1A bHLH transcription activator regulates defense signaling by nucleo-cytosolic trafficking in rice显示文摘Crosstalk between plant hormone signaling pathways is vital for controlling the immune response during pathogen invasion.Salicylic acid(SA)and jas-monic acid(JA)often play important but antagonistic roles in the immune responses of higher plants.Here,we identify a basic helix-loop-helix transcription acti-vator,OsbHLH6,which confers disease resistance in rice by regulating SA and JA signaling via nucleo-cytosolic trafficking in rice(Oryza sativa).OsbHLH6 expression was upregulated during Magnaporthe oryzae infection.Transgenic rice plants overexpressing OsbHLH6 display increased JA responsive gene expression and enhanced disease susceptibility to the pathogen.Nucleus-localized OsbHLH6 activates JA signaling and suppresses SA signaling;however,the SA regulator OsNPR1(Nonexpressor of PR genes 1)sequesters OsbHLH6 in the cytosol to alleviate its effect.Our data suggest that OsbHLH6 controls disease re-sistance by dynamically regulating SA and JA signaling.Fanwei Meng Chao Yang Jidong Cao Huan Chen Jinhuan Pang Qiqi Zhao Zongyi Wang Zheng Qing Fu Jun Liu 2020Journal of Integrative Plant Biology2020,62,10:10
2Effect of chitooligosaccharides on cyclin D1,bcl-xl and bcl-2 mRNA expression in A549 cells using quantitative PCR显示文摘Chitooligosaccharide (COS) is derived from the chemical and enzymatic hydrolysis of chitosan and has been reported to have potent antitumor activity.Our study investigated the effects of five chitooligomers ranging from the dimer form to the hexamer form (chitobiose,chitotriose,chitotetraose,chitopentaose,chitohexaose) on the expression of cyclin D1,bcl-2 and bcl-xl mRNA in A549 cells using reverse transcription quantitative real-time PCR.We demonstrated that,of the five chitooligomers used,chitohexaose (COS6) had the most potent inhibitory effect on A549 cell proliferation.COS6 also significantly down regulated cyclin D1 and bcl-xl mRNA expression levels.Our data suggested that COS6 exerts its antitumor activity by two different mechanisms:(1) COS6-mediated inhibition of Cyclin D1 levels leads to suppression of tumor cell proliferation;and (2) COS6-mediated down-regulation of the pro-survival protein,Bcl-xl,promotes the apoptosis of tumor cells.LI Xian WANG Ju CHEN XiaoJia TIAN JinHuan LI LiHua ZHAO MingYan JIAO YanPeng ZHOU ChangRen 2011Chinese Science Bulletin2011,56,15:5
3Efficient inversions and duplications of mammalian regulatory DNA elements and gene clusters by CRISPR/Cas9显示文摘The human genome contains millions of DNA regulatory elements and a large number of gene clusters,most of which have not been tested experimentally.The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated nuclease 9(Cas9)programed with a synthetic single-guide RNA(sgRNA)emerges as a method for genome editing in virtually any organisms.Here we report that targeted DNA fragment inversions and duplications could easily be achieved in human and mouse genomes by CRISPR with two sgRNAs.Specifically,we found that,in cultured human cells and mice,efficient precise inversions of DNA fragments ranging in size froma few tens of bp to hundreds of kb could be generated.In addition,DNA fragment duplications and deletions could also be generated by CRISPR through trans-allelic recombination between the Cas9-induced double-strand breaks(DSBs)on two homologous chromosomes(chromatids).Moreover,junctions of combinatorial inversions and duplications of the protocadherin(Pcdh)gene clusters induced by Cas9 with four sgRNAs could be detected.In mice,we obtained founders with alleles of precise inversions,duplications,and deletions of DNA fragments of variable sizes by CRISPR.Interestingly,we found that very efficient inversions were mediated by microhomology-mediated end joining(MMEJ)through short inverted repeats.We showed for the first time that DNA fragment inversions could be transmitted through germlines in mice.Finally,we applied this CRISPR method to a regulatory element of the Pcdha cluster and found a new role in the regulation of members of the Pcdhg cluster.This simple and efficient method should be useful in manipulating mammalian genomes to study millions of regulatory DNA elements as well as vast numbers of gene clusters.Jinhuan Li Jia Shou YaGuo Yuanxiao Tang YonghuWu Zhilian Jia Yanan Zhai Zhifeng Chen Quan Xu Qiang Wu 2015Journal of Molecular Cell Biology2015,7,4:4

5Tet2 Regulates Osteoclast Differentiation by Interacting with Runx1 and Maintaining Genomic 5-Hydroxymethylcytosine(5hmC)显示文摘As a dioxygenase, Ten-Eleven Translocation 2(TET2) catalyzes subsequent steps of 5-methylcytosine(5 mC) oxidation. TET2 plays a critical role in the self-renewal, proliferation,and differentiation of hematopoietic stem cells, but its impact on mature hematopoietic cells is not well-characterized. Here we show that Tet2 plays an essential role in osteoclastogenesis. Deletion of Tet2 impairs the differentiation of osteoclast precursor cells(macrophages) and their maturation into bone-resorbing osteoclasts in vitro. Furthermore, Tet2^(-/-) mice exhibit mild osteopetrosis, accompanied by decreased number of osteoclasts in vivo. Tet2 loss in macrophages results in the altered expression of a set of genes implicated in osteoclast differentiation, such as Cebpa, Mafb, and Nfkbiz. Tet2 deletion also leads to a genome-wide alteration in the level of 5-hydroxymethylcytosine(5 hmC) and altered expression of a specific subset of macrophage genes associated with osteoclast differentiation. Furthermore, Tet2 interacts with Runx1 and negatively modulates its transcriptional activity. Our studies demonstrate a novel molecular mechanism controlling osteoclast differentiation and function by Tet2, that is, through interactions with Runx1 and the maintenance of genomic 5 hmC. 6Chromosome doubling mediates superior drought tolerance in Lycium ruthenicum via abscisic acid signaling显示文摘Plants are continuously affected by unfavorable external stimuli,which influences their productivity and growth.Differences in gene composition and expression patterns lead homologous polyploid plants to exhibit different physiological phenomena,among which enhanced environmental adaptability is a powerful phenotype conferred by polyploidization.The mechanisms underlying the differences in stress tolerance between diploids and autotetraploids at the molecular level remain unclear.In this research,a full-length transcription profile obtained via the singlemolecule real-time(SMRT)sequencing of high-quality single RNA molecules for use as background was combined with next-generation transcriptome and proteome technologies to probe the variation in the molecular mechanisms of autotetraploids.Tetraploids exhibited an increase in ABA content of 78.4%under natural conditions and a superior stress-resistance phenotype under severe drought stress compared with diploids.The substantial differences in the transcriptome profiles observed between diploids and autotetraploids under normal growth conditions were mainly related to ABA biosynthesis and signal transduction pathways,and 9-cis-epoxycarotenoid dioxygenase 1(NCED1)and NCED2,which encode key synthetic enzymes,were significantly upregulated.The increased expression of the ABREbinding factor 5-like(ABF5-like)gene was a pivotal factor in promoting the activation of the ABA signaling pathway and downstream target genes.In addition,ABA strongly induced the expression of osmotic proteins to increase the stress tolerance of the plants at the translational level.We consider the intrinsic mechanisms by which ABA affects drought resistance in tetraploids and diploids to understand the physiological and molecular mechanisms that enhance abiotic stress tolerance in polyploid plants.Shupei Rao Yuru Tian Xinli Xia Yue Li Jinhuan Chen 2020Horticulture Research2020,7,1:2
7The coloring mechanism of a novel golden variety in Populus deltoides based on the RGB color mode显示文摘Compared to flower color and red leaf mutants,the mechanism of yellow leaf formation in woody plants is more complicated and less understood.Poplars are common and widely distributed perennial trees.Here,we obtained a golden leaf mutant poplar variety(JHY)and compared it with its original green leaf poplar(Populus sp.Linn.‘2025’)in terms of phenotype,pigment content,the transcriptomes and metabolomes.Through transcriptome sequencing,we found that the chlorophyll degradation genes,and the genes in anthocyanin metabolism in JHY were up-regulated compared to L2025 and the carotenoid synthesis genes were down-regulated.Further based on HPLC-ESI-MS/MS technology,16 differentially expressed anthocyanins were identified,14 reddish anthocyanins of which were significantly up-regulated.According to these results,we proposed a coloring mechanism of JHY based on the RGB color mode.The yellow color of carotenoids and the red color of anthocyanins overlay each other,combined with a reduction in chlorophyll,turning the leaves golden.This study systematic analyzes the causes of golden leaf formation through the omics integration approach,which will provide reference for the breeding of golden leaf trees.Yuru Tian Shupei Rao Qianqian Li Meng Xu Aike Wang Hechen Zhang Jinhuan Chen 2021Forestry Research2021,1,1:2

20Coupling Analysis of Tourism Development and Rural Revitalization in Yannanfei Tea Field Resort of Guangdong Province显示文摘By deconstructing the dimensions of rural tourism and rural revitalization and constructing a coupled conceptual model,a coupled evaluation model is constructed.Yannanfei Tea Field Resort in Guangdong is taken as a research case for empirical analysis,to explore the coupled development trend of the two.The results show that①the tourism development of Guangdong Yannanfei Tea Field Resort has a good synergistic relationship with rural revitalization.The two complement and promote each other,and rural tourism has significantly promoted the development of rural revitalization.②The coupling development of tourism development and rural revitalization in Guangdong Yannanfei Tea Field Resort is divided into coupling imbalance period and coupling coordination period.The results indicate that the coordination between the two has not yet reached a level of good coordination and high-quality coordination.It is recommended that the resort should fully utilize the opportunity of rural revitalization strategy,increase investment in rural tourism,promote the transformation and upgrading of the resort,impel the integration of tea and tourism,and develop new business model of tea-tourism rural vacation.CHEN Haiming LIU Jun ZENG Ziye FENG Jinhuan WU Xinyi WU Zhuohua CHEN Yiyu 2023Journal of Landscape Research2023,15,6:0
