| 1 | Experimental study of an insert and its influence on churning losses in a high-speed electro-hydrostatic actuator pump of an aircraft显示文摘The speed of an Electro-Hydrostatic Actuator(EHA) pump can recently reach 20000 r/min, and its churning losses increase obviously with an increasing speed, which results in low efficiency and thus increasing heat in aircraft EHA systems. In order to reduce churning losses at high speeds, more attention should be given to the design of an insert. In this paper, the effect of an insert with different design parameters on churning losses is investigated through Computational Fluid Dynamics(CFD) simulation and experiments by calculating the difference between churning losses torques of the test pump with and without the insert based on a high-speed churning losses test rig.Analytical results show that the gap between the insert and the cylinder is critical for churning losses reduction. It is found that the churning losses of the test pump can be reduced with a decreasing gap between the cylinder block and the insert at high speeds. This is because the insert can decrease the turbulence occurrence at high speeds. | 2 | Nb2C MXene-Functionalized Scaffolds Enables Osteosarcoma Phototherapy and Angiogenesis/Osteogenesis of Bone Defects显示文摘Early surgical resection and chemotherapy of bone cancer are commonly used in the treatment of bone tumor,but it is still highly challenging to prevent recurrence and fill the bone defect caused by the resection site.In this work,we report a rational integration of photonic-responsive two-dimensional(2D)ultrathin niobium carbide(Nb2C)MXene nanosheets(NSs)into the 3D-printed bone-mimetic scaffolds(NBGS)for osteosarcoma treatment.The integrated 2D Nb2C-MXene NSs feature specific photonic response in the second near-infrared(NIR-II)biowindow with high tissue-penetrating depth,making it highly efficient in killing bone cancer cells.Importantly,Nb-based species released by the biodegradation of Nb2C MXene can obviously promote the neogenesis and migration of blood vessels in the defect site,which can transport more oxygen,vitamins and energy around the bone defect for the reparative process,and gather more immune cells around the defect site to accelerate the degradation of NBGS.The degradation of NBGS provides sufficient space for the bone remodeling.Besides,calcium and phosphate released during the degradation of the scaffold can promote the mineralization of new bone tissue.The intrinsic multifunctionality of killing bone tumor cell and promoting angiogenesis and bone regeneration makes the engineered Nb2C MXeneintegrated composite scaffolds a distinctive implanting biomaterial on the efficient treatment of bone tumor. | Junhui Yin Shanshan Pan Xiang Guo Youshui Gao Daoyu Zhu Qianhao Yang Junjie Gao Changqing Zhang Yu Chen | 2021 | Nano-Micro Letters2021,13,2: | 5 |
| 3 | Effects of splined shaft bending rigidity on cylinder tilt behaviour for high-speed electrohydrostatic actuator pumps显示文摘High-speed axial piston pumps are hydraulic power supplies for electro-hydrostatic actuators(EHAs). The efficiency of a pump directly affects the operating performance of an EHA, and an understanding of the physical phenomena occurring in the cylinder/valve plate interface is essential to investigate energy dissipation. The effects of the splined shaft bending rigidity on the cylinder tilt behaviour in an EHA pump need to be considered, because the deflection and radial expansion of a steel shaft rotating at a high speed cannot be ignored. This paper proposes a new mathematical model to predict the cylinder tilt behaviour by establishing a quantitative relationship between the splined shaft deflection, the cylinder tilt angle, and the tilt azimuth angle. The moments exerted by the splined shaft are included in the equilibrium equation of the cylinder. The effects of solid and hollow splined shafts equipped in an EHA pump prototype are compared at variable speeds of 5000–10,000 r/min. With a weight saving of 29.7%, the hollow shaft is experimentally found to have almost no influence on the volumetric efficiency, but to reduce the mechanical efficiency by 0.6–2.4%. The results agree with the trivial differences of the simulated central gap heights of the interface between the two shafts and the enlargement of the simulated tilt angles by the hollow shaft. | 5 | Self-assembled PEG-b-PDPA-b-PGEM copolymer nanoparticles as protein antigen delivery vehicles to dendritic cells:preparation,characterization and cellular uptake显示文摘Antigen uptake by dendritic cells(DCs)is a key step for initiating antigen-specific T cell immunity.In the present study,novel synthetic polymeric nanoparticles were prepared as antigen delivery vehicles to improve the antigen uptake by DCs.Well-defined cationic and acid-responsive copolymers,monomethoxy poly(ethylene glycol)-block-poly(2-(diisopropyl amino)ethyl methacrylate)-block-poly(2-(guanidyl)ethyl methacrylate)(mPEG-b-PDPA-b-PGEM,PEDG)were synthesized by reversible addition-fragmentation chain transfer polymerization of 2-(diisopropylamino)ethyl methacrylate and N-(tert-butoxycarbonyl)amino ethyl methacrylate monomers,followed by deprotection of tert-butyl protective groups and guanidinylation of obtained primary amines.1H NMR,13C NMR and GPC results indicated the successful synthesis of well-defined PEDG copolymers.PEDG copolymers could self-assemble into nanoparticles in aqueous solution,which were of cationic surface charges and showed acid-triggered disassembly contributed by PGEM and PDPA moieties,respectively.Significantly,PEDG nanoparticles could effectively condense with negatively charged model antigen ovalbumin(OVA)to form OVA/PEDG nanoparticle formulations with no influence on its secondary and tertiary structures demonstrating by far-UV circular dichroism and UV-vis spectra.In vitro antigen cellular uptake by bone marrow DCs(BMDCs)indicated using PEDG nanoparticles as antigen delivery vehicles could significantly improve the antigen uptake efficiency of OVA compared with free OVA or the commercialized Alum adjuvant.Moreover,as the surface cationic charges of OVA/PEDG nanoparticle formulations reduced,the uptake efficiency decreased correspondingly.Collectively,our work suggests that guanidinylated,cationic and acid-responsive PEDG nanoparticles represent a new kind of promising antigen delivery vehicle to DCs and hold great potential to serve as immunoadjuvants in the development of vaccines. | Pan Li Junhui Zhou Pingsheng Huang Chuangnian Zhang Weiwei Wang Chen Lia Deling Kong | 2017 | Regenerative Biomaterials2017,4,1: | 2 |

| 14 | Tilianin extracted from Xiangqinglan(Herba Dracocephali Moldovicae)inhibits apoptosis induced by mitochondrial pathway and endoplasmic reticulum stress in H9c2 cells after oxygen-glucose deprivation/reoxygenation显示文摘OBJECTIVE:To investigate the efficacy of tilianin extracted from Xiangqinglan(Herba Dracocephali Moldovicae)on apoptosis of H9c2 cell after oxygenglucose deprivation/reoxygenation(OGD/R)and the mechanism.METHODS:Tilianin was obtained from Beijing Inluck Science and Technology Development Co.Ltd.,with purity≥98%.The OGD/R model was established in H9c2 cells.Flow cytometry detected the mitochondrial membrane potential,apoptosis rates,mitochondrial reactive oxygen species(ROS)and calcium ion concentration.Succinate dehydrogenase(SDH)activity,succinate content and levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and interleukin-1β(IL-1β)were detected with enzyme-linked immunosorbent assay.Western blot measured protein levels.RESULTS:Tilianin significantly reduced the apoptotic rates,ROS levels,calcium ion concentration,succinate content,and,levels of TNF-α,IL-6 and IL-1βof OGD/R cells,while significantly increased the membrane potential and SDH activity in mitochondria.Western blot analysis showed that tilianin significantly up-regulated pCalmodulin-dependent protein kinaseⅡand voltagedependent anion selective channel levels in OGD/R cells,while significantly down-regulated p-protein kinase B,Bcl-2-associated X,and dynamin-related protein 1 levels related to apoptosis in the mitochondrial pathway.Moreover,tilianin significantly up-regulated B-cell lymphoma-2 and mitochondrial protein 2 related to the inhibition of apoptosis.Furthermore,tilianin downregulated phosphorylated-apoptosis signal-regulated kinase 1,phosphorylated-p38 and C/EBP homologous protein related to endoplasmic reticulum stress.CONCLUSIONS:Tilianin may inhibit OGD/R-induced H9c2 cell apoptosis mediated by mitochondrial pathway and endoplasmic reticulum stress,thus protecting cardiomyocytes. | JIANG Wen ZHANG Wei ZHANG Yuxiang YANG Hao PAN Xiaomei CHEN Qiang CHEN Junhui | 2023 | Journal of Traditional Chinese Medicine2023,43,1: | 0 |
| 15 | C19orf66 Inhibits Japanese Encephalitis Virus Replication by Targeting-1 PRF and the NS3 Protein显示文摘The Japanese encephalitis serogroup of the neurogenic Flavivirus has a specific feature that expresses a non-structural protein NS1'produced through a programmed-1 ribosomal frameshifting(-1 PRF).Herein,C19orf66,a novel member of interferon-stimulated gene(ISG)products,exhibited significant activity of antagonizing Japanese encephalitis virus(JEV)infection.Overexpression of C19orf66 in 293T cells significantly inhibited JEV replication,while knock-down of endogenous C19orf66 in HeLa cells and A549 cells significantly increased virus replication.Notably,C19orf66 had an inhibitory effect on frameshift production of JEV NS1'.The inhibition was more significant when C19orf66 and JEV NS1-NS2A were co-expressed in the 293T cells.Both C19orf66-209 and C19orf66-Zinc^(mut) did not significantly change the NS1'to NS1 ratio and had weaker antiviral effects than C19orf66.Similarly,C19orf66-209 and C19orf66-Zinc^(mut) had no significant effect on the expression of the JEV NS3 protein,whose expression was down-regulated by C19orf66 via the lysosome-dependent pathway.These findings suggest that C19orf66 may possess at least two different mechanisms of antagonizing JEV infection.This study identified C19orf66 as a novel interferon-stimulated gene product that can inhibit JEV replication by targeting-1 PRF and the NS3 protein.The study provides baseline information for the future development of broad-spectrum antiviral agents against JEV. | Du Yu Yundi Zhao Junhui Pan Xingmiao Yang Zhenjie Liang Shengda Xie Ruibing Cao | 2021 | Virologica Sinica2021,36,6: | 0 |
| 16 | Japanese Encephalitis Virus NS2B-3 Protein Complex Promotes Cell Apoptosis and Viral Particle Release by Down-Regulating the Expression of AXL显示文摘Japanese encephalitis virus(JEV) is a flavivirus transmitted by mosquitoes that causes severe encephalitis in humans and animals. It has been suggested that AXL, a transmembrane protein, can promote the replication of various flaviviruses,such as dengue(DENV), Zika(ZIKV), and West Nile(WNV) viruses. However, the effect of AXL on JEV infection has not yet been determined. In the present study, we demonstrate that AXL is down-regulated after JEV infection in the late stage. JEV NS2B-3 protein specifically interacted with AXL, and promoted AXL degradation through the ubiquitin–proteasome pathway. AXL-degradation increased cell apoptosis by disrupting phosphatidylinositol 3-kinase(PI3 K)/Akt signal transduction. In addition, the degradation of AXL promoted JEV release to supernatant, whereas the virus in the cell lysates decreased. The supplementation of AXL ligand Gas6 inhibited the JEV-mediated degradation of AXL. | 19 | Prediction of Lifespan of Tropical Cyclones Landing at China in Spring:A Case Study of Northward-moving-cluster Tracks显示文摘The recurving cluster of tropical cyclones(TCs)landing at China in spring has the characteristics of high frequency,strong intensity,severe influence and long lifespan,a better understanding of tropical cyclone tracks and their underlying mechanisms and more accurate prediction are thus of great value for the prevention and mitigation of TC-related disasters.Based on the best track dataset of typhoon during 1951-2018 from Shanghai Typhoon Institute(STI)of Chinese Meteorological Administration,the tracks of tropical cyclones landfalling over the Chinese coast during 1951-2018 are grouped into three clusters through finite mixture model.The ENSO index,PDO index and the 74 circulation indices of the National Climate Center are analyzed by the classification and regression tree(CART)algorithm for the first TC cluster(the northward-moving cluster).The prediction model based on the results of CART for spring track lifespan is built by a random sampling of the data during the 50 years(about 75%)as a model of the training set,and the training accuracy is 82.46%.The 17-year(about 25%)remaining data are used for testing,with a prediction accuracy of 75%.This study suggests that the finite mixture model algorithm produces a better classification of the northward-going-cluster tracks of TC landfalling over China.In addition,the CART algorithm,which is used for the classification of track lifespan prediction model,not only shows high accuracy,but also the results are easily explained and understood.It provides a novel idea for forecasting the lifespan of tropical cyclones landing at China. | Junhui LI Dawei SHI Yaying PAN Wei WANG | 2020 | Meteorological and Environmental Research2020,11,2: | 0 |
