteoclasts(OCs)with that of OC precursors(OCPs)by RNA sequencing,we identified a new factor,semaphorin 3A(Sema3A),which is an osteoprotective factor involved in the lymphatic expansion of GSD.Compared to OCPs,OCs enhanced the growth,migration,and tube formation of lymphatic endothelial cells(LECs),in which the expression of Sema3A is low compared to that in OCPs.In the presence of recombinant Sema3A,the growth,migration,and tube formation of LECs were inhibited,further confirming the inhibitory effect of Sema3A on LECs in vitro.Using an LEC-induced GSD mouse model,the effect of Sema3A was examined by injecting lentivirus-expressing Sema3A into the tibiae in vivo.We found that the overexpression of Sema3A in tibiae suppressed the expansion of LECs and alleviated bone loss,whereas the injection of lentivirus expressing Sema3A short hairpin RNA(shRNA)into the tibiae caused GSD-like phenotypes.Histological staining further demonstrated that OCs decreased and osteocalcin increased after Sema3A lentiviral treatment,compared with the control.Based on the above results,we propose that reduced Sema3A in OCs is one of the mechanisms contributing to the pathogeneses of GSD and that expressing Sema3A represents a new approach for the treatment of GSD.Dongfang ZHANG Hao XU Chi QIN Kangming CAI Jing ZHANG Xinqiu XIA Jingwen BI Li ZHANG Lianping XING Qianqian LIANG Wensheng WANG 2024Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2024,25,1:0
20Correction of a CADASIL point mutation using adenine base editors in hiPsCsandbloodvessel organoids显示文摘Cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy (CADASIL) is a monogenic small vessel disease caused by mutations in the NOTCH3 gene. However, the pathogenesis of CADASIL remains unclear, and patients have limited treatment options. Here, we use human induced pluripotent stem cells (hiPSCs) generated from the peripheral blood mononuclear cells of a patient with CADASIL carrying a heterozygous NOTCH3 mutation (c.1261C>T, p.R421C) to develop a disease model. The correction efficiency of different adenine base editors (ABEs) is tested using the HEK293T-NOTCH3 reporter cell line. ABEmax is selected based on its higher efficiency and minimization of predicted off-target effects. Vascular smooth muscle cells (VSMCs) differentiated from CADASIL hiPSCs show NOTCH3 deposition and abnormal actin cytoskeleton structure, and the abnormalities are recovered in corrected hiPSC-derived VSMCs. Furthermore, CADASIL blood vessel organoids generated for in vivo modeling show altered expression of genes related to disease phenotypes, including the downregulation of cell adhesion, extracellular matrix organization, and vessel development. The dual adeno-associated virus (AAV) split-ABEmax system is applied to the genome editing of vascular organoids with an average editing efficiency of 8.82%.
